In a machine milking system in which calves were not present, oxytocin concentration (OT), cortisol release (CORT), milk yield and residual milk were evaluated for cows of three genetic groups: 1/2Gir x 1/2Holstein (n=6, genetic group F2), 1/4Gir x 3/4Holstein (n=6, genetic group F3) and Holstein (n=6, genetic group H). Group H had higher milk yield than groups F2 and F3, whereas OT was similar among groups. The increase in OT during milk-ing was greater for H and F3 than for F2. Residual milk for F2 was' higher than for F3 and H. The CORT for F2 was higher than for cows of the other two genetic groups. Cows from F2 and F3 were more stressed than H cows during machine milking, but the Gir x Holstein groups did release suffficient OT to induce an effective milk letdown response.

An ultrasonic density-measurement cell is experimentally characterized as a function of temperature. The measurement of propagation velocity and density of several liquids is performed in the 15 degrees C-40 degrees C temperature range. Results are compared to the tabulated values in the case of distilled water, showing an accuracy of 0.07% for the propagation velocity. The cell was tested with distilled water, alcohol, and homogenized milk, and density values are compared to those obtained with a pycnometer, showing 0.2% accuracy in density measurement for stabilized temperature and 0.4% accuracy under thermal gradient conditions.

Corn plants were ensiled at the milk, milk-early dough, medium dough, and semi-hard dough stages. The intake and digestibility trials showed that: a) silage DM and NFE contents increased and ADF, cellulose, and lignin contents decreased with plant maturity; b) corn plant maturity had little effect on the digestibility of the silages, but the milk and medium dough stages produced the best results; c) no consistent effect of maturity was observed in DM and CP daily intakes (mean values 41.86 and 26.77/g/kg(75), respectively). Nevertheless, daily digestible dry matter and digestible protein intakes (g/kg(75)) were higher with medium-dough grains (31.74 and 1.97g, respectively) than with the milk (23.27 and 1.45g, respectively) and the milk-early dough stages (22.60 and 1.28g, respectively). However, similar intakes were observed with the medium-dough and semi-hard dough stages. Total digestible nutrient contents of the silages were affected little by corn plant maturity. Nevertheless, statistical differences were Found between the medium dough (70.09%) and the milk-early dough stages (63.07%).

The effect of milk processing on the microstructure of probiotic low-fat yogurt was studied. Skim milk fortified with skim milk powder was subjected to three treatments prior to innoculation: thermal treatment at 85 degrees C for 30 min, high hydrostatic pressure at 676 MPa for 5 min, and combined treatments of high hydrostatic pressure (HHP) and heat. The processed milk was then fermented by using two different starter cultures containing Streptococcus thermophilus, Lactobacillus delbrueckii ssp. bulgaricus, Lactobacillus acidophilus, and Bifidobacterium longum. The microstructure of heat-treated milk yogurt had fewer interconnected chains of irregularly shaped casein micelles, forming a network that enclosed the void spaces. on the other hand, microstructure of HHP yogurt had more interconnected clusters of densely aggregated protein of reduced particle size, with an appearance more spherical in shape, exhibiting a smoother more regular surface and presenting more uniform size distribution. The combined HHP and heat milk treatments led to compact yogurt gels with increasingly larger casein micelle clusters interspaced by void spaces, and exhibited a high degree of cross-linking. The rounded micelles tended to fuse and form small irregular aggregates in association with clumps of dense amorphous material, which resulted in improved gel texture and viscosity. The experiment was carried out to evaluate the performances of cross breed Saanen dairy goats submitted to two strategies of supplementation with concentrate 30% of daily requirements in net energy (NRC, 1981) supplied by concentrate, from third week until the end of lactation (ESC. 1) or 60% of requirements from the 3rd to the 13th week of lactation and 15% from 14th until the end of lactation (ESC.2); and evaluate the animal response to these strategies of supplementation with concentrate in a semi-confined or pasture systems. Thirty one animals were used in a completely randomized design. The system of production affected the total intake of concentrate and total milk production; at pasture system the values for these two variables were higher. The supplementation with concentrate affected the total intake of concentrate and total production of milk and the relation of milk/consumption of concentrate. The highest value for relation of milk/consumption of concentrate and the least for the total consumption of concentrate were obtained at ESC. 1. Body weight affected of the system of production, resulting in a higher value to the system of pasture. Weight and body condition had different responses to he supplementation with concentrate according to the system of production. The semi-confined system ESC.2 resulted in a higher value to the body condition; the body weight was no affected. At pasture system the highest values for weight and body condition were obtained in the supplementation with concentrate ESC. 1. Reproductive parameters did not affect supplementation with concentrate and system of production.

