Este trabalho teve como objetivos padronizar a metodologia do teste de tetrazólio e avaliar a aplicabilidade deste para estimar a viabilidade de sementes de Gleditschia amorphoides. Inicialmente, foram avaliados os seguintes tratamentos de pré-condicionamento: semente intacta, escarificação mecânica, escarificação seguida de 24 ou 48 horas de embebição em água, com e sem posterior retirada do tegumento. em seguida, as sementes foram submetidas a 1, 3 ou 6 horas de coloração em solução de 2, 3, 5 trifenil cloreto de tetrazólio às concentrações de 0,025; 0,050; 0,075 ou 0,10% a 35ºC, no escuro. Sementes escarificadas e embebidas por 48 horas, com retirada do tegumento, imersas em solução de tetrazólio a 0,075% por 3 horas apresentaram coloração ideal, possibilitando a identificação das sementes em viáveis e inviáveis. Utilizando o protocolo acima descrito, avaliou-se a adequação do teste de tetrazólio em estimar a viabilidade de sementes de Gleditschia amorphoides através da comparação com o teste de germinação. A comparação não resultou em diferenças significativas entre eles. O teste de tetrazólio utilizando solução a 0,075% por 3 horas pode ser utilizado na estimativa da viabilidade de sementes de Gleditschia amorphoides.

O presente trabalho teve como objetivo avaliar a sensibilidade in vitro e in vivo de um isolado de Clavibacter michiganensis subsp. michiganensis (Cmm) aos produtos cloridrato de kasugamicina, fulusulfamide e oxitetraciclina. Para o teste in vitro, foram avaliados os halos de inibição formados ao redor de discos de papel de filtro umedecidos com os produtos, nas concentrações de 0, 1, 10, 100 e 1000 mig mL-1, 24 h após a instalação do ensaio. Quanto à avaliação dos produtos in vivo, dois ensaios foram instalados sob condições de casa-de-vegetação, com plantas de tomateiro cultivar Ângela Hiper, através da inoculação das plantas por dois métodos: pulverização foliar e ferimento no caule. Nos dois ensaios, as plantas foram pulverizadas com os produtos cloridrato de kasugamicina a 0,06 mL L-1, fulusulfamide a 0,025, 0,05 e 0,075 mL L-1 e oxitetraciclina a 0,40 g L-1, duas vezes antes e duas vezes após a inoculação, em intervalos médios de 5-7 dias. Foram avaliados a incidência de folíolos doentes (inoculação foliar) e a severidade dos sintomas nas plantas inoculadas por ferimento no caule. Verificou-se que o isolado de Cmm foi sensível in vitro ao cloridrato de kasugamicina, ao fulusulfamide e à oxitetraciclina, respectivamente a partir das concentrações de 1000, 100 e 10 mig mL-1. Com relação aos ensaios in vivo, apenas oxitetraciclina propiciou menor incidência de folíolos doentes nas plantas inoculadas através de pulverização foliar; nenhum produto teve êxito em controlar a doença nas plantas inoculadas por ferimento no caule. O fulusulfamide, em todas as concentrações, foi fitotóxico aos folíolos das plantas de tomateiro.

OBJETIVO: determinar as características mecânicas de molas T e T com helicóides, quando da incorporação de variações na liga metálica (aço inoxidável e beta-titânio), na intensidade de dobras de pré-ativação (0º e 40º/180º) e na secção transversal do fio utilizado para a construção destas molas (0,017 x 0,025 e 0,019 x 0,025). METODOLOGIA: foram submetidas ao ensaio mecânico 80 molas para fechamento de espaços, sendo estas centralizadas em um espaço de 21mm. As magnitudes de força horizontal, proporção momento/força e relação carga/deflexão produzidas pelos corpos de prova foram quantificadas utilizando-se um transdutor de momentos acoplado ao módulo indicador digital para extensiometria e adaptado a uma máquina universal de ensaios Instron. As molas foram submetidas a uma ativação total de 5mm, sendo registrados os valores a cada 1mm de ativação. No tratamento estatístico dos dados obtidos, foi realizada a análise de variância, sendo esta complementada pelo teste de comparações múltiplas de Tukey, considerando o nível de significância de 5%. RESULTADOS E CONCLUSÕES: os resultados demonstraram que, de maneira geral, as molas T com helicóides produziram menores magnitudes de força horizontal e relação carga/deflexão do que as molas T. Na presença de pré-ativação, as molas produziram altas proporções momento/força, enquanto, na ausência de dobras de pré-ativação, as mesmas geraram baixas proporções momento/força. Dentre todas as variáveis analisadas, aquela que apresentou uma maior influência na força horizontal e na relação carga/deflexão produzidas pelas molas foi a liga metálica. Já a proporção momento/força mostrou ser influenciada em maior grau pela pré-ativação das molas de fechamento.

