The aim of this study was to investigate if the Na+-channel activating alkaloid veratrine is able to change the oxidative and m-ATPase activities of a fast-twitch glycolytic muscle (EDL, extensor digitorum longus) and slow-twitch oxidative muscle (SOL, soleus) in mice. Oxidative fibers and glycolytic fibers were more sensitive to veratrine than oxidative-glycolytic fibers 15, 30 and 60 min after the i.m. injection of veratrine (10 ng/kg) with both showing an increase in their metabolic activity in both muscles. In EDL, the m-ATPase reaction revealed a significant (p < 0.001) decrease (50%) in the number of type IIB fibers after 30 min while the number of type I fibers increased by 550%. Type I fibers decreased from 34% in control SOL to 17% (50% decrease) in veratrinized muscles, with a 10% decrease in type IIA fibers within 15 min. A third type of fiber appeared in SOL veratrinized muscle, which accounted for 28% of the fibers. Our work gives evidence that the changes in the percentage of the fiber types induced by veratrine may be the result, at least partially, from a direct effect of veratrine on muscle fibers and else from an interaction with the muscle type influencing distinctively the response of a same fiber type. Based on the results obtained in the present study the alterations in EDL may be related to the higher number of Na+ channels present in this muscle whereas those in SOL may involve an action of veratrine on mitochondria. Although it is unlikely that the shift of enzymes activities induced by veratrine involves genotypic expression changes an alternative explanation for the findings cannot be substantiated by the present experimental approach. Fiber types distribution in the diagastric muscle of tufted capuchin monkey was studied by means of NADH-TR, myosin-ATPase, after alkaline and acid preincubations and SDH histochemical reactions. Three different types of fibers were found presenting an equal distribution. The percentage and types of fibers were as follow: 18.2 % SO (Slow Oxydative), 38.4 % FOG (Fast Oxydative Glycolytic) and 43.4 % FG (Fast Glycolytic). FG fibers revealed the largest area. The relatively high concentration of fast twitch (81.2 %) seems to indicate this muscle is involved with the acceleration and fast speed of jaw movements. Aerobic metabolism represented by SO + FOG fibers (56.6 %) suggests that this muscle possesses an additional role than that related to the lowering of the jaw.

As revealed by the NADH-diaphorase and myosine ATPase, the M. extensor carpi radialis longus of the rat possesses at least 3 main kinds of fibres, with different distribution on the superficial and deep portions of the muscle. The superficial portion revealed that 67.68 % are FG (fast-twitch-glycolytic) fibres, 14.72 % are FOG (fast-twitch-oxidative) fibres and 17.60 % are SO (slow-twitch-glycolytic) fibres. Already the deep portion revealed that 71.29 % are SO (slow-twitch-glycolytic) fibres, 17.46 % are FOG (fast-twitch-oxidative-glycolytic) fibres and 11.25 % are FG (fast-twitch-glycolytic) fibres. The miosine ATPase reaction was used to demonstrate contracting characteristics. These findings suggest that the movements of fast contraction of the M. extensor carpi radialis longus are predominant.

The objective of this paper was to study the effect of sympathetic innervation on morphological and histochemical aspects of skeletal muscle tissue. Rabbit masseter muscle was studied using histochemical and immunohistochemical methods for periods of up to 18 months post-sympathectomy. The morphological and enzymatic characteristics of control masseter muscles were similar on both the left and right sides. The main features were muscle fibres with a mosaic pattern and a predominance of type IIa fibres, followed by type I. Type IIb fibres showed very low frequency. Sympathectomized animals showed varying degrees of metabolic and morphological alterations, especially 18 months after sympathectomy. The first five groups showed a higher frequency of type I fibres, whilst the oldest group showed a higher frequency of type lib fibres. In the oldest group, a significant variation in fibre diameter was observed. Many fibres showed small diameter, atrophy, hypertrophy, splitting, and necrosis. Areas with fibrosis were observed. Thus cervical sympathectomy induced morphological alterations in the masseter muscles. These alterations were, in part, similar to both denervation and myopathy. These findings indicate that sympathetic innervation contributes to the maintenance of the morphological and metabolic features of masseter muscle fibres.

