The transit time of seeds ingested by frugivorous animals has important implications for the spatial distribution of seeds and their dispersal distance. Nevertheless, this parameter is rarely included in seed dispersal studies. In this paper, we provide information about the transit time of seeds of nine species of plants ingested by individuals of six species of captive birds (Turdus albicollis, T. amaurochalinus, T. leucomelas, T. rufiventris, Stephanophorus diadematus and Saltator similis). We found that (1) seeds are regurgitated quickly through the digestive tract of birds than defecated seeds, and (2) large seeds (e.g., > 5 mm in diameter for Turdus spp.) are regurgitated rather than defecated. These results corroborate other studies, but the relationship between seed size and transit time seems to be quite complex and variable, requiring more detailed studies on this important aspect of the ecology of seed dispersal and digestive physiology of frugivorous birds.

As sementes da palmeira juçara (Euterpe edulis Mart.) são recalcitrantes, apresentando baixa longevidade e sensibilidade à desidratação e ao armazenamento em temperaturas baixas. Neste trabalho foram estudadas condições de temperatura mais adequadas ao armazenamento temporário destas sementes com e sem a polpa. Frutos maduros foram colhidos em 24 plantas provenientes da coleção de palmeiras do Instituto Agronômico (IAC) localizada em Ubatuba, estado de São Paulo, e encaminhadas, em embalagem impermeável, à Faculdade de Ciências Agronômicas da UNESP, Campus de Botucatu (SP). Metade dos frutos foi despolpada e outra metade foi mantida com polpa, sendo ambas armazenadas em sacos fechados de polietileno (20 µm de espessura) mantidos em temperaturas de 5; 10; 15 e 20-30ºC. Amostras para os testes de qualidade foram retiradas aos 0; 3; 6; 9 e 12 dias após a colheita dos frutos. As sementes armazenadas com polpa foram despolpadas imediatamente antes da instalação dos testes. Foram avaliados o grau de umidade das sementes, porcentagem de germinação, comprimento e matéria seca das plântulas. Os resultados mostraram que há efeito positivo de pós-amadurecimento em sementes de Euterpe edulis. Um período de armazenamento de 9 a 12 dias, após a colheita e antes da semeadura, favoreceu a germinação e o vigor das sementes de juçara. Os efeitos foram maiores para sementes armazenadas sem polpa do que com polpa. Temperaturas na faixa de 5 a 20-30ºC são igualmente adequadas para o armazenamento temporário de sementes sem polpa. No entanto, para sementes com polpa, a temperatura de armazenamento não deve exceder a 20ºC, visto que um decréscimo na germinação e no vigor e um acréscimo no número de botões germinativos apodrecidos e sementes mortas foram observados na temperatura de 20-30ºC.

A falta de sincronismo de florescimento entre as linhagens auto incompatíveis em um campo de produção de sementes híbridas de couve flor pode além de reduzir a produção de sementes comprometer a pureza genética das mesmas. Com o objetivo de estudar o efeito da coincidência de florescimento entre linhagens de couve-flor na produtividade e qualidade de sementes híbridas, foi realizado o presente experimento. Os tratamentos consistiram em seis diferentes épocas de semeadura, espaçadas a cada quinze dias, de uma linhagem de verão auto-incompatível que foi polinizada por uma linhagem de inverno que não apresenta auto-incompatibilidade. Observou-se a coincidência do florescimento das diferentes épocas de semeadura com a linhagem polinizadora. Foram avaliadas as seguintes características: área foliar média, número de flores por planta, número de síliqüas por planta, número de sementes por planta (peso e número), peso médio de 1000 sementes e foi determinado o número de sementes por síliqüa. Foi realizado ainda, o teste padrão de germinação e determinada a pureza genética das sementes para cada tratamento. A coincidência da época de florescimento entre as linhagens de couve-flor afetou diretamente a produtividade e a qualidade genética das sementes híbridas produzidas, sendo que, quanto maior foi o nível de coincidência, maior foi o número de sementes formadas por síliqüa e menor a percentagem de sementes contaminantes. Entretanto, não teve influência na qualidade fisiológica das mesmas.

