Drug therapy involving bone tissue diseases is difficult, calling for the design of specific drugs. The present paper is a brief review of a new site-directed system termed ODDS (osteotropic drug delivery system), based on a latenciation process, using bisphosphonates as bone carriers. This is an important tool for the rational prodrug design for obtaining selective drugs.

OBJETIVO: Estudar as alterações agudas da interface entre os parafusos e o tecido ósseo vertebral, considerando o diâmetro do orifício-piloto em relação ao diâmetro interno do parafuso. MÉTODOS: O estudo foi realizado em carneiros e a segunda vértebra cervical selecionada para o estudo. Foram utilizados parafusos corticais de 3,5mm de diâmetro externo (2,4mm de diâmetro interno) e 14mm de comprimento. Foram formados quatro grupos experimentais: 1) orifício-piloto de 2,0mm e colocação do parafuso de 3,5mm; 2) orifício-piloto de 2,5mm e colocação do parafuso de 2,5mm; 3) orifício-piloto de 2mm sem colocação do parafuso; e 4) orifício-piloto de 2,5mm sem colocação do parafuso. RESULTADOS: O grupo 1 (orifício-piloto menor do que o diâmetro interno do parafuso) apresentou maior densidade óssea no interior da rosca do implante e maior densidade óssea na área em espelho do que o grupo 2 (orifício-piloto maior que o diâmetro interno do parafuso). A superfície de contato entre o tecido ósseo e o implante foi maior no grupo 2 em relação ao grupo 1. A comparação da densidade óssea na área em espelho entre os grupos com e sem implante (1 e 3) e (2 e 4) mostrou que a densidade óssea era maior nos grupos com o implante. Não foi observada diferença estatística entre as comparações dos grupos experimentais. CONCLUSÃO: A utilização de orifício-piloto de menor diâmetro em relação ao diâmetro interno do parafuso apresentou tendência a provocar maior compactação do osso ao redor do implante.

As lesões tendíneas nas extremidades distais dos membros estão entre as mais freqüentes alterações do aparelho locomotor na rotina clínico-cirúrgica humana e animal e, não raro, necessitam de terapias adjuvantes para seu completo retorno às funções fisiológicas. O ultra-som terapêutico (UST) é a modalidade mais utilizada nas clínicas de reabilitação para tratar lesões tendíneas, mas devido à falta ou a divergências de estudos específicos sobre seus efeitos no tecido ósseo, sua utilização sobre as regiões distais dos membros, ricas em protuberâncias ósseas e áreas desprovidas de cobertura muscular, sempre preocuparam os profissionais da área médica. No intuito de esclarecer os efeitos do UST sobre o tecido ósseo, seis cães receberam tratamento ultra-sônico contínuo, de 1MHz, durante cinco minutos diários, por um período de 20 dias sobre a região craniodistal do rádio e da ulna. A intensidade do UST aplicada foi de 0,5W cm-2 no membro torácico direito, ficando o membro contralateral como controle. A região distal de ambos os membros torácicos foi radiografada para análise de densitometria óssea em imagens radiográficas, antes do início da terapia e ao final do tratamento. Não houve alterações significativas de densidade mineral óssea entre os membros tratados e os controles. Conclui-se que dentro dos parâmetros utilizados no experimento a utilização do UST em regiões ósseas protuberantes ou desprovidas de cobertura muscular pode ser feita com segurança.

O trabalho foi conduzido no Setor de Ovinocultura do Departamento de Zootecnia da UFLA, em Lavras, com o objetivo de estudar o crescimento alométrico dos tecidos ósseo, muscular e adiposo dos cortes comerciais em cordeiros. Foram utilizados 24 cordeiros machos inteiros da raça Santa Inês. Os animais foram submetidos a regime de confinamento e receberam alimentação ad libitum. O abate ocorreu quando os animais atingiram os pesos vivos de 15, 25, 35 e 45 kg. Após a carcaça ter sido limpa e resfriada, foram obtidos os cortes comerciais a partir da meia carcaça esquerda. A quantidade dos diferentes tecidos foram obtidos por intermédio da dissecação de perna, lombo, costeleta, costela/ fralda e paleta. O estudo do desenvolvimento relativo da composição tecidual foi feito por meio do modelo de HUXLEY (1932). Constatou-se crescimento heterogônico negativo (b < 1) para o tecido ósseo e heterogônico positivo (b > 1) para o tecido adiposo de todos os cortes. Quanto ao tipo de crescimento do tecido muscular da costela/fralda e paleta, foi verificado crescimento isogônico (b=1) deste tecido em relação aos cortes.

