Diagnosis of Neospora caninum infection in dogs is based on serological assays such as the indirect fluorescent antibody test (IFAT) and enzyme-linked immunosorbent assays (ELISA). This study evaluated two serological tests (IFAT and ELISA) for the detection of IgG antibodies to N. caninum in 300 serum samples of dogs through the optimization of cut off titers by using the two-graph receiveroperating characteristic (TG-ROC) curve. In addition, the identification of major cross-reactive antigens with Toxoplasma gondii was investigated by inhibition ELISA and immunoblotting (IB) assays. IFAT and ELISA results showed 74% agreement, with a good negative concordance (P-neg=0.83), but a poor positive concordance (P-pos=0.42). The great majority (86%) of sera with positive concordant results (IFAT+/ELISA+) recognized at least two out of three N. caninum immunodominant antigens, particularly the 29-32 and 35-37 kDa bands. Optimization of cut off titers in IFAT and ELISA was performed considering the reactivity to at least two out of three N. caninum immunodominant antigens as infection markers, obtaining a titer of 50 for IFAT and 200 for ELISA. Seropositivity to N. caninuin was significantly associated with T gondii-seropositive samples, particularly in ELISA (55.4%). Inhibition ELISA curves for N. caninum showed a partial heterologous inhibition, indicating some degree of cross-reactivity between N. caninum and T gondii antigens. Inhibition IB assays showed a moderate heterologous inhibition for N. caninum antigens above 45-50 kDa. These results indicate that ELISA should be used critically when crude tachyzoite antigen preparations are employed, due to possible cross-reactivity with other related parasites as T gondii. Also, the cut off dilution of 1:50 in IFAT showed to be the most appropriated for N. caninum serology in dogs. Therefore, we suggest that N. caninum immunodominant antigens, specially the 17 and 29-32 kDa proteins, should be selected markers in serological assays for canine neosporosis. (c) 2006 Elsevier B.V. The objective of this work was to evaluate sweet potato cultivars with starch processing capability, on two cultivation periods, in the western region of Parana (Brazil). Sweet potato was grown at Marechal Candido Rondon, Parana State (24 degrees 33'40 '' S and 54 degrees 4'12 '' W), at a mean annual temperature between 14 and 28 degrees C. The experiment was designed in complete randomized blocks, in a factorial scheme of 4 cultivars (CNPH 003, CNPH 004, Brazlandia Roxa, and Brazlandia Rosada) and 2 cultivation periods (115 and 183 days), with four repetitions. The evaluated parameters were the root productivities (t ha(-1)) and the physical and chemical root composition (humidity, pH, acidity, starch, total soluble sugars and fibers). Data was submitted to all analysis of variance (Anova) and the means were compared by the Tukey test at 5% of probability, Results indicated that sweet potato yield at the cultivars was higher during the 183 day cultivation period (average of 9.14 t ha(-1)) than during the 115 day cultivation period (average of 4.25 t ha(-1)). Thus the CNPH 003 cultivar seems to be a better indication for starch extraction, due to the lowest fiber content in the root, the driest mass content and the largest yield of starch per area.

The objective of the present research was to determine the allelopathic potential of Ipomoea fistulosa plants on germination and growth of seedlings. In laboratory, lettuce and tomato seeds were placed to germinate in the presence of water or Ipomoea fistulosa leaf extract, in the concentrations of 5, 10 e 15% (p/v). In greenhouse, seeds were placed to germinate in trays containing Plantmax (R) substrate and Plantmax (R) substrate + Ipomoea fistulosa dry leaf residue, at 3% (w/w). The aqueous extract inhibited germination, raised the mean germination time of the seeds and reduced the radicle and hypocotyl growth in all used concentrations. In greenhouse, the plant height, leaf number, and dry mass accumulation also were reduced in the presence of the leaf residue in the substrate. The results suggest the presence of allelochemicals in Ipomoea fistulosa and demonstrate the highest sensitivity of lettuce that tomato to these compounds.

Currently, the cultivation of the cotton plant is based on a scale production model, characterized by high yields and intensive use of fertilizers, agrochemicals and mechanization. The objective of this study was to evaluate the influence of different row spacings on cotton crops, with or without growth regulator, and their effects in crop development and yield. The experimental design was in completely randomized blocks, in a 3x3 factorial scheme with 4 replications, using three row spacings: 0.45, 0.70 and 0.90 m. Three growth regulator conditions were tested: a) application split into four stages, b) single application at 70 d.a.e, and c) no regulator application. The project was conducted in the city of Selviria, Mato Grosso do Sul state, in November 2005. It was verified that the application of growth regulator is efficient in the limitation of plant height. The highest yield and boll mass was found in the split application of the regulator. The number of bolls and reproductive branches per plant was higher in the wider row spacings.

