Post-transcriptional gene silencing (PTGS) is a conserved surveillance mechanism that identifies and cleaves double-stranded RNA molecules and their cellular cognate transcripts. The RNA silencing response is actually used as a powerful technique (named RNA interference) for potent and specific inhibition of gene expression in several organisms. To identify gene products in Eucalyptus sharing similarities with enzymes involved in the PTGS pathway, we queried the expressed sequence tag database of the Brazilian Eucalyptus Genome Sequence Project Consortium (FORESTs) with the amino acid sequences of known PTGS-related proteins. Among twenty-six prospected genes, our search detected fifteen assembled sequences encoding products presenting high level of similarity (E value < 10 -40) to proteins involved in PTGS in plants and other organisms. We conclude that most of the genes known to be involved in the PTGS pathway are represented in the FORESTs database. Copyright by the Brazilian Society of Genetics.

Fungi are a diverse group of organisms with an overall global number of 1.5 M up to 3.3 M species on Earth. Besides their ecological roles as decomposers, fungi are important in several aspects of applied research. Here, we review how culture collections may promote the knowledge on diversity, conservation and biotechnological exploitation of fungi. The impact of fungi diversity on biotechnological studies is discussed. We point out the major roles of microbial repositories, including fungal preservation, prospecting, identification, authentication and supply. A survey on the World Data Center for Microorganisms (WDCM) powered by the World Federation for Culture Collections and on the Genetic Heritage Management Council (CGEN) database revealed that 46 Brazilian culture collections registered in these databases are dedicate to preserving fungi. Most of these culture collections are located in the Southeast of Brazil. This scenario also demonstrates that Brazil has many collections focused on fungal strains, but the lack of up-to-date information in WDCM as well as of a solid national platform for culture collections registration do not allow accurate assessment of fungal preservation. © 2013 Elsevier Inc. The influence of different media and incubation temperatures on the quantification of microbial populations in sorghum, eucalyptus and forest soils was evaluated. Microbial growth was compared by using complex (tryptone soybean agar, TSA, casein-starch, CS, and Martin) and saline (Thorton, M3, Czapeck) media and incubation temperatures of 25 and 30&DEG; C. Higher numbers of total bacterial. and fungal colony-forming units (CFU) were observed in sorghum soils, and of spore-forming and Gram-negative bacteria in forest soils than other soils. Actinomycetes counts were highest in forest soil when using CS medium at 30&DEG; C and in sorghum soil at 25&DEG; C in M3 medium. Microorganism counts were dependent on the media and incubation temperatures. The counts at temperatures of 30&DEG; C were significantly higher than at 25&DEG; C. Microbial quantification was best when using TSA medium for total. and spore-forming bacteria, Thorton for Gram-negative bacteria, M3 for actinomycetes, and Martin for fungi. © 2005 Elsevier GmbH. Crosses between resistant and susceptible soybean cultivars were performed and the F2 populations were obtained to study the inheritance of soybean resistance to powdery mildew and to estimate the number and action of genes related to resistance. The reaction to powdery mildew was studied in a greenhouse and pots carrying plants with symptoms were distributed among the pots carrying the genotypes to be tested as a source of inoculum. Individual plants were scored according to the method of Yorinori (1997), with modifications, and classified as resistant or susceptible. The results showed that adult soybeans plants can present resistance to powdery mildew, which is controlled by one major gene with a dominant effect.

A compactação é um dos fatores mais agravantes para a qualidade do solo, porém o seu efeito na comunidade e atividade enzimática microbiana não tem sido suficientemente estudado. Seis níveis de compactação foram obtidos pela passagem de tratores com diferentes pesos em um Latossolo Vermelho, e a densidade final foi medida. Amostras de solo foram coletadas nas profundidades de 0-10 e 10-20 cm, após a colheita do milho. O efeito da compactação foi evidente em todos os parâmetros estudados, mas nem sempre foi significativo. A contagem das bactérias totais reduziu significativamente em 22-30 %, e a das nitrificantes, em 38-41 %, no solo com maior densidade em relação ao controle. Contudo, a população de fungos aumentou de 55 a 86 %, e a das bactérias desnitrificantes, de 49 a 53 %. A atividade da desidrogenase diminuiu de 20 a 34 %; a da urease, de 44 a 46 %; e a da fosfatase, de 26 a 28 %. O conteúdo de matéria orgânica e o pH do solo diminuíram na camada 0-0,10 em relação à de 0,10-0,20 m e influíram possivelmente na redução das contagens microbianas exceto das bactérias desnitrificantes, e na atividade das enzimas, menos a da urease. Esses resultados indicam que a compactação do solo teve influência na comunidade de microrganismos aeróbios e na sua atividade. Esse efeito pode alterar a ciclagem de nutrientes e diminuir a produção da planta.

