A citometria de fluxo é um método emergente na medicina veterinária que permite a identificação e quantificação de células em suspensão. Apesar do custo elevado e da necessidade de técnicos especializados para realização da avaliação citofluorométrica, esta técnica tem uma ampla aplicação na hematologia veterinária, incluindo a avaliação de células-tronco hematopoiéticas, eritrócitos, leucócitos e plaquetas. O grande potencial de aplicação clínica da citometria de fluxo inclui a quantificação das células CD34+ como indicação da capacidade de reconstituição hematopoiética após o transplante de células-tronco e das subpopulações linfocitárias para avaliação da resposta imune frente aos transplantes e às doenças que acometem os animais. Projetos científicos sobre a avaliação citofluorométrica das células-tronco e subpopulações linfocitárias de cães têm sido desenvolvidos com sucesso por pesquisadores da área de hematologia veterinária na Universidade Estadual Paulista (UNESP), Campus de Jaboticabal. Portanto, no futuro, a citometria de fluxo deve se tornar uma técnica de rotina em laboratórios veterinários no Brasil.

As células-tronco hematopoéticas promovem a reconstituição hematopoética e de outros tecidos, estando presentes no embrião, sangue periférico, medula óssea e sangue do cordão umbilical. Os modelos experimentais de células-tronco em cães têm propiciado informações relevantes para transplantes de células-tronco em humanos. A capacidade de reconstituição hematopoética e da plasticidade das células-tronco de cães permite o emprego do modelo canino em várias propostas científicas e terapêuticas, que propiciam informações pré-clínicas ao homem. O objetivo desta revisão bibliográfica é relatar a importância das células-tronco hematopoéticas de cães, sendo que a sua principal aplicação clínica é o transplante das células-tronco.

Lymphoma represents the most prevalent hematopoietic malignancy in small animal medicine. It is highly responsible to chemotherapy and therefore several protocols are used as therapeutic tools. For that reason, the bone marrow transplantation, enshrined in human medicine through initial trial in canine patients, has increasingly become the focus of studies in order to make it a reality also in veterinary medicine. First, the treatment with the chosen chemotherapy protocol is made. As complete remission of lymphoma is observed, it must be initiated the bone marrow harvesting. The obtained material is subjected to the processes of erythrocyte depletion, plasma depletion, cryoprotectants addition, total nuclear cells counting, hematopoietic progenitor quantification, analysis of cell viability and freezing. Following that, with radiotherapy or application of cyclophosphamide, the conditioning phase of the patient who is receiving the transplantation is carried out. The bags containing hematopoietic stem cells are then thawed and transplanted into the receptor organism. Support with hematopoietic stem cells allows the use of lethal doses of chemotherapy or radiotherapy and has been shown to considerably raise the disease remission time and survival rate of the canine patients

Introdução: As exigências nutricionais para o paciente transplantado são conhecidas, mas há poucas evidências sobre o consumo alimentar e o seu efeito no estado nutricional. O objetivo deste estudo é avaliar o efeito do transplante de células-tronco hematopoéticas (TCTH) no estado nutricional de pacientes orientados e monitorados quanto ao consumo alimentar durante os primeiros 100 dias do procedimento. Método: Participaram do estudos 27 indivíduos que realizaram TCTH, com idade entre 20 e 60 anos, 13 mulheres e 14 homens, índice de massa corporal médio de 27+/- 5kg/m². Os pacientes receberam orientações nutricionais individualizadas quanto ao consumo de alimentos. Nos momentos pré-TCTH, D+30, D_60 e D+100 (D+n = dias decorridos do TCTH) foram monitorados o consumo de alimentos, a partir de recordatório do consumo de 24 horas e o estado nutricional a partir dos indicadores antropométricos. Resultados: Após 100 dias do transplante, foi constatado que os 27 indivíduos analisados mantiveram estáveis o consumo alimentar e o estado nutricional. Conclusão: A orientação nutricional proporcionou a preservação do estado nutricional após o TCTH, a qual se deu à custa de incremento no consumo de energia e nutrientes oriundos de suplementos alimentares e também de alimentos não tão saudáveis.

