Foregut in D. hominis (Linnaeus Jr., 1781) as the majority of the larval Diptera somatic tissue, is made up of polytenic cells, and grows at the expenses of the polytenization of its nuclei followed by the increase in size of each cell. The oesophagus, of ectodermic origem, is interiorly covered by a chitinous squamous epithelium that rests upon a very thin basal lamina. This sheet is surrounded by thick muscle bundles. The oesophagus intussuscepts the midgut forming the cardia. The cardia, with three epithelial layers: two internal ones, of ectodermal origin and one external of endodermic origin. At the anterior portion of the cardia, between these two types of epithelium, there is a cluster of small, non polytenic cells, forming the imaginal disk of the foregut. Metamoiphosis begins at the end of the larval period with signs of nuclear degeneration of all the polytenic cells, as well as the increase in number of the imaginal disk ones. The oesophagic portion intussuscepted into the cardia, everts; its cells suffer apoptosis and are replaced by the new cells growing from the imaginal disk. The external layer cells also degenerate and are pinched off into the lumen of the very anterior portion of the midgut. The newly formed oesophagus intussuscepts de novo to form the two internal layers of the adult cardia. At the same time the midgut regenerative cells grow anteriorly to form the new external layer of the adult cardia.

In vitro antimicrobial susceptibility of strains of Pseudomonas aeruginosa by standard diffusion disk test and a modified method, by the addition Tris-EDTA, was evaluated. Increase in sensitivity of agent using modified method was observed mainly in aminoglycosides (amikacin, gentamicin, tobramycin), quinolones (ofloxacin and norfloxacin) and cephalosporins (cefoperazone and ceftazidime) groups. by standard diffusion disk test and a modified method, by the addition Tris-EDTA, was evaluated. Increase in sensitivity of agent using modified method was observed mainly in aminoglycosides (amikacin, gentamicin, tobramycin), quinolones (ofloxacin and norfloxacin) and cephalosporins (cefoperazone and ceftazidime) groups.

Pesquisou-se a ocorrência de Escherichia coli toxigênica, em queijo produzido com leite não pasteurizado, na Região Centro Oeste do Brazil. Foram utilizados 50 queijos adquiridos em diferentes supermercados. As amostras isoladas foram classificadas por sorogrupo, avaliadas em relação à sensibilidade para 13 agentes antimicrobianos e submetidas à reação em cadeia da polimerase para a presença de genes característicos de E. coli verotoxigênica (VTEC) e enterotoxigênica (ETEC). E. coli foi recuperada em 48(96,0%) dos queijos. Foram identificados os sorogrupos O125 (6,0%), O111 (4,0%), O55 (2,0%) e O119 (2,0%). Três (6,0%) amostras de E. coli foram classificadas como VTEC e uma (2,0%) como ETEC. Os maiores índices de resistência foram verificados para: cefalotina (60,0%), ácido nalidíxico (40,0%), doxiciclina (33,0%), tetraciclina (31,0%) e ampicilina (29,0%).

Os mecanismos que causam o amolecimento e a perda na textura post-mortem da carne de matrinxã foram determinados por meio das mudanças na microestrutura do músculo, imediatamente após a morte e depois de 12 horas de estocagem a -3°C. As observações na microestrutura, realizadas com microscópio eletrônico de transmissão, foram semelhantes aos resultados obtidos na força de ruptura do músculo medidos com o texturômetro. Os valores da força da ruptura foram menores para a carne após o resfriamento. Observou-se que as fibras do colágeno do tecido conectivo pericelular se desintegraram e que as do colágeno do tecido conectivo do miocommata conservaram sua arquitetura e integridade. Houve pouca degradação da linha Z. Isso sugere que o amolecimento post-mortem da carne de mantrinxã, durante a estocagem a -3°C, é causado pela degradação do tecido conectivo pericelular.

O óleo essencial e o extrato etanólico obtidos a partir de folhas de Myrtus communis foram avaliados quanto 'a atividade inibitória frente a vários microrganismos. Os halos de inibição de crescimento foram avaliados através das técnicas de template e difusão em disco para as linhagens de Staphylococcus aureus, Staphylococcus epidermidis, Escherichia coli, Bacillus subtilis e Serratia marcescens. Os resultados obtidos evidenciaram que o óleo de M. communis apresentou atividade antibacteriana superior ao extrato etanólico frente a todas as bactérias. A atividade demonstrada tanto para o óleo como para o extrato etanólico frente a algumas linhagens, foi melhor evidenciada pela técnica template. E. coli foi resistente ao óleo e extrato etanólico testados.