The effects of agricultural-pastoral and tillage practices on soil microbial populations and activities have not been systematically investigated. The effect of no-tillage (NT), no-tillage agricultural-pastoral integrated systems (NT-I) and conventional tillage (CT) at soil depths of 0-10, 10-20 and 20-30 cm on the microbial populations (bacteria and fungi), biomass-C, potential nitrification, urease and protease activities, total organic matter and total N contents were investigated. The crops used were soybean (in NT, NT-I and CT systems), corn (in NT and NT-I systems) and Tanner grass (Brachiaria sp.) (in NT-I system); a forest system was used as a control. Urease and protease activities, biomass-C and the content of organic matter and total N were higher (p < 0.05) in the forest soil than the other soils. Potential nitrification was significantly higher in the NT-I system in comparison with the other systems. Bacteria numbers were similar in all systems. Fungi counts were similar in the CT and forest, but both were higher than in NT. All of these variables were dependent on the organic matter content and decreased (p < 0.05) from the upper soil layer to the deeper soil layers. These results indicate that the no-tillage agricultural-pasture-integrated systems may be useful for soil conservation.

This paper reports a study regarding the distribution of lead in the organic matter fractions of a loamy oxisol treated with stable manure, and its effect on the soil microbial biomass. The experimental design was a completely randomized factorial with three replicates. Treatments were four lead (Pb) rates (0, 200, 400, and 600 mg kg(-1) soil), two levels of manure (0 and 30 Mg ha(-1)) and two sampling times (30 and 60 days after lead application). Total and soil lead soluble in Mehlich 1 extractant, and Ph in the different fractions of the organic matter (fulvic acid, humic acid, and residual) were evaluated along with the soil microbial biomass. Data showed that lead, applied as PbCl2, was concentrated in the residual fraction. The application of manure at the rate of 30 Mg ha(-1) did not affect lead distribution in the fractions until 60 days after incorporation. Lead extracted by Mehlich 1 increased as function of the quantity applied. Manure stimulated the growth of the soil microbial biomass which was reduced by the 200 mg kg(-1) doses of lead, with manure application, increasing, however, with larger rates. Mehlich 1 extracted only part of the lead present in the residual fraction.

Cateteres venosos centrais inseridos em pacientes internados em unidade de terapia intensiva foram avaliados por métodos microbiológicos (cultura semi-quantitativa) e microscopia eletrônica de varredura a fim de detectar adesão microbiana e correlacionar com a cultura de sangue. Durante o período de estudo, foram avaliados 59 pacientes com cateter venoso central. A idade dos pacientes, sexo, sítio de inserção e tempo de permanência do cateter foram anotados. O cateter era de poliuretano não tunelizado e de único lúmen. O sangue para cultura foi coletado no momento da remoção do cateter. de 63 pontas de cateteres, 30 (47,6%) foram colonizadas e a infecção encontrada em 5 (23,8%) cateteres. A infecção foi mais prevalente em 26 pacientes (41,3%) com cateteres inseridos em veia subclávia do que nos 3 (3,2%) inseridos em veia jugular. A infecção foi observada com mais freqüência em cateteres com tempo de permanência maior do que sete dias. Os microrganismos isolados incluíram 32 estafilococos coagulase-negativa (29,7%), 61 bactérias Gram-negativas (52,9%), 9 estafilcocos coagulase-positiva (8,3%) e 3 leveduras (2,7%). Como agentes causais de infecções em unidade de terapia intensiva foram isolados E. aerogenes, P. aeruginosa, A. baumannii. Os antimicrobianos com maior atividade in vitro contra as bactérias Gram-negativas foram o imipenem e contra as Gram-positivas vancomicina, cefepime, penicilina, rifampicina e tetraciclina. As análises por microscopia eletrônica de varredura revelaram biofilmes sobre a superfície de todos os cateteres examinados.

Este trabalho teve por objetivo estudar os efeitos de diferentes sistemas de uso e manejo na densidade do solo nas suas propriedades químicas e na atividade microbiana em um Latossolo Vermelho distrófico (Oxisol). As amostras de solo foram retiradas de parcelas dos seguintes tratamentos: cerrado denso preservado, pastagem de Brachiaria decumbens degradada (20 anos), plantio direto com rotação de culturas (8 anos) e sistema convencional com rotação de culturas anuais (10 anos). O delineamento experimental utilizado foi o inteiramente casualizado, com dez repetições. O uso contínuo de plantio direto resultou em mais alta taxa de C-biomassa microbiana e menor perda relativa de carbono pela respiração basal, podendo determinar, desta forma, maior acúmulo de C no solo a longo prazo. Proporcionou, ainda, melhoria na densidade aparente e nas propriedades químicas do solo. Assim, o sistema plantio direto, com manejo de culturas, mostrou ser uma alternativa para a conservação e manutenção das condições físicas e do potencial produtivo de solos de cerrado.
