The fucose mannose ligand (Leishmania donovani FML)-saponin vaccine has earlier shown its immunoprophylactic potential against visceral leishmaniasis in the CB hamster (87.7% of parasite load reduction), Balb/c (84.4%) and Swiss albino mouse (85-93%) models. In this investigation its specific immunotherapeutic efficacy against L. donovani infection in Balb/c mice was studied. The effects of vaccine treatment on the Immoral response, delayed type of hypersensitivity to promastigote lysate (DTH), cytokine levels in sera and reduction of the liver parasitic load of L. donovani infected mice, were examined. The types and subtypes of anti-FML antibodies increased significantly in the vaccinees over the saline and saponin controls. As expected for a saponin vaccine, the highest ratios were found in relation to IgG1, IgG2a and IgG2b (4.4, 5 and 2.5, respectively). The DTH response and the in vitro ganglion cell proliferative response against FML antigen were also significantly higher than controls (P < 0.005). Concomitantly, an impressive and specific decrease of liver parasitic burden was detected only in vaccine-treated animals (94.7%). Our results indicate that the therapeutic FML-vaccine has a potent effect on modulation of the murine infection leading to the reduction of parasitic load and signs of disease, being a new potential tool in the therapy and control of visceral leishmaniasis. Young poultry are very susceptible to Salmonella Enteritidis (SE) infections because of the absence of complete intestinal flora colonization and an immature immune system. This study evaluated the role of passive immunity on the resistance of young birds against early infections caused by SE. The progeny of broiler breeders vaccinated with an oil-emulsion bacterin was compared to the progeny of unvaccinated birds. Efficacy was determined by challenging birds at 1 and 14 days of age with SE Nal Spc strain, phage type 4. After challenge at 1 day of age, the progeny of vaccinated birds presented a significantly lower number (log(10)) of SE Nal Spc reisolation (P < 0.05) in liver (2.21), spleen (2.31), and cecal contents (2.85) compared with control groups (2.76, 3.02, and 6.03, respectively). The examination of the internal organs, 3 days after infection, revealed that 28% of the birds (7/25) from vaccinated breeders were positive, whereas 100% (25/25) of the chicks derived from unvaccinated birds were positive. Birds challenged at 14 days of age presented a lower number of positive samples compared with those challenged at 1 day of age, and the progeny of vaccinated birds presented statistically lower numbers (log(10)) of colony-forming units/ml of SE Nal Spc only in the cecal contents compared with nonvaccinated breeder progeny (2.11 vs. 2.94). Age seems to influence the susceptibility of birds to SE infections: in control groups, the number of positive birds at 14 days of age (9/25) was lower when compared with the group infected at 1 day of age (25/25). The number of positive fecal samples of the progeny of vaccinated birds was significantly lower (36) than those of the control group (108) after challenge at 1 day of age. Unchallenged progeny of vaccinated birds presented passive antibodies detectable by enzyme-linked immunosorbent assay (ELISA) up to 21 days of age. on the other hand, antibodies of the control group were detected by ELISA 14 days after challenge. These results show a significant contribution of breeder vaccination by increasing the resistance of the progeny against early SE infections. However, the bacteria were not completely eliminated, suggesting that additional procedures are needed to effectively control SE infections.

