'Profusion Cherry' is a dwarf zinnia with prospect for pot use in Brazil. The success of flowering potted plants depends on its performance during transport and on the period of time that it performs well indoors. Benzyladenine application may retard leaf and flower senescence, increasing postproduction longevity and quality. Senescent flowers removal by consumers, to give a fresh appearance to home flowering potted plant, could influence source-sink relationship and postproduction. This study evaluated the effect of benzyladenine and senescent flowers removal on postproduction performance of 'Profusion Cherry', and observed the senescence symptoms. When plants, produced in greenhouse at São Paulo State, Brazil, had 4 to 5 open flowers, they were sprayed to runoff a single time (20ml/pot) with benzyladenine (0.4, 0.6, 0.8 or 1.0 mmol) and placed into plastic trays, without sleeve. The experimental design was a randomized blocks with 6 treatments (control, four benzyladenine concentrations and senescent flowers removal), 4 replications (2 pots per experimental unit), totalising 12 potted plants in each plastic tray (block). To simulate highway transport, plants remained for 4 days in a dark chamber, at 20.0 °C without irrigation. To include vibration, each plastic tray, was placed in an incubator shaker for 3hr a day, at 60 rpm, 25°C and darkness. After simulated transport, plants remained indoors (10h.day-1 with 18 μmol.m-2.s-1 PPF, 21.5 to 27.0°C and, 14 h.day-1 at darkness, 18.5 to 24.0°C) during 21 days. Plants performed well during simulated transport, and also indoors for two weeks. For most of evaluated parameters there was not significant effect of benzyladenine concentrations by Tukey's test. Senescent flowers removal did not delayed senescence or improved plants quality. The symptoms associated with the loss of decorative life were ray florets color fading and wilting, without abscission of flowers or petals.

Sebastiania commersoniana (Euphorbiaceae) is a tree species of riparian forests in Brazil. Seeds of this species released from mature fruits have heteromorphy in coat colour: dark (dark-brown to black), striated (dark with light-grey stria) and clear (light-grey to whitish). In this work two experiments were carried out in order to study the effect of temperature on seed germination in interaction with coat colour. Germination final percentage and speed index were evaluated. In the first experiment seeds of the three colours were submitted to constant (30°C) and alternating (20-35°C) temperatures. For all the seed colours, best results were obtained at alternating temperature. Physiological quality of striated seeds was greater than those clear seeds and dark seeds were intermediate. In the second experiment striated and clear seeds were submitted to three amplitudes of alternating temperature: 5°C (20-25°C and 25-30°C), 10°C (20-30°C and 25-35°C) and 15°C (20-35°C). Both germination percentage and speed were higher in striated seeds and for the two seed colours, best results were obtained at 20-30°C and 20-35°C. These temperatures are recommended for the germination test.

The morphology and phenology of Sirodotia huillensis was evaluated seasonally in a central Mexican first-order calcareous stream. Water temperature was constant (24-25°C) and pH circumneutral to alkaline (6.7-7.9), and calcium and sulfates were the dominant ions. The gametophyte stages were characterized by the presence of a distinctive mucilaginous layer, a marked difference in phycocyanin to phycoerythrin ratio between female and male plants, and the presence of a carpogonia with a large trichogyne (>60 μm). Occasionally three capogonia were observed on a single basal cell. The 'Chantransia' stages were morphologically similar to those described for the other members of Batrachospermales. A remarkable observation was the formation of dome-shaped structures, consisting of prostrate filaments that are related with the development of new gametophytes. Chromosome numbers were n = 4 for fascicle cells, cortical filament cells and dome-shaped cells, and 2n = 8 for gonimoblast filament cells and 'Chantransia' stage filaments. Gametophytes and 'Chantransia' stages occurred in fast current velocities (60-170 cm/s) and shaded (33.1-121 μmol photons/m2/s) stream segments. The population fluctuated throughout the study period in terms of percentage cover and frequency: the 'Chantransia' stages were most abundant in the rainy season, whereas gametophytic plants had the highest frequency values during the dry season. These results were most likely a result of fluctuations in rainfall and related changes in current velocity. Some characteristics of this population can be viewed as probable adaptations to high current velocities: the mucilaginous layer around plants that reduces drag; potential increase in fertilization by the elongate and plentiful trichogynes and abundant dome-shaped structures producing several gametophytes.

Differential scanning calorimetry (DSC) was used to determine phase transitions of freeze-dried plums. Samples at low and intermediate moisture contents, were conditioned by adsorption at various water activities (0.11≤a w≤0.90) at 25°C, whereas in the high moisture content region (a w>0.90) samples were obtained by direct water addition, with the resulting sorption isotherm being well described by the Guggenheim-Anderson-deBoer (GAB) model. Freeze-dried samples of separated plum skin and pulp were also analysed. At a w≤0.75, two glass transitions were visible, with the glass transition temperature (T g) decreasing with increasing a w due to the water plasticising effect. The first T g was attributed to the matrix formed by sugars and water. The second one, less visible and less plasticised by water, was probably due to macromolecules of the fruit pulp. The Gordon-Taylor model represented satisfactorily the matrix glass transition curve for a w≤0.90. In the higher moisture content range T g remained practically constant around T g′ (-57.5°C). Analysis of the glass transition curve and the sorption isotherm indicated that stability at a temperature of 25°C, would be attained by freeze dried plum at a water activity of 0.04, corresponding to a moisture content of 12.9% (dry basis). © 2006 SAGE Publications.

