Mesenchymal stem cells (MSCs) are a heterogeneous population of cells that proliferate in vitro as plastic-adherent cells, have fibroblast-like morphology and can differentiate into bone, cartilage and fat cells. Therapeutic potential of MSCs have been studied in experimental models, such as rabbit, in Laboratory of Cell Engineering of Botucatu. However, no specific markers have been reported for expanded rabbit MSCs, which hampers the isolation of pure MSC populations by immunophenotypic characterization. Thus, the objective of this study was to produce monoclonal antibodies (mAbs) to rabbit MSCs. MSCs derived from rabbit bone marrow (BM) were isolated, cultured, expanded ex vivo, and immunized into three BALB/c mices, and spleen cells subsequently harvested were used to generate hibridoma cell lines secreting antibodies against MSCs. Hybridoma cells were screened by flow cytometry and antibody-producing cells were subjected to subsequent rounds of retests. MSC1-160 obtained the best positivity for IgG expression and was cloned by limiting dilutions and micromanipulation. Ascitic fluid from ten best clones was purified by affinity chromatography in Protein A-sepharose CL-4B column and purification control was performed by electrophoresis in agarose gels. The purified IgG were tested against rabbit MSCs, obtaining high positivity by flow Cytometry. In conclusion, we developed 10 mAbs, MSC1-160 A20, A30, A41, A47, A55, A60, A63, A69, A81, and A82, that recognize rabbit MSC cell surface antigens showing potential for immunophenotypic characterization of rabbit MSC cell lines

Breast cancer has received an increasing attention because it is one of the most common cancer type and a leading cause of morbity and mortality among women worldwide. This disease has been considered as a heterogeneous condition, demonstrating a large spectrum of clinical and histopathological variability. In the last two decades, several studies have been conducted to identify new molecular markers of cancer cells, including the alterations of DNA methylation, which is the major epigenetic mechanism associated with the control of gene expression. The hypermethylation of promoter-associated CpG islands contributes to the loss of function of several cancer-related genes, including those encoding to the estrogen receptor (ESR) and progesterone receptor (PGR). This study aimed to determine the methylation patterns of CpG islands of the genes encoding the estrogen receptor α (ESR1 gene, promoters A and B), estrogen receptor β (ESR2 gene) and progesterone receptor (PGR gene, promoter A and B) in 15 cell lines derived from breast cancer. The DNA methylation analysis was based on the “Methylation Specific-Polymerase Chain Reaction” (MSP), which provides a qualitative assessment of the methylation status of a specific CpG island. The results revealed heterogeneous data: the promoter region of ESR1A showed complete methylation in one cell line (BT549) and only two cell lines showed partial methylation (MDA-MB-231 and MDA-MB-453), while the others lineages presented unmethylated alleles. The promoter region of isoform ESR1B was unmethylated in the cell lines BT549, SKBR3 and T47D; partial methylation were observed in the cell lines MDA-MB- 231, MCF-7 and ZR-75-30, while the others cell lines presented complete methylation. All lineages showed complete or partial methylation of the ESR2 gene. The methylation pattern of the promoter A of the PGR ...(Complete abstract click electronic access below)

Snakes from Bothrops genus are responsible for more than 90% of the ophidian accidents in Brazil. One of the main complications from this kind of accident is muscular necrosis, which is related to the action of phospholipases A2 and metalloproteases, two groups of enzymes found in the venom of these animals. Although this complication cannot be solved by serum therapy administration, a great number of studies have been performed with the attempt to know the pharmacological sites of these toxins aiming, in the future, the development of complementary treatments to serum therapy. This work proposes structural studies of bothropic phospholipases A2 (PLA2s) in the presence of ions relevant to their activity, using the X-ray crystallography technique. Recently, it was demonstrated ions, as manganese, calcium and others, interfere in the biological activity of the PLA2s. Particularly, Lys49-PLA2s in the presence of manganese ions have miotoxicity reduced. Asp49-PLA2s show catalytic activity dependent of calcium, although structural studies with a miotoxic Asp49-PLA2, BthTX-II, suggest a possible catalytic mechanism independent of calcium. Therefore, co-crystallization of BthTX-II in the presence of calcium ions and of PrTX-I in the presence of manganese ions were performed. Comparative structural studies among obtained results and others already published in the literature were performed aiming a better understanding of the structure-function relationship of these toxins. The BthTX-II with the presence of calcium do not show this ion in the loop of coordination of calcium, presence necessary to develop the catalyses. After comparison of this model with the native one, only one distortion was found, but no apparent relationship with the residues responsible for its activity. In the PrTX-I structure, regions candidates of manganese ions were also found... (Complete abstract click electronic access below)

