In this study, it was demonstrated that β-galactosidase can be deactivated and reactivated with EDTA and divalent metal ions. The enzyme was deactivated after 20 minutes in EDTA solution. Maximal deactivation at the lowest EDTA concentration (10-3 mol.L-1) occurred in the presence of Tris-HCl buffer (pH 7.0). The enzyme recovered 50% of its initial activity after 10 minutes at Mg2+concentrations higher than 0.1 mmol.L-1. Experimental concentrations of 0.1 mmol.L-1 Mn2+ and 1.0 mmol.L-1 Co2+ were sufficient to reactivate the enzyme to around 300% of the control activity for the Mn2+ ion and nearly 100% for the Co2+ ion. The enzyme gradually lost its activity when the Co2+ concentration was 10-2 mol.L-1. Ni2+ and Zn2+ were unable to restore the catalytic activity. Km app and Vmax app were 1.95 ± 0.05 mmol.L-1 and 5.40 ± 0.86x10-2 mmol.min-1.mg-1, with o-NPG as substrate. Optimal temperature and pH were 34oC and 7.5. The half-life (t1/2) at 30°C was 17.5 min for the holoenzyme and 11.0 min for the apoenzyme. With respect to pH variation, the apoenzyme proved to be more sensitive than the holoenzyme. RESUMO Efeito de íons metálicos divalentes na atividade e estabilidade da β-galactosidase isolada de Kluyveromyces lactis Este estudo demonstra como a β-galactosidase pode ser desativada e reativada usando EDTA e íons metálicos divalentes. A enzima foi desativada após 20 minutos na presença de EDTA. Desativação máxima para a menor concentração de EDTA (10-3 mol.L-1) ocorreu na presença do tampão Tris-HCl. A enzima recuperou 50% de sua atividade inicial após 10 minutos na presença de Mg2+ em concentrações superiores a 0,1mmol.L-1. Concentrações de 10-4 e 10-3mol.L-1 de Mn2+ e Co2+ foram suficientes para reativar a enzima em 300% comparado ao controle de íons Mn2+ e aproximadamente 100% para íons Co2+. A enzima perdeu gradualmente a sua atividade quando a concentração foi de 10-2 mol.L-1. Ni2+ e Zn2+ foram incapazes de restabelecer a atividade catalítica. Km app e Vmax app foram 1,95 ± 0,05 mmol.L-1 e 5,40 ± 0,86 x 10-2 mmol.min-1.mg-1. A temperatura e pH ótimos foram 34ºC e 7,5. A meia vida da holoenzima foi de 17,5 min a 30ºC e para a apoenzima foi de 11,0 min a 30ºC. Quanto à variação de pH, a apoenzima provou ser mais sensível que a holoenzima. Urban homegardens are green areas of households within the city limits and they have the potential to provide families with a cheap alternative for diet improvement, and to complement the income of the families who sell cultivated products. This research analyzes the contributions of old urban homegardens on food consumption and household economy. Data related to homegardens composition were collected by interviews and by collecting cultivated plants. Diets were assessed through a retrospective method (last 24 hours food recall) and administered every two months, during a year, to include seasonal variations. The diet of the sampled population was found to be dependent on certain foods, indicating a narrow food niche (Levins index = 25.9; Levins standardized index = 0.23). Variations in interviewees' diet are related to the consumption of fruits and vegetables, which are partly supplied by homegardens. Spices and teas consumed were obtained from homegardens, revealing its importance in food consumption and health. Among the 98 species found in homegardens, only 38% appeared in the interviewees' diet, indicating an under-exploitation of these homegardens. Our study found that the main role of homegardens is to supply variation in the diet, contributing to the consumption of different types of products.

