Indirect drug susceptibility tests of Mycobacterium tuberculosis was done to investigate the accuracy and feasibility of a broth microdilution method (BMM) for determining minimal inhibitory concentrations of conventional drugs against M. tuberculosis. Test drugs included isoniazid (H), rifampicin (R), ethambutol (E), streptomycin (S) and pyrazinamide (Z). Fifty isolates of M. tuberculosis from patients who had never received drug therapy, and H37Rv strain for control, were evaluated in the system. When comparing this method with the gold standard proportional method in Lowenstein-Jensen medium, sensitivity of 100% for all drugs and specifities of 91, 100, 96, 98 and 85% were observed respectively for H, R, E, S and Z. The BMM was read faster (14-20 days) than the proportional method (20-28 days). The microdilution method evaluated allows the testing of multiple drugs in multiple concentrations. It is easy to perform and does not require special equipment or expensive supplies. In contrast to radiometric method it does not use radioactive material.

A eficácia antimicrobiana de conservantes empregados em formulações cosméticas foi avaliada usando Phenova® e Imidazolinidil uréia que inibiram o crescimento de Bacillus subtilis no extrato de Achillea millefolium L. e Nipagin®/ Nipasol® 0,2% em propilenoglicol não apresentaram efeito microbicida.

Resistance in Mycobacterium tuberculosis to isoniazid (INH) is caused by mutations in the catalase-peroxidase gene (katG) , and within the inhA promoter and/or in structural gene. A small percentage (~ 10%) of INH-resistant strains do not present mutations in both of these loci. Other genes have been associated with INH resistance including the gene encoding for NADH dehydrogenase (ndh) . Here we report the detection of two ndh locus mutations (CGT to TGT change in codon 13 and GTG to GCG change in codon 18) by analyzing 23 INH-resistant and in none of 13 susceptible isolates from Brazilian tuberculosis patients. We also detected two isolates without a mutation in ndh, or any of the other INH resistance-associated loci examined, suggesting the existence of additional, as yet to be described, INH resistance mechanisms.

Mycobacterium tuberculosis is responsible for over 8 million cases of tuberculosis (TB) annually. Natural products may play important roles in the chemotherapy of TB. The immunological activity of Davilla elliptica chloroform extract (DECE) was evaluated in vitro by the determination of hydrogen peroxide (H2O2), nitric oxide (NO), and tumor necrosis factor-alpha (TNF-alpha) release in peritoneal macrophages cultures. DECE was also tested for its antimycobacterial activity against M. tuberculosis using the microplate alamar blue assay. DECE (50, 150, 250 µg/ml) stimulated the production of H2O2 (from 1,79 ± 0,23 to 7,27 ± 2,54; 15,02 ± 2,86; 20,5 ± 2,1 nmols) (means ± SD), NO (from 2,64 ± 1,02 to 25,59 ± 2,29; 26,68 ± 2,41; 29,45 ± 5,87 µmols) (means ± SD) and TNF-alpha (from 2,44 ± 1,46 to 30,37 ± 8,13; 38,68 ± 1,59; 41,6 ± 0,90 units/ml) (means ± SD) in a dose-dependent manner and also showed a promising antimycobacterial activity with a minimum inhibitory concentration of 62,5 µg/ml. This plant may have therapeutic potential in the immunological and microbiological control of TB.

Podridões radiculares causadas por espécies de Pythium são um importante problema em cultivos hidropônicos. Sintomas de subdesenvolvimento são observados nas plantas parasitadas pelo patógeno, sendo muitas vezes não diagnosticados pelo produtor. O objetivo do trabalho foi avaliar o controle biológico da podridão radicular causada por Pythium aphanidermatum e a promoção de crescimento por Pseudomonas chlororaphis 63-28 e Bacillus subtilis GB03, reconhecidos agentes de controle biológico de doenças de plantas. A inoculação das plantas com P. aphanidermatum ocasionou o subdesenvolvimento, sendo essa diminuição de 20%. A adição dos agentes de biocontrole na solução nutritiva teve um efeito positivo no aumento da massa (6% a 13%), no número de folhas (4% a 7%) e no teor de clorofila (3%) das plantas de alface. Entretanto, maiores estudos devem ser realizados para melhorar a capacidade de controle da doença e de promoção de crescimento pelos agentes de biocontrole estudados no cultivo de alface hidropônica.

Cento e dez amostras de 11 diferentes marcas de leite ultra alta temperatura (UAT), comercializadas em São José do Rio Preto - SP, foram submetidas à contagem de microrganismos heterotróficos mesófilos viáveis e à pesquisa de bactérias do grupo do Bacillus cereus. A população de microrganismos mesófilos variou de <1,0´10² UFC/ml a >1,0´10(6) UFC/ml. Bactérias do grupo do Bacillus cereus foram verificadas em 13 (11,8%) amostras. Os resultados evidenciaram elevada população de microrganismos indicadores mesófilos.