In this work we have:investigated the growth and differentiation of bone marrow stem cells in mice bearing Ehrlich ascites tumor-and treated with three dose-regimens of Dicyclopentadienyldichlorotitanium (IV) (DDCT). We also: studied the presence of colony stimulating factors In the serum of PDCT-treated animals as well-as the effects-of the drug on the survival of the tumor-bearing mice. The-results demonstrated that the myelosuppression developed in the tumor-bearing animals is prevented by the administration:of 1, 2 or 3 doses of 15 mg/kg DDCT. In the treatment with three doses, however, 23 % of the animals died. Moreover, DDCT treatment in normal animals resulted in increased numbers of CFU-GM. We observed the presence of stimulating factors in the serum of drug-treated animals which induced the growth and differentiation of bone marrow progenitor cells from normal animals in vitro. on the other hand, in vitro addition of the drug to these cultures had no effect. Thus, we conclude that the drug protects against the myelosuppression induced by the tumor and that this protection may be related to an indirect action of the drug. Tendinitis is an important disease that leads to lameness and decreased performance in equine athletes and results in high costs associated with therapy due to a long recovery period and a high rate of recurrence. Although, several treatments for equine tendinitis have been described, few are effective in significantly improving the quality of the extracellular matrix and reducing the rate of recurrence. The use of cell therapy with mesenchymal stem cells (MSCs) derived from various sources has received much attention because of its therapeutic potential for equine tendinitis. In this paper, we review patents on stem cell therapy and the specific use of MSCs for the treatment of equine tendinitis. © 2013 Bentham Science Publishers.

Colorectal cancer (CRC) is a disease whose genesis may include metabolic dysregulation. Cancer stem cells are attractive targets for therapeutic interventions since their aberrant expansion may underlie tumor initiation, progression, and recurrence. To investigate the actions of metabolic regulators on cancer stem cell-like cells (CSC) in CRC, we determined the effects of soybean-derived bioactive molecules and the anti-diabetes drug metformin (MET), alone and together, on the growth, survival, and frequency of CSC in human HCT116 cells. Effects of MET (60 μM) and soybean components genistein (Gen, 2 μM), lunasin (Lun, 2 μM), β-conglycinin (β-con, 3 μM), and glycinin (Gly, 3 μM) on HCT116 cell proliferation, apoptosis, and mRNA/protein expression and on the frequency of the CSC CD133(+)CD44(+) subpopulation by colonosphere assay and fluorescence-activated cell sorting/flow cytometry were evaluated. MET, Gen, and Lun, individually and together, inhibited HCT116 viability and colonosphere formation and, conversely, enhanced HCT116 apoptosis. Reductions in frequency of the CSC CD133(+)CD44(+) subpopulation with MET, Gen, and Lun were found to be associated with increased PTEN and reduced FASN expression. In cells under a hyperinsulinemic state mimicking metabolic dysregulation and without and with added PTEN-specific inhibitor SF1670, colonosphere formation and frequency of the CD133(+)CD44(+) subpopulation were decreased by MET, Lun and Gen, alone and when combined. Moreover, MET + Lun + Gen co-treatment increased the pro-apoptotic and CD133(+)CD44(+)-inhibitory efficacy of 5-fluorouracil under hyperinsulinemic conditions. Results identify molecular networks shared by MET and bioavailable soy food components, which potentially may be harnessed to increase drug efficacy in diabetic and non-diabetic patients with CRC.

The graft-versus-host disease is the major cause of morbidity and mortality in patients who have undergone hematopoietic stem cell transplantation. Aiming at contributing to the understanding of the role of myeloid and plasmacytoid dendritic cells, and natural killer cells in chronic graft-versus-host disease, we examined biopsies of jugal mucosa of 26 patients with acute myeloid leukemia who had undergone allogenic hematopoietic stem cell transplantation. Half of these patients developed oral chronic graft-versus-host disease. Microscopic sections were immunohistochemically stained for anti-CD1a, anti-CD123 and anti-CD56. We calculated the number of immunostained cells in the corium per square millimeter and applied the Mann-Whitney test. Results showed a statistically significant increase of myeloid dendritic cells (CD1a+; p=0,02) and natural killer cells (CD56; p=0,04) in patients with oral chronic graft-versus-host disease. CD123 immunostaining showed no statistical difference between groups. It was concluded that myeloid dendritic cells and natural killer cells participate in the development of oral chronic graft-versus-host disease.
