Coagulase-negative staphylococci (CoNS) are the microorganisms most frequently isolated from clinical samples and are commonly found in neonatal blood cultures. Oxacillin is an alternative treatment of choice for CoNS infections; however, resistance to oxacillin can have a substantial impact on healthcare by adversely affecting morbidity and mortality. The objective of this study was to detect and characterise oxacillin-resistant CoNS strains in blood cultures of newborns hospitalised at the neonatal ward of the University Hospital of the Faculty of Medicine of Botucatu. One hundred CoNS strains were isolated and the mecA gene was detected in 69 of the CoNS strains, including 73.2% of Staphylococcus epidermidis strains, 85.7% of Staphylococcus haemolyticus strains, 28.6% of Staphylococcus hominis strains and 50% of Staphylococcus lugdunensis strains. Among these oxacillin-resistant CoNS strains, staphylococcal cassette chromosome mec (SCCmec) type I was identified in 24.6%, type II in 4.3%, type III in 56.5% and type IV in 14.5% of the strains. The data revealed an increase in the percentage of CoNS strains isolated from blood cultures from 1991-2009. Furthermore, a predominant SCCmec profile of the oxacillin-resistant CoNS strains isolated from neonatal intensive care units was identified with a prevalence of SCCmec types found in hospital-acquired strains.

Staphylococcus are not usually studied in the oral cavity, when this happens, they are considered to belong to transitory microflora. Individuals that present periodontal disease represent possibles reservoirs of these opportunist bacteria in the oral cavity. The use of antibiotics whether for treatment of periodontal disease or due to hospital infections, may predispose the increase of the Staphylococcus spp. in the oral cavity because they easily become resistant to antibiotics, resulting in superinfection. The study was made with 88 patients, minimum age- 25 years old, presenting chronical periodontitis, with, at least, two sites having a probing pocket bigger or equal to 5mm. After anamnese and clinical periodontal examination samples were taken from the periodontal pocket using paper cones and from the oral cavity using mouth rinse. Of the total patients 37,50% presented Staphylococcus spp. in the periodontal pocket and 61,36% in lhe oral cavity; 27,27% presented bacteria in the two sites, not necessarily of the same specie. S. epidermidis was the most prevailing specie in periodontal pocket (15,9%) and oral cavity (27,27%). Positive for S. aureus in the periodontal pocket were 4,5% and for the oral cavity 25%, and 3,4% were positive for the two sites. There was not found significative statistical difference referring to the presence of the microorganisms as to age, smoking habit and increase of the probing depth. The majority of the isolated Staphylococcus samples showed resistance to the tested antibiotics, indicating that the drugs as an adjunct to periodontal therapy, must be seen with caution

Objectives: To evaluate the antimicrobial activity of Arctium lappa L. extract on Staphylococcus aureus, S. epidermidis, Streptococcus mutans, Candida albicans, C. tropicalis and C. glabrata. In addition, the cytotoxicity of this extract was analyzed on macrophages (RAW 264.7).Design: By broth microdilution method, different concentrations of the extract (250-0.4 mg/mL) were used in order to determine the minimum microbicidal concentration (MMC) in planktonic cultures and the most effective concentration was used on biofilms on discs made of acrylic resin. The cytotoxicity A. lappa L. extract MMC was evaluated on RAW 264.7 by MTT assay and the quantification of IL-1 beta and TNF-alpha by ELISA.Results: The most effective concentration was 250 mg/mL and also promoted significant reduction (log(10)) in the biofilms of S. aureus (0.438 +/- 0.269), S. epiderrnidis (0.377 +/- 0.298), S. mutans (0.244 +/- 0.161) and C. albicans (0.746 +/- 0.209). Cell viability was similar to 100%. The production of IL-beta was similar to the control group (p > 0.05) and there was inhibition of TNF-alpha (p < 0.01).Conclusions: A. lappa L. extract was microbicidal for all the evaluated strains in planktonic cultures, microbiostatic for biofilms and not cytotoxic to the macrophages. A teicoplanina é um complexo antibiótico glicopeptídico derivado do Actinoplanes teichomyceticus, ativo contra bactérias Gram-positivas resistentes a outros antibióticos. Seu espectro de ação é similar ao da vancomicina, sendo, porém mais ativo para Streptococcus faecalis e Clostridium difficile. Seu uso é indicado para profilaxia de endocardite, peritonite, osteomielite e para septicemia estafilocócica. No Brasil, a teicoplanina é comercializada sob a forma farmacêutica de pó liofilizado, que deve ser reconstituído antes da administração. O medicamento de referência é o Targocid®, produzido pelo laboratório Sanofi-Aventis, em duas apresentações, 66 mg/mL e 133 mg/mL. A teicoplanina, bem como a vancomicina, inibe a sntese da parede celular bacteriana, pois a molécula se liga ao precursor da parede D-alanil-D-alanina, formando um complexo, impedindo a ligação à porção terminal do peptidoglicano, que é o alvo das enzimas transglicolase e transpeptidase. Desse modo, não há incorporação de aminoácidos aos glicopeptídeos integrantes da parede celular das bactérias Gram-positivas. No estudo de validação, foram aplicados os parâmetros de linearidade, intervalo, preciso, sensibilidade, especificidade, exatidão e robustez. O método desenvolvido e validado para a quantificação de teicoplanina pó liofilizado foi: Ensaio microbiológico por turbidimetria na faixa de concentração de 20,0 a 80,0 μg/mL, utilizando o micro-organismo Staphylococcus epidermidis ATCC 12228 IAL 2150. Os parâmetros estudados para a validação do método turbidimétrico atenderam a todas as especificações para a adequada quantificação de teicoplanina na forma farmacêutica pó liofilizado

