Estudaram-se os efeitos da rufiação e da manipulação do sistema genital (estocadas) sobre a fertilidade de éguas inseminadas com sêmen fresco (exp. 1) e resfriado/transportado a 14ºC/3,6 horas (exp. 2).

Ninety-seven Mangalarga Marchador mares were allocated in a randomized experimental design, with different number of replicates per treatments. The objective of this experiment was to study the effect of raw (T3) or diluted, cooled semen transported to the breeding farms 1 (T1) and 3 (T2), on mare fertility and different aspects of reproductive efficiency. The mares were inseminated, with semen of only one stallion (Farm 2), on alternate days using different inseminating volumes, according to number of inseminated mares per day using only one ejaculate. For transportation, semen diluted in a lactose-egg yolk modified extender and the MSP-2 container were used. There was no difference among treatments in relation to conception rates of mares inseminated with raw or diluted, cooled and transported semen. Regardless of the farm, the total fertility obtained with cooled semen was 80% (52/65). The following variables were not affected by the treatments: cycles/mare, cycles/pregnant mare, cycles/pregnancy, pregnancy/cycle, number of inseminations/mare, number of inseminations/pregnant mare, number of inseminations/open mare and efficiency of pregnancy. However, the breeding farm had an effect on water and semen temperature in the container, time between collection and insemination, according to time and distance covered. Based on the results, cooling and transporting equine semen, using an extender, lactose-egg yolk without glycerine and the MSP-2 container should be recommended for short distances. The use of raw semen immediately after collection is also recommended.

One hundred and one estral cycles of 92 Mangalarga Marchador breed mares were analyzed to study the effect of different spermatic concentrations on different reproductive characteristics. The mares were inseminated with diluted equine semen, cooled at 20 degrees C and transported: T1-concentrations below 250 x 10(6) (177.58 +41.74 x 10(6) viable spermatozoa per inseminate dose), T2-concentrations between 250 and 350 10(6) sptz (315.51 +/- 20.78 x 10(6)), and T3-concentrations above 350 x 10(6) sptz (477.71 +/- 136.83 x 10(6)). The conception rates in the first cycle were 43.75 (7/16), 57.89 (11/19)and 54.55% (18/33) for treatments 1, 2 and 3,respectively. After five cycles, the conception rates were 36.00 (9/25), 44.12 (15/34) and 54.76% (23/42) for treatments 1,2 and 3, respectively. Concentrations of 177.58 +/- 41,74 x 10(6) viable spermatozoa perinseminate dose had moderate fertility index in a commercial program using cooled and transported semen.

A cytogenetic study of 22 mares with fertility has showed that four of them had 63,X/64,XX mosaicism. The X-chromosome has presented the expected interstitial heterochromatic C - banding located in the long arm, besides of the usual centromeric band. The great variation of clinic signs observed in mares with mosaicism, could be due to the type of zygote or the time the mosaicism occured.

The effects of extender and farms on the fertility of sixty-five Mangalarga Marchador breed mares were studied. The mares were randomly assigned to a 2 x 2 factorial experiment, with two breeding farms (H1 and H2) and two extenders (glycine-egg yolk-T1 and lactose-egg yolk modified-T2), from only one stallion, with different number of replicates. The mares were inseminated, on alternated days, with diluted, cooled, transported semen, in a special container (MSP-2). There were no differences in the conception rate at first cycle, regardless of the breeding farm or extender used. Also. there was no difference among breeding farm or extender concerning the conception rate/cycle and the following reproductive characteristics: number of cycles/pregnancy, number of inseminations/mare, inseminations/pregnant mare, inseminations/open mare, water temperature in the container, temperature of semen in the container, time between collection and insemination and efficiency of pregnancy. The use of lactose-egg yolk modified extender for the dilution, cooling and transport of stallion semen between farms is recommended. The new container MSP-2 showed to be a option for the transportation between farms, for the same purpose.