A lectin-like protein from the seeds of Acacia farnesiana was isolated from the albumin fraction, characterized, and sequenced by tandem mass spectrometry. The albumin fraction was extracted with 0.5 M NaCl, and the lectin-like protein of A. farnesiana (AFAL) was purified by ion-exchange chromatography (Mono-Q) followed by chromatofocusing. AFAL agglutinated rabbit erythrocytes and did not agglutinate human ABO erythrocytes either native or treated with proteolytic enzymes. In sodium dodecyl sulfate gel electrophoresis under reducing and nonreducing conditions, AFAL separated into two bands with a subunit molecular mass of 35 and 50 kDa. The homogeneity of purified protein was confirmed by chromatofocusing with a pI=4.0+/-0.5. Molecular exclusion chromatography confirmed time-dependent oligomerization in AFAL, in accordance with mass spectrometry analysis, which confers an alteration in AFAL affinity for chitin. The protein sequence was obtained by a liquid chromatography quadrupole time-of-flight experiment and showed that AFAL has 68% and 63% sequence similarity with lectins of Phaseolus vulgaris and Dolichos biflorus, respectively.

Parkia platycephala lectin 2 was purified from Parkia platycephala (Leguminosae, Mimosoideae) seeds by affinity chromatography and RP-HPLC. Equilibrium sedimentation and MS showed that Parkia platycephala lectin 2 is a nonglycosylated monomeric protein of molecular mass 29 407 ± 15 Da, which contains six cysteine residues engaged in the formation of three intramolecular disulfide bonds. Parkia platycephala lectin 2 agglutinated rabbit erythrocytes, and this activity was specifically inhibited by N-acetylglucosamine. In addition, Parkia platycephala lectin 2 hydrolyzed β(1-4) glycosidic bonds linking 2-acetoamido-2-deoxy-β-d-glucopyranose units in chitin. The full-length amino acid sequence of Parkia platycephala lectin 2, determined by N-terminal sequencing and cDNA cloning, and its three-dimensional structure, established by X-ray crystallography at 1.75 Å resolution, showed that Parkia platycephala lectin 2 is homologous to endochitinases of the glycosyl hydrolase family 18, which share the (βα) 8 barrel topology harboring the catalytic residues Asp125, Glu127, and Tyr182. © 2006 The Authors.

Magnolia ovata seeds have been reported as desiccation sensitive. In order to test if the drying rate would affect the assessment of storage behaviour of these seeds, the effect of different drying rates and storage times on the viability was tested. Seeds were dried over activated silica gel (fast drying) or salt solutions for different periods (slow drying) and stored at -20°C. Partial drying transiently increased the final germination and the germination speed index, but further drying resulted in reduction of these parameters. Drying rate affected the final germination and vigour. Seeds that were slow-dried to 0.10 g H 2O ̇ g -1 dw retained high viability when compared with seeds desiccated to the same water content level by the fast drying method, although their vigour was reduced. Only slow-dried seeds could be stored at -20°C for 90 d without reduction of viability. These data suggested that the storage behaviour of seeds of M. ovata seeds should be classified as intermediate.

Annona emarginata (Schldtl.) H. Rainer (araticum-de-terra-fria) is used as a rootstock for several species of Annonaceae. It is suggested that these seeds should be sown immediately after extraction and, therefore, they could be intolerant to desiccation. There are several mechanisms involved with desiccation tolerance. Soluble sugars, for example, can accumulate and act as osmoprotectants for the membrane system during desiccation. The aim of this study is to assess desiccation tolerance in A. emarginata seeds. In addition, we examined the soluble sugars involved in desiccation tolerance. Finally, we determined the effect of gibberellic acid (GA4+7) and N-(phenylmethyl)-aminopurine in promoting the germination of seeds with different water contents. The experiment consisted of a randomized 4×5 factorial design (desiccation levels × concentration of growth regulators). After drying, seeds containing 31 (control), 19, 12 and 5% water were incubated in different concentrations of GA4+7 N-(phenylmethyl)-aminopurine (0, 250, 500, 750 and 1000 mg L-1) for 60 hours. The experiment was conducted in a germination chamber with alternating temperature and photoperiod of 20oC for 18 hours of darkness and 30oC for 6 hours of light. We analyzed electrical conductivity, germination rate, mean germination time, germination speed, frequency and uniformity of germination, percentage of dormant seeds and soluble sugar profile in intact seeds through high-performance liquid chromatography (HPLC). The data were subjected to analysis of variance, and the means were compared using Tukey's test at a threshold of p<0.05. The results showed that seeds of A. emarginata appears to be desiccation tolerant and, also, that sucrose increases when seed water content is reduced to values as low as 12%, exogenous GA4+7+N-(phenylmethyl)-aminopurine improves its germinability.

