Prostatic lesions such as prostatic intraepithelial neoplasia (PIN) and proliferative inflammatory atrophy (PIA) are studied in human and canine species due to their malignance potential. The plasminogen activator (PA) system has been suggested to play a central role in cell adhesion, angiogenesis, inflammation, and tumor invasion. The urokinase-type plasminogen activator receptor (uPAR) is a component of the PA, with a range of expression in tumor and stromal cells. In this study, uPAR expression in both canine normal prostates and with proliferative disorders (benign prostatic hyperplasia-BPH, proliferative inflammatory atrophy-PIA, prostatic intraepithelial neoplasia-PIN, and carcinoma-PC) was evaluated by immunohistochemistry in a tissue microarray (TMA) slide to establish the role of this enzyme in extracellular matrix (ECM) remodeling and in the processes of tissue invasion. A total of 298 cores and 355 diagnoses were obtained, with 36 (10.1%) normal prostates, 46 (13.0%) with BPH, 128 (36.1%) with PIA, 74 (20.8%) with PIN and 71 (20.0%) with PC. There is variation in the expression of uPAR in canine prostate according to the lesion, with lower expression in normal tissue and with BPH, and higher expression in tissue with PIA, PIN and PC. The high expression of uPAR in inflammatory and neoplastic microenvironment indicates increased proteolytic activity in canine prostates with PIA, PIN, and PC.

The oral cancer model in hamsters shows many simílarities with developmental oral cancer in humans. The proliferating capacity is one the most characteristics of neoplásica ce/Is and detection of these ce/Is allow us, throughout of its counting, to achieve an estimated tumour growing index, with a consequent repercussion about prognostic and in the treatment of those lesions. 40 golden Syrian hamsters, both genders, aged between two to six mouth and weight 150g in average were used. The left síde of tangue of each animal was painted for eight consecutive weeks, with a solution of DMBA. Tongues were removed, fixed in 10% buffered fonnal solution. The histological slides were stained by AgNOR technique and by PCNA and β-catenin immunohistochemical antibodies. Statistical analyzes were performed by ANOVA one-way test and Tukey test. We may conclude that an association between AgNOR and PCNA might indicate the higher proliferating activity of the analyzed celIs. The experimental carcinogenesis model in hamster tongue is an available methodology for immunohistochemistry study. And finally, PCNA and β-catenin immunohistochemical antibodies may be used to analyze possible premalignant areas in oral leukoplakia

The present study compared the expression of cytokeratins CK6, CK16 and CK19 and pan-cytokeratin (PAN) in oral mucosa cells between smokers and nonsmokers in order to determine the stage of cell differentiation and to consequently infer proliferative activity and expressions indicative of a potential for malignant differentiation. Thirty smokers and 30 non-smokers seen at the clinics of FOSJC-UNESP were screened. Smears were obtained from the left lateral border of the tongue with a cytobrush and slides were processed for immunohistochemistry using the antibodies reported Conventional microscopy was used for qualitative analysis. The results were analyzed statistically by the Z test, Fisher's exact test and comparison of two proportions (plus-4 confidence interval method). The expression of CK6 (p=0.002), CK16 (p=0.003), CK19 (p=0.0001) and PAN (p=0.008) was higher in oral mucosa smears from smokers compared to non- smokers. ln conclusion, increased epithelial proliferation is observed in the oral mucosa of smokers as demonstrated by the increased expression of CK6 and CKJ6, and these cells present alterations in epithelial maturation

Zidovudine (AZT) is the drug most commonly used in AIDS treatment, isolated or in combination with other antiretroviral agents, but it has certain limitations due to its therapeutic dose-dependent haematological toxicity. In addition, it has low oral bioavailability, since it undergoes pre-systemic metabolism. The nasal route has been used as an alternative route for drug administration, because it can promote its direct absorption to blood circulation, avoiding hepatic metabolism. However, this route presents as a factor limiting the mucociliary clearance mechanisms that remove quickly the formulation of the nasal cavity. To prolong the residence time of formulations, in this direction, has been proposed the development of mucoadhesive systems. Among the various existing systems, the use of chitosan (QS), as mucoadhesive polymer, has been widely exploited in the preparation of nanoparticles (NPs). The objective of this study was to develop and characterize QS’s NPs for intranasal administration of AZT. For both NPs have been developed by ionic crosslinking of QS with sodium tripolyphosphate (TPP). These NPs were characterized by studies of particle size distribution, zeta potential, morphology, mucoadhesion tests, assessing the ability of encapsulation of the drug and permeation profile of AZT. The evaluation of AZT in the NPs was determined by UV-Vis spectroscopy. Mucoadhesion measures were made using a texture analyzer, using a mucin disk and porcine mucous membrane , and permeation assay were conducted using porcine nasal mucous membrane adapted to the Franz cell. These results suggest that the systems in hand have great potential for nasal AZT administration