Objective - To investigate the effects of inhalation and total IV anesthesia on pituitary-adrenal activity in ponies. Animals - 9 healthy ponies: 5 geldings and 4 mares. Procedure - Catheters were placed in the cavernous sinus below the pituitary gland and in the subarachnoid space via the lumbosacral space. After 72 hours, administration of acepromazine was followed by induction of anesthesia with thiopentone and maintenance with halothane (halothane protocol), or for the IV protocol, anesthesia induction with detomidine and ketamine was followed by maintenance with IV infusion of a detomidine-ketamine-guaifenesin combination. Arterial blood pressure and gas tensions were measured throughout anesthesia. Peptide and catecholamine concentrations were measured in pituitary effluent, peripheral plasma, and CSF. Peripheral plasma cortisol, glucose, and lactate concentrations also were measured. Results - Intravenous anesthesia caused less cardiorespiratory depression than did halothane. ACTH, metenkephalin, arginine vasopressin, and norepinephrine pituitary effluent and peripheral plasma concentrations were higher during halothane anesthesia, with little change during intravenous anesthesia. Pituitary effluent plasma β-endorphin and peripheral plasma cortisol concentrations increased during halothane anesthesia only. Dynorphin concentrations did not change in either group. Hyperglycemia developed during intravenous anesthesia only Minimal changes occurred in CSF hormonal concentrations during anesthesia. Conclusion - The pituitary gland has a major role in maintaining circulating peptides during anesthesia. Compared with halothane, IV anesthesia appeared to suppress pituitary secretion.

Six or 7-day-old equine embryos were divided into 4 groups; Group 1, n = 15, Day 7 embryos destined for immediate transfer; Group 2, n = 15, Day 6 embryos destined for deep-freezing with glycerol plus sucrose as cryoprotectant; Group 3, n = 10, Day 6 embryos destined for deep-freezing with glycerol plus 1,2-propanediol as cryoprotectant and Group 4, n = 3, fresh embryos destined for ultrastructural analysis. All the frozen/thawed embryos were transferred to recipient mares, except 3 embryos in Group 3 that were subjected to ultrastructural analysis. After thawing the cryoprotectants were removed by successive dilutions in PBS + 15% v:v fetal calf serum (FCS) containing decreasing concentrations of the cryoprotectants. Pregnancy was diagnosed ultrasonographically in 53.3%, 13.3% and 0% of the mares in Groups 1, 2 and 3 respectively. Ultrastructural analysis showed differences between frozen/thawed and fresh embryos. In the former, embryonic cells were deformed and showed dilation of the intercellular and perivitelline spaces, a decrease of desmosome number in the junctional complexes, few microvilli on the apical surface of the trophectoderm and an almost total absence of pinocytotic vesicles. Most of the mitochondria showed regions containing dilation and irregularities on the cristae, which appeared electron-dense. The results obtained with Groups 2 and 3 embryos showed that the cryoprotectants employed were not effective in protecting the embryos against damage during freezing and thawing. Indeed, the ultrastructural changes observed in the Group 3 embryos explained the absence of any established pregnancies in this group of mares.

Chronic hypertrophic pododermatitis cases in six horses of different breeds, aging 14 months to 19 years are described. The lesion begun with a infiltrative tissue in the frog and sole regions of the hoof, characterized by a fast and disorganized growth, with a papillary aspect, white colored in the roots and dark on the extremity, with a necrotic secretion and an extremely fetid odor. Microscopically, an exuberant epidermic proliferative tissue was observed, intermingled with little connective tissue. The horses were divided into two treatment groups. In the first group, including three young mares and a foal, showing lesions in only one limb, a surgical resection of the invasive mass was performed, followed by cauterization of the remaining edges and subsequent daily local application of antiseptic substances. In three of these horses, recurrence of the initial lesion occurred, with fast growth of hyperplasic tissue, affecting almost all the frog and half of the sole. Two horses developed contraction deformities of the hoof. In the second group, one male and one female, each with lesions in two limbs, after surgical debridement of the tissue, the animals received daily applications of picric acid 5%, associated to local use of oxitetracyclin. Although one of these cases required a second surgical intervention for removal of the mass, the horses showed after a period of two to three months total absence of the infiltrative tissue. The use of local picric acid 5% and oxitetracyclin associated to previous surgical debridement showed to be more efficient than the use of antiseptic substances in the treatment of chronic hypertrophic pododermatitis.