The objective of this research was to investigate the potential of xylanase production by Aspergillus japonicus and to determine the effects of cultivation conditions in the process, aiming toward optimization of enzyme production. The best temperature, as well as the best carbon source, for biomass production was determined through an automated turbidimetric method (Bioscreen-C). The enzyme activity of this fungus was separately evaluated in two solid substrates (wheat and soybean bran) and in Vogel medium, adding other carbon sources. Temperature effects, cultivation time, and spore concentrations were also tested. The best temperature for enzyme and biomass production was 25°C; however, the best carbon source for growth (determined by the Bioscreen C) did not turn out to be a good inducer of xylanase production. Maximum xylanase activity was achieved when the fungus was cultivated in wheat bran (without the addition of any other carbon source) using a spore concentration of 1 × 107 spores/mL (25°C, pH 5.0, 120 h). A. japonicus is a good xylanase producer under the conditions presented in these assays. © 2006 Academic Journals.

This study aimed to evaluate the effect of different plant growth regulators on essential oil yield in Salvia officinalis L. plants. The experiment was held in a greenhouse and the experimental design was completely randomized, with 5 treatments and three replications. The Treatments consisted in the application of gibberellic acid (GA3), benzylaminopurine (BAP), 2-chloroethyl phosphonic acid (ethephon), and Stimulate at 2%, compared with control plants (water). Applications of plant growth regulators were performed in three consecutive periods, in turn, 15, 25 and 35 days after transplanting of seeding germinating in the light at 25°C. The dry mass yield of the aerial part and the oil essential content were determined 131 days after the transplant. The aerial part of the plants was dried in an oven at 35°C; after dry mass determination, the oil was extracted by hydrodistillation and its volume was determined. Plants treated with GA 3 and Stimulate showed increase in essential oil content, while plants treated with BAP and ethephon showed decrease in essential oil volume when compared with the control plants.

Paracoccidioides brasiliensis is a dimorphic fungus that causes paracoccidioidomycosis, the most prevalent human deep mycosis in Latin America. The dimorphic transition from mycelium to yeast (M-Y) is triggered by a temperature shift from 25°C to 37°C and is critical for pathogenicity. Intracellular Ca 2+ levels increased in hyphae immediately after temperature-induced dimorphism. The chelation of Ca 2+ with extracellular (EGTA) or intracellular (BAPTA) calcium chelators inhibited temperature-induced dimorphism, whereas the addition of extracellular Ca 2+ accelerated dimorphism. The calcineurin inhibitor cyclosporine A (CsA), but not tacrolimus (FK506), effectively decreased cell growth, halted the M-Y transition that is associated with virulence, and caused aberrant growth morphologies for all forms of P. brasiliensis. The difference between CsA and FK506 was ascribed by the higher levels of cyclophilins contrasted to FKBPs, the intracellular drug targets required for calcineurin suppression. Chronic exposure to CsA abolished intracellular Ca 2+ homeostasis and decreased mRNA transcription of the CCH1 gene for the plasma membrane Ca 2+ channel in yeast-form cells. CsA had no detectable effect on multidrug resistance efflux pumps, while the effect of FK506 on rhodamine excretion was not correlated with the transition to yeast form. In this study, we present evidence that Ca 2+/calmodulin-dependent phosphatase calcineurin controls hyphal and yeast morphology, M-Y dimorphism, growth, and Ca 2+ homeostasis in P. brasiliensis and that CsA is an effective chemical block for thermodimorphism in this organism. The effects of calcineurin inhibitors on P. brasiliensis reinforce the therapeutic potential of these drugs in a combinatory approach with antifungal drugs to treat endemic paracoccidioidomycosis. The purpose of this study was to evaluate the effect of pre-heating resin composite photo-cured with light-curing units (LCU) by FT-IR. Twenty specimens were made in a metallic mold (4 mm diameter × 2 mm thick) from composite resin-Tetric Ceram® (Ivoclar/Vivadent) at room temperature (25°C) and pre-heated to 37, 54, and 60°C. The specimens were cured with halogen curing light (QTH) and light emitted by diodes (LED) during 40 s. Then, the specimens were pulverized, pressed with KBr and analyzed with FT-IR. The data were submitted to statistical analysis of variance and Kruskal-Wallis test. Study data showed no statistically significant difference to the degree of conversion for the different light curing units (QTH and LED) (p > 0.05). With the increase of temperature there was significant increase in the degree of conversion (p < 0.05). In this study were not found evidence that the light curing unit and temperature influenced the degree of conversion. © 2010 Pleiades Publishing, Ltd.

