Foram submetidas a PCR-Ribotipagem e aos testes de sensibilidade in vitro frente a 12 antimicrobianos 77 estirpes de Staphylococcus aureus isoladas em amostras de leite procedentes de 40 vacas da raça holandesa que apresentaram mastite subclínica, em uma propriedade rural localizada no Estado de São Paulo, Brasil. Os resultados obtidos revelaram quatro diferentes padrões de resistência a antimicrobianos, sendo observada a predominância de resistência à lincomicina entre 19 (24,7%) estirpes de S.aureus. As 58 (75,3%) estirpes restantes foram sensíveis aos 12 antimicrobianos testados. A PCR-ribotipagem revelou a ocorrência de nove padrões genotípicos distintos, além de apresentar uma capacidade discriminatória maior (D = 0,82) que a obtida nos antibiogramas (D = 0,42). Entre as 19 estirpes resistentes aos antimicrobianos, 14 (73,7%) foram agrupadas em três padrões de ribotipagem e, destas, 13 (92,9%) apresentaram resistência à eritromicina e à lincomicina, isoladamente ou em associação. O grande número de ribotipos e de padrões de resistência a antimicrobianos observados nesta propriedade demonstrou que há grande heterogeneidade genética em populações naturais de S. aureus, fato este que deve ser levado em consideração em programas de controle da mastite bovina.

O experimento foi conduzido na UNESP - Jaboticabal com o objetivo de obter informações sobre o rendimento de matéria seca (RMS) e composição química das forrageiras de inverno, aveia preta (Avena strigosa Schreb) e triticale (X Triticosecale Wittmack), em plantio direto sob palhada de área com milheto (Pennisetum americanum (L.) K. Schum.) ou híbrido sorgo sudão (Sorghum bicolor (L.) Moench x Sorghum sudanense (Piper) Stapf) submetidas a pastejo. A forragem foi rebaixada por vacas da raça Holandês. O delineamento experimental utilizado foi em blocos casualizados, em arranjo de parcelas subdivididas, sendo analisado nas parcelas as espécies forrageiras e nas sub-parcelas as épocas de pastejo, com quatro repetições. Os tratamentos testados foram: AS: Aveia Preta sobre palhada de sorgo; AM: Aveia preta sobre palhada de milheto; TS: Triticale sobre palhada de sorgo; e, TM: Triticale sobre palhada de milheto. As pastagens apresentaram RMS semelhantes e, em relação às épocas de avaliação observou-se menor rendimento no segundo corte. Foram observados maiores teores de PB e de FDA na forragem colhida no primeiro corte e de hemicelulose no segundo corte, enquanto os valores de FDN não foram diferentes. A forragem de triticale apresentou menor conteúdo de PB e maiores teores de constituintes da parede celular.

Foram utilizadas 19.565 lactações vitalícias de vacas da raça Holandesa para avaliar o impacto dos motivos de descarte sobre a rentabilidade vitalícia de vacas leiteiras, mais especificamente sobre a receita líquida vitalícia (RLV), eficiência vitalícia (EF), receita do leite menos o custo de alimentação (RLMA) e receita líquida por dia de vida produtiva (RLD). A taxa geral de descarte foi igual a 31,39%, valor que pode ser considerado alto e estaria diretamente relacionado com a vida produtiva curta encontrada para estes rebanhos, 25,57 meses. A principal causa de descarte foi problema reprodutivo (21,57%), seguido por injúrias e outras causas (21,17%) e baixa produção (17,20%). Os descartes involuntários foram a grande maioria, representando 75% do total. As diferentes medidas econômicas estudadas sofreram impacto significativo do motivo de descarte, em que as vacas removidas por venda para fins leiteiros foram as mais rentáveis ao produtor.

