Bioactive components in rice vary depending on the variety and growing condition. Fat-soluble components such as gamma-oryzanol, tocopherols, tocotrienols, carotenoids, and fatty acids were analyzed in brown, sugary brown, red, and black rice varieties using established high-performance liquid chromatography (HPLC) and GC methodologies. In addition, these colored rice varieties were further analyzed using a high-resolution liquid chromatography-mass spectrometry/mass spectrometry (LC-MS/MS) (LTQ-Orbitrap XL) to identify the [M-H](-) ions of gamma-oryzanol, ranging from m/z 573.3949 to 617.4211. The highest content of tocopherols (alpha-, 1.5; gamma-, 0.5 mg/100 g) and carotenoids (lutein 244; trans-beta carotene 25 mu g/100 g) were observed in black rice; tocotrienols (alpha-, 0.07; gamma-, 0.14 mg/100 g) in red rice, and gamma-oryzanol (115 mg/100 g) in sugary brown rice. In all colored rice varieties, the major fatty acids were palmitic (16:0), oleic (18:1n-9), and linoleic (18:2n-6) acids. When the gamma-oryzanol components were further analyzed by LC-MS/MS, 3, 10, 8, and 8 triterpene alcohols or sterol ferulates were identified in brown, sugary brown, red, and black rice varieties, respectively. Such structural identification can lead to the elucidation of biological function of each component at the molecular level. Consumption of colored rice rich in beneficial bioactive compounds may be a useful dietary strategy for achieving optimal health.

This study aimed at evaluating the oils extracted from seeds originating from agro-industrial waste, in order to identify the presence of bioactive compounds. Therefore, determinations of fatty acid profile, triacylglycerols, tocopherol composition, phytosterols, phenolic compounds, total carotenoids, and antioxidant capacity were performed in the oils of grape, guava, melon, passion fruit, pumpkin, soursop, and tomato seeds. Antioxidant capacity analysis was performed by the methods DPPH• , ABTS•+, FRAP, and β-carotene/linoleic acid, besides measure of oxidative stability in the oils. The oils showed to be predominantly unsaturated with high percentage of linoleic essential fatty acid (38.8 to 79.4%), besides presenting significant quantities of tocopherols, phytosterols, and phenolic compounds. Tomato and guava oils showed better results in the antioxidant capacity tests and pumpkin oil had higher induction period in the oxidative stability test (65.3 h). The results obtained in this study collect information that enables the use of new alternative sources of vegetable oils, obtained from agroindustrial waste, which may serve as raw material for food, chemical, and pharmaceutical industries.

Aloe vera (Aloe barbadensis Miller), popularly known in Brazil as babosa, has a long history of use as medicinal plant for different therapeutic purposes. The components of the plant extract are present in various products of human use, mainly for nutritional and cosmetics purposes. However, some studies suggest that this extract might also have carcinogenic activity. The aloe vera extract is a complex mixture of bioactive compounds. The study of isolated compounds may contribute to elucidate contradictory results about the effects related to the consumption of the plant, as well as their mechanisms of action. One of the most important compound from Aloe vera is aloe-emodin, which is a secondary metabolite generated in the intestinal tract. Putative antimicrobial and antitumor effects were previously attributed to aloe-emodin. Although the exposure of urothelial cells to aloe-emodin was already reported in the literature, only one study showed its effects on urothelial cells, suggesting that aloe-emodin inhibits the viability of T24 cancer cells due to apoptosis induction. Since there is no sufficient information about the effects of aloe-emodin on urothelial cells, and low efficiency in the treatment of bladder cancer currently, the present study aims to evaluate the hypothesis that the treatment with aloe-emodin could impact the behavior of other urothelial cell lines in vitro. Therefore, the in vitro IC50 exposure of aloe-emodin to human immortalized neoplastic urothelial cells will be determinated in order to verify possible differences in the behavior of urothelial cells in vitro treated with aloe-emodin in comparison with untreated cells. The association between tridimensional scaffolds to cells of interest has provided excellent perspectives for obtaining viable complex tissues in vitro, such as skin, resulting in impressive advances in the field of tissue engineering applied to regenerative therapies. The use of multipotent mesenchymal stromal cells in the treatment of dermo-epidermal wounds is particularly promising due to several relevant properties of these cells, such as high capacity of proliferation in culture, potential of differentiation in multiple skin cell types, important paracrine and immunomodulatory effects, among others. Membranes of chitosan complexed with xanthan may be potentially useful as scaffolds for multipotent mesenchymal stromal cells, given that they present suitable physico-chemical characteristics and have adequate tridimensional structure for the adhesion, growth, and maintenance of cell function. Therefore, the purpose of this work was to assess the applicability of bioactive dressings associating dense and porous chitosan-xanthan membranes to multipotent mesenchymal stromal cells for the treatment of skin wounds. The membranes showed to be non-mutagenic and allowed efficient adhesion and proliferation of the mesenchymal stromal cells in vitro. In vivo assays performed with mesenchymal stromal cells grown on the surface of the dense membranes showed acceleration of wound healing in Wistar rats, thus indicating that the use of this cell-scaffold association for tissue engineering purposes is feasible and attractive.