O objetivo deste trabalho foi comparar in vitro a efetividade de três métodos de diagnóstico da lesão de cárie em 26sítios de interesse, dispostos na face palatina de 20 coroas de dentes caninos permanentes de humanos. Quatro examinadores receberam orientação teórico-prática para o diagnóstico, através de três métodos: inspeção visual; táctil (inspeção visual - sonda exploradora com ponta romba); e videoscópico (sistema AcuCam). Para a validação dos dados, os dentes foram seccionados paralelamente ao seu longo eixo por um disco de diamante e analisados histologicamente em um microscópio estereoscópico com aumento de 40X. Diagnósticos compatíveis com o exame histológico apresentaram percentual de acerto no método de inspeção visual de 40,4%, no exame táctil de 38,5% e no videoscópico de 41,3%, resultados que não demonstraram diferença estatisticamente significante. Concluiu-se, pelos resultados obtidos, que os três métodos apresentaram acuidade semelhante no diagnóstico da lesão de cárie; dentre os métodos estudados, o exame ao videoscópio foi o que mais se aproximou do histológico, seguido pelo visual; o mais distante foi o táctil. Constatou-se também uma elevada porcentagem de erro de diagnósticos (59,9%), o que conseqüentemente acarretaria uma conduta terapêutica inadequada

Aproximadamente, 15% das discopatias em cães acometem a região cervical, sendo a dor o principal sinal clínico. Descreve-se a ocorrência de protrusão de disco cervical em 17 cães, agrupados segundo a raça, sexo, peso, idade, assim como a distribuição quanto à duração dos sintomas, acometimento dos discos intervertebrais (DIV), tempo de recuperação e porcentagem de sucesso, em relação à condição neurológica presente antes da cirurgia. A raça Dachshund representou 29,5% (n=5), cães sem raça definida, Poodle e Cocker Spaniel Inglês, 17,6% cada (n=9), Pinscher, 11,8% (n=2) e Dálmata, 5,9% (n=1). Destes, 58,8% eram machos (n=10) e 41,2%, fêmeas (n=7), com peso entre 2 e 29kg, e idade média igual a 5,8 anos. O quadro neurológico desses animais correspondia à dor e ataxia, com exceção de um cão Dálmata, 11 anos de idade, que apresentava tetraparesia. A duração dos sinais variou de 2 a 90 dias. Os DIV mais acometidos foram C2/3 (40%), C3/4 (25%), C4/5 (15%), C5/6 (10%) e C6/7 (10%), sendo que alguns animais apresentavam lesões múltiplas. O procedimento foi padrão para todos os animais, através da fenestração e curetagem de todos os DIV abordados pelo acesso ventral, ou seja, de C2/3 até C6/7, empregando-se para isso instrumental usado para remoção de tártaro dentário (curetas Gracey, curetas McCall, extratores de tártaro S.S.White e McCall). O tempo médio de recuperação foi de 9 a 38 dias, sendo que 100% deles recuperaram totalmente as funções neurológicas. Conclui-se que a fenestração ventral apresenta excelentes resultados no tratamento das discopatias cervicais, desde que bem selecionados os pacientes, inclusive, com respeito aos diagnósticos diferenciais.

Comparou-se a sensibilidade microbiana in vitro de isolados de Rhodococcus equi pelo teste padrão de difusão com discos, com o modificado, pela adição de 5% de dimetilsulfóxido-DMSO. Observou-se aumento da sensibilidade do R. equi no teste com DMSO, frente a aminoglicosídeos (canamicina, amicacina, estreptomicina) e ao cloranfenicol, enquanto para a eritromicina e derivados ß-lactâmicos (penicilina G, cefalosporinas, amoxicilina, oxacilina), constatou-se redução da sensibilidade do agente.

