O objetivo deste trabalho foi desenvolver um método de análise de resíduos do herbicida fluridone em peixes. O herbicida foi extraído do peixe (partes comestíveis), concentrado em colunas de florisil e então analisado em cromatógrafo líquido com detector de UV visível a 313 nm. Os coeficientes de correlação obtidos para linearidade do detector e do método foram de 0,99983 e 0,99987, respectivamente. Com o método descrito foi possível extrair, separar e identificar com eficiência o herbicida fluridone das amostras de peixe, o qual apresentou médias de percentagens de recuperação variando de 60 a 73%. O limite de detecção foi de 20 mg kg-1.

Este artigo traz os primeiros resultados sobre a resposta ao estresse em jundiá (Rhamdia quelen) expostos a diferentes cores de luz. O jundiá é uma espécie para produção no sul da América do Sul - sendo assim apta a cultivo em qualquer região de clima temperado e subtropical. Com o objetivo de desenvolver um manejo de luz para o jundiá durante sua manutenção em tanques internos e de entender a relação entre as cores de luz e o bem-estar da espécie, alevinos foram exposto por 10 dias à luz branca, azul e verde. No 10° dia de exposição às diferentes cores de luz, um estressor agudo foi imposto. Uma hora após a aplicação do estressor, os peixes foram amostrados e a resposta ao estresse medida por meio da determinação da concentração sérica de cortisol. Nossos resultados mostraram que a luz verde parece ser a pior alternativa para iluminar instalações internas ou de transporte de jundiás. Os resultados também sugerem que a cor da luz afeta a resposta ao estresse dos jundiás, conhecimento que pode ser útil no manejo da espécie.

Amostras de músculo de carpa comum (Cyprinus carpio) e de água de três viveiros fertilizados com dejetos de suínos e de um viveiro em que os peixes foram alimentados com ração comercial para peixes foram analisadas durante seis meses. O objetivo foi observar a presença de bactérias patogênicas, como Salmonella, Staphylococcus aureus e coliformes totais e fecais, em peixes comercializados in natura no município de Chapecó, Santa Catarina. Foram aferidos quinzenalmente, na água dos quatro viveiros, oxigênio dissolvido, pH, turbidez, alcalinidade total, dureza total, amônia, nitrato e ortofosfato, bem como a quantidade de coliformes totais e fecais. A análise microbiológica músculo dos peixes demonstrou ausência de Salmonella em todas as amostras. A contagem dos demais microrganismos manteve-se inferior ao limite máximo permitido pela legislação vigente, em todas as amostras. Portanto, os peixes analisados estavam aptos ao consumo humano, pois atenderam aos padrões recomendados pela Divisão Nacional de Vigilância Sanitária de Alimentos (DINAL). A comparação entre os viveiros fertilizados com dejetos de suínos e o viveiro fertilizado com ração comercial para peixes não apresentou diferença significativa, porém apresentou efeito significativo sobre oxigênio dissolvido, turbidez, alcalinidade total e amônia. A matéria orgânica foi responsável pelos maiores valores, com exceção da turbidez, que foi menor no viveiro fertilizado com ração comercial para peixes. Entretanto, após análise de todos os parâmetros físico-químicos e microbiológicos da água dos viveiros, pode-se constatar que todos os viveiros se enquadraram nas classes II ou III da classificação de águas interiores estabelecida pelo CONAMA.

Considerable interest is currently focused on fish haemoglobins in order to identify the structural basis for their diversity of functional behavior. Hoplosternum littorale is a catfish that presents bimodal gill (water)/gut (air) -breathing, which allows this species to survive in waters with low oxygen content. The hemolysate of this fish showed the presence of two main haemoglobins, cathodic and anodic. This work describes structural features analyzed here by integration of molecular modeling with small angle X-ray scattering. Here is described a molecular model for the cathodic haemoglobin in the unliganded and liganded states. The models were determined by molecular modeling based on the high-resolution crystal structure of fish haemoglobins. Background: Hematology tests are useful to evaluate physiologic disturbances in fish and can provide important information for the diagnosis and prognosis of disease. Objectives: the primary purpose of this study was to define reference intervals for thrombocytes and leukocytes in healthy channel catfish (Ictalurus punctactus). In addition, the morphologic, cytochernical, and ultrastructural features of blood cells were assessed. Methods: Blood samples (0.5 mL were collected into EDTA from 40 clinically healthy catfish on a commercial fish farm in Jaboticabal, Brazil. Thrombocyte, total WBC, and differential WBC counts were determined and reference intervals were calculated as the 25-95th percentiles of data. Thrombocyte and leukocyte morphology was assessed in blood smears stained with May Griinwald-Giemsa-Wright and ultrastructurally by transmission electron microscopy. Cytochemical staining patterns were described using periodic acid-Schiff (PAS), peroxidase, nonspecific esterase, alkaline phosphatase, and toluidine blue. Results: Reference intervals were as follows: thrombocytes 58,802-99,569/mu L; total WBCs 27,460-41,523/mu L; lymphocytes 5380-11,581/mu L; monocytes 2949-7459/mu L; neutrophils 12,529-22,748/mu L, and basophils 736-2003/mu L. Neutrophils were positive for peroxidase and PAS; monocytes were positive for nonspecific esterase; and basophils were positive with toluidine blue. Conclusion: the morphologic and staining features of neutrophils and monocytes of channel catfish are similar to those of mammals, and the presence of basophils in this species was verified. These reference intervals and morphologic findings provide a foundation for future investigations on the functions and alterations of blood cells in channel catfish.

