O provável fator de início de tradução 5A (eIF5A) é altamente conservado de arqueas a mamíferos e sofre uma modificação pós-traducional única e essencial chamada hipusinação. Este fator já foi relacionado ao transporte nucleocitoplasmático, à degradação de mRNA e à proliferação celular. Dados recentes restabelecem uma função para eIF5A na tradução e sugerem a sua atuação na etapa de elongação ao invés de início, como originalmente proposto. Uma vez que o envolvimento de eIF5A com a degradação de mRNA ainda não foi elucidado, tornou-se interessante estudar qual a natureza desta relação. O metabolismo de mRNA é um processo complexo que envolve as etapas da tradução, repressão da tradução e degradação de mRNA. Na primeira parte deste trabalho, foi avaliada a existência de interação genética sintética entre os mutantes tif51A-1 e tif51A-3 e mutantes de fatores envolvidos com a repressão da tradução e/ou degradação de mRNA. Foi revelada uma supressão parcial do fenótipo de termossensibilidade com nocautes dos genes SBP1, DHH1 e PAT1, que codificam fatores ativadores da remoção do capacete de metilguanosina e repressores da tradução. Por outro lado, uma interação sintético doente (“synthetic sick”) entre os mutantes tif51A-1 e xrn1Δ foi observada.Os dados obtidos reforçam o envolvimento de eIF5A com a elongação da tradução, mostram que o efeito de eIF5A na degradação de mRNA é secundário e sugerem uma função para eIF5A como ativador da tradução. Na segunda etapa são apresentados os resultados do rastreamento de supressores extragênicos do mutante tif51A-1 através de deleções genômicas induzidas por transposon. The objective of the present work was to investigate the potential of cyanobacteria isolated from different environments in decolorizing eleven different types of textile dyes. For inoculum preparation 50 ml of BG-11 medium were used for the cyanobacteria Leptolyngbia CENA103, Leptolyngbia CENA104 and Phormidium autumnale UTEX1580 and 50 ml of SWBG-11 medium for Phormidium sp., Leptolyngbya sp. and Synecochoccus sp. Test tubes containing 10 ml of liquid medium and 0.02% of each dye (remazol, indigo blue, indanthrene blue RCL, drimaren blue CL-R, dispersol blue C-2R, drimaren red CL-5B, dispersol red C- 4G, indanthrene red FBB, drimaren yellow CL-R, palanil yellow 3G and indanthrene yellow 5GF) were inoculated with cyanobacteria. A spectrophotometer was used to verify the maximum absorbance of each dye and the percentage of decolorization and also thin layer chromatography (TLC). The results showed that all the tested cyanobacteria were capable to remove more than 50% of some dyes. The present study confirmed the capacity of cyanobacteria in decolorize and possibly degrade structurally different textile dyes, suggesting the possibility of their application in bioremediation studies. The data are promising, and will lead to further studies of dye degradation and its toxicicity.

This study aimed to evaluate the force degradation of synthetic elastics over time using dynamic biomechanical tests in order to find clinical parameters of what size of elastic and frequency of exchange should be used in orthodontic therapy. It was used 240 elastics of Dental Morelli, sizes 1/8", 3/16", 1/4 "and 5/16", medium force, divided in four groups. In each group was measured forces at different times, from immediate to 72 hours after immersion in water at 37°C and incubated stretched to 600% of its initial inner diameter. Tensile tests were performed on a universal testing machine EMIC DL2000 submerged in distilled water maintained at 37°C. Data were analyzed using SPSS version 16.0, by the Analysis of Variance (One-Way) with Tukey´s post test. There was a greater increase in pattern of force decay of immediate measurement until 24 hours in all sizes of synthetic elastics, from which it was observed a decrease in force progressively smaller, with the exception of 5/16´´ elastic with a maximum force decay occurred at 12h. It was concluded that: the elastic synthetic Dental Morelli suffered significant force degradation over time, with reduction of approximately 70% of the initial force during 24 hours of stretching, followed by a progressively smaller decrease; the amount of stretch for a ideal force in intermaxillary therapies depends on the distance between the points of attachment of elastic, being necessary to consult the tables to choose the size, force and frequency of replacement.

In 2001, it was estimated that pesticide used worldwide exceeded 2.27 billion kilograms, over 35%, of which, were herbicides. Brazil is considered one of the leaders in the production of sugarcane and mainly ethanol as fuel. The monoculture of sugarcane requires the usage of a range of pesticides, among these, the herbicides diuron and tebuthiuron. The degradation products most studied (DCA and DCPU) are diuron's, especially for toxicological characteristics of this herbicide that is identified as carcinogen and suspected to be endocrine disruptor in mammals. After optimization of the chromatographic separation using HPLC-UV, the analytical curve was constructed in solvent and subsequently in the matrix (surface water). The extraction method contains the usage of SPE (solid phase extraction) (Strata-X, 200 mg/6 mL), applicating 1L of sample and elution with 5 mL of acetonitrile / methanol (50:50, v/v). Analysis by HPLC/UV was performed in gradient mode, acetonitrile/water (70/30-74/26 by 1 min, 74/26 - 78/22 till 3.2 min, returning to initial conditions and remaining this way until 10 min), 018 column (Phenomenex, 4.6 mm diameter, 250 mm long and 5pm particle size) and detection at 254 nm. Tests F and t were performed to verify the presence of the matrix effect. There was matrix effect to all analytes, ranging from -33% (DCA) and 38% (tebuthiuron). Thereby the method was optimized and validated for analysis of diuron, tebuthiuron, and DCPU DCA in surface water using HPLC/UV. The data obtained show that in order to assure the analytical reliability desired the use of the analytical curves in the matrix for the quantification of these analytes in water is required.
