LASSBio-767 [(-)-3-O-acetyl-spectaline] and LASSBio-822 [(-)-3-O-tert-Boc-spectaline] were recently described as cholinesterase inhibitors derived from the natural piperidine alkaloid (-)-spectaline, obtained from the flowers of Senna spectabilis (Fabaccae). We investigated their mechanism of inhibition of acetylcholinesterase and their efficacy in reversing scopolamine-induced amnesia. Competition assays with the substrate acetylthiocholine showed a concentration-dependent reduction in rat brain cholinesterase V-max without changes in apparent K-m. The kinetic data for LASSBio-767 and LASSBio-822 were best fit by a model of simple linear noncompetitive inhibition with K-i of 6.1 mu M and 7.5 mu M, respectively. A dilution assay showed a fast and complete reversal of inhibition, independent of incubation time. Simulated docking of the compounds into the catalytic gorge of Torpedo acetylcholinesterase showed interactions with the peripheral anionic site, but not with the catalytic triad. Anti-amnestic effects in mice were assessed in a step-down passive avoidance test and in the Morris water maze 30 min after injection of scopolamine (1 mg/kg i.p.). Saline, LASSBio-767, or LASSBio-822 was administered 15 min before scopolamine. Both compounds reversed the scopolamine-induced reduction in step-down latency at 0.1 mg/kg i.p. LASSBio-767 reversed scopolamine-induced changes in water maze escape latency at 1 mg/kg i.p. or p.o., while its cholinergic side effects were absent or mild up to 30 mg/kg i.p. (LD50 above 100 mg/kg i.p.). Thus, the (-)-spectaline derivatives are potent cholinergic agents in vivo, with a unique profile combining noncompetitive cholinesterase inhibition and CNS selectivity, with few peripheral side effects. O crambe é uma cultura promissora para produção de biodiesel, principalmente pelo alto conteúdo de óleo de suas sementes. No entanto, não há metodologias estabelecidas para avaliar a qualidade fisiológica das sementes desta espécie e lotes não podem ser comparados, especialmente por testes de vigor, como o de envelhecimento acelerado. O objetivo da presente pesquisa foi avaliar o efeito da alta temperatura e do período de exposição durante o teste de envelhecimento acelerado na qualidade fisiológica e atividade enzimática de sementes de crambe. Dois lotes de sementes de crambe, cultivar Brilhante, foram analisados por meio dos testes de teor de água, massa de 1.000 sementes, germinação, primeira contagem, condutividade elétrica, atividade enzimática (peroxidase e superóxido dismutase) e comprimento de plântulas. As avaliações foram conduzidas antes e após o envelhecimento acelerado, que foram testadas diferentes temperaturas (38, 40 e 42ºC) e períodos de exposição (24, 48 e 72h). O delineamento experimental foi o inteiramente casualizado com quatro repetições. Os dados foram submetidos à análise de variância e as médias foram comparadas pelo teste de Tukey (p < 0.05). O teste de Dunnet (p < 0.05) foi utilizado para comparar os valores da testemunha (antes do envelhecimento acelerado) com cada valor médio individualmente. O teste de correlação linear simples também foi aplicado. Conclui-se que a interação temperatura x período de exposição afeta a qualidade fisiológica das sementes e a atividade enzimática e que as melhores condições durante o teste de envelhecimento acelerado são dependentes do genótipo.

Esse trabalho visou investigar o efeito da idade de aves sobre a digestibilidade dos nutrientes da soja integral extrusada (SIE), soja integral tostada à vapor (SITV) e farelo de soja com óleo (FSO) na produção de enzimas digestivas do pâncreas. Cinco ensaios de digestibilidade foram conduzidos com frangos de corte de uma, duas, três, quatro e seis semanas de idade. Foi utilizada a metodologia de coleta total de excretas. A atividade das enzimas amilase e tripsina pancreática aumentou linearmente com a idade das aves, assim como o crescimento alométrico do pâncreas. A maior taxa de crescimento ocorreu na segunda semana, coincidindo com a fase de maior aumento da atividade das enzimas digestivas. Entretanto, para a atividade da lipase, o efeito da idade foi diferente para cada alimento. Para aves alimentadas com SITV, a atividade dessa enzima cresceu linearmente com a idade, enquanto nas alimentadas com SIE, FSO e ração, o efeito foi quadrático. Os coeficientes de digestibilidade da matéria seca e extrato etéreo e os valores de energia metabolizável dos tipos de sojas variaram em proporções diferentes em função da idade. Também, observou-se correlação positiva entre a digestibilidade do extrato etéreo e a atividade de lipase. Os valores de energia metabolizável aparente corrigida (EMAn) e verdadeira corrigida (EMVn) determinados para a SIE apresentaram comportamento quadrático em função da idade, ocorrendo aumento da energia metabolizável (EM) até a 3ª semana de idade e diminuindo a partir da 4ª semana. Entretanto, a em da SITV, FSO e da ração não foi afetada pela idade da ave. O aproveitamento da energia dos alimentos varia com a idade das aves, em função de sua dependência da atividade enzimática.

