Seasonal variation of abundance of Ecballocystis pulvinata Bohlin in a stream of central-southern region of Parana State. The temporal dynamics of E. pulvinata and its relation to abiotic variables were evaluated monthly during January to December 2007 in a stream located in the central-southern part of Parana State (25 degrees 11'16 '' S - 51 degrees 021'25 '' W). The abundance of the population in terms of percentage cover was obtained by means of the quadrat technique and the following stream water variables were measured: current velocity, irradiance, depth, substrate type, temperature, oxygen saturation, conductivity, pH and turbidity. The abundance of E. pulvinata was considered high (X = 26.7%), with more development (22-46%) during autumn/winter. The low values (0-0.08) of niche width and the few associations with environmental variables shows that the population evaluated is a generalist with regards to the environment in which it occurs. Factors most strictly related were the water flow and the particle size of the substrate, which positively correlated to the seasonal pattern of the population investigated.

Objective To assess the viability of the early diagnosis of fetal gender in material plasma before 7 weeks of pregnancy by real-time polymerase chain reaction (real-time PCR), starting at 5 weeks of pregnancy.Method peripheral blood was collected from pregnant women, starting at 5 weeks of gestation. After centrifugation, plasma was separated for fetal DNA extraction. DNA was analyzed by quantitative real-time PCR for two genomic regions, one on the Y chromosome (DYS-14) and the other shared by both sexes (beta-globin), by the TaqMan Minor Groove Binder (MGB) probe assay. The results of the examinations were compared to fetal gender determined after delivery.Results A total of 79 examinations of fetal DNA in maternal plasma were performed for 52 pregnant women. Accuracy according to gestational age was 92.6% (25 of 27 cases) at 5 weeks, and 95.6% (22 of 23 cases) at 6 weeks. These results also demonstrate that fetal DNA is present at low concentrations in maternal plasma at 5 weeks (8.5 genome equivalents (GE)/mL) and 6 weeks (34.1 GE/mL) of pregnancy.Conclusion Quantitative real-time PCR and TaqMan MGB probes specific for the detection of fetal gender in maternal plasma starting at 5 weeks of gestation have good sensitivity and excellent specificity. OBJECTIVE To (1) analyze possible relationships between motile sperm organelle morphology examination (MSOME) and sperm chromatin status, aneuploidy incidence, and patient's age; (2) determine the effects of sperm morphologic abnormalities on intracytoplasmic sperm injection (ICSI) outcomes; and (3) identify the benefits of intracytoplasmic morphologically selected sperm injection (IMSI) in patients with high DNA fragmentation rate.METHODS The study was performed in 50 patients undergoing ICSI cycles. The MSOME, sperm DNA fragmentation, and sperm aneuploidy incidence were performed in 200 sperm cells of each patient. Regression models were used to assess the relationships among sperm morphology and sperm aneuploidy, sperm DNA fragmentation, patient's age, and ICSI outcomes. In cycles with patients showing a high incidence of DNA fragmentation, oocytes were split into 2 groups according to the sperm selection method: Standard-ICSI (n = 82) and IMSI (n = 79). Fertilization and high-quality embryo rates were compared between the groups.RESULTS A close relationship between sperm DNA fragmentation and the presence of vacuoles in the MSOME was noted. The patient's age was correlated to the presence of vacuoles. No correlation between sperm aneuploidy and IMSI was observed. Vacuolated cells were negatively correlated with fertilization, pregnancy, and implantation. In patients with a high incidence of sperm DNA fragmentation, fertilization and high-quality embryo rates were similar when comparing IMSI and Standard-ICSI.CONCLUSIONS Our data demonstrate a correlation between paternal age and the incidence of nuclear vacuoles, as well as an effect of large and small vacuoles on late embryo development. A toxoplasmose é uma das zoonoses mais difundidas no mundo, causada pelo Toxoplasma gondii, um protozoário parasita intracelular obrigatório. Uma alta porcentagem de animais apresenta anticorpos específicos causados por exposição prévia, levando a uma infecção crônica. Os felídeos são os hospedeiros definitivos e outros animais homeotérmicos, incluindo os primatas, são os hospedeiros intermediários. Este estudo objetivou determinar a prevalência da infecção por T. gondii em macacos-prego (Cebus apella nigritus) de vida livre da Estação Ecológica localizada na Mata de Santa Teresa, Ribeirão Preto, SP, Brasil. Anticorpos anti-T. gondii foram pesquisados pelo método de aglutinação direta modificada (MAT) em amostras de soro de 36 macacos-prego, utilizando-se o título oito como de corte. Dos animais estudados, 3/36 (8,33%; IC95% 3,0-21,9%) apresentaram anticorpos anti-T. gondii, todos com título 32. Nenhuma diferença significativa (P>0,05) foi observada com relação ao sexo (1/3 machos e 2/3 fêmeas), e à idade (1/3 jovens e 2/3 adultos). Assim, estes resultados demonstram alta prevalência de anticorpos anti-T. gondii em primatas no estado de São Paulo.