O manejo da palhada da cana-de-açúcar com cana colhida sem queima associado a variedades mais adaptadas podem influenciar a produção de colmos e a qualidade do caldo. Assim, desenvolveu-se o presente trabalho com objetivo de avaliar a influência de diferentes variedades de cana-de-açúcar submetida a sistemas de manejo de colheita da cana colhida sem queima sobre a produção e a qualidade do caldo da cana-de-açúcar. Os tratamentos foram os sistemas de manejo da palhada da soqueira colhida mecanicamente: palha sem triturar/sem cultivo (PST/SC); palha sem triturar/com cultivo com escarificador (PST/CC); palha triturada/com cultivo com escarificador (PT/CC), cultivado com 18 variedades de cana-de-açúcar (RB83-5486, RB85-5536, RB85-5546, RB84-5230, SP81-3250, SP80-3280, SP76-112, SP86-155, SP87-344, SP87-365, SP87-396, SP86-042, SP85-3877, SP84-5560, SP88-725, SP88-817, SP83-2847 Q-138). A qualidade do caldo da cana-de-açúcar varia em função das variedades e sistema de manejo da palhada da soqueira da cana colhida sem queima. O uso do cultivo da entrelinha com escarificador comparado ao sistema sem cultivo aumentou a produção de colmos e a tonelada de sacarose aparente por ha, entretanto, reduziu ATR e a sacarose aparente das soqueiras da cana colhida sem queima. A trituração da palhada nos tratamentos com uso do cultivo da entrelinha com escarificador não é importante na produção e na qualidade de colmos das soqueiras da cana colhida sem queima.

Purpose: To evaluate the bond strength between human dentin and composites, using two light-activated single-bottle total-etch adhesive systems with different pHs combined with chemically activated and light-activated-composites. The tested hypothesis was that the dentin bond strength is not influenced by an adhesive system of low pH, combined with chemically activated or light-activated composites. Material and Method: Flat dentin surfaces of twenty-eight human third molars were allocated in 4 groups (n=7), depending on the adhesive system: (One Step Plus-OS and Prime & Bond NT-PB) and composite (light-activated Filtek Z-100 [Z100] and chemically activated Bisfil 2B [B2B]). Each adhesive system was applied on acid-etched dentin and then one of the composites was added to form a 5 mm-high resin block. The specimens were stored in tap water (37 degrees C/24 h) and sectioned into two axes, x and y. This was done with a diamond disk under coolant irrigation to obtain beams with a cross-section area of approximately 0.8 mm(2). Each specimen was then attached to a custom-made device and submitted to the microtensile test (1 mm.min(-1)). Data were analyzed using two-way ANOVA and Tukey's tests (p<0.05). Results: the anticipated hypothesis was not confirmed (p<0.0001). The bond strengths (MPa) were not statistically different between the two adhesive systems when light-activated composite was used (OS+Z100 = 24.7 +/- 7.1(a); PB+Z100 = 23.8 +/- 5.7(a)). However, with use of the chemically activated composite (B2B), PB (7.8 +/- 3.6(b) MPa) showed significantly lower dentin bond strengths than OS (32.2 +/- 7.6(a)). Conclusion: the low pH of the adhesive system can affect the bond of chemically activated composite to dentin. on the other hand, under the present conditions, the low pH did not seem to affect the bond of light-activated composites to dentin significantly.

A simple and inexpensive way to fabricate arrays of gold microelectrodes is proposed. Integrated circuit chips are sawed through their middle, normal to the longest axis, leading to destruction of the silicon circuit and rupture of the gold wires that interconnect it with the external terminals. Polishing the resulting rough surface converts the tips of the wires embedded in the chip halves into arrays of gold microdisks of about 25 mu m diameter. The number of active microelectrodes (MEs), of an array depends on the number of pins in the chip, n, being typically (n/2)-4. These MEs can be used individually or externally interconnected in any combination. X-ray images of the chips and micrographs of the resulting surface of the polished arrays have revealed variable distances between neighbor MEs, which are, however, larger than 10 times the radius of the disks. This feature of the MEs prevents diffusional cross-talk between electrodes. The use of these microdisk electrodes for analytical purposes exhibits sigmoidal voltammograms, and chronoamperometric experiments confirm the nonlinear i vs. t(1/2) plots, typical for processes where radial diffusion prevails. Satisfactory uniformity was observed for the response of each electrode of an array, indicating similarity of geometry and disk areas. The potentialities of these MEs were demonstrated by the determination of cadmium at ppb levels using square wave voltammetry with preconcentration. Due to the relative ease with which these MEs can be manufactured and their good performance in (chemical) analysis, wide applications in electrochemistry and electroanalysis is envisioned.

