Enzymatic production by thermophilic fungi using agricultural wastes and ruminant diet as substrates

Purpose: To characterize the vitreous intrinsic proteoglycans, investigate their dynamics, and examine their role in the supramolecular organization of the vitreous. Methods: Vitreous from normal rabbits was collected and processed for observation with the transmission electron microscope after treatment with glycosidases. Also, rabbits were injected intravitreally with [S-35]-sodium sulfate and sacrificed at several time intervals after the injection. Proteoglycans (PGs) were assayed in the vitreous supernatant or in whole samples extracted with guanidine hydrochloride by polyacrylamide or agarose gel electrophoresis, followed respectively by fluorography or autoradiography, and ion-exchange chromatography and gel-filtration chromatography, combined with glycolytic treatment of the samples. The sulfated glycosaminoglycans (GAGs) were characterized by agarose gel electrophoresis after treating vitreous samples with protease and specific glycosidases. Results: the electron microscopic study revealed a network with hyaluronic acid ( HA) as thin threads coating and connecting collagen fibrils. The elimination of the HA coat showed chondroitin sulfate granules (8-25 nm) arranged at regular intervals on the fibril surface. The chondroitinase ABC digestion, besides removing the granules, also caused the formation of thicker bundles of the collagen fibrils. The PG and GAG analysis indicated that there are three renewable PGs in the vitreous ( e. g., one heparan-and two chondroitin-sulfate ones). Conclusions: At least one of the chondroitin sulfate PGs is involved in the interactions that occur in the vitreous structure, mainly by providing adequate spacing between the collagen fibrils, a condition that is probably required for the transparency of the vitreous.

Acetone and ethanol extracts of the tubercula and several compounds isolated from Aristolochia pubescens (Willd) were bioassayed on velvebean caterpillars, Anticarsia gemmatalis (Hubner), for evaluation of the insecticidal activities. of the extracts subjected to bioassay, the acetone extract showed the highest activity. (-)-Cubebin did not show activity against soybean caterpillars, whereas aristolochic acid and ent-kaur-15-en-17-ol increased the larval period. These compounds, and (+)-eudesmin and (+)-sesamin, reduced the viability of this period, giving rise to malformed adults. These extracts and compounds are therefore potential botanical insecticide agents for the control of velvetbean caterpillars in soybean crops. (C) 2003 Society of Chemical Industry.

A specific and sensitive high-performance liquid chromatographic procedure was developed for the assay of sparfloxacin in raw material and tablets. It was also found that the excipients in the commercial tablet preparation did not interfere with the assay. The method validation yielded good results and included the range, linearity, precision, accuracy, specificity and recovery. This method can also be applied to stability studies. Purine nucleoside phosphorylase (PNP) catalyzes the reversible phosphorolysis of nucleosides and deoxynucleosides, generating ribose 1-phosphate and the purine base, which is an important step of purine catabolism pathway. The lack of such an activity in humans, owing to a genetic disorder, causes T-cell impairment, and thus drugs that inhibit human PNP activity have the potential of being utilized as modulators of the immunological system to treat leukemia, autoimmune diseases, and rejection in organ transplantation. Besides, the purine salvage pathway is the only possible way for apicomplexan parasites to obtain the building blocks for RNA and DNA synthesis, which makes PNP from these parasites an attractive target for drug development against diseases such as malaria. Hence, a number of research groups have made efforts to elucidate the mechanism of action of PNP based on structural and kinetic studies. It is conceivable that the mechanism may be different for PNPs from diverse sources, and influenced by the oligomeric state of the enzyme in solution. Furthermore, distinct transition state structures can make possible the rational design of specific inhibitors for human and apicomplexan enzymes. Here, we review the current status of these research efforts to elucidate the mechanism of PNP-catalyzed chemical reaction, focusing on the mammalian and Plamodium falciparum enzymes, targets for drug development against, respectively, T-Cell and Apicomplexan parasites-mediated diseases.

