Objectives: To evaluate the antimicrobial activity of Arctium lappa L. extract on Staphylococcus aureus, S. epidermidis, Streptococcus mutans, Candida albicans, C. tropicalis and C. glabrata. In addition, the cytotoxicity of this extract was analyzed on macrophages (RAW 264.7).Design: By broth microdilution method, different concentrations of the extract (250-0.4 mg/mL) were used in order to determine the minimum microbicidal concentration (MMC) in planktonic cultures and the most effective concentration was used on biofilms on discs made of acrylic resin. The cytotoxicity A. lappa L. extract MMC was evaluated on RAW 264.7 by MTT assay and the quantification of IL-1 beta and TNF-alpha by ELISA.Results: The most effective concentration was 250 mg/mL and also promoted significant reduction (log(10)) in the biofilms of S. aureus (0.438 +/- 0.269), S. epiderrnidis (0.377 +/- 0.298), S. mutans (0.244 +/- 0.161) and C. albicans (0.746 +/- 0.209). Cell viability was similar to 100%. The production of IL-beta was similar to the control group (p > 0.05) and there was inhibition of TNF-alpha (p < 0.01).Conclusions: A. lappa L. extract was microbicidal for all the evaluated strains in planktonic cultures, microbiostatic for biofilms and not cytotoxic to the macrophages. Fungi isolated from marine organisms have been shown to produce several interesting secondary metabolites with important biological activities. Such chemical diversity may be associated to environmental stress conditions and may represent an important source of NCE for bioprospection. Quinolactins belong to a rare fungi-alkaloid class with a unique N-methyl-quinolone moiety fused to a lactam ring and present several bioactivities1. Fungi strain Dm1 was isolated from red alga Dichotomaria marginata, collected from Brazil SE coast, and was grown in sterile rice solid media at 26oC 2, which was then extracted with MeOH. The MeCN fr. from the MeOH extract was chromatographed over Sephadex LH-20 and fr. 4 afforded quinolactin (QL) alkaloids B1, B2 and A, whereas fr. 5 afforded quinolactin D1 after purification by HPLC-DAD. Structural determination of pure compounds was based on HRMS, UV, and NMR spectral analyses, in addition to comparison with literature data and Antimarin® databank. UV data indicated the presence of similar chromophores with λmax at ca. 247 and 320nm. HRMS and tandem MS analyses using both negative and positive ion modes for the isolated compounds indicated their molecular formula and structural features, as for QL B1: C15H16O2N2 [M+H 257], which showed one fragment at m/z 214 [-CHNO]; QL B2: C15H16O3N2 [M+H 273], with product ions at m/z 230 [-CHNO.] and m/z 186 [-C4H9NO.]; for QL A: C16H18N2O2 [M+H 271], which presented one ion at m/z 214, due to loss of fragment (-C4H9) from the molecular ion; and for QL D1: C16H18N2O3 [M+H 287], with product ions at m/z 186 [-CHNO] and m/z 230 [-C4H9]. Such data suggested fragmentation proposals, e.g. for Quinolactin B1 (Fig. 1), which confirmed the structures of the isolated quinolactins, and may represent an important contribution for the sustainable exploration of marine biodiversity.

This study aimed to investigate the antitumor and cytotoxicity activities of Kielmeyera coriacea and Pyrostegia venusta extracts. Therefore, the hydroalcoholic extracts of P. venusta flowers and K. coriacea leaves were prepared. The extracts were evaporated and the dry extracts were diluted at concentrations of 1.0, 0.1, 0.01 and 0.001 mg/ml for carrying out the bioassays. Artemia salina eggs were incubated in saline solution at 28°C for 24 h. The larvae were treated with different extracts concentrations and the mortality was evaluated after 24 and 48 h. Five discs of potato were placed in Petri dishes and 50 μl of inoculum of Agrobacterium tumefaciens were added to it at 28°C for 24 h incubation. So, 50 μl of the extracts in different concentrations were added. Positive and negative controls were made. The P. venusta and K. coriacea extracts did not show statistically significant acute toxicity. K. coriacea extract showed (mean% of tumor ± standard deviation) 15.30 ± 3.24, 6.34 ± 3.82, 7.57 ± 2.92 and 5.77 ± 2.85 and P. venusta showed 25.82 ± 5.15, 38.40 ± 8.28, 15.75 ± 4.44 and 13.38 ± 7.92, with their concentrations for the antitumor bioassay, and the positive control showed 25.80 ± 6.14. According to the obtained results it was established that the K. coriacea and P. venusta extracts showed antitumor activity but did not show significant cytotoxic activity in A. salina test.
