In 1956, Africanized bees began to spread in the American continent from southern Brazil, where original African bees mated with European bees. A few years later, in 1990, these Africanized bees reached the United States and were found in Texas. Currently, these hybrid bees are found in several North American states and will probably reach the Canadian border in the future. Although the presence of Africanized bees had produced positive effects on Brazilian economy, including improvement in crop pollination and in honey production, turning Brazil into a major exporter, the negative impacts-such as swarming, aggressive behavior, and the ability to mass attack-resulted in serious and fatal envenomation with humans and animals. Victims of bee attacks usually develop a severe envenomation syndrome characterized by the release of a large amount of cytokines [interleukins (IL) IL-1, IL-6, IL-8], and tumor necrosis factor (TNF). Subsequently, such cytokines produce an acute inflammatory response that triggers adverse effects on skeletal muscles; bone marrow; hepatic and renal functions; and cardiovascular, central nervous, and immune systems. Finally, the aim of the present review is to study historical characteristics and current status of Africanized bees' spread, the composition of their venom, the impact of the bees on the Brazilian economy and ecology, and clinical aspects of their stings including immune response, and to suggest a protocol for bee sting management since there is no safe and effective antivenom available.

The Dufour's gland is found closely associated with the sting apparatus of all female hymenopterans, playing multiple roles among bees. In some species of Bombus the gland may be involved in production of nestmate recognition pheromones, but in B. terrestris its function is not certain yet. The morphology of the :Dufour's gland of B. terrestris queens and the ultrastructural features of its cells were studied in different ages and behavioural stages using routine transmission electron microscopy. Measurements of the length and the diameter of the gland in the same conditions were also made. The Dufour's gland of the queen increases significantly in size (both in length and in diameter) with age and reproductive activity the ultrastructural features of the gland show electrondense material that comes from the haemolymph. This material is also present in the intercellular spaces, and is conducted to the subcuticular space, to be released directly into the glandular lumen. Hence at least part of the secretion is probably taken up directly from the haemolymph. The ultrastructural features indicate a more active phase of the gland corresponding to the period of egg-laying of the queen, and a decrease in activity when the queen is in hibernation as well as after the competition point. In conclusion, the gland is probably involved in reproduction, more specifically, in the marking off eggs.

A series of experiments was performed to assess possible roles of the honey bee Dufour gland secretion. Bioassays with extracts of queen and worker glands from two colonies were made under artificial conditions, in which nestmate and non-nestmate forager workers were tested. The results demonstrate that forager workers display behavioral responses when exposed to Dufour gland extracts of nestmates, but remain indifferent when exposed to non-nestmate extracts. Also, the results demonstrate that forager workers are attracted by virgin queen gland extracts, and repelled by forager worker extracts. The data demonstrate that the Dufour gland secretion is colony- and caste-specific. The attractant remains in the nest. The repellent effect of forager worker extracts is interpreted as an alarm-like pheromone. The attractant effect of virgin queen extracts could be useful in the swarming process to attract scout bees to the eventual founder virgin queen or to attract old foragers to the virgin queen that remains in the nest.

Different modes of cell death have been revealed in the regressing hypopharyngeal glands of worker honey bees. The hypopharyngeal gland, which is well developed in young nursing bees to produce protein for larval food, was seen to regress naturally in foraging adult worker bees. A range of techniques including histology, cytochemistry, in situ TUNEL, Annexin V and Comet assays indicated that cells within the gland demonstrate progressive symptoms of apoptosis, necrosis and a vacuolar form of programmed cell death. The latter mode of cell death did not display chromatin margination, but was accompanied by an enhanced level of autophagic and hydrolytic activity in which a cytosolic source of acid phosphatase became manifest in the extra-cisternal spaces. Normal and annexin-positive cells were found to occur in the younger nursing bees, whilst necrosis and an aberrant vacuolar type of apoptosis predominated in the older foraging bees. The relevance of these results to the classification of programmed cell death is discussed. (C) 2000 Academic Press.

The number of sensilla coeloconica, ampullacea and campaniformia of flagellomeres 3 to 10 of the antennae of workers of four honey bee types (Italian, Caucasian, African and Africanized) was studied by scanning electron microscopy. Comparisons of the four bee types showed that only African and Africanized honey bees did not differ from one another with respect to the number of sensilla coeloconica and ampullacea of flagellomere 10. African and Africanized honey bees and Caucasian and Italian honey bees also did not differ from one another in terms of flagellomere 9. In the other flagellomeres there were no differences among bee types. Italian and Caucasian honey bees differed from Africanized honey bees in terms of number of sensilla campaniformia on flagellomere 6, and Caucasian honey bees differed from African and Africanized honey bees in terms of flagellomere 3. Five significant but random correlation values were obtained between number of antennal sensilla and defensive behaviour in Africanized honey bees. Thus, there is no relationship between antennal structures and defence behaviour.