Os objetivos neste experimento foram avaliar os efeitos da inclusão de extrato de orégano (EO) como aditivo promotor de crescimento nas rações sobre o desempenho, o sistema imune (peso e tamanho da bursa de Fabricius, peso do baço e do timo), as características anatomo-fisiológicas do trato gastrointestinal (altura de vilosidade, profundidade de cripta e suas relações), a microbiologia do ceco e o pH do duodeno e do ceco de frangos de corte. Foram utilizados 1.440 pintos de corte machos Cobb 500, em duas fases de criação (1 a 21 e 1 a 42 dias de idade), distribuídos em delineamento inteiramente casualizado, com seis tratamentos e oito repetições de 30 aves. Utilizou-se ração basal (RB) para as três fases de criação (1 a 21, 22 a 35 e 36 a 42 dias de idade), constituindo os seguintes tratamentos: T1 - RB; T2 - RB com antibiótico (25 ppm de bacitracina de zinco); T3 - RB com 0,025% EO; T4 - RB com 0,050% EO; T5 - RB com 0,075% EO; e T6 -RB com 0,100% EO. Observou-se que os tratamentos não influenciaram o desempenho e os pHs dos conteúdos duodenal e cecal das aves nas duas fases de criação. As variáveis de imunidade e avaliação anatomo-fisiológica do trato gastrointestinal aos 21 dias não apresentaram diferenças. Apenas o peso do baço e a altura de vilosidade aos 42 dias de idade foram influenciados pelos tratamentos. Houve redução no número de bactérias no ceco das aves à medida que se elevou o conteúdo do extrato de orégano nas rações, indicando que houve ação antimicrobiana dos componentes deste extrato. Na condição em que foi realizado o experimento, o uso do extrato de orégano como aditivo promotor de crescimento não ocasionou efeito diferente dos demais tratamentos (antibiótico e testemunha).

Thirty-seven samples of human milk (colostrum) from donors living in the Ribeirao Preto region were analyzed to determine the levels of organochlorine pesticide residues. Donors were classified into two groups, i.e., occupationally exposed and non-exposed to pesticides. Other factors such as age, previous lactations, race, smoking habit, occupation, family income and educational level were also considered. Analysis was performed by preliminary lipid extraction followed by fractional partition on a column and finally by gas chromatography with an electron capture detector. Lindane was found in 32% of the samples in amounts of less than 0.001 mg/kg; heptachlor was found in 65% of the samples at mean levels of 0.001 mg/kg, i.e., a level five-fold lower than that established by FAO/WHO (1970) for cow's milk. Aldrin and endrin were not detected in any of the samples. Dieldrin was detected in only one sample at a level of 0.038 mg/kg, which is considered high. DDT and DDE amounts are reported as total DDT and at least one of these compounds was present in every sample. Amounts detected in donors occupationally exposed to pesticides ranged from 0.008 to 0.455 mg/kg (mean, 0.149 mg/kg), i.e., three times the limit established by FAO/WHO (1970), while values for donors who had not been exposed ranged from 0.002 to 0.072 mg/kg (mean, 0.025 mg/kg), i.e., half the limit. Considering the level of acceptable daily intake proposed by FAO/WHO (1973), lactents ingested 1% of the acceptable intake of lindane (all donors), 30% of the acceptable intake of heptachlor (all donors), 60% of the acceptable intake of DDT (non-exposed donors), and 3.7 times the acceptable intake of DDT (exposed donors). Comparing the present results with those obtained 10 years ago, the total DDT level in human milk is decreasing in this part of the country. The mean amount of organochlorine residues in non-exposed women's milk was one of the lowest levels among those recorded in the literature. DDT levels of occupationally exposed women's milk were comparable with those reported for developed countries and lower than those detected in Latin American countries. When the results of this survey are considered in relation to the advantages of breast-feeding, the risk-benefit balance is still favorable to breast-feeding. However, given the lack of long-term epidemiological studies, undesirable or harmful long-lasting effects cannot be excluded.