Dados de 39.578 controles leiteiros de 3.766 primeiras lactações de vacas da raça Holandesa, ocorridas de 1994 a 2002, foram analisados com os objetivos de estimar parâmetros genéticos para as produções de leite no dia do controle (PLDC) e para a produção até 305 dias de lactação (P305) e comparar estes dois critérios de seleção. Os componentes de variância foram estimados pelo método da máxima verossimilhança restrita, sob modelo animal univariado ou bivariado. Para as PLDC, os modelos incluíram o efeito aleatório genético aditivo, o efeito fixo de grupo contemporâneo e, como covariáveis, a idade da vaca ao parto (efeitos linear e quadrático) e os dias em lactação (efeito linear). Para a P305, foi utilizado o mesmo modelo, substituindo dias em lactação por duração da lactação. Os grupos de contemporâneos foram formados por ano, mês do controle e rebanho (para as PLDC) e por ano, época do parto e rebanho (para a P305). As herdabilidades estimadas para a P305 foram de 0,27 e 0,25 para as análises univariadas e bivariadas, respectivamente. Para as PLDC, as herdabilidades variaram de 0,11 a 0,31. Para o modelo bivariado (pelo qual avaliaram-se simultaneamente P305 e as PLDC), as herdabilidades para os controles (PLDC) foram menores, variando de 0,08 a 0,25. As maiores estimativas ocorreram para as produções do 4º e 5º controles, correspondendo aos 2º e 3º meses de lactação. As correlações genéticas entre P305 e os controles individuais foram positivas e elevadas, variando de 0,83 a 1,00. Os resultados indicaram que a seleção direta para P305, como tradicionalmente realizada, implicaria maiores ganhos genéticos para a produção de leite (PL) na maioria dos controles quinzenais. Além disso, a seleção direta para as produções parciais poderia proporcionar ganhos correlacionados também para a P305, mas estes ganhos seriam menores que os obtidos via seleção direta.

Avaliou-se a influência do manejo de aleitamento nos níveis de cortisol, no metabolismo e na produção de leite de vacas leiteiras. Utilizaram-se 18 vacas holandesas e seus bezerros alocados em três tratamentos: T1: as vacas foram separadas dos bezerros 6 horas após o parto, sendo mantidas com suas crias 60 minutos por dia nos três primeiros dias; T2: as vacas foram separadas dos bezerros 6 horas após o parto; T3: as vacas foram mantidas com seus bezerros durante os três primeiros dias de lactação, exceto no momento das ordenhas. Foram coletadas amostras de sangue 168 horas antes do parto, no dia do parto (0 hora) e 24, 48, 72 e 96 horas após o parto para a determinação de glicose, triglicerídeos, proteína total e cortisol. A produção leiteira foi mensurada duas vezes ao dia e a porcentagem de leite residual foi estimada nas duas ordenhas posteriores à desmama. As concentrações de cortisol foram maiores ao parto, contudo, não houve diferença significativa entre os tratamentos. Durante o período experimental, as vacas separadas dos bezerros apresentaram maior produção de leite, porém, após este período, não houve diferença entre os tratamentos. Após a desmama, a porcentagem de leite residual foi maior em T1 e T3, demonstrando que o uso de ordenha mecânica exclusiva após um período de amamentação prejudicou a ejeção do leite, mesmo em vacas especializadas. Os níveis de glicose foram menores em T2 às 72 horas e as concentrações de triglicerídeos foram menores em T3 às 72 e 96 horas. Os manejos estudados não influenciaram os níveis de cortisol e o metabolismo das vacas e nem prejudicaram a produção leiteira após o período colostral.

The purpose of this study was to estimate the repeatability of transferable embryos in Holstein cows and to evaluate the effects of the year, season and order of the superovulation on the number of total structures, transferable embryos, non-transferable embryos and ovules. Four hundred and eighty-six superovulations were used in the analysis. The year of superovulation affected significantly all traits (P<0.01); however, we did not find effects of the season of the year. Superovulation order affected the total structures (P<0.01) and non-transferable embryos (P<0.05). The repeatability of the transferable embryo was 0.28 +/- 0.05. Positive correlations were found among total structures and transferable embryos (0.73) and total structure and ovule (0.51).