1. This study aimed to link basic ethnobiological research on local ecological knowledge (LEK) to the conservation of Brazilian streams, based on two case studies: original data on LEK of fishermen about freshwater fish in the Negro River, Amazon, and previously published data about LEK of farmers on the ecological relationship between forest and streams in the Macabuzinho catchment, Atlantic Forest.2. Information was obtained from fishermen through interviews using standard questionnaires containing open-ended questions. Informants for interview were selected either following some defined criteria or applying the 'snowball' method.3. Fishermen's LEK about the diets and habitats of 14 fish species in the Negro River provided new biological information on plant species that are eaten by fish, in addition to confirming some ecological patterns from the biological literature, such as dependence of fish on forests as food sources.4. In the Atlantic Forest, a comparison between farmers' LEK and a rapid stream assessment in the farmers' properties indicated that farmers tended to overestimate the ecological integrity of their streams. Farmers recognized at least 11 forest attributes that correspond to the scientific concept of ecosystem services. Such information may be useful to promote or enhance dialogue among farmers, scientists and managers.5. These results may contribute to the devising of ecosystem management measures in the Negro River, aimed to conserve both rivers and their associated floodplain forests, involving local fishermen. In the Atlantic Forest, we proposed some initiatives, such as to allow direct economic use of their forests to conciliate conflicting perceptions of farmers about ecological benefits versus economic losses from reforestation. Despite their cultural, environmental and geographical differences, the two study cases are complementary and cost-effective and promising approaches to including LEK in the design of ecological research. Colour preference of individual juvenile rainbow trout Oncorhynchus mykiss was tested at 1 and 12 degrees C, and also at 12 degrees C after a 42 day growth experiment under white, blue, green, yellow or red ambient colour. All experiments were carried out under controlled laboratory conditions and the preference was assessed by the location of the fish in a preference tank with four chambers. Rainbow trout showed a preference for blue and green at 1 degrees C and for green at 12 degrees C. After the growth experiment the fish reared in blue tanks preferred blue and green but green was the most preferred colour for the fish reared in green, yellow and red tanks. Yellow and especially red chambers were avoided, irrespective of the ambient colour during the growth trial. The final mass of fish reared in the red aquaria was significantly smaller than that of the fish in green tanks. In addition, when the data of the preference tests were correlated with the data of the growth experiment using mean values of the four tested colours, a very good linear relationship was observed between the preference (i.e. visit frequency in coloured compartments) and growth rate as well as food intake. When considering the results both from the preference and growth trials it is suggested that green is the best environmental colour for rearing juvenile rainbow trout while rearing in a red environment cannot be recommended. Monthly collections of fishes using a trawl net were carried out from August 1996 to December 1997 with the objective of evaluating the composition and diversity of fish species in the littoral area of the transition zone between Veados Stream and Jurumirim Dam (Upper Paranapanema River, São Paulo State). Diversity and similarity indices of ichthyofauna at the sites were determined and correlated with abiotic factors such as dissolved oxygen, temperature, pluviometric precipitation and hydrological level. Twenty-six (26) fish species (the majority small-sized) of the family Characidae (Tetragonopterinae and Cheirodontinae) were registered. Representatives of the orders Siluformes, Gymnotiformes, Perciformes and Synbranchiformes occurred in lower abundance. Significant differences were found in the site diversity indices, but not in the abiotic factors. Significant correlations between the abundance of the ichthyofauna and the variations in the abiotic factors were not detected. The presence of fish larvae in the samples indicates the importance of the transition zone as a reproduction area of the fish species registered in this study.