Caseins comprise make up about 80% of the total protein content of milk and present polymorphism with change in the amino acid sequence. Within this abundance of proteins, kappa-casein is noteworthy, since it has been associated with differences in milk yield, composition and processing. The objective of this study was to observe the existence of polymorphism in the kappa-casein gene in female buffaloes. For this purpose, blood samples from 115 female buffaloes, collected with vacutainer by needle punctionure of the jugular vein, were used. for genomic DNA extraction was done from blood samples. The PCR-RFLP and SSCP techniques demonstrated that the studied animals were monomorphic for the kappa-casein gene. Only allele B was observed in these animals, which was present in homozygosis. Therefore, it was not possible to quantify the gene action on milk yield and its constituents. The monomorphism observed in the population studied would allow the development of a method to identify mixtures of cow and buffalo milk in mozzarella cheese production, especially because, in cattle, the kappa-casein gene is polymorphic. Mastitis is an inflammatory process in the udder that can affect the quality and quantity of milk produced causing economic losses and risks for health. Considering the somatic cell count (SCCs) as indicator of udder health and the milk yield (MY) of buffaloes from São Paulo State, this study aimed to quantify the related losses in milk due to somatic cells count (SCC). 9404 sources of information from 2198 lactations that occurred between 1997 and 2004 were analysed. There was no relation between MY and the SCCs in the buffaloes at first parity. For the second parity in the months 1, 2, 5, 6 and 7 of lactation, there was a negative and significant relationship between SCCs and MY. For parities of three or more there was a significant and negative regression coefficient during every month of lactation betrween MY and SCC. The average losses varied from 0,18 to 2,2 milk liters per unit of SCCs. The results indicated large losses observed in the miltiparous buffaloes and that this category needs received special attention in terms of udder health. The effect of farm, parity and year, must be considered comparison between animals.

CAE is caused by a lentivirus. The animals are mainly infected when taking contaminated colostrums and/or milk. This study proposed a CAE control strategy without sacrificing contaminated mothers. Thirty-nine female kids, born to CAE seropositive mothers were isolated from their mothers at birth and fed heat-treated colostrums and pasteurized milk from seronegative goats up to two months of age. All kids were submitted to three-monthly serological tests from birth to 12 months; seropositives were segregated from the herd. The control group consisted of 12 kids born to seropositive mothers that remained with their mothers. Diagnosis was the same, but seropositve animals were not segregated. At the end of 12 months, 34 (87%) animals from the experimental group remained seronegative with 76% to 98% confidence limits; in control group animals, the accumulated negativity rate was 17%, with 0% and 38% confidence limits. These results show that the proposed plan is viable to assure disease control in contaminated herds and that without it contamination can pass to animals born to infected goats.

Aiming to compare milk yield, residual milk and behavioral responses of Holstein cows during milking, fourteen experimental cows were mechanically milked twice a day, and evaluated during the first six weeks of lactation. Milk yield was measured daily, and residual milk volume weekly. Body (ribs region), udder and teat temperatures and respiratory frequency were measured twice a week. Animal behavior at the milking parlor was registered using ethograms and interviews with the milkers. During the first weeks of lactation, a higher percentage of kicks, over-steps, milking cups being knocked down, urinations and vocalizations in primiparous cows (p < 0.05) was observed, which were also considered more reactive by the parlors compared to multiparous cows. Multiparous cows produced more milk than primiparous (p < 0.05). However, there were no significant differences in residual milk percentage (p > 0.05), which suggests adaptation of both groups to the milking environment.

