Um experimento foi realizado com leitões desmamados visando avaliar os efeitos de oito tratamentos, correspondentes à combinação dos fatores, apresentações de ração (seca e úmida), tipos de ração (sem e com edulcorante) e tipos de água de consumo (sem e com edulcorante) sobre o desenvolvimento morfológico e enzimático intestinal. Foram avaliadas a altura das vilosidades (V), a profundidade das criptas (C) e a relação altura das vilosidades e profundidade das criptas (V:C) do duodeno e do jejuno, o conteúdo de proteína na mucosa do jejuno, e as atividades enzimáticas (UA) da maltase, sacarase e dipeptidase. Utilizaram-se 32 leitões machos castrados, Large White x Landrace, desmamados aos 21 dias, submetidos aos tratamentos durante oito dias. O delineamento experimental foi em blocos casualizados, com oito tratamentos, fatorial 2 x 2 x 2, com quatro repetições. Foram observadas diferenças para a altura das vilosidades do duodeno para os grupos tratados com dieta úmida e ração com edulcorante e para a relação V:C do duodeno para os animais que receberam ração com edulcorante. Houve diferença para a UA da sacarase e da maltase para os grupos que receberam água sem edulcorante. As características histológicas foram favorecidas pelas dietas úmidas, o que pode sugerir que esta apresentação foi menos agressiva ao trato digestivo. Para os demais fatores os resultados contrariaram as expectativas de que o tratamento água com edulcorante melhoraria a qualidade do epitélio intestinal. A fase crítica pela qual os leitões foram submetidos, possivelmente, comprometeu os resultados.

The preventative intestinal anti-inflammatory activity of paepalantine, an isocoumarin isolated from the capitula of Paepalarithus bromelioides, was tested in the trinitrobenzenesulphonic acid (TNBS) model of rat colitis. This was performed in two different experimental settings, i. e. when the colonic mucosa is intact or when the mucosa is in process of recovery after an initial insult. The results obtained revealed that the paepalantine pretreatment, at doses of 5 and 10 mg/kg, significantly attenuated the colonic damage induced by TNBS in both situations, as it was evidenced both histologically and biochemically. This beneficial effect was associated with an improvement in the colonic oxidative status, since paepalantine prevented the glutathione depletion that occurred as a consequence of the colonic inflammation. In addition, the intestinal anti-inflammatory effect exerted by this isocoumarin was associated with an inhibition of colonic nitric oxide activity, which is upregulated as a consequence of the inflammatory process. In conclusion, the preventative effect exerted by paepalantine in the TNBS model of rat colitis is probably related with its antioxidant properties.

INTRODUÇÃO: A recidiva local no câncer colorretal tem como principal causa o implante de células tumorais nas anastomoses. 11-15 Dessa maneira, lavagem química do lúmen intestinal é preconizada para evitar tanto o implante quanto à recidiva local. 11-28 em estudos prévios constatamos que a solução bicarbonatada de ácido acetilsalicílico tem efeitos citolíticos e anti-tumorais in-vitro.31 OBJETIVOS: Avaliar a toxicidade da solução de aspirina na mucosa colônica de coelhos com o objetivo de usá-la no preparo intestinal de portadores de câncer colorretal. MATERIAIS E MÉTODOS: Foram utilizados 20 coelhos. Um clampe vascular foi colocado acima do cólon sigmóide. Os animais foram submetidos a um enema com 50 ml da solução de aspirina ou soro fisiológico de acordo com o grupo. Os animais foram sacrificados ao término do procedimento ou tardiamente de acordo com o grupo. RESULTADOS: A solução de aspirina não altera a mucosa colônica de coelhos. CONCLUSÃO: O uso da solução bicarbonatada de ácido acetilsalicílico no preparo intestinal de portadores de câncer colorretal é clinicamente possível.

1. The interaction between experimental protein deprivation and natural intestinal infection by Giardia lamblia was studied in terms of its effects on the intraepithelial lymphocyte (IEL) population and morphology of the jejunal mucosa of rats of different ages.2. Young, adult and old male Wistar rats received a protein-deficient diet (2% casein) or a control diet (20% casein) for 42 days. Mucosal height and the number of lymphocytes located among 500 consecutive epithelial cells (EC) along the villi or crossing the basement membrane were determined in PAS-stained jejunal fragments.3. The number of IEL increased progressively with animal age, from 14 to 25 per 100 epithelial cells, with significant differences between age ranges. However, the number of IEL did not differ between control and protein-deficient rats in any of the age groups. The proportion of lymphocytes crossing the basement membrane was approximately two-fold greater in young (2.8/100 EC) and adult (5.8/100 EC) protein-deficient animals than in their respective controls (1.6 and 2.8/100 EC). The intensity of parasite colonization was moderate, from 3 to 5/100 EC and did not differ between groups. The pattern of morphologic changes of jejunal mucosa in protozoal infection did not differ between control and protein-deficient animals in any of the three age groups.4. We conclude that intestinal infection with Giardia lamblia probably stimulated the local immune response, masking the reduction of the IEL population induced by protein deficiency. The increase in lymphocyte numbers with age may be related to prolonged antigenic stimulation promoted by infection.

A trial was carried to verify the effect of increasing levels (0, 10, 20, 30 and 40%) of canola meal in diets of broiler chickens on performance and mucous morphology, during the starting period (1 - 21 days). One thousand 1-d male chicks were assigned to a randomized experimental assay, with five treatments, four replicates and 50 birds per experimental unit. At the 1 st, 21 st and 41 st days, the birds and the rations were weighed. At the 21 st day, two birds of each experimental unit were sacrificed for collection of a duodenum segment to evaluate the intestinal morphometry. The results demonstrated that increasing levels of canola meal resulted in linear reduction on weight gain, weight and feed intake and worst feed conversion. During the growing period (21 to 41 days), when all birds received similar diet, average weight and weight gain decreased, as the canola meal levels increased, while feed conversion and feed intake were similar. Data regarding mucous morphology, submitted to the regression analysis, demonstrated that the crypt depth increased, as the canola meal levels increased. It is possible to include up to 20% of canola meal without damages on broiler performance and digestive tract.

Coumarins represent an important class of phenolic compounds with multiple biological activities, including inhibition of lipidic peroxidation and neutrophil-dependent anion superoxide generation, anti-inflammatory and immunosuppressor actions. All of these proprieties are essential for that a drug may be used in the treatment of inflammatory bowel disease. The present study examined intestinal anti-inflammatory activity of coumarin and its derivative, the 4-hydroxycoumarin on experimental ulcerative colitis in rats. This was performed in two different experimental settings, i.e. when the colonic mucosa is intact or when the mucosa is in process of recovery after an initial insult. The results obtained revealed that the coumarin and 4-hydroxycoumarin, at doses of 5 and 25 mg/kg, significantly attenuated the colonic damage induced by trinitrobenzenesulphonic acid (TNBS) in both situations, as evidenced macroscopically, microscopically and biochemically. This effect was related to an improvement in the colonic oxidative status, since coumarin and 4-hydroxycoumarin prevented the glutathione depletion that occurred as a consequence of the colonic inflammation. © 2008 Pharmaceutical Society of Japan.
