Selection of the best source of carbon for production of recombinants enzymes in liquid fermentation

A partial pseudo-ternary phase diagram has been studied for the cethyltrimethylammonium bromide/isooctane:hexanol:butanol/potassium phosphate buffer system, where the two-phase diagram consisting of the reverse micelle phase (L-2) in equilibrium with the solvent is indicated. Based on these diagrams two-phase systems of reverse micelles were prepared with different compositions of the compounds and used for extraction and recovery of two enzymes, and the percentage of enzyme recovery yield monitored. The enzymes glucose-6-phosphate dehydrogenase (G6PD) and xylose redutase (XR) obtained from Candida guilliermondii yeast were used in the extraction procedures. The recovery yield data indicate that micelles having different composition give selective extraction of enzymes. The method can thus be used to optimize enzyme extraction processes. Tuberculosis (TB) remains the leading cause of mortality due to a single bacterial pathogen, Mycobacterium tuberculosis. The reemergence of TB as a potential public health threat, the high susceptibility of human immunodeficiency virus-infected persons to the disease, the proliferation of multi-drug-resistant strains (MDR-TB) and, more recently, of extensively drug resistant isolates (XDR-TB) have created a need for the development of new antimycobacterial agents. Amongst the several proteins and/or enzymes to be studied as potential targets to develop novel drugs against M. tuberculosis, the enzymes of the shikimate pathway are attractive targets because they are essential in algae, higher plants, bacteria, and fungi, but absent from mammals. The mycobacterial shikimate pathway leads to the biosynthesis of chorismate, which is a precursor of aromatic amino acids, naphthoquinones, menaquinones, and mycobactins. Here we report the structural studies by homology modeling and circular dichroism spectroscopy of the shikimate dehydrogenase from M. tuberculosis (MtSDH), which catalyses the fourth step of the shikimate pathway. Our structural models show that the MtSDH has similar structure to other shikimate dehydrogenase structures previously reported either in presence or absence of NADP, despite the low amino acid sequence identity. The circular dichroism spectra corroborate the secondary structure content observed in the MtSDH models developed. The enzyme was stable up to 50 degrees C presenting a cooperative unfolding profile with the midpoint of the unfolding temperature value of similar to 63-64 degrees C, as observed in the unfolding experiment followed by circular dichroism. Our MtSDH structural models and circular dichroism data showed small conformational changes induced by NADP binding. We hope that the data presented here will assist the rational design of antitubercular agents.

Esterases are a group of enzymes that are reportedly associated with acaricide resistance in Riphicephallus (Boophilus) microplus. A comparative analysis was made of the esterase patterns in malathion and deltamethrin-sensitive, tolerant and resistant tick groups, using non-denaturing polyacrylamide gel electrophoresis. Electrophoretical profiles revealed four bands of esterase activity against alpha-naphthyl acetate; which were dubbed EST-1 to EST-4. The EST-3 and EST-4 were detected in all strains and were classified as carboxylesterases (CaEs). The EST-2, classified as an acetylcholinesterase (AChE), was detected in all groups, but its staining intensity increased from susceptible to resistant groups, indicating an altered production according to the degree of resistance. EST-1, which was also classified as an AChE, was detected exclusively in tolerant and resistant groups to both acaricides, but displayed greater activity in the malathion-resistant group. These data suggest that these AChEs may represent an important detoxification strategy developed to overcome the effects of acaricides. (C) 2008 Elsevier B.V. All rights reserved.

Microbial xylanolytic enzymes have a promising biotechnological potential, and are extensively applied in industries. In this study, induction of xylanolytic activity was examined in Aspergillus phoenicis. Xylanase activity induced by xylan, xylose or beta-methylxyloside was predominantly extracellular (93-97%). Addition of 1% glucose to media supplemented with xylan or xylose repressed xylanase production. Glucose repression was alleviated by addition of cAMP or dibutyryl-cAMP. These physiological observations were supported by a Northern analysis using part of the xylanase gene ApXLN as a probe. Gene transcription was shown to be induced by xylan, xylose, and beta-methylxyloside, and was repressed by the addition of 1% glucose. Glucose repression was partially relieved by addition of cAMP or dibutyryl cAMP.

Seasonality of pathogenic fungi in mites of rubber tree plantations adjacent to fragments of Cerrado

Fungos são os patógenos mais freqüentemente observados em populações de ácaros, auxiliando o controle destes em diferentes culturas. Vinte e cinco amostras de folhas foram coletadas de cultivos de seringueiras vizinhas a dois fragmentos de vegetação de Cerrado. em cada seringal, 25 plantas foram selecionadas para coletas e sete folhas ao redor da copa foram coletadas até a altura de sete a oito metros do solo. Aproximadamente 250 indivíduos de Calacarus heveae Feres, Phyllocoptruta seringueirae Feres e Tenuipalpus heveae Baker, coletados aleatoriamente, foram montados. Hirsutella thompsonii Fisher foi observado nas três espécies de ácaros e T. heveae foi a espécie mais infectada. Os maiores níveis de infecção ocorreram de novembro a fevereiro (estação chuvosa). Na estação seca, os níveis de infecção ficaram abaixo dos 5%. H. thompsonii tem potencial para ser usado como micoacaricida na estação chuvosa.

Estudos sobre a ecologia de organismos envolvidos no processo de produção são necessários para o desenvolvimento de uma agricultura sustentável, e hoje, a sustentabilidade está intimamente ligada à rentabilidade de produção. Objetivou-se com este trabalho verificar a ocorrência de fungos micorrízicos arbusculares em plantas daninhas de lavouras brasileiras e avaliar o potencial de solubilização de fosfato inorgânico da microbiota associada. 36 espécies de plantas daninhas foram avaliadas quanto à ocorrência de micorrizas, e 11 foram selecionadas para avaliação do potencial de solubilização total e relativa de fosfato. Todas as espécies apresentaram colonização por micorrizas, inclusive um exemplar da família Brassicaceae, geralmente enquadrada como não micorrizada. Na maioria das espécies, os tipos morfológicos de arbúsculos e enovelados de hifas foram observados, sendo os enovelados mais comuns entre as gramíneas. Fungos endofíticos do tipo dark septate foram visualizados na maioria das plantas. As plantas daninhas apresentaram distintos potenciais de solubilização de P na rizosfera. Amaranthus retroflexus, Bidens pilosa e Leonotis nepetaefolia apresentaram elevados valores de solubilização relativa de fosfato. Este é o primeiro relato de micorrizas e da atividade de solubilização de fosfato em plantas daninhas no Brasil.

In an attempt to isolate Paracoccidioides brasiliensis from nature 887 samples of soil from Botucatu, SP, Brazil, were collected cultured in brain heart infusion agar supplemented with dextrose, in potato dextrose agar and in yeast extract starch dextrose agar, all with antibiotics, at 25º and 37ºC. Five thermo-dependent dimorphic fungi morphologically resembling P. brasiliensis were isolated; two from armadillo holes; further studies of the biology, antigenicity and genetic features of the five dimorphic fungi are necessary to clarify their taxonomy and their possible relation to P. brasiliensis. In addition, 98 dematiaceous fungi and 581 different species of Aspergillus spp. were also isolated. Our findings emphasize that armadillos and their environment are associated with thermo-dimorphic fungi and confirm the ubiquity of pathogenic dematiaceous fungi and Aspergillus spp.