The objective of this work was to evaluate the nutritional composition (raw protein, ether extract, raw fiber, FDA, FDN and ash) of the POS 09/100 strain of Pleurotus ostreatus grown in three combinations of residues (pseudo-stem, leave and pseudo-stem + leaf) and four banana tree cultivars (Thap maeo, Prata ana, Pelipita and Caipira). The basidiomata harvested were submitted to dehydration in a forced ventilation stove with temperature adjusted to 40 degrees C until reaching constant weight, being then grinded in a knife mill. The method proposed by Kjeldahl (1883) was used to determine total nitrogen. The Weende method (AOAC, 1997) was employed to determine crude fiber, acid-detergent fiber (ADF) and neutral detergent fiber (NDF). According to the results obtained, crude protein, ash, ADF and NDF contents varied with the kind of residue and banana tree cultivar; the substrate prepared with leaves of banana tree (Prata Ana cultivar) yielded the highest crude protein (35.4%) and ash (6.7%) contents in the basidiomata of P. streams; the substrate prepared with pseudo-stein of banana tree (Prata Ana cultivar) provided the highest ADF (31.6%) and NDF (41.9%) contents in the basidiomata of P. ostreatus. Ether extract and crude fiber contents of P. ostreatus didn't vary in function of the treatments, with averages of 2.45 and 11.27%, respectively.

Renata G. Vieira R.G. & Acqua Coutinho S.D. 2009. Phenotypical characterization of Candida spp. isolated from crop of parrots (Amazona spp.). Pesquisa Veterinaria Brasileira 29(6):452-456. Curso de Pos-Graduagdo em Imunopatologia Veterinaria, Universidade Paulista, Rua Agariba 48, São Paulo, SP 05053010, Brazil. E-mail: selene@uol.com.brThe purpose of this study was to characterize Candida isolates from crop of parrots. Forty baby parrots of genus Amazona, species aestiva and amazonica that were apprehended from wild animal traffic were used: 18 presented ingluvitis and 22 other alterations, but showing general debilitation. Samples were seeded on Sabouraud dextrose agar with chloramphenicol after be obtained by the introduction of urethral probe through the esophagus. Based on morphology and biochemical reactions (API 20C) Candida was confirmed; it was still searched the production of proteinase and phospholipase, virulence factors for Candida species. Candida spp. were isolated from 57.5% parrots, being 72.2% from birds with ingluvitis and 45.5% from without ones. Twenty-five strains of Candida were isolated, 60% and 40%, respectively from parrots with and without ingluvitis, and were speciated: 28% C. humicola, 24% C. parapsilosis, 20% C. guilliermondii, 20% C. famata, and 8% C. albicans. These results demonstrate that C. albicans is not the most frequent species isolated, and it is the first report that shows C. guilliermondii, C. famata, and C. humicola causing infection in parrots. Many isolates presented filamentation (76%), 100% produced proteinase and 68% phospholipase. The observation of Candida spp. producing virulence factors reinforce the pathogenic role of these yeasts in the cases studied.

The evaluation of micronutrient availability in agricultural soil treated with sewage sludge was developed in Field with maize cultivation on a Dystrophic Red Latosol. The experiment used a randomized complete block design, with the treatments in a split-plot scheme, in time, with six doses of sewage sludge (main-plot treatment) in relation to the five sampling periods for the soil (subplot treatment), with four replications. The sewage sludge was applied in the superficial layer (0-20 cm). The micronutrients Fe, Cu, Mn and Zn were extracted from the soils using a DTPA (diethylenetriaminepentaacetic acid) solution. The same micronutrients were analyzed in the plant tissue after the nitric-perchloric decomposition of the maize samples by spectrometric atomic absorption. The results showed an increasing amount of Fe, Mn, Zn, Cu, Cation Exchange Capacity (CEC) and a decreasing pH in the soil. The increasing doses of sewage sludge increased the Fe, Mn, Cu and Zn availability in the soil solution, and the uptake amount by maize plants were always greater than the control values.

The response surface methodology as a tool for assessing the production of alginate and polyhydroxybutirate by Azotobacter vinelandii. Alginate is a polysaccharide extracted from cell walls of brown algae and used in the food, pharmaceuticals and biotech industries. Production is concentrated on the cultivation of brown seaweed, but several bacteria of the genus Pseudomonas and Azotobacter produce alginate. The chemical structure of alginates produced by algae is similar to those synthesized by A. vinelandii. The bacteria also produce intracellular polymers such as polyhydroxybutyrate (PHB), known as bioplastic. This work studied the simultaneous alginate and PHB production by A. vinelandii using sucrose and different parameters of fermentation in an orbital shaker. The optimal values for the production of these compounds were determined by the MSR. The first experiment was a 2(6-2) factorial design. The second was based on significant variables of the first, resulting in a full 3(3-0) factorial design. From the first to the second, an increase was observed in the PHB productivity from 12 to 45 mg g(-1) cell h(-1) and alginate from 100 to 1,600 mg g(-1) of cell h(-1). The productivity of both compounds was in the maximum incubation temperature of 62 degrees C, in the shortest time of incubation (18h) and the sucrose concentration, 11 g L(-1). In both experiments the PHB extracted presented purity of 94%.

The purpose of this work was to analyze the variability of pluvial precipitation at the Mid-Paranapanema Hydric Resources Management Unit (UGRI II-17), both spatially and temporally. To that effect, 33 pluviometric series were used, obtained from the National Water Agency (ANA), for the period from 1940 to 2000. The averages, standard deviation, upper and lower quartiles, maximum rainfalls, within the analyzed period, as well as the anomalies in these series, were calculated. The anomalies of the 1982/1983 El Nino, as well as the classification of homogeneous areas inside the basin were also calculated. Variability was observed from one year to the other, with rainier years - for example, 1982 and 1983 - and drier years - such as 1985 and 2000.