Biological security and the absence of residues in meat Effectiveness, and/or eggs must be considered when recommending options for the control Alphitobius diaperinus in poultry production environments. This research study evaluated the effectiveness of cypermethrin+chlorpyrifos+citronellal in the control of A. diaperinus, including analysis for the presence of residues of this compound in poultry carcasses (experimental farm). Two studies were carried out under laboratory conditions. One used paper filters a four dilutions of the compound, and the other used a container including with pulverized broiler litter and the compound. The analysis of carcasses for residues was conducted in broilers that raised in a broiler house treated (floor and/or litter) with the compound at a dilution of 1:800. Birds were regularly sacrificed, submitted to necropsy, and liver, muscle and fat fragments were collected. Gas chromatography was used to identify the possible presence of any chemical residue in these samples. High effectiveness rates against A.diaperinus were observed in the two laboratory studies, as well as the absence of residues in the carcasses. This compound, can be recommended as a valuable used in the studied concentrations, alternative for the control and treatment of A. diaperinus.

Objective. To evaluate bacterial growth inhibition, mechanical properties, and compound release rate and stability of copolymers incorporated with anthocyanin (ACY; Vaccinium macrocarpon). Methods. Resin samples were prepared (Bis-GMA/TEGDMA at 70/30 mol%) and incorporated with 2 w/w% of either ACY or chlorhexidine (CHX), except for the control group. Samples were individually immersed in a bacterial culture (Streptococcus mutans) for 24 h. Cell viability (n = 3) was assessed by counting the number of colony forming units on replica agar plates. Flexural strength (FS) and elastic modulus (E) were tested on a universal testing machine (n = 8). Compound release and chemical stability were evaluated by UV spectrophotometry and (1)H NMR (n = 3). Data were analyzed by one-way ANOVA and Tukey's test ( α = 0.05). Results. Both compounds inhibited S. mutans growth, with CHX being most effective (P < 0.05). Control resin had the lowest FS and E values, followed by ACY and CHX, with statistical difference between control and CHX groups for both mechanical properties (P < 0.05). The 24 h compound release rates were ACY: 1.33 μg/mL and CHX: 1.92 μg/mL. (1)H NMR spectra suggests that both compounds remained stable after being released in water. Conclusion. The present findings indicate that anthocyanins might be used as a natural antibacterial agent in resin based materials.

Colorectal cancer (CRC) is a disease whose genesis may include metabolic dysregulation. Cancer stem cells are attractive targets for therapeutic interventions since their aberrant expansion may underlie tumor initiation, progression, and recurrence. To investigate the actions of metabolic regulators on cancer stem cell-like cells (CSC) in CRC, we determined the effects of soybean-derived bioactive molecules and the anti-diabetes drug metformin (MET), alone and together, on the growth, survival, and frequency of CSC in human HCT116 cells. Effects of MET (60 μM) and soybean components genistein (Gen, 2 μM), lunasin (Lun, 2 μM), β-conglycinin (β-con, 3 μM), and glycinin (Gly, 3 μM) on HCT116 cell proliferation, apoptosis, and mRNA/protein expression and on the frequency of the CSC CD133(+)CD44(+) subpopulation by colonosphere assay and fluorescence-activated cell sorting/flow cytometry were evaluated. MET, Gen, and Lun, individually and together, inhibited HCT116 viability and colonosphere formation and, conversely, enhanced HCT116 apoptosis. Reductions in frequency of the CSC CD133(+)CD44(+) subpopulation with MET, Gen, and Lun were found to be associated with increased PTEN and reduced FASN expression. In cells under a hyperinsulinemic state mimicking metabolic dysregulation and without and with added PTEN-specific inhibitor SF1670, colonosphere formation and frequency of the CD133(+)CD44(+) subpopulation were decreased by MET, Lun and Gen, alone and when combined. Moreover, MET + Lun + Gen co-treatment increased the pro-apoptotic and CD133(+)CD44(+)-inhibitory efficacy of 5-fluorouracil under hyperinsulinemic conditions. Results identify molecular networks shared by MET and bioavailable soy food components, which potentially may be harnessed to increase drug efficacy in diabetic and non-diabetic patients with CRC.