The objective of the present research was to determine the allelopathic potential of Ipomoea fistulosa plants on germination and growth of seedlings. In laboratory, lettuce and tomato seeds were placed to germinate in the presence of water or Ipomoea fistulosa leaf extract, in the concentrations of 5, 10 e 15% (p/v). In greenhouse, seeds were placed to germinate in trays containing Plantmax (R) substrate and Plantmax (R) substrate + Ipomoea fistulosa dry leaf residue, at 3% (w/w). The aqueous extract inhibited germination, raised the mean germination time of the seeds and reduced the radicle and hypocotyl growth in all used concentrations. In greenhouse, the plant height, leaf number, and dry mass accumulation also were reduced in the presence of the leaf residue in the substrate. The results suggest the presence of allelochemicals in Ipomoea fistulosa and demonstrate the highest sensitivity of lettuce that tomato to these compounds.

Five species of smooth-hound sharks genus Mustelus (Family Triakidae) are know in the western South Atlantic, as follows: Mustelus canis (Mitchell 1815); Mustelus fasciatus (Garman 1913); Mustelus higmani Springer & Lowe 1963; Mustelus norrisi Springer 1939; and Mustelus schmitti Springer 1939. In the present paper, new data on the anatomy, morphometrics and meristic characters are given. Taxonomic aspects and comparison between the species are discussed. Most general body morphologic measurements and proportions are useless as a tool for species identification, since many of them show remarkable intraspecific variations. Head proportions and structures related seem to be a more adequate procedure to identify the species of Mustelus. The labial folds proportions, internasal distance and orbit diameter were the most useful character to separate the western South Atlantic species. The buccopharingeal pattern of denticles as well as tooth counts not was useful to distinguish the Mustelus species from western South Atlantic adequately, due great intraspecific variation.

Silages from three cultivars of triticale (X Triticosecale wittimack) were evaluated at the UNESP, Jaboticabal, Brazil. The cultivars FCA VJ-CB-01, FCA VJ-CB-02, and FCA VJ-CB-03 were harvested for silage in three growing stages of maturity; beginning of flowering, (S1), milk stage (S2), and dough stages (S3). Data were analyzed by randomized complete block with three replications. The DM (%) values increased while the CP (%DM) and buffering capacity (me HCl/100 g DM) decreased with plant development. Silages of plants harvested at S3 stage had higher pH and N-NH3 values compared to the S1 and S2 silages. The soluble carbohydrates contents (%DM) were higher at the S2 stage (16.9%) and were not different at the S1 (8.7%) and S3 (9.2%) stages. The crude energy contents (Kcal/kg MS) increased while the ADF, NDF, cellulose, and hemicellulose (DM%) decreased due to the presence of dough grains. This was not observed with the lignin contents. The IVDMD values were 66.3, 60.1 and 58.9%, for plants harvested at the S1, S2, and S3 stages, respectively. The results showed that there was no difference among for chemical composition, crude energy, and for IVDMD.

This paper describes the chemical variability of the Late Precambrian Itu Rapakivi Province (IRP), State of São Paulo, SE Brazil, based on 187 selected analyses from the Itu, São Francisco, Sorocaba, Campina do Veado and Sguario/ Correa granites. The IRP has an almost uniform petrographic character conferred by the overall dominance of subalkaline biotite granites. Monzogranites (adamellites), granodiorites, quartz syenites, quartz monzonites are rare to very rare rock types and tonalites and quartz diorites are almost restricted to enclaves. Typical chemical features are the high FeO*/MgO ratio, a clustering of the K2O values between 4.5 and 6.0 wt.% and K2O/Na2O ratios which define the IRP as mildly potassic although more potassic rocks also occur. The overal Peacock Alkalinity Index is 54 defining the Province as alkali-calcic. In the Shand diagram the data cluster near the metaluminous/peraluminous boundary. Relationships between Nb, Rb and Y stress the within plate character of the IRP and the relationships between Rb, Ba and Sr reveal the importance of feldspar fractionation in magma evolution. The data also show an interbody and an intrabody chemical variability due to the variation in the composition of the crustal magma protoliths, as assigned by K/Rb relations. The presence of several magmatic cycles which built up the major intrusions reflects a magma ascent from collecting chambers successively drained and recharged, a feature in agreement with the clear link between the bodies and long lived, successively reactivated, transcurrent faults. Most of the chemical features of the IRP correspond to those of the classical Finnish rapakivi granites.
