Mesenchymal stem cells (MSCs) are a heterogeneous population of cells that proliferate in vitro as plastic-adherent cells, have fibroblast-like morphology and can differentiate into bone, cartilage and fat cells. Therapeutic potential of MSCs have been studied in experimental models, such as rabbit, in Laboratory of Cell Engineering of Botucatu. However, no specific markers have been reported for expanded rabbit MSCs, which hampers the isolation of pure MSC populations by immunophenotypic characterization. Thus, the objective of this study was to produce monoclonal antibodies (mAbs) to rabbit MSCs. MSCs derived from rabbit bone marrow (BM) were isolated, cultured, expanded ex vivo, and immunized into three BALB/c mices, and spleen cells subsequently harvested were used to generate hibridoma cell lines secreting antibodies against MSCs. Hybridoma cells were screened by flow cytometry and antibody-producing cells were subjected to subsequent rounds of retests. MSC1-160 obtained the best positivity for IgG expression and was cloned by limiting dilutions and micromanipulation. Ascitic fluid from ten best clones was purified by affinity chromatography in Protein A-sepharose CL-4B column and purification control was performed by electrophoresis in agarose gels. The purified IgG were tested against rabbit MSCs, obtaining high positivity by flow Cytometry. In conclusion, we developed 10 mAbs, MSC1-160 A20, A30, A41, A47, A55, A60, A63, A69, A81, and A82, that recognize rabbit MSC cell surface antigens showing potential for immunophenotypic characterization of rabbit MSC cell lines

Melatonin is an endoleamine that has anti-inflammatory, immunomodulating and antioxidants properties. But there is a contradiction between the antioxidant effects of melatonin and glutathione (GSH). Therefore, the main objective of this work was to study the effect of melatonin on the oxidation of GSH and the effect of GSH on the oxidation of melatonin by peroxyl radicals generated by thermolysis of 2,2 -Azobis(2- amino-propane)-dihydrochloride (AAPH). The influence of the reaction conditions and the identity of the products of oxidation were also studied. The main products obtained during the oxidation of melatonin were its monohydroxylated derivative and N1-acetyl- N2-formyl-5-methoxykynuramine (AFMK), which is the product obtained by oxidative cleavage of the melatonin indole ring. By studying the buffer type, pH and the presence or absence of dissolved oxygen in the reaction system, it was observed that, the yield of AFMK was higher when the pH or the concentration of oxygen was increased. Comparing the reactivity of both molecules GSH and melatonin, it can be seen that intermediates radicals generated during the oxidation of melatonin are able to oxidize GSH itself. We propose that this chemical property could justify the recent reports that demonstrated the inability of melatonin to inhibit the oxidation of GSH in cells

Plumbism is considered the oldest occupational disease. Among the pathophysiological effects associated with lead (Pb) are cardiovascular disorders. Many diseases that develop later in life are determined during the early stages of life, under the influence of exposure and preferred diet of the mother. Still, one should consider that many environmental contaminants at levels not harmful can determine pathophysiological processes if physical or chemical stressors and/or pathological conditions are present. In this context, the intrauterine malnutrition may represent an additional risk factor in exposure to Pb during pregnancy and lactation. For these reasons, the objective of this study was to evaluate the cardiovascular risk of weaned rats that have suffered perinatal exposure to Pb and intrauterine malnutrition, alone or in combination. After mating, female rats were divided into control (ctrl, ad libitum), food restriction (RA, the same diet 50% of consumption in the control group during pregnancy), exposed to Pb (500 ppm Pb in drinking water during pregnancy and lactation) and association (As, received the last two procedures in combination). Cumulative concentration-effect curves (CECs) to CaCl2 and noradrenaline (NA) were obtained in rings with and without endothelium of the same thoracic aorta from male weaned rats (23-25 days old). Maternal weight, litter weight, weight and number of pups at birth, anogenital distance, arterial blood pressure (ABP) and weight of tissues (kidney, liver, aorta, left ventricle) were evaluated. Changes in vascular reactivity were assessed by the maximum response (MR) and 50% effective concentration (EC50). Data were presented as mean ± SEM. Statistical analysis was performed by multifactorial analysis of variance and Tukey's post test.