Quantitative real time PCR was performed on genomic DNA from 40 primary oral carcinomas and the normal adjacent tissues. The target genes ECGFB, DIA1, BIK, and PDGFB and the microsatellite markers D22S274 and D22S277, mapped on 22q13, were selected according to our previous loss of heterozygosity findings in head and neck tumors. Quantitative PCR relies on the comparison of the amount of product generated from a target gene and that generated from a disomic reference gene (GAPDH-housekeeping gene). Reactions have been performed with normal control in triplicates, using the 7700 Sequence Detection System (PE Applied Biosystems). Losses in the sequences D22S274 (22q13.31) and in the DIA1 (22q13.2-13.31) gene were detected in 10 out of 40 cases (25%) each. Statistically significant correlations were observed for patients with relative copy number loss of the marker D22S274 and stages T3-T4 of disease (P=0.025), family history of cancer (P = 0.001), and death (P = 0.021). Relative copy number loss involving the DIA1 gene was correlated to family history of cancer (P<0.001), death (P=0.002), and consumption of alcohol (P=0.026). Log-rank test revealed a significant decrease in survival (P=0.0018) for patients with DIA1 gene loss. Relative copy number losses detected in these sequences may be related to disease progression and a worse prognosis in patients with oral cancer.

The biosynthesis of (2S)-2-methyl-2-(4'-methyl-3-pentenyl)-8-(3-methyl-2-butenyl)-2H-1-benzopyran-6-carboxylic acid (gaudichaudianic acid), the major metabolite in leaves and roots of Piper gaudichaudianum Kunth (Piperaceae), has been investigated employing [1(-13) C]-D-glucose as precursor. The labelling pattern in the isolated gaudichaudianic acid was determined by quantitative 13 C NMR spectroscopy analysis and was consistent with involvement of both mevalonic acid and 2-C-methyl-D-erythritol-4-phosphate pathways in the formation of the dimethylallyl- and geranyl-derived moieties. The results confirmed that both plastidic and cytoplasmic pathways are able to provide isopentenyl diphosphate units for prenylation of p-hydroxybenzoic acid. (c) 2007 Elsevier Ltd. The FML antigen of Leishmania donovani, in combination with either Riedel de Haen (R), QuilA, QS21 saponins, IL12 or BCG, was used in vaccination of an outbred murine model against visceral leishmaniasis (VL). Significant and specific increases in anti-FML IgG and IgM responses were detected for all adjuvants, and in anti-FML IgG1, IgG2a and IgG2b and delayed type of hypersensitivity to L. donovani lysate (DTH), only for all saponins and IL12. The QS21-FML and QuilA-FML groups achieved the highest IgG2a response. QuilA-FML developed the strongest DTH and QS21-FML animals showed the highest serum IFN-gamma concentrations. The reduction of parasitic load in the liver in response to each FML-vaccine formulation was: 52% (P < 0.025) for BCG-FML, 73% (P < 0.005) for R-FML, 93% (P < 0.005) for QuilA-FML and 79.2% (P < 0.025) for QS21-FML treated animals, respectively. Protection was specific for R-FML and QS21-FML while the QuilA saponin treatment itself induced 69% of LDU reduction. The FML-saponin vaccines promote significant, specific and strong protective effects against murine visceral leishmaniasis. BCG-FML induced minor and non-specific protection while IL 12-FML, although enhancing the specific antibody and IDR response, failed to reduce the parasitic load of infected animals. The effect of uroguanylin (UGN) oil K(+) and H(+) secretion in the renal tubules of the rat kidney was studied using in vivo stationary microperfusion. For the study of K(+) secretion, a tubule was Punctured to inject a column of FDC-green-colored Ringer's solution with 0.5 mmol KCI/L 10(-6)(mol UGN/L, and oil was Used to block fluid flow. K(+) activity and transepithelial potential differences (PD) were measured with double microelectrodes (K(+) ion-selective resin vs. reference) in the distal tubules of the same nephron. During perfusion, K(+) activity rose exponentially, from 0.5 mmol/L to stationary concentration, allowing for the calculation of K(+) secretion J(K)). JK increased from 0.63 +/- 0.06 nmol.cm(-2).s(-1) in the control croup to 0.85 +/- 0.06 in the UGN group (p < 0.01). PD was -51.0 +/- 5.3 mV in the control group and -50.3 +/- 4.98 mV in the UGN group. In the presence of 10(-7) mol iberiotoxin/L, the UGN effect was abolished: JK was 0.37 +/- 0.038 nmol-cm(-2).s(-1) in the absence of, and 0.38 +/- 0.025 in the presence of, UGN. indicating its action oil rnaxi-K channels. In another series of experiments, renal tubule acidification was studied, using similar method: proximal and distal tubules were perfused with solutions containing 25 mmol NaHCO(3)/L. Acidification half-time was increased both in proximal and distal segments and, as a consequence, bicarbonate reabsorption decreased in the presence of UGN (in proximal tubules, from 2.40 +/- 0.26 to 1.56 +/- 0.21 nmol-cm(-2).s(-1)). When the Na(+)/H(+) exchanger was inhibited by 10(-4) mol hexamethylene amiloride (HMA)/L, the control and UGN groups were not significantly different. In the late distal tubule, after HMA, UGN significantly reduced J(HCO3)(-). indicating all effect of UGN oil H(+)-ATPase. These data show that UGN stimulated J(K)(+) by actin, oil maxi-K channels. and decreased J(HCO3)(-) by acting on NHE3 in proximal and H(+)-ATPase in distal tubules.

