Utilizou-se técnica de microimunodifusão dupla em gel de ágar para a medida quali e quantitativa de anticorpos circulantes anti - P. brasiliensis, comparando-se os resultados com o macrométodo. Todos os 103 soros de pacientes portadores de paracoccidioidomicose foram positivos no micrométodo contra 87% de positividade no macrométodo. Os 83 soros de pacientes sem paracoccidioidomicose foram negativos em ambas as reações. Os títulos dos soros positivos tenderam a ser mais elevados no micrométodo, que forneceu bandas de precipitação mais nítidas e fáceis de serem lidas. O micrométodo é de realização simples, utiliza pequena quantidade de material e possibilita o teste simultâneo de 102 soros. Acreditamos que ele poderá substituir o macrométodo, especialmente em laboratórios de grande rotina sorológica.

Background: Cerebrospinal fluid (CSF) is produced in the cerebral ventricles through ultrafiltration of plasma and active transport mechanisms. Evaluation of proteins in CSF may provide important information about the production of immunoglobulins within the central nervous system as well as possible disturbances in the blood-brain barrier. Objective: the objective of this study was to measure the concentration and fractions of protein in CSF samples using a membrane microconcentrator technique followed by electrophoresis, and to compare the protein fractions obtained with those in serum. Methods: CSF samples from 3 healthy dogs and 3 dogs with canine distemper virus infection were concentrated using a membrane microconcentrator having a 0.5 to 30,000 d nominal molecular weight limit (Ultrafree, Millipore, Billerica, MA, USA). Protein concentration was determined before and after concentration. Agarose gel electrophoresis was done on concentrated CSF samples, serum, and serial dilutions of one of the CSF samples. Results: Electrophoretic bands were clearly identified in densitometer tracings in CSF samples with protein concentrations as low as 1.3 g/dL. The higher CSF protein concentration in dogs with distemper was mainly the result of increased albumin concentration. Conclusion: the microconcentrating method used in this study enables characterization of the main protein fractions in CSF by routine electrophoresis and may be useful for interpreting the underlying cause of changes in CSF protein concentrations

Relationships of several reproductve traits and milk yield were studied in 716 Gyr cows at Sant'Ana da Serra farm, Mococa, State of São Paulo, a humid tropical climate region. Mean age at first calving was 49.8 +/- 0.4 months, with a coeficient of cariation (C. V.) of 20.8%. Only year of parturition significantly affected age at first calving (P < 0.01). Heritability, estimated from paternal half-sib correlations, was 0.91 +/- 0.20, a value considered unrealistically high. Overall mean gestation length was 287.7 +/- 0.5 days (C. V. = 3.3%) for 419 observations, with month and year of parturition having significant effects. Heritability estimate was 0.30 +/- 0.14. Overall mean dry period for 1.276 observations was 238 +/- 3 days (C. V. = 48%); repeatability estimate was 0.19 +/- 0.06. Estimated annual genetic trend for dry period was -0.6 days, phenotypic trend was 11.5 days, and environmental trend, 12.1 days. Month and year of parturition and cow age had no significant effect on dry period. Highest milk yield was obtained at fourth lactation. The gross correlation between milk yield and gestation period was 0.11, and between previous dry period and subsequent milk yield, 0.16. Normal gestations of 285 to 290 days were associated with higher milk yields. Milk yield increased as dry period advanced from 30 to 390 days, and declined as dry period continued beyond 390 days. Delays in first mating and a more extensive dry period decreased reproductive efficiency in the herd studied.

The effect of concentration on the structure of SnO2 colloids in aqueous suspension, on their spatial correlation and on the gelation process was studied by small angle x-ray scattering (SAXS). The shape of the experimental SAXS curves varies with suspension concentration. For diluted suspensions ([SnO2] less than or equal to 0.13 mol L-1), SAXS results indicate the presence of colloidal fractal aggregates with an internal correlation length xi congruent to 20 Angstrom, without any noticeable spatial correlation between them. This suggests that the aggregates are spatially arranged without any significant interaction like in ideal gas structures. For higher concentrations ([SnO2] = 0.16, 0.32, and 0.64 mol L-1), the colloidal aggregates are larger (xi = 24 Angstrom) and exhibit a certain degree of spatial correlation between them. The pair correlation function corresponding to the sol with the highest concentration (0.92 mol L-1) reveals a rather strong short range order between aggregates, characteristic of a fluid-like structure, with an average nearest-neighbor distance between aggregates d(1) = 125 Angstrom and an average second-neighbor distance d(2) = 283 Angstrom. The pair distribution function remains essentially invariant during the sol-gel transition, suggesting that gelation involves the formation of a few points of connection between the aggregates resulting in a gel network constituted by essentially linear chains of clusters..