Objective-To determine the capacity of inflammatory mediators tumor necrosis factor-alpha (TNF-alpha), interleukin-8 (IL-8), platelet-activating factor (PAF), lipopolysaccharide (LPS), and leukotoxin to prime, activate, or alter deformability of adult bovine neutrophils.Sample Population-Blood collected from 5 healthy adult Holstein cows.Procedure-Isolated neutrophils or whole brood was incubated with TNF-alpha, IL-8, PAF, LPS, or leukotoxin, and neutrophil chemiluminescence, degranulation, deformability, shape change, CD11b expression, and size distribution was measured.Results-incubation with TNF-alpha, IL-8; PAF, and IFS primed neutrophils for oxygen radical release but caused minimal oxygen radical release by themselves. None of the inflammatory mediators induced degranulation. Incubation with TNF-alpha and PAF resulted in a decrease in neutrophil deformability and induced shape change in neutrophils. incubation with PAF consistently resulted in an increase in neutrophil size as measured by use of flow cytometry. Only IL-8 caused an increase in expression of CD11b by neutrophils.Conclusions and Clinical Relevance-Inflammatory mediators tested had minimal effects on neutrophil oxygen radical production or degranulation but did prime neutrophils for oxygen radical production. Incubation with PAF and TNF-alpha caused a decrease in neutrophil deformability and altered neutrophil shape and size. Results of our study indicate that PAF- and TNF-alpha-induced changes in neutrophil deformability and size may cause integrin- and setectin-independent trapping of neutrophils in the lungs of cattle with pneumonic pasteurellosis.

Milk, fat, and protein yields of Holstein cows from the States of New York and California in the United States were used to estimate (co)variances among yields in the first three lactations, using an animal model and a derivative-free restricted maximum likelihood (REML) algorithm, and to verify if yields in different lactations are the same trait. The data were split in 20 samples, 10 from each state, with means of 5463 and 5543 cows per sample from California and New York. Mean heritability estimates for milk, fat, and protein yields for California data were, respectively, 0.34, 0.35, and 0.40 for first; 0.31, 0.33, and 0.39 for second; and 0.28, 0.31, and 0.37 for third lactations. For New York data, estimates were 0.35, 0.40, and 0.34 for first; 0.34, 0.44, and 0.38 for second; and 0.32, 0.43, and 0.38 for third lactations. Means of estimates of genetic correlations between first and second, first and third, and second and third lactations for California data were 0.86, 0.77, and 0.96 for milk; 0.89, 0.84, and 0.97 for fat; and 0.90, 0.84, and 0.97 for protein yields. Mean estimates for New York data were 0.87, 0.81, and 0.97 for milk; 0.91, 0.86, and 0.98 for fat; and 0.88, 0.82, and 0.98 for protein yields. Environmental correlations varied from 0.30 to 0.50 and were larger between second and third lactations. Phenotypic correlations were similar for both states and varied from 0.52 to 0.66 for milk, fat and protein yields. These estimates are consistent with previous estimates obtained with animal models. Yields in different lactations are not statistically the same trait but for selection programs such yields can be modelled as the same trait because of the high genetic correlations.

Blood constituents of Nelore, Hosltein and buffalo cows were determined during pregnancy, at the time of calving and at post-partum. It was verified that. 1. The pregnancy and post-partum periods had no influence in the erythrocyte values, but at calving the red blood cell counts and hemoglobin levels of Holstein and buffalo cows were lower than for Nelore cattle; 2. leukocyte counts were similar among groups; 3. total protein levels of Nelore cows were lower than Holstein and buffalo. The albumin levels were rite lowest at rite time of calving compared to late pregnancy; 4. glicose levels were lower ill buffalo cows during pregnancy as compared with Holstein cows. The glicemia of Nelore cows was lower as compared to Holstein cow's irt late pregnancy; 5. urea and creatinine levels were higher in buffalo cows than cattle. The urea and creatinine levels were greater in buffalo cows with maximum values at the post-partum and at the parturition, respectively; 6. bilirrubin levels were higher in bovine than buffalo cows; 7. aspartate aminotransferase activity was greater in buffalo cows and increased at the time of calving; 8. alkaline phosphatase activity was increased during pregnancy and decreased after the time of calving; 9. gammnglutamyltransferase activity,vas the highest for buffalo cows after calving; IO. calcium levels were the highest for Holstein cows at the post-partum and the phosphorus levels were higher in buffalo cows, which had the highest magnesium levels at the parturation.