The phylogenetic proximity of primates to humans, along with their behavioral, biochemical, and anatomical similarities, make such animals more interesting experimental models for biomedical researches, as compared to classical laboratory animals. Another aspect that has called the attention of researchers is the differentiated quadrupedalism present in some primates. The tufted capuchin monkey uses the ground and tree branches as its support for locomotion, showing various postures while performing this task. On the basis of this information, we have decided to study the rectus abdominis muscle of the tufted capuchin monkey, with the following goals: the frequency and area of fiber types; its possible compartmentalization; and identify if this muscle is better adapted to phasic or postural activities. To do this, samples were removed from 4 regions of the rectus abdominis muscle of 6 adult male tufted capuchin monkeys, and were submitted to reaction with m-ATPase, (with alkaline and acid pre-incubation), NADH, and H.E.. Results showed: a statistically significant difference (P<0.05) for both frequency and area, between fiber types FG and FOG and FG and SO, but did not show a statistically significant difference between fibers FOG and SO, in all studied regions; similarity in frequency and area of a same fiber type (FG, FOG, and SO) among the studied regions. Based on these data, it was concluded that: the rectus abdominis muscle of the tufted capuchin monkey does not show fiber compartmentalization, since the distribution and size patterns of the different fiber types are similar in the studied regions; there is a predominance of fast twitch fibers (FG + FOG) over slow twitch fibers (SO), for frequency and area, which characterizes the muscle as being more dedicated to phasic than to postural activities. © 2006 Sociedad Chilena de Anatom.

This experiment evaluated the growth of breast and leg muscle fibers of domestic fowl raised in two enclosure sizes (SE: Small Enclosure, 1.125 m2/10 birds; LE: Large Enclosure, 5.25 m2/10 birds). In breast muscles, the number of fibers per area decreased over time and higher values were observed in broilers housed in SE compared to LE. The fiber size increased with age and was greater in LE than SE at 56 days of age, suggesting greater hypertrophic growth of fibers in breast muscle for broilers maintained in LE. In leg muscles, the muscle cross-sectional area was greater for broilers raised in LE than SE at 56 days of age and decreased from 42 to 56 days of age in broilers raised in SE, suggesting leg muscle atrophy in these birds. The Fast Glycolytic (FG), Fast Oxidative-Glycolytic (FOG) and Slow Oxidative (SO) fibers grew until 42 days of age in both enclosure sizes. The area of FOG fibers was greater in broilers raised in LE than those in SE at 28 and 56 days of age; in LE-raised broilers, the SO area was greater at 28, 42 and 56 days of age, suggesting that the muscles of broilers housed in LE are more oxidative. The BW gain was greater for broilers raised in LE than SE, whereas BW, feed intake and feed conversion were not influenced by enclosure size. Thus, the enclosure space affected hypertrophic growth and metabolic characteristics of breast and leg muscle fibers. © Asian Network for Scientific Information, 2012.

The aim of this work was to characterize the distribution of myofibers in the gluteus medius muscle of inactive horses of the Brasileiro de Hipismo (BH) breed at different ages by means of histochemical analyses, according to sex and depth of the biopsy. A total of 78 inactive horses (9 castrated males, 35 stallions, and 34 females) of the BH breed, aged 1 to 4 years, were used. A percutaneous muscle biopsy was obtained with a 6.0-mm Bergstrom-type needle, which allowed the removal of muscle fragments at depths of 20 and 60 mm. Myofiber types were determined based on myofibrillar adenosine triphosphatase (mATPase) and nicotinamide dinucleotide tetrazolium reductase (NADH-TR) techniques. Morphometry of the fibers was determined based on cross-sectional area (CSA), mean frequency (F), and relative cross-sectional area (RCSA). The current study demonstrated that BH horses 3 and 4 years of age show a greater percentage of, and area occupied by, type IIA fibers and lower percentage of type IIX fibers in the gluteus medius muscle compared with horses 1 and 2 years of age. No difference was found between sexes in the frequency of and area occupied by the different fiber types at any of the depths and ages studied. In this study, females showed a greater CSA for all fibers in comparison with males, at 1 year of age. The results of the current study indicate that the gluteus medius muscle of inactive BH horses shows modifications in its structural and biochemical composition during the growth of the animals, leading to a better oxidative capacity.

The effect of thermal-shock cycles on the mechanical properties of fiber-metal laminates (FMLs) has been evaluated. FML plates were composed by two AA2024 Al sheets (1.6 mm thick) and one composite ply formed by two layers of unidirectional glass fiber epoxy prepreg and two layers of epoxy adhesive tape of glass fiber reinforced epoxy adhesive. The set was manufactured by hand layup and typical vacuum bag technique. The curing cycle was in autoclave at 125 +/- 5 degrees C for 90 min and an autoclave pressure of 400 kPa. FML coupons taken from the manufactured plate were submitted to temperature variations between -50 and +80 degrees C, with a fast transition between these temperatures. Tensile and interlaminar shear strength were evaluated on samples after 1000 and 2000 cycles, and compared to nonexposed samples. 2000 Cycles corresponds to typical C Check interval for commercial aircraft maintenance programs. It was observed that the thermal-shock cycles did not result in significant microstructural changes on the FML, particularly on the composite ply. Similarly, no appreciable effect on the mechanical properties of FML was observed by the thermal-shock cycles. (c) 2012 Elsevier Ltd. All rights reserved.