Dormancy caused by the tegument's impermeability to water is a feature of Schizolobium parahyba seeds. So that the best methodology for overcoming the species's dormancy may be determined, the seeds were submitted to mechanical scarification treatments, using sandpaper and scissors, on the opposite side of the hilum. Chemical scarification was also undertaken with sulfuric (H2SO4) and hydrochloric (HCl) acids for 30 minutes. Seeds were distributed in groups of 25 units in four petri plates. The experiment was undertaken in a BOD germination chamber at 25°C and 120 μmol.m-2.s-1. Data were used to calculate the germination capacity (G%), the Germination Velocity Index (IVG) and Mean Germination Time (TM). Imbibation curve was also calculated to analyze the dormancy type of the seeds by using the three-phase model where phase FI was completed in 6 hours and FIII was initiated after 12 hours of imbibation. Results show that whereas pre-germinative treatments trigger S. parahyba germination, mechanical scarification is the most effective method to overcome the species's dormancy.

Understanding basic information on weed biology contributes to the implementation of appropriate management and control strategies. Thus, this work was developed to evaluate the germination of Spermacoce latifolia Aubl. seeds, an important weed in reforestation areas. The seeds were subjected to dormancy break treatments, in which the mechanical scarification, chemical treatment (H2SO 4 and KNO 3), heat treatment, and control, were evaluated. Three more tests were done determinate the effects of temperature (10, 15, 20, 25, 30 and 35°C), light filters (absence of light and red, green, yellow, distant red, blue, orange and clear light) and water availability (0.0, -0.2, -0.4, -0.6, -0.8 and -1.0 MPa) on the seeds germination and vigor. The treatments were arranged in a completely randomized design with four replications, and the experimental plot was constituted by gerbox plastic boxes with 50 seeds. The mechanical scarification (sanding) provided the best dormancy break, indicating that Spermacoce latifolia seeds have tegument water impermeability. The seeds showed higher germination percentage under conditions of mild water stress (-0.2 MPa) and the optimal temperature was 25°C. The blue light reduced seed germination.

The longtailed mealybug Pseudococcus longispinus (Targioni Tozzetti, 1867) (Hemiptera: Pseudococcidae) has been reported attacking coffee crops causing fruit fall in the State of Minas Gerais, Brazil. The knowledge of the population dynamic of this pest is then necessary to implement control measures. Its development was studied at temperatures of 15, 20, 25, 30 and 35°C, determining the thermal requirements. The insects were confined inside a Petri dish containing a foliar disc of 4 cm diameter of Coffea arabica L., cultivar 'Acaiá Cerrado'. The temperature affected the P. longispinus development and survival. Few insects survived at temperatures of 15 and 30°C, and 100% of mortality was obtained at 35°C. The duration of the nymphal stage was reduced when the temperature was increased from 20 to 25°C, with a survival rate of 80% at both temperatures. The thermal parameters varied according to the development stage of the mealybug and the base temperature was fixed at 8.0 °C for the nymphal stage of females, a thermal constant of 422.1 day degrees and number of generations increased with rising temperature. The optimal temperature for the insect development was 25°C.

Mutant of dwarf rice (Oryza sativa L.) were treated with three different concentrations of gibberellic acid originated from two sources (Gibberellin and Pro-Gibb) and submitted to the determination of bigger height, in relation to the soil leveI. Mensurations were realized in each seven days at greenhouse conditions under mean temperature of 25°C. lt was concluded that concentrations of 10 mg L-1 of gibberellin originated from Pro-Gibb or Gibberellin was enough to promote a differentiated performance of the test plant. For the study of leaf anatomy Pro-Gibb presented, under all tested concentration, increase of mesophyll thickness in relation to Gibberellin. In this analysis, source and interaction between source and concentration applied promoted significant variations between the mean data.

Several geothermometers have been employed to determine the expected temperature in the groundwater reservoirs, most part of them based on the chemical equilibrium between thermal groundwaters and minerals which constitute the aquifer framework. Based on this principle, groundwater from Guarani Aquifer System, which presents temperatures upon to 38°C, classified as hyperthermal by the Brazilian Mineral Water Code, had their chemical composition evaluated with the main purpose to determine the expected temperatures in the reservoir based on the application of several geothermometers. These groundwaters can be classified as Na-HCO3 type evolving to Na-HCO3-Cl-SO4 type due the increase in sulphate and chloride concentrations. Measured groundwater temperatures show a good correlation with the depth of the aquifer, indicating an average geothermal gradient of 27.7°C/km. The lowest geothermal gradients, bellow 25°C/km, are observed in the central portion of the study area; meanwhile the highest gradients are observed in NE and W portions of the study area. Quartz equilibrium based geothermometers return temperature over the measured temperatures, meanwhile chalcedony geothermometer returns temperatures near the measured in the wells, except for the wells located in the area which presents the lowest geothermal gradients. The geothermometers based on the equilibrium between Na/K have returned temperatures diverse from the measured at the field, meanwhile the geothermometer based on the equilibrium between Na/K/Ca has presented good match with groundwater which present temperatures under 50°C, due the thermodynamic equilibrium between these waters and feldspars. Geothermometers show evidences about mixtures between groundwaters from GAS and underlying units, especially in wells located in the SW portion of the study area.