In materials science, the search for technological improvements have become one of the main subject of study of researchers. This is especially true in the case of materials with reduced sizes, in the nanometer scale. Important phenomena to be studied in these cases are the desorption and adsorption on two-dimensional materials, such as graphene. These phenomena are of great importance in the study of interactions between organic films, synthesis or catalysis of reactions on surfaces and even in the creation of nanoscale devices [1, 2, 3, 4]. Between the most important topics related to these phenomena are the storage of gases in low-dimensional systems and the study of nanostructured fuel cells or batteries. In this context we used two different parametrizations for the reactive force field ReaxFF to study the potential barriers and reaction barriers of our system. First we made a study about the Reaction Barriers and Energy Barriers for bonds between graphene and the following atoms: sulfur, fluorine, hydrogen, nitrogen and oxygen. It is important to have this information in order to make it possible to understand how these atoms react with the graphene sheet. Subsequently, we calculate reaction barriers for mixed structures where fluorine is a fixed element bonded to graphene and other element is simultaneously bonded to graphene. This other element (N, O, H or S) is varied in its possible relative positions (ortho, meta and para in relation to fluorine in either: the same side and in the opposite side of the graphene membrane)

The cinematographic language is notedly plural and heterogeneous. There are many filmic elements that “narrate” all the time, in different ways. Besides enumerating some of them, this study seeks to analyse if the criticism is attentive to all of these tools or if it sticks only to some of them. It’s also reflects if this activity uses more objectives or subjectives aspects as basis for its conclusions. Here, a very specific field was chosen: the study is done by a comparative analysis of the reception of Brazilian’s and European’s criticism from two Lars Von Trier’s films, Antichrist and Melancholia. After explaining how the narrative elements are used in the cinema, it’s demonstrated that, in general, the criticism is negligent and does not analyze all of those tools. In some cases, the analysis of some elements is done in a superficial way, so others aspects can be highlighted

Nowadays technological trend is based on finding materials that could support low weight with satisfactory mechanical properties and for this reason composite material became a very attractive topic in research projects all over the world. Due to its heterogenic properties, this type of material shows scatter in mechanical test results, especially in cyclic loading. Therefore it is important to predict its fatigue strength behaviour by statistic analysis, once fatigue causes approximately 90% of the failure in structural components. The present work aimed to investigate the fatigue behaviour of the Twill/Cycom 890 composite, which is carbon fiber reinforced with polymeric resin as matrix and manufactured via RTM process (Resin Transfer Molding). All samples were tested in different tensile level in triplicate in order to associate these values. The statistical analysis was conducted with Two-Parameter Weibull Distribution and then evaluated the fatigue life results for the composite. Weibull graphics were used to determine the scale and shape parameters. The S-N curve for the Twill/Cycom composite was drawn and indicated the number of cycles to occur the first damages in this material. The probability of failure was associated with material reliability, as shown in graphics for the different tensile levels and fatigue life. In addition, the laminate was evaluated by ultrasonic inspection showing a regular impregnation. The fractographic analysis conducted by SEM showed failure mechanisms for polymeric composites associated to cyclic loadings ... (Complete abstract click electronic access below)