Soil tillage may influence CO2 emissions in agricultural systems. Agricultural soils are managed in several ways in Brazil, ranging from no tillage to intensive land preparation. The objective of this study was to determine the effect of common soil tillage treatments (disk harrow, reversible disk plow, rotary tiller and chisel plow tillage systems) on the intermediate CO2 emissions of a dark red latosol, located in southern Brazil. Different tillage systems produced significant differences in the CO2 emissions, and the results indicate that the chisel plow produced the highest soil carbon loss during the 15 days period after tillage treatments were performed. Emissions to the atmosphere increased as much as 74 g CO2 m(-2), at the end of a 2-week period, in the plot where the chisel plow treatment was applied, in comparison to the non-disturbed plot. The results indicate that the total increase on the intermediate term soil CO2 emissions due to tillage treatments in southern Brazil is comparable to that reported for the more humid and cooler regions. The code STATFLUX, implementing a new and simple statistical procedure for the calculation of transfer coefficients in radionuclide transport to animals and plants, is proposed. The method is based on the general multiple-compartment model, which uses a system of linear equations involving geometrical volume considerations. Flow parameters were estimated by employing two different least-squares procedures: Derivative and Gauss-Marquardt methods, with the available experimental data of radionuclide concentrations as the input functions of time. From the pathways of entry of radionuclides in the human (or animal) body, ingestion is the most important because it is closely related to life-long alimentary (or dietary) habits. Those radionuclides which are able to enter the living cells by either metabolic or other processes give rise to localized doses which can be very high. The evaluation of these internally localized doses is of paramount importance for the assessment of radiobiological risks and radiological protection. The time behavior of trace concentration in organs is the principal input for prediction of internal doses after acute or chronic exposure. The General Multiple-Compartment Model (GMCM) is the powerful and more accepted method for biokinetical studies, which allows the calculation of concentration of trace elements in organs as a function of time, when the flow parameters of the model are known. However, few biokinetics data exist in the literature, and the determination of flow and transfer parameters by statistical fitting for each system is an open problem.Restriction on the complexity of the problem: This version of the code works with the constant volume approximation, which is valid for many situations where the biological half-live of a trace is lower than the volume rise time. Another restriction is related to the central flux model. The model considered in the code assumes that exist one central compartment (e.g., blood), that connect the flow with all compartments, and the flow between other compartments is not included.Typical running time: Depends on the choice for calculations. Using the Derivative Method the time is very short (a few minutes) for any number of compartments considered. The aim of this study was to evaluate the effect of ageing in distilled water on the hardness and compressive strength of a direct composite resin Z100, a feldspatic porcelain (Noritake) and three indirect composites (Artglass, Solidex and Targis). For the Vickers hardness tests, five disk-shaped specimens (2 x 4 mm) of each material were prepared according to the manufacturers' instructions. The hardness tests were conducted using a Vickers diamond indentor. Compressive strength measurements were recorded on cylindrical specimens with a diameter of 6 mm and a length of 12 mm. The compression tests were carried out with a constant cross-head speed of 0.5 mm min(-1) on a mechanical test machine. For each material, 10 specimens were tested after 7 days of dry storage at 37 +/- 1 degreesC and 10 specimens were tested after water storage at 37 +/- 1 degreesC for 180 days. Noritake porcelain specimens showed higher hardness values than the composites. Among the composite materials, Z100 promoted the highest VHN values, regardless of the ageing periods. The results showed that Solidex and Z100 had the highest compressive strength values. Ageing in water reduced the hardness for all composites, but had no long-term effect on the compressive strength.

The impact of tillage systems on soil CO2 emission is a complex issue as different soil types are managed in various ways, from no-till to intensive land preparation. In southern Brazil, the adoption of a new management option has arisen most recently, with no-tillage as well as no burning of crops residues left on soil surface after harvesting, especially in sugar cane areas. Although such practice has helped to restore soil carbon, the tillage impact on soil carbon loss in such areas has not been widely investigated. This study evaluated the effect of moldboard plowing followed by offset disk harrow and chisel plowing on clay oxisolCO(2) emission in a sugar cane field treated with no-tillage and high crop residues input in the last 6 years. Emissions after tillage were compared to undisturbed soil CO2 emissions during a 4-week period by using an LI-6400 system coupled to a portable soil chamber. Conventional tillage caused the highest emission during almost the whole period studied, except for the efflux immediately following tillage, when the reduced plot produced the highest peak. The lowest emissions were recorded 7 days after tillage, at the end of a dry period, when soil moisture reached its lowest rate. A linear regression between Soil CO2 effluxes and soil moisture in the no-till and conventional plots corroborate the fact that moisture, and not soil temperature, was a controlling factor. Total soil CO2 loss was huge and indicates that the adoption of reduced tillage would considerably decrease soil carbon dioxide emission in our region, particularly during the summer season and when growers leave large amounts of crop residues on the soil surface. Although it is known that crop residues are important for restoring soil carbon, our result indicates that an amount equivalent to approximately 30% of annual crop carbon residues could be transferred to the atmosphere, in a period of 4 weeks only, when conventional tillage is applied on no-tilled soils.