PURPOSE: to investigate the effect of cigarette smoke exposure on body and tissue weight gain, serum parameters and milk yield during pregnancy and lactation in rats, and the impact on offspring from birth toil young adulthood. METHODS: 40 Wistar pregnant rats were randomly divided into: CG - not exposed to cigarette smoke and sacrificed at the end of pregnancy; CL - not exposed to cigarette smoke and sacrificed at the end of lactation; FG - exposed to cigarette smoke and sacrificed at the end of pregnancy; FL - exposed to cigarette smoke and sacrificed at the end of lactation. The offspring were separated by gender and divided according to their mothers' groups. Tissue weight, body weight and serum parameters were evaluated in rats and offspring. Milk yield per pup was calculated. RESULTS: body weight was decreased in FL during lactation (CL=267.0±7.2; FL=235.5±7.2 g*, *p<0.05). Adipose tissue was not detected in the CL and FL groups, and was reduced in FG compared to CG (CG=3.3±0.3; FG=2.4±0.3 g*, *p<0.05). Rats exposed to cigarette smoke had higher blood glucose levels (CG=113±17, CL=86±16, FG=177±21*, FL=178±23 mg/dL*, *p<0.05 CG versus FG e CL versus FL), CL and FL groups presented lower HDL-cholesterol with no change in total cholesterol. Finally, rats exposed to cigarette smoke had lower milk yield compared to unexposed rats (CL=6.7±0.4, FL=5.4±0.3 g*, *p<0.05). In offspring from the FG and FL groups, there was a decrease of body weight from birth to young adulthood, with no changes in gastrocnemius, liver or heart weights in any group, and adipose tissue was no detected in female offspring. There was an increase in blood glucose in offspring of both sexes from rats exposed to cigarette smoke (males: Pcg=107±10.5, Pcl=115±8.6, Pfg=148±16.8*, Pfl=172±11.2**; females: Pcg=109±27.2, Pcl=104±9.7, Pfg=134±20.0*, Pfl=126±13.3**; p<0.05 *Pcg versus Pfg and **Pcl versus Pfl). CONCLUSIONS: exposure to cigarette smoke provokes impairment of morphometric and serum parameters during pregnancy and lactation both in mothers and offspring, which is maintained during young adulthood.

Bovine mastitis is considered important disease causing major economic losses in dairy herds. Antimicrobial drug can promote resistance, chemical residues in food and environmental contamination. The purpose of this study was to evaluate the antibacterial activity in vitro of pomegranate extract on Staphylococcus aureus isolated from bovine milk, evaluate its antioxidant activity, and quantify levels of total phenols and fravonoides of the different extracts used. Aqueous extracts were used in nature and dry, from the peel of the fruit (EAC) and leaves (EAF). Additionally, it was evaluated the antioxidant activity (AA%), total phenols and flavonoids. Milk samples were inoculated, incubated, and the colonies were characterized as Staphylococcus aureus were adjusted to 1.0x106 UFC/mL in the 6 standard of the MacFarland scale. The sensitivity of the microbial isolates were determined in quintuplicate, in disk diffusion test. The minimum inhibitory concentration was determined by visible inhibition zone greater than 15mm. The results were evaluated by ANOVA, Tukey 5%, using the program SISVAR 5.3 - DEX/UFLA. Results implied that the aqueous extract from the bark of dried fruit was capable of inhibiting bacterial growth at concentrations of 3%. The other treatments only showed this activity, from the concentrations of 15%, 20% and 30% for dry EAF, in nature EAC and in nature EAF, respectively. Regarding the action antioxidant of dry EAC, was not correlated with total phenols and flavonoids. Probably other alkaloids substances present in the extract studied, may have been responsible for this activity. It is concluded that extracts of Punica granatum L., especially those obtained by the shell of the fruit, showed inhibitory activity against S. aureus, indicating your use potential for the control of bovine mastitis.

This study was conducted with the objective to evaluate the duration of exposure of animals to the evaporative cooling system in the waiting barn on the weather variables (temperature and relative humidity), animal behavior, production and quality aspects of milk from Girolando cows. Sixteen multiparous lactating cows were used having a genetic composition of 7/8 Holstein-Gir and mean milk production of 18 kg animal-1day-1. A 4 × 4 Latin square design was used. The experiment was conducted during the summer season (February-March 2009) and lasted 56 days. The treatments consisted of different periods of 10, 20 and 30 minutes of exposure to acclimatization and control. Time of 30 minutes of exposure to thermal conditioning system enabled the second milking (evening shift), providing an increase in milk production in the next milking of 0.640 kg animal-1. A higher percentage of time spent on food intake and rumination (17.31 and 30.19%) in 30 minutes treatment, compared to control (15.50 and 27.37%), respectively. There was no significant effect of treatments in the composition of milk.