The effects of La2O3 on the properties of (Zn, Co, Ta) doped SnO2 varistors were investigated in this study. The samples with different La2O3 concentrations were sintered at 1400 degrees C for 2 h and their properties were characterized by XRD, SEM, I-V and impedance spectroscopy. The grain size was found to decrease from 13 pm to 9 gm with increasing La2O3 content. The addition of rare earth element leads to increase the nonlinear coefficient and the breakdown voltage. The enhancement was expected to arise from the possible segregation of lanthanide ion due to its larger ionic radius to the grain boundaries, thereby modifying its electrical characteristics. Furthermore, the dopants such as La may help in the adsorption of O' to O '' at the grain boundaries characteristics. (c) 2006 Elsevier B.V. All rights reserved.

Sugarcane workers in Brazil are exposed to various genotoxic compounds, including polycyclic aromatic hydrocarbons (PAHs), derived from an incomplete combustion process of burnt sugarcane fields. The effects of the occupational exposure to sugarcane fields burning were measured in urine samples of sugarcane workers from the northwest of the State of São Paulo when exposed (harvesting) and when non-exposed (non-harvesting). The urinary levels of 1-hydroxypyrene (1-OHP) and the influence of the genetic polymorphisms CYP1A1, GSTM1, GSTT1 and GSTP1 were evaluated. Our results showed that the 1-OHP levels were significantly higher (P < 0.0000) in the exposed sugarcane workers (0.318 mu mol mol(-1) creatinine) than in the non-exposed workers (0.035 mu mol mol(-1) creatinine). In an unvaried analysis, no influence regarding the polymorphisms was observed. However, multivariate regression analysis showed that the CYP1A1*4 polymorphism in the exposed group, and age and the GSTP1 polymorphism in the non-exposed group significantly influenced urinary 1-OHP excretion levels (P < 0.10). The same group of sugarcane workers was significantly more exposed to PAHs during the harvesting period than during the non-harvesting period. (c) 2006 Elsevier B.V. All rights reserved.

The electrical and microstructural properties of SnO2-based varistors with the addition of 0.025 and 0.050 mol% of Fe2O3 have been characterised. Electric field (E) versus current density (J) curves showed that the effect of Fe2O3 addition is to increase both the non-linear coefficient and the breakdown voltage. Variations in the potential barrier height were inferred from impedance spectroscopy (IS) analysis. Through transmission electron microscopy (TEM), the presence of precipitates of secondary phases was confirmed. Samples with precipitates displayed poor electrical properties. Objective - To determine and compare the number, type, location, and distribution of apoptotic epidermal cells in the laminae of clinically normal horses and horses with laminitis.Sample Population - Formalin-fixed samples of digital lamellar tissue from 47 horses (including clinically normal horses [controls; n = 7], horses with acurte [4] and chronic [7] naturally acquired laminitis, and horses with black walnut extract-induced [11] or carbohydrate overload-induced [18] laminitis).Procedure - Blocks of paraffin-embedded lamellar tissues were stained for DNA fragmentation with the terminal deoxynucleotidyl transferase-mediated dUTP nick-end labeling (TUNEL) technique. Differential immunohistochemical staining for caspases 3 and 14 were used to confirm apoptosis.Results - the number of TUNEL-positive epidermal cells per 0.1 mm of primary laminae was significantly greater in the acute laminitis group than in the other groups. In the acute laminitis group, there were 17 and 1,025 times as many TUN EL-positive basal layer cells and keratinocytes, respectively, compared with the control group. Apoptosis of TUNEL-positive basal layer cells was confirmed by results of caspase 3 immunohistochemical staining. The TUNEL-positive keratinocytes did not stain for caspases 3 or 14.Conclusions and Clinical Relevance - the large number of apoptotic basal layer cells detected in the lamellar tissue of horses with acute naturally acquired laminitis suggests that apoptosis may be important in the development of acute laminitis. The role of the large number of TUNEL-positive keratinocytes detected in the interface of primary and secondary epidermal laminae of horses with acute laminitis remains to be elucidated.
