During the growth of Kluyveromyces marxianus var. marxianus ATCC 10022 on lactose, peaks of glucose, but not β-galactosidase activity, were detected in culture medium. Harvested and washed whole cells produced glucose and galactose from lactose, or ortho-nitro-phenol from the chromogenic substrate ortho-nitro-phenyl-β-D-galactopyranoside (ONPG), indicating that β-galactosidase is physically associated with cells. ONPG hydrolysis by whole cells presented a monophasic kinetics (Km 36.6 mM) in lactose exponential growth phase cells, but a biphasic kinetics (Km 0.2 and 36.6 mM) in stationary growth phase cells. Permeabilization with digitonin or disruption of cells from both growth phases led to monosite ONPG hydrolysis (Km 2.2 to 2.5 mM), indicating that β-galactosidase is not located in the periplasm. In addition, the energy inhibitors fluoride or arsenate, as well as the uncouplercarbonyl cyanide m-chlorophenylhydrazone (CCCP) prevented ONPG hydrolysis by whole cells. These findings indicate that energy coupled transmembrane transport is the rate-limiting step for intracellular ONPG cleavage. The taxonomic and physiologic implications of the exclusive intracellular location of β-galactosidase of K. marxianus var. marxianus ATCC 10022 are discussed. © 1996 Kluwer Academic Publishers.

Sour cassava starch is traditionally produced in Latin America for preparation of cheesebreads such as 'pan de yuca' and 'pandebono' in Colombia, and 'pao de queijo' in Brazil. The processing involved is described and improvements suggested. Criteria for quality assessment of sour cassava starch are based on consumers' requirements. Recommendations are made for improving the processing and product quality. Alternatives are given for extending the potential value of this traditional foodstuff.

The species is a very popularly used plant as woundwort. Leaves, top flowers and rhizomes are used. The objective of this work is a contribution to the germination study, photoblastism and more adequate temperature for reproduction of the species. The best temperature for the germination was found to be 20°C and no seeds germinated under continuous darkness, within 14 days; therefore, the seeds are positive photoblastics. The seed germination percentage was low, in relation to the mass of the achene produced, varying from 8,2 to 20,0%. The alternated temperatures of 20° and 30°C did not influence the seed germination and the average of the weight of 100 seeds was 0,026 1 g.

A combined methodology consisting of successive linear programming (SLP) and a simple genetic algorithm (SGA) solves the reactive planning problem. The problem is divided into operating and planning subproblems; the operating subproblem, which is a nonlinear, ill-conditioned and nonconvex problem, consists of determining the voltage control and the adjustment of reactive sources. The planning subproblem consists of obtaining the optimal reactive source expansion considering operational, economical and physical characteristics of the system. SLP solves the optimal reactive dispatch problem related to real variables, while SGA is used to determine the necessary adjustments of both the binary and discrete variables existing in the modelling problem. Once the set of candidate busbars has been defined, the program implemented gives the location and size of the reactive sources needed, if any, to maintain the operating and security constraints.

Salting have been a very utilized fish conservation method, however only in the last years the basic mechanism involved in salting fish has been understood. The objectives of this study were determine the addition in brine salt of rosemary leaves, rosemary extracts and tocopherol, and your action in the followed parameters: water activity (Aw), moisture, ash, salt content and TBARS. The results showed that the addition of antioxidants was difficulted the salt absorption, however didn't have differences between rosemary or tocopherol use. In the salting time of 3 hours the values of Aw and salt levels, was respectively: 0.77±0.01 and 14.42±1.69. for control treatment; 0.85±0.02 and 9.09±1.39for rosemary filtrate; 0.78±0.03 and 10.63±0.69 rosemary without filtrating and 0.85±0.02 and 11.96±1.78 tocopherol, showed that didn't grow indigenous bacterias. Lipid oxidation was evaluated by TBARS and the results showed the oxidative effect of salt and the pro oxidant effects of alls antioxidants used in brine salting.

It had been studied the dimensional variations Pinus caribæa var. hondurensis and Eucalyptus grandiswood, emphasizing the axial, radial, tangential and volumetric shrinkage. The results had shown that the volumetric shrinkage of Pinus and Eucalyptus varied according to an exponential curve, R V=R Vmaxe -ku. The total or maximum volumetric shrinkage showed a linear behavior with the specific gravity on basis of ovendry mass and volume and the same behavior as respect of the specific gravity, where the adjusted equations were, respectively: R vmax = 3,01 + 19,15,oo e R vmax = 1,96 + 24,28,ob. It was also verified that the tangential shrinkage was 1,3 times greater than the radial one and 9,0 times greater than the axial one.

