Differential scanning calorimetry (DSC) in association with chemical analysis was applied to assess the maturity reached by the organic fraction of Municipal Solid Wastes (MSW) subjected to composting processes with manual and fixed aeration and sampled at different composting times. Thermograms showed that the difference in the treatments, i.e., the manual aeration and the fixed aeration, had no relevant effect on the stabilization and maturation of OM in the substrates. Common thermal effects observed were: a low temperature endotherm assigned to dehydration and/or loss of peripheral polysaccharides chains; a medium temperature exotherm assigned to loss of peptidic structures, and a high temperature exotherm assigned to oxydation and polycondensation of aromatic nuclei of the molecule. Results obtained suggest that in the experimental conditions used, a shorter time of composting (about 30 d) appears adequate, in order to limit the extended mineralization of OM, whereas a prolonged composting time (up to 132 d) would produce a compost of poor quality with high ash content and low OM content.

In this work was developed an alternative methodology to separation of aquatic organic matter (AOM) present in natural river waters. The process is based in temperature decreasing of the aqueous sample under controlled conditions that provoke the freezing of the sample and separation of the dark extract, not frozen and rich in organic matter. The results showed that speed of temperature decreasing exerts strongly influence in relative recovery of organic carbon, enrichment and time separation of the organic matter present in water samples. Elemental composition, infrared spectra and thermal analysis results showed that the alternative methodology is less aggressive possible in the attempt of maintaining the integrity of the sample.

The interactions of tropical aquatic fulvic acids (AFA) with chlorine and formation of trihalomethanes were characterized by fluorescence spectroscopy. The aquatic humic substances (AHS) were isolated from a dark-brown stream (located in a environmental protection area near Cubatão city in São Paulo State, Brazil) by means of the collector XAD 8 according the procedure recommended by the International Humic Substances Society. The photoluminescence measurements were made by using a Perkin Elmer spectrometer; AHS, aquatic humic acids (AHA) and AFA samples were assayed. The interactions of AFA and chlorine were characterized by using different reaction times (1, 24, 48, 72 and 168 h) and chlorine concentrations (2.5, 5.0, 10.0 and 20.0 mg L-1). The relative fluorescence intensity for AFA was significantly decreased with the increasing of chlorine concentration and reaction time. The reduction of fluorescence intensity in the region of longer wavelength was interpreted as an indicative of interaction between condensed aromatic groups of AFA and chlorine.

We report the singular filtration properties of an ultrafiltration membrane made with mesoporous silica that exhibits cylindrical pores aligned mostly normal to the support. This membrane supported on tubular commercial macroporous alumina supports was prepared by the interfacial growth mechanism between stable silica-surfactant hybrid micelles made of the association of silica oligomers with polyethyleneoxide-based (PEO) surfactants and sodium fluoride, a well-known silica condensation catalyst [Boissière et al., An ultrafiltration membrane made with mesoporous MSU-X silica, Chem. Mater. 15 (2003) 460-463]. It appears that the combined effect of the silica nature of the membrane, whose surface charge can be easily adjusted by changing the pH and the non-connected cylindrical shape of the pores provides a new behavior in the retention properties, as proved by the filtration of polyoxyethylene polymers (PEO) with different molecular weights. Depending on the filtration conditions, a rejection rate of 80% and a steep cut-off at 2000 Da can be obtained or, on the reverse, polymers three times bigger than the pore diameter can diffuse through the membrane. This new filtration mechanism, which opens up new modes of separation modes, is explained in the light of both topology of the porous network and pH-dependent interactions between PEO polymers and silica porous media. © 2004 Elsevier B.V. Fenton 's reagent was used as coagulant agent to treat water with high true color (100 ± 5 HU) caused by the introduction of humic substances extracted from peat, using dissolved air flotation. The pair value of coagulant dosage x coagulation pH was optimized to posterior construction of coagulation diagrams, reaching apparent color removal efficiency slightly superior to 60%. It was tried to simulate a treatment with complete cycle, carrying out an experiment with sand filtration after flotation, obtaining an effluent with excellent quality, presenting remnant apparent color, turbidity and absorbance of 253.7 nm less or equal to 2 HU, 0.40 TU and 0.009 cm -1, respectively, and residual total iron < 0.005 mg/L and DOC < 0.001 mg/L.