Study the semi-quantitative and quantitative technique in the diagnosis of catheter-related infections in newborns and to determine oxacillin resistance in Staphylococcus isolated. It was analyzed 353 catheter tips from 273 newborns in the Neonatal Unit of Hospital FMB. To confirm the diagnosis of infection, were analyzed the clinical data of newborns, the presence of at least one positive blood culture and growth of ≥ 1000 CFU/mL on quantitative culture and/or ³15 UFC on semiquantitative culture, with the same microorganism isolation (species and drug sensitivity) in blood culture and no other focus of infection except the catheter. The disk diffusion technique was used to check similarity of strains and resistance to oxacillin. Of the 353 tips analyzed, 39 were included in this study as the inclusion criteria. The semiquantitative culture was positive in 26 (66.7%) catheters and quantitative culture was positive in 24 (61.5%). Of 273 patients, 19 (6.9%) had a diagnosis of catheter-related bloodstream Infection (CR-BSI). Of the 19 episodes of CR-BSI, S. epidermidis was the predominant etiological agent (84.2%). The resistance to the antibiotic methicillin was found in 14 (73.7%) strains of Staphylococcus. The semiquantitative method was more sensitive (79%) compared with the quantitative method (63%). The use of antibiotics may have influenced the sensitivity of the quantitative method as the microorganisms present in the lumen are exposed to higher concentrations of antibiotics administered via the catheter

Coleus blumei Benth (Lamiaceae), popularly known in Brazil as heart-hurt or coleus-of-Java is an ornamental plant widely used due to the color of its leaves. Several species of the genus Coleus present compounds with antioxidant, antimicrobial and allopathic activities due to the presence of chemicals such coleonol, forskolin and rosmarinic acid. Therefore, this study aimed to carry out phytochemical study and biological tests to evaluate the antibacterial, antioxidant and allopathic activities of the 70% ethanol extract and its fractions ethyl acetate, dichloromethane and end aqueous fraction leaf of Coleus blumei Benth. The plant extract was prepared by turbolisis from the powder of dried leaves, as a solvent for extraction using 70% ethanol. The ethanol extract was fractionated with solvents dichloromethane and ethyl acetate. The phytochemical study identified the presence of saponins, free anthraquinones, flavonoids and anthocyanidins. In evaluating the antibacterial dichloromethane fraction showed activity against all microorganisms tested both in agar diffusion test and microdilution test. The 70% ethanol extract showed activity against the micro-organisms S. aureus and S. epidermidis in agar diffusion test and against all microorganisms tested in microdilution test. The ethyl acetate fraction showed activity against the micro-organism S. aureus in the agar diffusion test and against all microorganisms tested in microdilution test. The end aqueous fraction showed no activity against any micro-organism tested. In the test of evaluation activity allopathic dichloromethane fraction showed greater inhibition of seed germination and growth of tomato (Lycopersicon esculentum Mill), followed by 70% ethanolic extract, the ethyl acetate fraction and the end aqueous fraction.