A utilização de microrganismos para fins de controle biológico de doenças em plantas tem sido muito estudada no mundo. No caso do nematóide de cisto da soja (Heterodera glycines), uma das principais doenças da soja, o controle biológico é de grande importância devido à ausência de controle químico eficiente. Para se estudar o efeito de Bacilllus subtilis sobre H. glycines, foram conduzidos alguns experimentos de casa de vegetação e laboratório. Nos trabalhos de laboratório, verificou-se, em câmara de eclosão, que a presença de B. subtilis reduz a eclosão de ovos de H. glycines estimulados com exsudatos de sementes de soja. Foi observado também que o tratamento de raiz de soja com a bactéria inibiu a migração de larvas juvenis de H. glycines para a planta em comparação à raiz não tratada com a bactéria. Nos ensaios de casa de vegetação, utilizando-se vasos com solo infestados com ovos de H. glycines, observou-se uma redução de fêmeas na raiz de soja quando o solo ou sementes foram tratadas previamente com formulação pó-molhável ou calda contendo B. subtilis, respectivamente. Com base nesses resultados e sabendo-se que H. glycines apresenta dependência de estímulo de exsudatos vegetais para eclosão e orientação das larvas, pode-se afirmar que B. subtilis interfere nesse estímulo prejudicando o desenvolvimento do ciclo do nematóide.

Avaliou-se a resposta imune celular e humoral de camundongos inoculados com vírus rábico de rua e submetidos aos imunomoduladores Onco-BCG, avridina e Propionibacterium acnes. Os animais submetidos ao tratamento com P.acnes apresentaram um maior percentual de sobrevivência quando comparados aos dos demais tratamentos. Foram observados menores níveis de IFN-g nos animais infectados, sugerindo imunossupressão viral. O teste do Coxim Plantar não foi eficaz para a detecção da resposta de hipersensibilidade retardada na metodologia utilizada, contrariamente ao MIF. A sobrevivência dos animais não apresentou correlação com os níveis de anticorpos soroneutralizantes, concentração de IFN-g e resposta ao MIF.

Tuberculosis remains the leading cause of mortality arising from a bacterial pathogen ( Mycobacterium tuberculosis). There is an urgent need for the development of new antimycobacterial agents. The aromatic amino-acid pathway is essential for the survival of this pathogen and represents a target for structure-based drug design. Accordingly, the M. tuberculosis prephenate dehydratase has been cloned, expressed, purified and crystallized by the hanging-drop vapour-diffusion method using PEG 400 as a precipitant. The crystal belongs to the orthorhombic space group I222 or I2(1)2(1)2(1), with unit-cell parameters a = 98.26, b = 133.22, c = 225.01 angstrom, and contains four molecules in the asymmetric unit. A complete data set was collected to 3.2 angstrom resolution using a synchrotron-radiation source.

An understanding of isoniazid (INH) drug resistance mechanism in Mycobacterium tuberculosis should provide significant insight for the development of newer anti-tubercular agents able to control INH-resistant tuberculosis (TB). The inhA-encoded 2-trans enoyl-acyl carrier protein reductase enzyme (InhA) has been shown through biochemical and genetic studies to be the primary target for INH. In agreement with these results, mutations in the inhA structural gene have been found in INH-resistant clinical isolates of M. tuberculosis, the causative agent of TB. In addition, the InhA mutants were shown to have higher dissociation constant values for NADH and lower values for the apparent first-order rate constant for INH inactivation as compared to wild-type InhA. Here, in trying to identify structural changes between wild-type and INH-resistant InhA enzymes, we have solved the crystal structures of wild-type and of S94A, I47T and I21V InhA proteins in complex with NADH to resolutions of, respectively, 2.3 angstrom, 2.2 angstrom, 2.0 angstrom, and 1.9 angstrom. The more prominent structural differences are located in, and appear to indirectly affect, the dinucleotide binding loop structure. Moreover, studies on pre-steady-state kinetics of NADH binding have been carried out. The results showed that the limiting rate constant values for NADH dissociation from the InhA-NADH binary complexes (k(off)) were eleven, five, and tenfold higher for, respectively, I21V, I47T and S94A INH-resistant mutants of InhA as compared to INH-sensitive wildtype InhA. Accordingly, these results are proposed to be able to account for the reduction in affinity for NADH for the INH-resistant InhA enzymes. (c) 2006 Elsevier Ltd. All rights reserved.

The resumption of tuberculosis led to an increased need to understand the molecular mechanisms of drug action and drug resistance, which should provide significant insight into the development of newer compounds. Isoniazid (INH), the most prescribed drug to treat TB, inhibits an NADH-dependent enoyl-acyl carrier protein reductase (InhA) that provides precursors of mycolic acids, which are components of the mycobacterial cell wall. InhA is the major target of the mode of action of isoniazid. INH is a pro-drug that needs activation to form the inhibitory INH-NAD adduct. Missense mutations in the inhA structural gene have been identified in clinical isolates of Mycobacterium tuberculosis resistant to INH. To understand the mechanism of resistance to INH, we have solved the structure of two InhA mutants (121V and S94A), identified in INH-resistant clinical isolates, and compare them to INH-sensitive WT InhA structure in complex with the INH-NAD adduct. We also solved the structure of unliganded INH-resistant S94A protein, which is the first report on apo form of InhA. The salient features of these structures are discussed and should provide structural information to improve our understanding of the mechanism of action of, and resistance to, INH in M. tuberculosis. The unliganded structure of InhA allows identification of conformational changes upon ligand binding and should help structure-based drug design of more potent antimycobacterial agents.