A protein extract containing a plant lipase from oleaginous seeds of Pachira aquatica was tested using soybean oil, wastewater from a poultry processing plant, and beef fat particles as substrate. The hydrolysis experiments were carried out at a temperature of 40°C, an incubation time of 90 minutes, and pH 8.0-9.0. The enzyme had the best stability at pH 9.0 and showed good stability in the alkaline range. It was found that P. aquatica lipase was stable in the presence of some commercial laundry detergent formulations, and it retained full activity up to 0.35% in hydrogen peroxide, despite losing activity at higher concentrations. Concerning wastewater, the lipase increased free fatty acids release by 7.4 times and promoted the hydrolysis of approximately 10% of the fats, suggesting that it could be included in a pretreatment stage, especially for vegetable oil degradation.

The external morphology of seeds and post-germination developmental stages of Angelonia salicariifolia Bonpl. (Scrophulariaceae) were investigated using scanning electron microscopy. Some structural features of the seed exotesta and seedling in Angelonia are presented for the first time and are of potential taxonomic value for this neotropical genus. The seeds are very small (0.9-1.7 mm long and 0.5-0.9 mm wide), ovate, with a reticulate-crested exotesta, reticules arranged uniformly in longitudinal rows, with a high density of microcilia-like projections on the cell wall of the reticule base and on the edge of the crests. The hilum is located beside the micropyle at the narrow end of the seed. Germination is epigeal. During germination the radicle develops, followed by elongation of the hypocotyl and primary root. At this stage dense root hairs develop on the lower part of the hypocotyl. The apical bud-located between the cotyledons-begins to develop after the cotyledons have unfolded. The cotyledons are equal in size, sessile and ovate. The seedlings have two types of trichomes, one characteristic of the cotyledons and first pair of leaves (glandular, sessile, four-celled head with quadrangular shape) and the other characteristic of the hypocotyl and epicotyl (stalked, erect, elongate and three-celled with dome-shaped unicellular head). Patógenos em sementes de milho (Zea mays) causam sérios problemas, como a perda de sua capacidade germinativa. O objetivo do trabalho foi determinar qual o melhor tempo para infecção das sementes de milho com Fusarium graminearum, para posterior avaliação dos danos causados pelo fungo na germinação e vigor das mesmas. As sementes foram colocadas sobre meio de BDA contendo o patógeno e incubadas por 4, 8, 16 e 32 h. Após os respectivos períodos de incubação, estas foram submetidas ao teste de sanidade (papel de filtro), com duas variações, sem e com assepsia superficial, usando hipoclorito de sódio a 1% de cloro ativo, por 3 min. Determinado o melhor tempo para infecção, outras sementes foram infetadas com o patógeno, para realização dos testes de germinação e vigor (envelhecimento acelerado e teste de frio) com uma mistura de sementes sadias (colocadas sobre o meio BDA) e sementes inoculadas, resultando em 0, 20, 40, 60, 80 e 100% de sementes infetadas com o fungo em estudo. Os resultados obtidos mostraram que o período de incubação de 32 h foi suficiente para se obter sementes infetadas. Com relação à germinação, não houve diferenças significativas entre os diferentes níveis de infecção, provavelmente devido ao alto vigor das sementes de milho testadas. Quanto aos testes de vigor, os níveis de infecção diferiram significativamente da testemunha, apesar de não terem diferido entre si.

The amount of solar energy made available for the production of a sabid seed varied as a function of the time of the year, the face of the plant in which, the position in the plant on which and the position in the pod in which it was produced.Variation in solar energy availability as a consequence of the time of the year was a direct consequence of latitude. At 21degrees5'22 S the highest amounts of Global Solar Radiation (GSR) reaching the site where the experiment was conducted took place during the months from November through February. During these months there were no marked differences between any two of the amounts of GSR reaching faces North (N), South (S), West (W) East (E). From February through November (period during which the sabid plants of this study flowered and the resulting seeds matured and were harvested) the total GSR's were the lowest and marked differences were found between faces N and S, with face N receiving much more GSR than face S. During that period, faces W and E received practically the same amount of GSR and it was much less than that received by face N and much more than the one received by face S.The amount of biological energy made available for the development of a seed seemed also to vary according to a dry matter partitioning strategy by the plant -the central third of the plant seemed to be the one receiving the highest amounts of energy, followed either by the upper or the lower third of the plant- it was not very clear which third of the plant immediately followed the central one. The partitioning of biological energy at the pod level also seemed to follow a strategy by which the central seeds would be the ones to receive more, followed by the proximal seeds and these by the distal ones.This availability of energy seemed to have a direct effect on seed size, weight and on the percentage of seeds which showed a degree of dormancy deep enough to prevent their germinating under the conditions of a standard germination test.The implications of these results for the improvement of methods for the overcoming of dormancy of sabia seeds are discussed.