Objective: To associate changes of body composition, muscle strength (MS) and plasma hormones (PH) in resistance-training protocol in sedentary postmenopausal women (PMW).Design: This randomized controlled trial, Brazilian 43 PMW (45-70-year-old) able for physical exercises were selected after they have accomplished medical and ethical criteria. They were assigned in two groups: RT, resistance training (n = 22); and CT, not trained control (n = 2 1); with supervision sessions of two to three exercise for large and one exercise for smaller groups in three series of 8-12 rep. (60-80% 1RM) for each exercise. The training period lasted 16 weeks and was preceded by low-load exercise (40-50% 1RM) adaptation period of 4 weeks (3/(times week)). Body weight, height, body mass index (BMI), and composition (BIA) along with fast-PH (FSH, LH, estrachol, cortisol, IGF-1 and testosterone) were assessed before (MO) and after (M 16) the 4 weeks period with the MS (1RM) determined also at 8 weeks (W). The values were correlated by Person's test and the means compared by Student's t-test and ANOVA.Results: At baseline both groups were similar in age, time of PMW, body composition, MS and fast-PH. However after 16 weeks, RT presented higher BMI (2. 1 %), IGF- 1 (37.8%) and MM gain (1.8 +/- 0.8 kg) than CT. MM correlated positively with IGF-1 (r = 0.45, p < 0.05) and MS progressively increased in all exercise greater in pectoral than legs and upper arms.Conclusion: Former sedentary postmenopausal women submitted to resistance training gained MM and MS irrespectively of fat mass changes but significantly associated with IGF-1 increase. O objetivo deste trabalho foi avaliar o efeito da temperatura ambiente sobre as características morfométricas das fibras musculares do músculo flexor longo do hálux de frangos de corte alimentados com a mesma quantidade de ração (pair-feeding), isolando-se o efeito do consumo voluntário de alimento. Foram utilizados 16 frangos de corte machos, Ross, distribuídos em um delineamento inteiramente casualizado com dois tratamentos (estresse pelo calor e frio), com sete e nove repetições cada, respectivamente. Aos 43 dias de idade, o peso médio das aves estressadas pelo calor (1255 g) foi maior que o das aves estressadas pelo frio em pair-feeding (1086 g). O músculo das aves submetidas ao estresse pelo calor e pelo frio apresentou 22,82 e 27,93% de fibras brancas (FG - Fast Glycolytic), 52,76 e 47,67% de intermediárias (FOG - Fast Oxidative Glycolytic) e 24,42 e 24,40% de vermelhas (SO - Slow Oxidative), respectivamente. O diâmetro das fibras FG foi maior no músculo das aves submetidas ao calor (48,69 mm), quando comparado ao das aves submetidas ao frio (37,74 mm). A freqüência e o número dos tipos de fibras no músculo não diferiram entre as aves estressadas pelo calor e pelo frio. O estresse pelo frio associado à limitação no consumo de alimento determinou redução no crescimento e alterou a composição dos tipos de fibras no músculo flexor longo do hálux de frangos de corte, reduzindo o tamanho das fibras do tipo FG e mantendo o tamanho das fibras SO e FOG, as quais apresentam metabolismo oxidativo, associado à maior produção de calor.

Objetivando-se estudar o efeito da somatotropina bovina recombinante (rbST) sobre a freqüência de distribuição e o diâmetro das fibras musculares do músculo Semitendinosus, 36 bezerros mestiços ½Angus-Nelore, com idade inicial de 63 ± 17 dias e pesando 76,8 ± 14,7 kg, criados em pastagem de Brachiaria decumbens e suplementados em creep feeding, foram submetidos a dois tratamentos até a desmama (217 dias): 18 bezerros receberam 1,4 mg/kg de rbST (Boostin®) a cada 14 dias e 18 receberam solução salina (controle). As amostras de músculo foram coletadas aos 117 (biópsia) e aos 217 dias de idade, quando foram abatidos cinco animais por tratamento. Os animais suplementados apresentaram maior diâmetro para as fibras do tipo glicolítica de contração rápida (FG) aos 117 dias e tendência de aumento aos 217 dias e não diferiram em relação ao grupo controle quanto ao diâmetro das fibras oxidativas-glicolíticas de contração rápida (FOG) e oxidativas de contração lenta (SO) e à frequência de FG, FOG e SO aos 117 e 217 dias de idade. Independentemente da aplicação de rbST, houve significativo aumento do diâmetro das fibras SO e FOG, tendência de aumento de diâmetro das fibras FG, maior frequência de SO e redução da frequência de FG entre 117 e 217 dias de idade. A utilização de somatotropina exógena possibilitou maior hipertrofia das fibras musculares brancas de contração rápida em bezerros suplementados em creep feeding durante a fase de cria, sem interferir na frequência de distribuição dos tipos de fibras no músculo Semitendinosus.