O trabalho foi realizado com objetivo de realizar um estudo alométrico dos tecidos ósseo, muscular e adiposo na meia carcaça esquerda de cordeiros em crescimento. Foram utilizados 36 cordeiros machos inteiros, sendo 24 Santa Inês e 12 Bergamácia. Os animais foram confinados em gaiolas individuais e alimentados ad libitum. O abate ocorreu quando os animais atingiram os pesos vivos de 15, 25, 35 e 45 kg. Após a carcaça ter sido limpa e resfriada, foram obtidos os cortes comerciais a partir da meia carcaça esquerda. A quantidade dos diferentes tecidos foi obtida a partir da dissecação da perna, do lombo, da costeleta, costela/fralda e paleta. A quantidade total dos tecidos ósseo, muscular e adiposo foi obtida pela adição das quantidades de cada corte comercial. O estudo do desenvolvimento relativo da composição tecidual foi feito por meio do modelo de HUXLEY (1932). Constatou-se crescimento heterogônico negativo (b < 1) para o tecido ósseo, crescimento isogônico (b =1) para o tecido muscular e heterogônico positivo (b > 1) para a gordura, em relação ao peso da ½ carcaça.

Ingestion of transgalactosylated oligosaccharides (TOS) and other non-digestible oligosaccharides (NDOs) induces a significant increase in Bifidobacterium, Lactobacillus and some desirable species of Streptococcus populations in the gut of human and other animals (prebiotic effect). This change in the intestinal flora is responsible for several beneficial physiological effects such as a decrease of putrefactive products in the feces, lower blood cholesterol content, higher Ca2+ absorption, a smaller loss of bone tissue in ovariotomized female rats and a lower incidence of colon cancer. beta-Galactosidase from Penicillium simplicissimum, a strain isolated from soil, showed high galactosyltransferase activity when incubated with a highly concentrated lactose solution. Optimum pH and temperature ranges for hydrolytic activity were 4.0-4.6 and 55-60 degrees C, respectively, for a lactose concentration of 5.0% (w/v). Maximal galactosyltransferase activity was obtained at pH 6.5 and 50 degrees C and TOS synthesis was positively associated with lactose concentration in the reaction medium. Thus, when 50 ml of a 60% (w/v) lactose solution was incubated with 26.6 U of beta-galactosidase under the best pH and temperature conditions for transferase activity, a final product with 30.5% TOS (183 mg ml(-1)), 27.5% residual lactose and 42.0% monosaccharides was obtained. 1. We investigated the effect of a persistent carrageenin- or nystatin-induced inflammatory reaction on heterotopic ossification produced by the subcutaneous implant of a demineralized bone matrix in female Swiss mice (25 to 35 g).2. Subcutaneous carrageenin injection (0.3 ml of a 2% solution in saline) into mice induced an inflammatory reaction characterized by a mature granuloma predominantly of macrophages containing particles of the irritant in their cytoplasm and which remained unchanged until the end of the experiment (40th day).3. Subcutaneous nystatin inoculation (30,000 IU in 0.3 ml saline) induced an inflammatory reaction consisting initially of macrophages (4th day) but later turning into an epithelioid granuloma (7th day) consisting predominantly of epithelioid cells and which was present up to the 2 lst day when it was gradually replaced by adipocytes up to the 30th day.4. An intramuscular implant of demineralized bone matrix (DBM, approximately 10 mg) induced the formation of cartilage and bone tissue and of hemopoietic bone marrow (heterotopic ossification) in 100% of the control animals (N = 5). An intramuscular DBM implant in animals that received carrageenin (N = 19) or nystatin (N = 21) induced heterotopic ossification in 100 and 57% (P<0.01)) of the animals, respectively.5. The response to a dorsal subcutaneous DBM implant was essentially negative in control animals (N = 5), whereas implants performed near the site injected with carrageenin (N = 28) or nystatin (N = 31) produced a response in 71 (P <0.01) and 36 % (P<0.01) of the animals, respectively. A DBM implant into the contralateral (control) dorsal subcutaneous tissue of the same animals that received carrageenin (N = 25) or nystatin (N = 29) resulted in heterotopic ossification in 64 (P<0.01) and 7% of the animals, respectively.6. The results suggest that the macrophages present in the mature granuloma induced by carrageenin somehow favored the development of metaplastic plates after subcutaneous DBM implant and that this effect may be systemic since the same response was observed in contralateral subcutaneous tissue.