This study evaluated the effect of different concentrations of selenium (Se) supplementation on cattle anti-rabies humoral immune response, serum Se concentrations and cortisol levels. Sixty uncastrated male Nelore calves from 10 to 12 months grazing on Brachiaria decumbens forage were studied. The animals were assigned to one of four groups (n = 15 each), which received non-supplemented diets (Gc) or supplemented with daily and individual Selenium ( Se) concentrations of 3.6 mg (G(3.6)), 5.4 mg (G(5.4)) or 6.4 mg (G(6.4)). The calves were immunized on day 0 with one dose of commercial liquid inactivated rabies vaccination. on days 15, 30, 60, 90 and 120, the cattle underwent the same stressing procedures used for vaccination in the corral. Cattle blood samples were collected after vaccination and stressing procedures to determine serum Se levels, rabies antibody titers and serum cortisol. Se levels were also determined in forage samples collected from the paddocks in which the cattle were held. Se concentration in B. decumbens was 0.04 mg of Se/kg dry matter. Baseline Se levels obtained on day 0 were higher in Gc than in G(5.4) and G(6.4) (P = 0.005). Serum Se levels decreased in Gc throughout the experiment (P < 0.004), increased in G(3.6) (P < 0.000) and G(5.4) (P < 0.000) and were kept high from day 60 on in group G(6.4) (P < 0.002). Rabies antibody titers did not differ among control and supplemented groups. However, 120 days after vaccination rabies antibody titers were kept above protective levels (>= 0.5 UI/mL) only in group G(3.6) (P < 0.00002), whereas they dropped in the other groups (P < 0.05). Serum cortisol levels did not differ among the experimental groups (P = 0.79), reached peak levels on day 90 and returned close to baseline levels on day 120. Se and cortisol levels were not markedly correlated. Serum cortisol and rabies antibody titers were correlated only in group G(6.4), on day 60 (R = 0.513; P = 0.05) and 120 (R = 0.644; P = 0.009). Serum Se and rabies antibody titers were correlated only in group G(6.4), on day 60 (R = -0.580; P = 0.023). In conclusion: a) the profile of Se variation is different among groups receiving different concentrations of this element; b) the supplementation dosage of 3.6 mg Se/animal/day is efficient to treat/prevent marginal Se deficiency; c) individual supplementation with daily concentrations of 3.6 mg Se enhances the maintenance of rabies antibody titers in cattle; d) individual supplementation with daily concentrations of 3.6; 5.4 and 6.4 mg Se are ineffective in reducing serum cortisol; e) repeated cattle handling in corrals stress animals that adapt to these procedures, although serum cortisol does not return to baseline levels by 120 days; and f) the stress generated by repeated management in cattle in the corral does not diminish antibody titers after vaccination against rabies.

This study evaluated the humoral immune response of a new rabies vaccine developed by the Instituto Butantan (potency of 3.27 UI/ml) in primovaccinated cattle and the effect of probiotic on this response. Thirty-four 15-month old Nelore cattle were randomly divided into 2 groups (17 animals/group). All the animals were vaccinated on day 0 (zero) and then animals in one group received probiotic added to a mineral mixture (GP) while the others were given only the mineral mixture (GC). Blood samples were collected on days 0, 75 and 150 for rabies neutralizing antibodies titers by seroneutralization assay on BHK21 cells (RFFIT). Protective antibody titers (>= 0.5 UI/mL) were found in 82.4% of the animals from GP and in 76.5% of the animals from GC and no statistical difference (p>0.05) between antibody titers in GP and GC was detected on days 75 and 150. It was also observed that in both groups antibody titers was decreased on day 150 (p<0.01). In conclusion, the tested rabies vaccine promotes efficient soroconversion and keeps antibody levels in primovaccinated cattle, but probiotic does not affect the humoral anti-rabies immune response.

Naive experimental groups of dogs, hamsters and guinea pigs were inoculated three times subcutaneously with unfed adult extract of the tick Rhipicephalus sanguineus and challenged with adult R. sanguineus to evaluate resistance. The acquisition of resistance was based on alterations of some reproductive and feeding performance parameters of female ticks such as female and egg mass weights, engorgement, pre-oviposition and incubation periods, larval hatchability rate and efficiency rates of female ticks in converting their food reservoir to eggs and larvae. Dogs did not develop resistance under these experimental conditions; guinea pigs and hamsters, to a lesser extent, acquired an effective immunity to ticks as demonstrated by the impairment of the reproductive and feeding performance. However, the resistance induced by inoculation of the extract in the rodents seemed not to be as efficient as that induced by successive infestations.

Most parasite-host relationships are characterized by the development of resistance by the host, thus limiting the number of parasites. However, some cases are very unusual. In the relationship of the domestic dog with the brown dog-tick Rhipicephalus sanguineus this does not occur, whereas guinea pigs develop efficient resistance. Sera from domestic dogs, crab-eating foxes and guinea pigs collected before and after infestation with R. sanguineus ticks, and after immunization with a whole tick adult or larval homogenate, were used in Western blot analysis to compare and identify potential important antigens from a tick larval homogenate. The same sera were tested in an indirect immunohistochemistry assay in an attempt to compare relevant antigenic sites on histological tick sections. The immunoblotting displayed antigens recognized only by the guinea pigs, as well as several shared antigens between host species, depending on the kind of immunization. Immunohistochemistry revealed probable antigenic sites on the cells and tissues of ticks, which varied depending on the kind of immunization (infestation or vaccination) and the animal species involved.