The process of milling wood is widely used in operations such as planing and manufacturing frames . Machines like planers , desengrossadeiras , routers , moldureiras and machining centers employ the milling process for cutting wood . In this work the process of milling CNC machining center of Eucalyptus grandis was studied because this is very much used in furniture , but without consistent studies on this process . This work a CNC machining center brand TECH Z1 for analysis of surface quality ( Ra ) in relation to the variation of cutting speed and feed in concordant and discordant tangential milling and face milling was used . We used Eucalyptus grandis . Four forward speeds ( 3, 5 , 7, and 9 m / min ) for four shear rates ( 5,9; 8,4; 10,9 and 13,4 m/s ) were used. Was used for testing a cutter finishing top speed steel with helical teeth 16mm in diameter . 6 repetitions for each test condition were performed . From the results it was observed that the best results for roughness Vc = 10,9 m / s were obtained for the milling concordant with the forward speed Vf = 7 m / min. As for Vf = 5 m / min the best finish was achieved with Vc = 8,4 m /s in discordant for milling . The feedrate and cutting influenced the roughness . The senses of concordant and discordant and cut the top and the top had a significant difference in roughness

The Biosusceptometry AC (BAC) is a research tool that has been extensively explored by the group Biomagnetism IBB-UNESP for monitoring of the gastrointestinal tract, its response to a known drug or in vivo performance of solid dosage forms. During this period the BAC, which has the characteristics of high sensitivity and low cost, has been developed primarily for recording signals contraction of activity and traffic human gastrointestinal tract. With the possibility of producing images with this instrumentation, it was possible to evaluate different situations in vitro and in vivo for physiological studies and pharmaceuticals. Considering the good performance of this system to produce planar images, the first aim of the BAC system tomography (TBAC) was to evaluate the system performance of BAC to produce tomographic images of phantoms ferromagnetic for a single channel system. All these applications were only possible because of their sensitivity to materials of high magnetic suscepitibility as ferrite, which allow to produce an electrical signal proportional to the variation of the magnetic flux generated by the presence of magnetic marker next to a first-order gradiometer. Measuring this variation at various points was possible to generate planar images that recently came to be produced in systems with multiple detectors, said multi-channels. From planar images, also producing tomographic images of simulators BAC bars in a system of 13 channels using only the center channel, with good results when applied to simple objects as one and two bars. When testing the resolution of the system with more elaborate forms the quality and resolution of images reconstructed is not satisfactory, which would be solved by increasing the spatial sampling rate and hence the acquisition time. This work is to analyze the behavior of context concentrated stresses generated around a nozzle connected to a pressure vessel. For this analysis we used the finite element method via a computer interface, the software ANSYS WORKBENCH. It was first necessary to study and intensive training of the software used, and also a study of the ASME Code, Section VIII, which is responsible for the standards used in pressure vessels. We analyzed three cases, which differ primarily in the variation of the diameter of the nozzle in order to analyze the variation of the stresses according to the variation of the diameters. The nozzle diameters were 35, 75 and 105 mm. After the model designed vessel, a pressure was applied on the innervessel of 0.5 MPa. For the smallest diameter, was found the lowest tensions concentrated. Varying between 1 and 223 MPa. Increasing the diameter of the nozzle resulted in increased tensions concentrated around the junction nozzle /vessel. The maximum stresses increased by 78% when the value was increased in diameter from 35 to 75 mm. Since the increase in diameter from 75 to 105 mm, the values of the tensions increased around 43%. Geophysics studies in areas impacted by petroleum derivatives describe abnormalities of both low and high electrical resistivity (the opposite of electrical conductivity), confirmed as contaminant phase by chemical analysis: this contradiction can be explained by degrading processes that naturally occur and create sub products that can change the environment conductivity. Monitoring the variation of the parameters mentioned serves as a comparative basis to the variation in geoelectrical parameters, which identified the correlation between the same contaminant parameters and the difference between their behavior studied apart, as well as its relations with the biodegradation process. The results are applied to the fuel distribution and storage sectors, leading to the diagnosis and monitoring of possible groundwater contamination scenarios, and the knowledge of the area exposure time to the contaminant, besides the better remediation alternative and impacts control. Among some conclusions, the most significant are the decrease in conductivity over time, so as the increase in Eh value in the gasoline contaminated tank, as well as the decrease in the pH value in the second tank with ethanol, which can be attributed to its degradation. Comparing the variations in both tanks, it is evident that Eh, pH and electrical conductivity do not behave temporally in a similar way, although some correlations between Eh and pH can be related.