GCTase production by a new strain of Bacillus alkalophillc CGII isolated from Brazilian wastewater of manioc flour industry was examined. The growth medium used was composed by 1.5% starch, 1.5% nitrogen and 1% Na 2CO3. Higher activity was obtained with starch, maltodextrin and galactose. When glucose was added to the medium, no enzyme production was observed. High enzyme activity and growth were reached when aeration was increased (88.6 U/mL). The enzyme characterization showed an optimum pH and temperature 8.0 and 55°C for starch hydrolyses, respectively. Mg+ and Ca++ showed small activation; however, Hg + and Cu+ showed a strong enzyme inhibition.

A newly-isolated thermophilic strain of the zygomycete fungus Rhizomucor pusillus 13.36 produced highly active dextrinogenic and saccharogenic enzymes. Cassava pulp was a good alternative substrate for amylase production. Dextrinogenic and saccharogenic amylases exhibited optimum activities at a pH of 4.0-4.5 and 5.0 respectively and at a temperature of 75°C. The enzymes were highly thermostable, with no detectable loss of saccharogenic or dextrinogenic activity after 1 h and 6 h at 60°C, respectively. The saccharogenic activity was inhibited by Ca2+ while the dextrinogenic was indifferent to this ion. Both activities were inhibited by Fe2+ and Cu2+ Hydrolysis of soluble starch by the crude enzyme yielded 66% glucose, 19.5% maltose, 7.7% maltotriose and 6.6% oligosaccharides. The thermophilic fungus Thermoascus aurantiacus 179-5 produced large quantities of a glucosidase which preferentially hydrolyzed maltose over starch. Enzyme production was high in submerged fermentation, with a maximal activity of 30 U/ml after 336 h of fermentation. In solid-state fermentation, the activity of the enzyme was 22 U/ml at 144 h in medium containing wheat bran and 5.8 U/ml at 48 h when cassava pulp was used as the culture medium. The enzyme was specific for maltose, very slowly hydrolyzed starch, dextrins (2-7G) and the synthetic substrate (α-PNPG), and did not hydrolyze sucrose. These properties suggest that the enzyme is a type II α-glucosidase. The optimum temperature of the enzyme was 70°C. In addition, the enzyme was highly thermostable (100% stability for 10 h at 60°C and a half-life of 15 min at 80°C), and stable within a wide pH range. The objective of this work was to study the effect of selective thinning on the genetic divergence in progenies of Pinus caribaea var. bahamensis, aiming to identify the most productive and divergent progenies for the use of improvement program. The test of progenies containing 119 progenies and two commercial controls were planted in March 1990, using 11 × 11 square lattice design, sextuple, partially balanced, disposed in lineal plots with six trees in the spacing of 3,0 × 3,0m. 13 years after planting thinning was realized (selection for DBH), with 50% selection intensity based on Multi-effect index, leaving three trees per plot in all the experiment. The evaluations were done at four situations: A (before the thinning); B (thinned trees); C (remaining trees after thinning) and D (one year after thinning). The analyzed traits were: height, diameter at breast height (DBH), volume, form of stem and wood density. The genetic divergence among the progenies was studied with aid of the canonical variables and of clustering of Tocher method, using the generalized distance matrix of Mahalanobis (D2) as estimate of the genetic similarity. The progenies were grouped in four groups in situation A, fourteen in the situation B, two in the situation C and three in the situation D. The selective thinning of the trees within of the progenies caused a change in the genetic divergence among the progenies, genetically homogenizing the progenies, as demonstrated by the generalized distances of Mahalanobis, clustering of Tocher' and canonical variables methods. The thinning made possible a high uniformity in respect to the relative contribution of the traits for the total genetic divergence. The techniques of clustering were efficient to identify groups of divergent progenies for the use hybridization and little divergent progenies for the use in backcross program.

The objective of this work was to study the effect of selective thinning on the estimates of genetic parameters in progenies of Pinus caribaea var. hondurensis. The progeny test was installed in June 1986, following a 10 × 10, triple square lattice design, containing ten trees in linear plots in 3.0 × 3.0m spacing. Twelve years after the planting, a selective thinning based on Multi-effect index (selection for DBH) was performed, leaving six trees per plot. The assessments were done in four situations: A (before thinning); B (among thinned trees); C (among remnant trees at 13 years of age) and D (two years after the thinning). The following traits were analyzed: total height (H), diameter at breast height (DBH), volume (VOL), stem form (FOR), foxtail (FT), wood density at 1.3 m (WD1) and wood density at the half height (WD2). The thinning led to increase in the estimates of variability among progenies for VOL and decrease for ALT and DBH. There was a high and positive genetic correlation among the growth traits, mainly between DBH and VOL. The latter remained unchanged after thinning. The narrow-sense heritabilities at the individual level, showed a slight increase while at the family mean level had a reduction with the thinning. The estimates of breeding values increased with the thinning. This may lead to an easier identification of the best progenies in further selection.