Planning control programs, for diseases such as rabies requires information on the size and structure of the dog and cat population. In order to evaluate the dog population of the urban area of Araçatuba city, S. Paulo State, Brazil, a survey was conducted using a questionnaire to interview members of households. Eighty-eight districts were visited (37,778 houses) and the interview was possible at 77.93% of these. Human population size evaluated was 113,157 inhabitants. Houses that owned animals represented 55.2%, 26,926 of the animals concerned were dogs and 5,755 were cats. Of the dogs, 56.64% were 1-4 year olds and males represented 56.2% of the total population. Dog: person ratio was estimated at 2.8 dogs to every 10 persons, almost 3 times the ratio hitherto estimated and used in the planning of rabies vaccination campaigns.

A method based on the use of resazurin (RSZ) is described to determine the number of viable Bradyrhizobium japonicum cells in culture medium. The observation of RSZ reduction can be done spectrophotometrically or visually. B. japonicum strains behaved differently when the reducing time was considered. This methodology can be used to determine the number of viable cells during the liquid culture stage of inoculant production.

We present an empirical investigation of the correcting coils behavior used to homogenize the field distribution of the race-track microtron accelerator end magnets. These end magnets belong to the second stage of the 30.0 MeV cw electron accelerator under construction at IFUSP, the race-track microtron booster, in which the beam energy is raised from 1.97 to 5.1 MeV. The correcting coils are attached to the pole faces and are based on the inhomogeneities of the magnetic field measured. The performance of these coils, when operating the end magnets with currents that differ by ±10% from the one used in the mappings that originated the coils copper leads, is presented. For one of the magnets, adjusting conveniently the current of the correcting coils makes it possible to homogenize field distributions of different intensities, once their shapes are practically identical to those that originated the coils. For the other one, the shapes are changed and the coils are less efficient. This is related to intrinsic factors that determine the inhomogeneities. However, we obtained uniformity of 0.001% in both cases. © 1998 The American Physical Society.

This work deals with the design of the Institute of Physics of the University of São Paulo (IFUSP) main racetrack microtron accelerator end magnets. This is the last stage of acceleration, comprised of an accelerating section (1.04 m) and two end magnets (0.1585 T), in which a 5.10 MeV beam, produced by a racetrack microtron booster has its energy raised up to 31.15 MeV after 28 accelerations. POISSON code was used to give the final configuration that includes auxiliary pole pieces (clamps) and auxiliary homogenizing gaps. The clamps create a reverse fringe field region and avoid the vertical defocusing and the horizontal displacement of the beam produced by extended fringe fields; PTRACE code was used to perform the trajectory calculations in the fringe field region. The auxiliary homogenizing gaps improve the field uniformity as they create a magnetic shower that provides uniformity of ±0.3%, before the introduction of the correcting coils that will be attached to the pole faces. This method of correction, used in the IFUSP racetrack microtron booster magnets, enabled uniformity of ±0.001% in an average field of 0.1 T and will also be employed for these end magnets. © 1999 The American Physical Society.

A historical chronology of smallpox vaccination in Brazil is presented, with emphasis on the State of Sao Paulo. We also present the scientific and philosophical concepts that influenced the regulation and practice of vaccination in Sao Paulo based on the historiographic bibliography, legislation about vaccination, and the debates in the state legislative body. Discovered by Jenner in 1796, the vaccine reached in Brazil in 1804 and was only used in the colonial capital, the city of Rio de Janeiro. In the 1890 decade, smallpox, side by side with yellow fever, typhoid fever and other pestilential diseases, was the major health problem in the State of Sao Paulo. There was also the fear that the vaccine might transmit syphilis, an Unfounded attitude since the product used in Sao Paulo (the 'animal vaccine') was elaborated from bovine serum. The immediate necessity to fight a highly lethal disease that threatened the State population and the coffee-growing business led to the abandonment of the fears and of the liberal principles in favor of the sanitary needs. The vaccine became compulsory in 1891 in the State of Sao Paulo and its application met no resistance on the part of the population, in contrast to the so-called 'Vaccine Revolt' that would occur in Rio de Janeiro in 1904.