In this work, a micellar system of benzathine penicillin G (BPG) in sodium deoxycholate (NaDC) was developed and evaluated physicochemically. The solubility profile of the drug in water and buffer solutions at various pH was determined, as well as its n-octanol/water partition coefficient. The Critical Micellar Concentration of NaDC and its ability to incorporate BPG were also assessed. The study was carried out at low and high ionic strength which was adjusted by the addition of sodium chloride. The results demonstrated the ability of the micellar system to incorporate BPG, as well as to increase its apparent solubility in water. The enhancement of the solubility of BPG by the presence of NaDC micelles could be analyzed quantitatively within the framework of the pseudo-phase model. Concentration analysis showed that the micellar system could attain up to 90% incorporation of BPG. The incorporated drug is expected to exhibit improved stability, since the antibiotic enclosed in the hydrophobic core of micelles is rather shielded from the aqueous external environment.

Alluvial paleosoil horizons from the 3 to 4 m high banks of the Paraná River were used to reconstruct the palaeoenvironmental history and palaeohydrological regime of the river. The bank consists of a Middle to Late Holocene sequence of sandy to silty clay layers of overbank deposits. The paleosoil horizon is located to a depth of 1.95 m from the top of the bank and is recognized as a guide horizon throughout the floodplain of the Upper Paraná River floodplain. Analyses of organic matter (δ13C and humic/fulvic acids), palynology (pollen and charcoal fragments), magnetic susceptibility, micromorphology, x-ray diffraction and 14C dating were conducted for samples from two representative profiles of the study area. Two phases were characterized in the history of the river: 1) An older phase, of stability in fluvial hydrology (stasis) with low frequency of floods, which produced conditions for soil development (14C 1700 ± 70 yr. BP). At this period, the predominance of herbaceous vegetation (determinated by pollen and δ13C analyses) suggests a climate less humid than the present one, and 2) A second phase, when climate changed to the present conditions of humidity (annual rain fall of 1600 rum) and characterized by the predominance of C3 plants. Under this new hydrological regime, the river developed an agradational floodplain, with a depositional sedimentary rate of 1.2 mm.y-1. © 2006 Gebrüder Borntraeger.

Polar Regions are the most important soil carbon reservoirs on Earth. Monitoring soil carbon storage in a changing global climate context may indicate possible effects of climate change on terrestrial environments. In this regard, we need to understand the dynamics of soil organic matter in relation to its chemical characteristics. We evaluated the influence of chemical characteristics of humic substances on the process of soil organic matter mineralization in selected Maritime Antarctic soils. A laboratory assay was carried out with soils from five locations from King George Island. We determined the contents of total organic carbon, oxidizable carbon fractions of soil organic matter, and humic substances. Two in situ field experiments were carried out during two summers, in order to evaluate the CO2-C emissions in relation to soil temperature variations. The overall low amounts of soil organic matter in Maritime Antarctic soils have a low humification degree and reduced microbial activity. CO2-C emissions showed significant exponential relationship with temperature, suggesting a sharp increase in CO2-C emissions with a warming scenario, and Q10 values (the percentage increase in emission for a 10°C increase in soil temperature) were higher than values reported from elsewhere. The sensitivity of the CO2-C emission in relation to temperature was significantly correlated with the humification degree of soil organic matter and microbial activity for Antarctic soils. © 2012 Antarctic Science Ltd.