The efficacy of the first ovulation of the breeding season was determined through the response of first pre-ovulatory follicle of the breeding season to hCG, embryo recovery rate, viability of recovered embryos, serum concentrations of progesterone, and response of first CL to PGF 2α. Thirteenth mares that were in vernal transition were accompanied until the first pre-ovulatory follicle was detected. At this moment, the ovulation was induced with 2,500 IU hCG (IV) and the mares were inseminated every other day until ovulation. Seven days after the ovulation, embryo recovery was performed and the progesterone concentration was determined. After detection of the first pre-ovulatory follicle of breeding season with ≥ 25mm, it took 14.92 ± 10.80 days for the follicle to reach the preovulatory size and 18.00 ± 11.08 days to ovulation. After administration of hCG, 11/13 mares ovulated in 48 hours. These follicles growth 2.19 ± 0.86 mm/day on average. Nine of 13 mares (69.2%) produced embryos and all were considered viable after morfological evaluation and fluorescence exams. The CL appeared competent producing 7.39 ± 2.11 ng/ml P 4 on average, and responding to PGF 2α. According to these results the first ovulatory cycle of the year can be utilized to produce viable embryos.

The aim of this study was to evaluate the pre-emptive effect of epidural ketamine S (+) (SK) or racemic ketamine (RK) administration, in post-incisional pain in horses. Were used in a blinded, randomized experimental study, sixteen mixed breed mares, 6±2 years old, weighting 273.2±42.0 kg. An epidural catheter was inserted 24 hours before the trials. The thigh region was shaved bilaterally, and mechanical cutaneous sensibility was measured using von Frey filaments (T-30). Using the left side as the control one, local anesthesia was performed at the right side. Twenty-five minutes later, SK was injected in G1 or RK in G2 through the epidural catheter. Five minutes after the ketamine injection, a 10 cm skin incision was made on the right side, and then sutured. Mechanical post-incisional pain was measured using von Frey filaments, at 1, 3 and 5 cm around the incision at 15 minutes intervals, for 2 hours, then 4, 6 and 8 hours after suturing. No changes were observed in the heart and respiratory rate and rectal temperature among groups or times of each group. Hind limb ataxia was observed in 62.5% and 12.5% of G1 and G2 respectively. SK and RK reduced cutaneous sensibility in the right and the left sides to mechanical postincisional pain during all time of experiment. Epidural SK and RK produce similar post-incisional analgesic effects, did not interfere in the cardio-respiratory parameters. The SK induces more intense ataxia in mares and presents a larger analgesic potency in the first 60 minutes after the administration.

The effect of physical exercise, training and vitamin E supplementation on electrocardiographic parameters was evaluated in eight untrained Arabian mares, divided into two groups: control (n=4) and supplemented with vitamin E (n=4) at the daily dose of 1.000 UI. Animals were submitted to an incremental exercise test (P1) on high-speed treadmill inclined +7%, after that to a training period of 20 days and later to a new incremental exercise test (P2). Analysis and interpretation of electrocardiographic tracings were performed regarding the rhythm, heart rate, duration and amplitude of waves and intervals, before, immediately after and 30 min after the incremental exercise. A reduction of the rest heart rate was observed after training. There was no effect of vitamin E supplementation on electrocardiographic parameters. Results indicated that the electrocardiogram was efficient in detention of cardiac alterations promoted by the physical exercise, but more studies are needed to elucidate its clinical meaning.