Based on in vitro experiments, Bos indicus embryos were more resistant to heat stress (HS) than Bos taurus embryos. To increase knowledge regarding differences between Bos indicus and Bos taurus in resistance to HS, the primary objective of this study was to determine if tolerance to HS is due to the breed, origin of the oocyte, sperm, or both. Additionally, the influence of the interval between ovary acquisition (in the abattoir) and oocyte aspiration in the laboratory, on early embryo development was ascertained. Oocytes were collected from Nelore and Holstein cows in an abattoir; 4.0 or 6.5 h later, oocytes were aspired in the laboratory, and then matured and fertilized using semen from Nelore (N), Gir (GIR), or Holstein (H) bulls. Ninety-six h post insemination (hpi), embryos with >= 16 cells were divided in two groups: control and HS. In the control group, embryos were cultured at 39 degrees C, whereas in the HS group, embryos were subjected to 41 degrees C for 12 h, and then returned to 39 degrees C. Rates of cleavage, and formation of morula and blastocysts were higher (P < 0.05) for oocytes aspirated at 4.0 versus 6.5 h after ovaries were acquired. Heat stress decreased rates of blastocyst formation for all breeds (N X N; H x H; and H X GIR) and in both time intervals (4.0 and 6.5 h). However, N X N had higher cleavage rate (P < 0.05) in both time intervals when compared with H X H and H X GIR. In addition, Nelore oocytes fertilized with Nelore semen (N X N) had higher blastocyst yields (P < 0.05) in the control and HS group, when compared with the other two breeds (H X H and H X GIR). We concluded that the breed of origin of the oocyte was more important than that of the sperm for development of thermotolerance, because bull breed did not influence embryo development after HS, and in vitro early embryonic development was impaired by increasing (from 4 to 6.5 h) the interval between ovary acquisition and oocyte aspiration. The experiment was carried out at Piracicaba, São Paulo, Brazil, from January to February 1993, with the objective of evaluating the behavior responses of Holstein cows, with constant or limited access to shade. The experimental design used was completely randomized. Twenty four dairy cows were used, at different lactation stages and production levels, kept in two free stall barns, with or without protection against solar radiation in south-east and north-west edge. The behavior parameters studied were: alimentation, rumination, rest time and frequency and water ingestion frequency. The protection of the free stall barn didn't affect the behavior responses. The alimentation, rumination and rest time, daily, were 3.4, 7.0 e 9.0 hours, respectively. The highest alimentation frequencies were before and after milking. The rumination was mainly during nocturnal period; the rest was more frequent during the period with higher solar radiation. The animals stayed more time in the shelter (13.4 vs 2.5 h/day). The highest daily water ingestion frequencies were in the hot time and next milking, mainly.

Aiming to compare milk yield, residual milk and behavioral responses of Holstein cows during milking, fourteen experimental cows were mechanically milked twice a day, and evaluated during the first six weeks of lactation. Milk yield was measured daily, and residual milk volume weekly. Body (ribs region), udder and teat temperatures and respiratory frequency were measured twice a week. Animal behavior at the milking parlor was registered using ethograms and interviews with the milkers. During the first weeks of lactation, a higher percentage of kicks, over-steps, milking cups being knocked down, urinations and vocalizations in primiparous cows (p < 0.05) was observed, which were also considered more reactive by the parlors compared to multiparous cows. Multiparous cows produced more milk than primiparous (p < 0.05). However, there were no significant differences in residual milk percentage (p > 0.05), which suggests adaptation of both groups to the milking environment.