Some divergencies in the literature about periodontal healing after surgical injury stimulated the development of this experiment. The root canals of dogs' teeth were negotiated and filled by the lateral condensation technique with two kinds of sealers: Sealapex and zinc oxide-eugenol cement. In the second session, the bone tissue was exposed and one cavity was made at the apical third of the root and another at the border between the coronal and middle thirds, both penetrating into the root canal. Six months later the animals were sacrificed and the specimens prepared for histopathologic analysis. The results showed that the kind of filling material and the level of the periodontal wound exposing the root canal can influence the healing process (P<0.01).

Osseointegration was an innovative treatment in dentistry during the last 3 decades. The success of osseointegration is related to factors such as material biocompatibility, adequate quality of bone tissue that allows implantation, surgical technique, and macrostructure and microstructure of implant. The osseointegrated implants are successfully applied in dental clinic including oral and facial rehabilitations mainly for patients submitted to mutilating surgeries. However, patients submitted to radiation therapy present risks to treatment with implants owing to adverse effects on bone tissue. Nowadays, the literature suggests different therapies to improve the success of osseointegration such as hyperbaric oxygen therapy that aims to prepare bone and adjacent tissues to receive the implant. Therefore, the purpose of this study was to present a literature review concerning indications, contraindications, successes, and difficulties with hyperbaric oxygen therapy associated to maxillofacial implants.

Inferior Alveolar Nerve (IAN) transposition is an option for prosthetic rehabilitation in cases of moderate or even severe bone reabsorption for patients that do not tolerate removable dentures. The aim of the present report is to describe an inferior alveolar nerve transposition with involvement of the mental foramen for implant placement. The surgical procedure was performed under local anesthesia, by the inferior alveolar, lingual and buccal nerve blocking technique. Centripetal osteotomy was performed, and bone tissue was removed, leaving the nerve tissue free in the foramen area. After that, transsection of the incisor nerve was performed, and lateral osteotomy was started from the buccal direction, toward the trajectory of the IAN. The procedure was concluded, by making use of a delicate resin spatula to manipulate the vascular-nervous bundle. The drilling sequence for placing the dental implants was performed, and autogenous bone was harvested using a bone collector attached to the surgical suction appliance. After the implants were placed, the bone tissue previously collected during the osteotomies and drilling processes was placed in order to protect the IAN from contact with the implants. The surgical protocol for inferior alveolar nerve transposition, followed by implant placement presented excellent results, with complete recovery of the sensitivity, seven months after the surgical procedure.

Natural or synthetic materials may be used to aid tissue repair of fracture or pathologies where there has been a loss of bone mass. Polymeric materials have been widely studied, aiming at their use in orthopaedics and aesthetic plastic surgery. Polymeric biodegradable blends formed from two or more kinds of polymers could present faster degradation rate than homopolymers. The purpose of this work was to compare the biological response of two biomaterials: poly(L-lactic acid)PLLA and poly(L-lactic acid)PLLA/poly(ethylene oxide)PEO blend. Forty four-week-old rats were divided into two groups of 20 animals, of which one group received PLLA and the other PLLA/PEO implants. In each of the animals, one of the biomaterials was implanted in the proximal epiphysis of the right tibia. Each group was divided into subgroups of 5 animals, and sacrificed 2, 4, 8 and 16 weeks after surgery, respectively. Samples were then processed for analysis by light microscopy. Newly formed bone was found around both PLLA and PLLA/PEO implants. PLLA/PEO blends had a porous morphology after immersion in a buffer solution and in vivo implantation. The proportion 50/50 PLLA/PEO blend was adequate to promote this porous morphology, which resulted in gradual bone tissue growth into the implant.

A case of peripheral ameloblastoma in a 57-years-old woman is presented, along with a discussion of the clinical and histological characteristics of the lesion. After clinical and radiographic examinations, and with a differential diagnosis of pyogenic granuloma, an excisional biopsy was performed and the material collected was sent for histological examination. On the basis of the histopathological diagnosis, a second operation was performed with a wide safety margin, including bone tissue, which did not show any involvement with the odontogenic neoplasm.

The advantages and disadvantages of maintaining the periodontal ligament (PDL) in immediate replantation as well as chemical treatment of the root surface have been a matter of discussion because the vitality of such tissue in surgery is always questioned. This study evaluated the effects of conserving the tooth in sodium fluoride and the removal of the PDL before replantation of incisors in rats. There was more cementum-dentin resorption in the group with the PDL. The group without the PDL showed more discreet resorption, repair occurred through the newly formed bone tissue in the PDL space and ankylosis was more extensive than in the group with the PDL.