Cell death that occurs during ovary differentiation in the honeybee worker's larval development accounts for ovariole reabsorption. From a morphological standpoint, three modes of death were detected. Germinative cells in the ovarioles die by an apoptotic-like process, whereas the somatic cells die by an autophagic process, type 11 cell death; and during pupation, stromatic and ovarian capsular cells die through cytoplasmic disintegration, releasing their components into the hemolymph. These modes of cell death are in part determined by the pattern of tissue organization within which the cell occurs. (C) 2002 Elsevier B.V. Ltd. All rights reserved.

The Dufour's gland of Bombus terrestris workers, of different ages and with varying degrees of ovary development, was studied with the aim to verify its involvement in reproduction. Measurements of the diameter and the length of the gland were made using an ocular micrometer adapted to a microscope. Transmission electron microscopy was used to study the morphology of the secretory cells and to analyze the secretory cycle. The glandular cells were considered to be near type II cells of NOIROT and QUENNEDEY (1974), a type that has not been described before in Hymenopterans. The results show that there is a correlation between the degree of ovary development and Dufour's gland activity of workers. The diameter of the gland and the secretory cell activity increased with increasing oocyte size in the ovary. Regressive conditions of the gland were observed, which are probably related to increasing worker age. To elucidate the production and releasing process of the secretion and to establish its precise function, a comparative analysis of the secretion process of the Dufour's gland of queens and workers is needed.

Biochemical studies revealed that the activity of some hydrolytic enzymes from the venom glands of honey bee Apis mellifera was higher in workers of 14 days of age than in those of 40 days. Among these enzymes, the highest activity was recorded for acid phosphatase, which was cytochemically detected throughout the length of the secretory filament and surrounding the canaliculi of the distal region of the reservoir. The acid phosphatase was considered to be a typical secretion product, since it was present in the cytoplasm as well as in the canaliculi of the secretory cells. (c) 2009 Elsevier Ltd. This research was developed in the center-urban area at the city of Dourados (MS), in Tibouchina granulosa trees, a plant popularly known as quaresmeira in Brazil. The floral visitors of this species were recorded in three daily periods: 7 - 8 a.m., 1 - 2 p.m. and 4 - 5 p. m. Bee visitors of those plants were collected with an entomological net when they landed on the flowers. The bees were anesthetized in a closed camera containing etila acetate, conserved in Dietrich ' s fixative and, soon afterwards, transferred to 70% ethyl alcohol for subsequent identification. Almost 300 specimens of Africanized Apis mellifera, Trigona spinipes and Tetragonisca angustula were collected. The presence of these three species of bees in the flowers of T. granulosa suggests that they can be considered the main floral visitors of that vegetal species. T. angustula workers did not visit the flowers of T. granulosa between 7 and 8 a.m., but they were the main floral visitors from 4 to 5 p.m. The workers of T. spinipes presented a very aggressive behavior against the Africanized A. mellifera workers on the quaresmeira flowers, defending the food sources with strength and efficiency and provoking a drastic reduction in the number of honeybee visits between 1 and 2 p.m. Nevertheless, they did not interfere in the opportunistic activity of foraging by T. angustula workers, mainly between 4 and 5 p.m. The most important result of this research was the detection of the Competitive Exclusion Principle between A. mellifera and T. spinipes species in relation to the foraging behavior, when they exploit the floral resources of T. granulosa.

Dufour glands of Apis mellifera and Melipona bicolor were studied under light and transmission electron microscopy, using the cytochemical techniques of mercury bromophenol blue for protein detection, imidazole-buffered osmium tetroxide selective staining of unsaturated lipids, lanthanum nitrate for intercellular junction identification and zinc-iodide-osmium tetroxide for cytoplasmic endomembrane visualization. The results in both species corroborated the lipid nature of the gland secretion and showed in A. mellifera the poverty of the synthetic machinery in the worker gland cells in comparison with the queen, as expected by previous biochemical analyses. The pathway of the exogenous compounds of the secretion is intracellular, since substances can penetrate the cell folds and intercellular junctions, but their access to the, gland lumen is barred by the apical intercellular junctions.