Esta pesquisa tem como objetivo de examinar as estratégias de expansão das firmas brasileiras do setor de carnes (bovina, suína e avícola) para além das fronteiras nacionais a partir da literatura sobre internacionalização de empresas. Essa literatura aponta fatores como a origem do capital, o grau de internacionalização da empresa, o setor de atuação, entre outros, como importantes condicionantes que afetam esse processo. No entanto, vários desses fatores não são diretamente aplicáveis aos casos brasileiros, caracterizado por uma internacionalização tardia e, em alguns casos, rápida. Para este estudo foram selecionadas três empresas nacionais do setor de carnes que apresentam diferentes estágios de internacionalização: JBS-Friboi, Marfrig, Brasil-Foods (Perdigão e Sadia). As informações selecionadas sobre cada empresas foram reunidas em dossiês corporativos, e analisadas através de estudos de casos, permitiram examinar e comparar as estratégias globais das empresas. Os resultados permitem concluir que as duas primeiras empresas demonstram uma estratégia agressiva de internacionalização (em certa medida a primeira fazendo o papel de líder e a segunda de seguidora e, por isso, complementares em posicionamento nos diferentes mercados regionais), enquanto a terceira se mantém nas etapas iniciais desse processo, fortemente vinculadas às exportações a partir do Brasil

Embora os núcleos das fibras musculares esqueléticas não se dividam, o músculo tem uma pequena capacidade de reconstituição. As células satélites são as responsáveis pela regeneração do músculo esquelético e ajustes induzidos pelo exercício. Células satélites são pequenas células miogênicas mononucleadas, fusiformes, dispostas paralelamente às fibras musculares dentro da lâmina basal que envolve as fibras e só podem ser identificadas no microscópio eletrônico. Essas células contribuem para o crescimento do músculo no embrião e no período pósnatal e são quiescentes no adulto. Têm potencial para, quando ativadas, se diferenciarem em mioblastos, se duplicarem ou migrarem para região lesionada e fundirem-se às células musculares acelerando o processo regenerativo. Vários são os fatores que estimulam essas funções (IGF-I, FGF, citocinas, etc.), sendo que o exercício pode potencializar a produção deles. A regeneração é uma adaptação que ocorre no músculo esquelético em resposta ao traumatismo. Este processo tem sido descrito desde o final do século XIX, mas somente nos últimos trinta anos foi realmente compreendida a capacidade regenerativa das fibras musculares esqueléticas. O reparo tecidual é comum a todos os tecidos do organismo e envolve ações integradas entre células, matriz e mensageiros químicos, visando restaurar, no menor período de tempo possível, a integridade do tecido e o equilíbrio biológico. A regeneração pode ser divida em três fases: fase inflamatória, fase proliferativa e fase de remodelagem. Este trabalho tem como objetivo realizar uma revisão sobre as células satélite e seu papel na regeneração muscular a fim de sistematizar o que pesquisadores de diversas tem publicado sobre o tema atualmente.

Escherichia coli O157:H7 is a foodborne pathogen associated with enteritis in humans, ranging from a mild to bloody diarrhea to hemolytic uremic syndrome, thrombotic thrombocytopenic purpura and even death. Large E. coli O157:H7 outbreaks have been reported worldwide and are frequently associated with consumption of undercooked beef. Cattle are a major reservoir of the pathogen, which is found in the intestinal tract of the animal. The carcasses can be contaminated with feces during the slaughter and production process. Ground beef remains the most common vehicle. The purpose of this study was to determine the E. coli O157:H7 importance associated to human illness and productivity losses to the meat industry, as well as identifying mechanisms of contamination related to beef and strategies to improve the safety of beef products

Proteins of animal origin are among the most consumed foods in the world, this scenario highlights the pig production, providing much of the daily protein intake worldwide. Brazil has significant numbers in the production and export of pork to stand among the first in the world marker. Several diseases such as brucellosis, can configure a threat to this sector. Brucellosis in pigs is an important disease and zoonosis once it is the cause of great economic losses by making the species vulnerable to sanitary barriers as well as putting at risk the health of attendants of farms, slaughterhouses employees and consumers. Pigs slaughtered in slaughterhouses approved by competent organs guarentee the safety of employees, in the flowchart of slaughter, and consumers, but illegal slaughterhouses pose risk to public health