Leachate samples from a sanitary landfill of Araraquara city and composting usine of Vila Leopoldina, São Paulo, Brazil were lyophilized to remove the water content. TG/DTG curves at different heating rates were recorded. The second step of the thermal decomposition of leachate from the Araraquara landfill (CB1), from the composting usine from Vila Leopoldina (CB2) from the organic phase extracted (FO) and aqueous phase (FA) were all kinetically evaluated using the non-isothermal method.By Flynn-Wall isoconversional method the following values were obtained: E=234 +/- 3.65 kJ mol(-1) and logA=29.7 +/- 0.58 min(-1) for CB1; E=129 +/- 1.66 kJ mol(-1) and logA=11.8 +/- 0.10 min(-1) for CB2; E=51.6 +/- 1.35 kJ mol(-1) and logA=6.09 +/- 0.09 min(-1) for FO and E=76.91 +/- 6.33 kJ mol(-1) and logA=8.88 +/- 0.7 min(-1) for FA with 95% confidence level. Applying the procedures of Malek and Koga, SB kinetic model (Sestak-Berggren) is the most appropriate to describe the decomposition of CB1, CB2, FO and FA.

In this study the relationship between the enzymatic susceptibility and the size of the com and cassava starch granules was studied. The starch granules were separated by size and classified according to their average diameter in: a) larger than 16 mum; b) between 15 and 10 mum and c) smaller than 10 mum. The starch granules of various sizes were hydrolyzed by bacterial alpha-amylase and fungal amyloglucosidase. The results showed a relationship between the enzymatic susceptibility and the size of the starch granules; smaller size of the starch granules resulted in a higher percentage of hydrolysis. A basic difference in the mode of action of enzymes on small and large granules was observed. Enzymatic attack on the large granules was characterized by considerable surface corrosion, mainly at the radial axis. For small granules, the enzymatic action occurred on the surface of the granules and was characterized by an erosion with solubilization of the granules. Chemical and physical analysis of the starches suggested that hydrolysis should occur mainly at the amorphous areas of the granules.

Submandibular glands of male rats were homogenized with 33 mM sodium potassium phosphate buffer, pH 6.5, containing 1 mM MgCl2 and 0.1 mM DTT and purified with ammonium sulphate, phosphocellulose chromatography, eluted with KC1 0.5 M, followed by Blue Sepharose CL-6B chromatography, eluted with NADH 0.5 mM. The enzyme kepts stable for 60 days when stored at -15-degrees-C in 33 mM phosphate buffer. In other experiment the enzyme was purified by oxamate-agarose chromatography from a crude extract of submandibular gland and the results obtained were better than by phosphocellulose and Sepharose CL-6B chromatography. The Km values for pyruvate. NADH, lactate and NAD+ were established. Sodium oxamate at 0.1 and 0.9 mM concentrations inhibited the LDH activity by 40 and 85%, respectively (competitive); with sodium oxalate the inhibition was of 30% (uncompetitive) and with 3-acetyl pyridine adenine dinucleotide was 80%.

Supercritical fluid extraction (SFE) from solids has proven to be technically feasible for almost any system; nonetheless, its economical viability has been proven for a restricted number of systems. A common practice is to compare the cost of manufacturing of vegetable extracts by a variety of techniques without deeply considering the huge differences in composition and functional properties among the various types of extracts obtained; under this circumstance, the cost of manufacturing do not favor SFE. Additionally, the influence of external parameters such as the agronomic conditions and the SFE system geometry are not considered. In the present work, these factors were studied for the system fennel seeds + CO2. The effects of the harvesting season and the degree of maturation on the global yields for the system fennel seeds + CO2 were analyzed at 300 bar and 40 degrees C. The effects of the pressure on the global yields were determined for the temperatures of 30 and 40 degrees C. Kinetics experiments were done for various ratios of bed height to bed diameter. Fennel extracts were also obtained by hydrodistillation and low-pressure solvent extraction. The chemical composition of the fennel extracts were determined by gas chromatography. The SFE maximum global yield (12.5%, dry basis) was obtained with dry harvested fennel seeds. Anethole and fenchone were the major constituents of the extract; the following fat acids palmitic (C16H32O2), palmitoleic stearic (C18H36O2), oleic (C18H34O2), linoleic (C18H32O2) and linolenic (C18H30O2) were also detected in the extracts. A relation between amounts of feed and solvent, bed height and diameter, and solvent flow rate was proposed. Fluoride glasses have been extensively studied due to their high transparency in the infrared wavelength. The crystallization kinetics of these systems has been studied using DTA and DSC techniques. Most of the experimental data is frequently investigated in terms of the Johnson-Mehl-Avrami (JMA) model in order to obtain kinetic parameters.In this work, DSC technique has been used to study the crystallization of fluorozirconate glass under non-isothermal conditions. It was found that JMA model was not fit to be applied directly to these systems, therefore, the method proposed by Malek has been applied and the Sestak-Berggren (SB) model seems to be adequate to describe the crystallization process.

A modified spectrophotometric method for serum glutamic-oxaloacetic transaminase (SGOT) assay was developed. A crude cell-free extract from Streptomyces aureofaciens which showed a high level of malate dehydrogenase (MDH) activity (E.C. 1.1.1.37) was used as the enzymatic indicator. The lyophilized microbial preparation was used without previous purification and was quite stable under refrigeration for one year. Serum sample assays using both the method utilizing the crude cell extract and an enzymatic commercial kit showed good correlation.