Heat stress has negative effects on pregnancy rates of lactating dairy cattle. There are genetic differences in tolerance to heat stress; Bos taurus indicus (B. t. indicus) cattle and embryos are more thermotolerant than Bos taurus taurus (B. t. taurus). In the present study, the effects of sire and sire breed on conception and embryonic/fetal loss rates of lactating Holstein cows during the Brazilian summer were determined. In Experiment 1, cows (n = 302) were AI after estrus detection or at a fixed-time with semen from one Gyr (B. t. indicus) or one Holstein sire (B. t. taurus). Pregnancy was diagnosed 80 days after AI. In Experiment 2, cows (n = 811) were AI with semen from three Gyr and two Holstein sires. Pregnancy was diagnosed at 30-40 and at 60-80 days after AI. Cows diagnosed pregnant at the first examination but non-pregnant at the second were considered as having lost their embryo or fetus. Data were analyzed by logistic regression. The model considered the effect of sire within breed, sire breed, days postpartum, period of lactation, and AI type (AI after estrus versus fixed-time). There was no effect of the AI type, days postpartum or milk production on conception or embryonic loss rates. The use of Gyr bulls increased pregnancy rate when compared to Holstein bulls [9.1% (60/657) versus 5.0% (23/456), respectively, P = 0.008; data from Experiments 1 and 2 combined]. Additionally, in Experiment 2, cows inseminated using semen from sire #4 (Gyr) had lower embryonic loss (10%) when compared with other B. t. indicus (35.3% and 40%) or B. t. taurus sires (18.2% and 38.5%, P = 0.03). In conclusion, the use of B. t. indicus sires may result in higher conception rates in lactating Holstein cows during summer heat stress. Moreover, sire can affect embryonic loss and selection of bulls according to this criterion may result in higher parturition rates in lactating Holstein cows. (c) 2006 Elsevier B.V. The objective was to determine the relationship among the diameter of ovarian follicles, ovulation rate, and gene expression of the LH receptor (LHR) in Nelore cattle. In Experiment 1, ovulation was synchronized in 53 Nelore cows. Three days after ovulation, ovaries were assessed with ultrasonography, all cows were given 6.25 mg LH im, and they were allocated into three groups, according to diameter of their largest ovarian follicle: G1 (7.0-8.0 mm); G2 (8.1-9.0 mm); and G3 (9.1-10.0 mm). For these three groups, ovulation rates were 9, 36, and 90%, respectively, (P < 0.03; each rate differed significantly from the other two). In Experiment 2, granulosa and theca cells were subjected to total RNA extraction, and gene expression of the LHR was determined by RT-PCR. Follicles were allocated in three groups based on their diameter (similar to the Experiment 1), which were denoted Groups A, B, and C. Expression of the LHR gene in granulosa cells was lower in Group A than Group C (P < 0.05). However, there were no significant differences among groups in expression of the LHR gene in theca cells. We concluded that ovulatory capacity in Nelore cattle was related to increased follicular diameter and expression of the LHR gene in granulosa cells. (C) 2012 Elsevier B.V. All rights reserved.

Avaliaram-se os efeitos da remoção temporária de bezerros (RTB) ou da aplicação de gonadotrofina coriônica eqüina (eCG) na taxa de prenhez (TP) de vacas Nelore lactantes tratadas com um dispositivo intravaginal liberador de progesterona (DILP). No experimento 1, 83 vacas Nelore e 102 mestiças (Nelore × Red Angus) foram distribuídas em três grupos. em dia aleatório do ciclo estral (D0), os animais do grupo 1 foram tratados com benzoato de estradiol (BE; 2,5 mg, IM, Estrogin®) e um DILP (1 g de progesterona, DIB®), removido no D9, quando também se administrou d-cloprostenol (150 µg, IM, Prolise®) e aproximadamente 12 horas após identificação do estro realizou-se a inseminação artificial. No grupo 2 (IATF), o tratamento foi semelhante ao aplicado no grupo 1, porém, administrou-se uma segunda dose de benzoato de estradiol (1 mg) no D10 e 30-36 horas mais tarde realizou-se a inseminação artificial em tempo fixo (IATF). No grupo 3 (IATF/RTB), os bezerros foram removidos a partir do D9 até a inseminação artificial com tempo fixo (IATF). As taxas de prenhez para as vacas Nelore e mestiças foram, respectivamente, de 7,69 e 41% (Controle); 23,52 e 59,57% (IATF); 69,44 e 55,81% (IATF/RTB). No experimento 2, 255 vacas Nelore lactantes foram distribuídas em três grupos. Os animais do grupo 4 (IATF) foram tratados com benzoato de estradiol (2 mg) e um DILP (1,9 g de progesterona, CIDR-B®) no D0 e no D8 sofreram remoção do DILP no D8 e administração de 25 mg de dinoprost (IM, Lutalyse®). No D9, foi aplicado benzoato de estradiol (1 mg) realizando-se a IATF 30-36 horas mais tarde. No grupo 5 (IATF/RTB), os bezerros foram removidos no D9 até a IATF. O grupo 6 (eCG) foi semelhante ao IATF, exceto pela aplicação de eCG no D9 (400 UI, IM, Novormon®). As taxas de prenhez foram de 50,57 (IATF), 53,57 (IATF/RTB) e 54,76% (eCG). A associação de RTB ao tratamento hormonal com DILP aumenta as taxas de prenhez, enquanto a adição de eCG ao tratamento não melhora as taxa de prenhez de vacas Nelore lactantes ciclando e em boa condição corporal.