Titanium oxide is a good candidate as new support for hydrotreating (HDT) catalysts, but has the inconvenience of presenting small surface area and poor thermal stability. To overcome these handicaps TiO2-Al2O3 mixed oxides were proposed as catalyst support. Here, the results concerning the preparation, characterization and testing of molybdenum catalyst supported on titania-alumina are presented. The support was prepared by sol-gel route using titanium and aluminum isopropoxides, chelated with acetylacetone (acac) to promote similar hydrolysis ratio for both the alcoxides. The effect of nominal complexing ratios [acac]/[Ti] and of sol aging temperature on the structural features of nanometric particles was analyzed by quasi-elastic light scattering (QELS) and N-2 adsorption isotherm measurements. These characterizations have shown that the addition of acac and the increase of aging temperature favor the full dispersion of primary nanoparticles in mother acid solution. The dried powder presents a monomodal distribution of slit-shaped micropores, formed by irregular packing of platelet primary particles, surface area superior to 200 m(2) g(-1) and mean pore size of about 1 nm. These characteristics of porous texture are preserved after firing at 673 K. The diffraction patterns of sample fired above 973 K show only the presence of anatase crystalline phase. The crystalline structure of the support remained unaltered after molybdenum adsorption, but the surface area and the micropore volume were drastically reduced. (C) 2002 Published by Elsevier B.V. Transparent thin films of nanocrystalline anatase were obtained by dip-coating process using an ethanolic suspension of redispersed nanoparticles. This suspension was prepared by sol-gel route and their redispersability achieved by surface grafting of para-toluene-sulfonic acid and acetylacetone. The effects of the acetylacetone content on the powder redispersibility and on the structural evolution of films were determined by small angle X-ray scattering, X-ray reflectometry and X-ray diffraction for different firing temperatures. The results demonstrated that the porous structure of the studied films consist of agglomerates of primary particles with two levels of porosity. The control of the amount of capping ligand allows for a fine-tuning of the average pore size of the dried films. Upon increasing the firing temperature up to 500 degrees C, progressive increase in apparent density, average pore size of films and average crystallite size of powders were observed. Structural, optical, electro and photoelectrochemical properties of amorphous and crystalline sol-gel Nb2O5 coatings have been determined. The coatings are n-type semiconductor with indirect allowed transition and present an overall low quantum efficiency (phi < 4%) for UV light to electric conversion. The photoconducting behavior of the coatings is discussed within the framework of the Gartner and Sodergren models. Improvement can be foreseen if Nb2O5 coatings can be made of 10-20 nm size nanoparticles.

This work presents a new route of preparation of zirconium ceramic foams based on the thermostimulated sol-gel process. This method produces gelled bodies with up to 90% of porosity in the wet gel and can be used to make complex-shaped components. Unfortunately, the shrinkage during the drying step allows to a catastrophic reduction (50%) of the foam porosity. To improve the foam stability we carried out a systematic study of the effect of gel foam aging on the drying process. Samples were aged in closed vessel at 25 C during different time period (from 6 to 240 h). The shrinkage and the mass loss during drying at 50 C were measured in situ, using a non-contact technique performed with a special apparatus. The results show that the total linear shrinkage decreases from 46% to 8% as the aging period increase from 6 to 240 h. This behavior is followed by a small change of total mass loss, from 42 to 54%. It indicates that by aging the structural stiffness of the foams increases due to secondary condensation reactions. Thus, by controlling the aging period, the porosity can be increased from 67 to 75% and the average size of mesopores of dried foams can be screened from 0.3 to 0.9 mum. Finally, these results demonstrate that the thermostimulated sol-gel transition provides a potential route to ceramic foams manufacture.

Dry matter intake (DMI) of coast-cross grazing by crossbred Holstein-Zebu and Zebu lactating cows was calculated using in vitro dry matter digestibility from extrusa (four esophageal fistulated cows) and fecal output estimate with mordent chromium. Pasture was rotationally grazed with three days grazing period and 27 days testing period, adopting a stocking rate of 1.6 and 3.2 cows/ha, during the dry and rainy season respectively. Voluntary DMI was estimated from degradation characteristics using different equations. Predicted coast-cross DMI varied with models. The prediction of tropical forages dry matter intake from equations based in ruminal degradation parameters needs farther investigation before being employed in practice.

This experiment was conducted in green house conditions to evaluate the DM accumulation in the shoots and in the roots of two cultivars of Lablab purpureus (L.) Sweet. A 2x3 factorial (two cultivars and three evaluation dates) was conducted according to a randomized complete block design with four replications, being the cultivars Highworth and Rongai evaluated at 42, 56, and 70 days after seedling emergence (DASE). The results indicated that the cvs. Highworth and Rongai have the same pattern of DM accumulation in the shoots. In the upper layer of the soil (0-0.20 in) it was found 38.83% and 43.64% of the DM accumulated in the roots down to 2.00 in depth, in the cvs. Highworth and Rongai, respectively. In the deepest layer (1.80-2.00 in) it was found 3.02% and 1.5% of the DM accumulated in the roots of the cvs. Highworth and Rongai, respectively. The root density showed a striking decrease upper layer from the soil (0-0.2 m) down to the depth of 0.60 0.80 in (from 10.83 to 1.75 cm.cm(-3) in the cv. Highworth and from 10.76 to 1.28 cm.cm(-3) in the cv. Rongai). At the bottom layer (1.80-2.00 in) the root density values were 0.98 cm.cm(-3) and 0.59 cm.cm(-3), respectively for the cvs. Highworth and Rongai. The root/shoot ratios were similar in both cvs. and decreased from 42 to 70 DASE showing that the cvs. evaluated had the same dynamics of DM accumulation.
