Bacterial artificial chromosomes (BAC) have been widely used for fluorescence in situ hybridization (FISH) mapping of chromosome landmarks in different organisms, including a few in teleosts. In this study, we used BAC-FISH to consolidate the previous genetic and cytogenetic maps of the turbot (Scophthalmus maximus), a commercially important pleuronectiform. The maps consisted of 24 linkage groups (LGs) but only 22 chromosomes. All turbot LGs were assigned to specific chromosomes using BAC probes obtained from a turbot 5x genomic BAC library. It consisted of 46,080 clones with inserts of at least 100 kb and < 5 % empty vectors. These BAC probes contained gene-derived or anonymous markers, most of them linked to quantitative trait loci (QTL) related to productive traits. BAC clones were mapped by FISH to unique marker-specific chromosomal positions, which showed a notable concordance with previous genetic mapping data. The two metacentric pairs were cytogenetically assigned to LG2 and LG16, and the nucleolar organizer region (NOR)-bearing pair was assigned to LG15. Double-color FISH assays enabled the consolidation of the turbot genetic map into 22 linkage groups by merging LG8 with LG18 and LG21 with LG24. In this work, a first-generation probe panel of BAC clones anchored to the turbot linkage and cytogenetical map was developed. It is a useful tool for chromosome traceability in turbot, but also relevant in the context of pleuronectiform karyotypes, which often show small hardly identifiable chromosomes. This panel will also be valuable for further integrative genomics of turbot within Pleuronectiformes and teleosts, especially for fine QTL mapping for aquaculture traits, comparative genomics, and whole-genome assembly.

The current nature of World Wide Web (Web), which highlights the collaborative participation of users in some digital information environments, lead to development of guidelines with a focus on Inclusive Digital Information Architecture for a different audiences in multiples informational ambiences. This research proposes guidelines for a inclusive digital information environment, aiming to identify the elements of accessibility that allow the promotion of inclusion digital information, in order to highlight the references of the Architecture of Digital Information, the international recommendations and the structures representation of information (specially accessibility attributes), with the look of Information Science and New Technologies of Information and Communication. The thesis is as to the need to develop methodologies with guidelines that contemplate the elements of digital accessibility focused on the task of the user. The research problem is that actual methodologies and recommendations used for the development of digital information environments do not cover all the elements of accessibility with a focus on user needs. The proposition is to present, through a methodological approach (exploratory and descriptive), a theoretical-methodological proposal to promote elements of digital accessibility to the development of digital information environments, for users with or without special needs. The overall objective of this study is to contribute to the development of inclusive digital information environments, in perspective of union of theories, recommendations and technologies for the development of digital accessibility guidelines. In a theoretical dimension, the research covers a critical review of the specialized literature in the areas of Information Science and Computer Science and, in the applied part, the analysis and development of a proposal of guidelines for the architecture of a digital information environment with accessibility.

A new selective sensor based on molecularly imprinted polymers (MIPs) was developed for the determination of hexazinone (HXZ) in environmental samples. MIPs were synthesized using a non-covalent approach, and selection of the monomers employed in the polymerization reaction was carried out by molecular modeling. Three functional monomers with high (2-vinylpyridine (MP17)) and intermediate (methacrylic acid (MP12) and acrylamide (MP5)) energies of binding to the template (HXZ) were selected for preparation of the MIPs, in order to conduct comparative studies and validate the theoretical data. For sensor construction, carbon pastes were modified with each MIP or NIP (non-imprinted polymer), and HXZ determination was performed using differential pulse adsorptive cathodic stripping voltammetry (DPAdCSV). All parameters affecting the sensor response were optimized. In HCl at pH 2.5, the sensor prepared with MP17 (5% w/w in the paste) showed a dynamic linear range between 1.9 × 10−11 and 1.1 × 10−10 mol L−1, and a detection limit of 2.6 × 10−12 mol L−1, under the following conditions: accumulation time of 200 s at a potential of −0.5V, scan rate of 50 mVs−1, pulse amplitude of 60 mV, and pulse width of 50 ms. The sensor was selective in the presence of other similar compounds, and was successfully applied to the analysis of HXZ in river water samples.

1. 1. Myoglobin from the subterranean reptile Amphisbaena alba was isolated for measurement of concentrations and physico-chemical properties. 2. 2. The concentrations (averaging 12.1 mg.g-1 wet weight in the temporal muscles and 5.8-6.0 in the muscles that motivate the wedge-shaped head which forms the burrowing tool) far exceed those earlier reported for reptiles and other terrestrial vertebrates. 3. 3. The myoglobin has a low O2 affinity compared to mammals (P50 = 2mmHg at 25°C). In the presence of the same myoglobin O2 tension as in mammals this appears to favour similar in vivo O2 saturations at the lower reptilian body temperature. 4. 4. The temperature sensitivity of P50 reflect a heat of oxygenation, ΔH near -13 kcal· mol-1. The myoglobin is monomeric and thus lacks cooperativity in O2 binding and there is no Bohr effect. 5. 5. The pattern of microheterogeneity is similar to that of myoglobin of terrestrial vertebrates but different to aquatic mammals and reptiles. The major and two minor components exhibit very similar O2 affinities. 6. 6. The concentrations and oxygen-binding characteristics of Amphisbaena myoglobin are discussed with regard to the flow of O2 to the mitochondria during digging activity in hypoxic burrow environments. © 1981.

1. 1. The oxygen consumption in workers of two simpatric leaf cutting ants, Atta laevigata and Atta sexdens rubropilosa was measured at different temperatures. 2. 2. In the temperature range between 5-35°C, with 5°C increments, the respiratory rates increased with temperature, but the R-T curves of both ants showed neither a marked drop at the low end nor a break at the high end; except between 30 and 35°C. 3. 3. The respiratory rates of A. s. rubropilosa were higher than those of A. laevigata and in the midrange of temperatures, the rates of A. laevigata increased faster than those of A. s. rubropilosa. 4. 4. Q10 values did not indicate regions of compensation for temperature in both ants, but suggested that adjustments may occur at high temperatures (25-35°C), as expected for tropical ants. 5. 5. Temperature variations did not alter significantly the slope of the curve relating oxygen consumption and body weight in both species. © 1982.

1. 1. The respiration of the different castes of two species of leaf-cutting ants, Atta laevigata and Atta sexdens rubropilosa was measured in both workers and sexed forms. 2. 2. The respiratory rates (μl O2/mg(live)/hr) were, in A. laevigata, 0.644 (gardeners), 0.594 (cutters), 0.354 (soldiers), 0.233 (males) and 0.085 (females); in A. s. rubropilosa, 0.803 (gardeners), 0.748 (cutters), 0.510 (soldiers), 0.375 (males) and 0.274 (females), significantly higher than A. laevigata. 3. 3. All castes considered, the b-values of the equation relating oxygen consumption and weight in A. laevigata (0.53) were significantly different from that in A. s. rubropilosa (0.73). © 1981.

1. 1. Temperature preference in experimental thermal gradient in the laboratory was determined and field preferred temperature was recorded, for Amphisbaena mertensi. 2. 2. Mean preferred temperature in 52 experiments averaged 21.4°C which was close to those recorded in 16 specimens measured in the field (21.1°C). 3. 3. Selected temperature varied from 12 to 27°C in the laboratory, and data taken in the field ranged from 18 to 26°C. © 1984.