Antimicrobial peptides (AMPs) isolated from several organisms have been receiving much attention due to some specific features that allow them to interact with, bind to, and disrupt cell membranes. The aim of this paper was to study the interactions between a membrane mimetic and the cationic AMP Ctx(Ile21)-Ha as well as analogues containing the paramagnetic amino acid 2,2,6,6-tetramethylpiperidine-1-oxyl-4-amino-4-carboxylic acid (TOAC) incorporated at residue positions n = 0, 2, and 13. Circular dichroism studies showed that the peptides, except for [TOAC13]Ctx(Ile21)-Ha, are unstructured in aqueous solution but acquire different amounts of α-helical secondary structure in the presence of trifluorethanol and lysophosphocholine micelles. Fluorescence experiments indicated that all peptides were able to interact with LPC micelles. In addition, Ctx(Ile21)-Ha and [TOAC13]Ctx(Ile21)-Ha peptides presented similar water accessibility for the Trp residue located near the N-terminal sequence. Electron spin resonance experiments showed two spectral components for [TOAC0]Ctx(Ile21)-Ha, which are most likely due to two membrane-bound peptide conformations. In contrast, TOAC2 and TOAC13 derivatives presented a single spectral component corresponding to a strong immobilization of the probe. Thus, our findings allowed the description of the peptide topology in the membrane mimetic, where the N-terminal region is in dynamic equilibrium between an ordered, membrane-bound conformation and a disordered, mobile conformation; position 2 is most likely situated in the lipid polar head group region, and residue 13 is fully inserted into the hydrophobic core of the membrane. © 2013 Vicente et al.

Antimicrobial peptides (AMPs) are a promising solution to face the antibiotic-resistant problem because they display little or no resistance effects. Dimeric analogues of select AMPs have shown pharmacotechnical advantages, making these molecules promising candidates for the development of novel antibiotic agents. Here, we evaluate the effects of dimerization on the structure and biological activity of the AMP aurein 1.2 (AU). AU and the C- and N-terminal dimers, (AU)2K and E(AU)2, respectively, were synthesized by solid-phase peptide synthesis. Circular dichroism spectra indicated that E(AU)2 has a coiled coil structure in water while (AU)2K has an α-helix structure. In contrast, AU displayed typical spectra for disordered structures. In LPC micelles, all peptides acquired a high amount of α-helix structure. Hemolytic and vesicle permeabilization assays showed that AU has a concentration dependence activity, while this effect was less pronounced for dimeric versions, suggesting that dimerization may change the mechanism of action of AU. Notably, the antimicrobial activity against bacteria and yeast decreased with dimerization. However, dimeric peptides promoted the aggregation of C. albicans. The ability to aggregate yeast cells makes dimeric versions of AU attractive candidates to inhibit the adhesion of C. albicans to biological targets and medical devices, preventing disease caused by this fungus. © 2013 Springer-Verlag Wien.

The study objective was to evaluate the influence of the addition of soybeans residues on the chemical properties of Eutrudox and Hapludox soils. Soybean leaves and stems were incubated for 0-200 days. The statistical model used was a 5×4 factorial (plantxincubation period) with three replications. Soils without addition of plants were used as controls. Total Organic Carbon (TOC), Soluble Carbon (SC), Total Carbohydrates (TC), Humic Acid (HA), Fulvic Acid (FA) and Humification Rate (HR) were determined. Higher values of chemical attributes (TOC, SC and TC) were found in the Eutrudox soil than in the Hapludox soil and these values increased significantly (p<0.05) after 50 days of incubation in relation to the initial period. The TOC, SC and TC increased in soils amended with soybean plants when compared to controls without plants. HA and FA contents and HR were not affected by the addition of soybean residues. Maximum HA contents were found after 100 days and maximum FA contents and HR were found after 200 days incubation in both soils. It can be concluded that the addition of soybean residues increased the soil chemical properties when compared to the controls. © 2013 Academic Journals Inc.

The objective of this work was to investigate the adsorption of micronutrients in humin and to verify the ability to release these elements in water. The first step was to determine the adsorption capacity of humin for several essential plant micronutrients and check the kinetic parameters. The order of adsorption was Zn < Ni < Co < Mn < Mo < Cu < Fe, whereas Zn showed maximum values of ca. 2.5 mg g-1 and Fe values of ca. 0.5 mg g-1 for systems containing 1 g of humin. Iron presented higher percentages of release (ca. 100%) and Co the lowest percentages (0.14%). The findings suggested that the use of humin enriched with micronutrients can be a promising alternative for the fertilization of agricultural soils, with the additional benefit of incorporating organic matter present in the form of humic substances into the soil and improving the agricultural productivity. © 2013 Sociedade Brasileira de Química.