Uterine leiomyomas (UL) are benign smooth muscle tumors from myometrium, representing the major indication for hysterectomies and a significant public health problem. Several genetics alterations have been associated with its development and pathogenesis. Although the initial factors that lead to the development of uterine leiomyomas are unclear, there are several evidences showing that ovarian steroids, estrogen and progesterone, are associated with these tumors.Recent reports, however, show others genes related with distinct signalization pathways besides sex hormones, which may be or not activated by these hormones, are associated with uterine leiomyomas development. In this study, the AHR, ESR1, ESR2, PGR and WNT7A gene transcripts expression was evaluated using quantitative real time RT-PCR in 58 UL samples compared to five normal myometrium tissues to explore the hormonal molecular basis of these tumors and associate these expression pattern with clinical and pathological features. The present study showed AHR, ESR1, ESR2, PGR and WNT7A down expression in 60%, 43%, 52%, 60% e 40% of the tumors, respectively, and a significant AHR loss of expression compared with the controls samples (P=0.0130). The comparison between the genes expression values revealed a positive correlation between the AHR and ESR1 transcripts (P<0.0001; r=0.6911) and between the genes ESR1 and WNT7A (P<0.0001; r=0.5655). The expression pattern was compared with clinical and pathological features. The WNT7A gene presented a differential expression pattern between the proliferative and secretory phase of menstrual cycle (P=0.0373) and the AHR and ESR1 genes were differentially expressed between women in reproductive and menopause years (P=0.0267; P=0.0415, respectively). Malformations and possible damages to the urogenital system can be originated in the embryonic period. Moreover, fire guns, knives and accidents, where there is the disruption of the urethra, also cause these lesions. The objective was to analyze the contribution of tissue engineering in the construction of neo-urethra, developed by bioengineering. We performed an urothelial ex vivo expansion of cells in 3D scaffolds (platelet gel matrix and acellular porcine aorta) to assess the contribution of this technique in the construction of a neo-urethra. Mechanical dissociation was made of the inner wall of 10 North Folk rabbit’s bladder, weighing 2.5 to 3.0 kg. After dissociation the cell content was centrifuged and obtained a pellet of urothelial cells. The pellet was ressuspended in culture medium DMEM F12 and cells were maintained in culture for 15 days. Immunohistochemical analysis characterized the urothelial culture. The cells were then implanted in the scaffold - platelet gel. In a second experiment using aortic porcine acellular matrix were implanted urothelial cells alone and urothelial cells on platelet gel, on the inner wall of the scaffold - aorta, with space for setting bordered by a urethral probe. The complex probe - cells - aorta and probe - cells in platelet gel - aorta, were sealed with suture material and culture were maintained in a humidified 37ºC incubator with 5% CO2 in air for 12 days to subsequent histological analysis of urothelium cell adhesion to the scaffolds. By observation under an optical microscope, we could see the growth of cells in the scaffold platelet gel, from a monolayer in to a three-dimensional structure. In the acellular porcine aortic matrix containing the platelet gel, we could observe a few quantity of urothelial cells adhered. However with the acellular porcine aortic matrix in which was implanted only the urothelial cells, we have obtained adhesion to the wall

Brazil is one of the largest producers of animal protein. In 2010, the production of meat (beef, pork and poultry) was estimated at 24.5 million tons and 75% was consumed in the country. The meat is a primary source of water and fat and contains between 20% and 35% protein, providing all essential amino acids and several micronutrients and vitamins. Due to the large consumption of this food, its quality is extremely important and the research of some indicators microorganism become essential in order to ensure the hygienic and sanitary quality, indicating contamination of fecal origin, with the possible presence of pathogens or to identify food spoilage. Thus, the aim of study was to determine the microbiological quality of 90 samples handled meat products and pre- prepared marketed in Botucatu city, according to the parameters required by ANVISA (RDC 12, 2001). Among the samples analyzed, all of them were negative for Staphylococcus coagulase positive and were within the limit allowed by ANVISA for the Clostridium sulfite reducer (up to 3 x 103 CFU/ g). The presence of Salmonella was confirmed in only one sample (1.1%), against to the legal parameters. About thermotolerant coliforms, 54.4% of the samples were outside the acceptable limit by law (up to 5 x 103 MPN/g). Therefore, the presence of these microorganisms indicates inadequate hygiene conditions of foodhandlers and equipment used in the, which are considered inadequate for consumption
