Aim: The purpose of this in vivo study was to compare the effectiveness of a new light cured resin based dicalcium/tricalcium silicate pulp capping material (TheraCal LC, Bisco), pure Portland cement, resin based calcium hydroxide or glass ionomer in the healing of bacterially contaminated primate pulps. Study design: The experiment required four primates each having 12 teeth prepared with buccal penetrations into the pulpal tissues with an exposure of approximately 1.0 mm. The exposed pulps of the primate teeth were covered with cotton pellets soaked in a bacterial mixture consisting of microorganisms normally found in human pulpal abscesses. After removal of the pellet, hemostasis was obtained and the pulp capping agents applied. The light cured resin based pulp capping material (TheraCal LC) was applied to the pulpal tissue of twelve teeth with a needle tip syringe and light cured for 15 seconds. Pure Portland cement mixed with a 2% Chlorhexidine solution was placed on the exposed pulpal tissues of another twelve teeth. Twelve additional teeth had a base of GIC applied (Triage, Fuji VII GC America) and another twelve had a pulp cap with VLC DYCAL (Dentsply), a light cured calcium hydroxide resin based material. The pulp capping bases were then covered with a RMGI (Fuji II LC GC America). The tissue samples were collected at 4 weeks. The samples were deminerilized, sectioned, stained and histologically graded. Results: There were no statistically significant differences between the groups in regard to pulpal inflammation (H= 0.679, P=1.00). However, both the Portland cement and light cured TheraCal LC groups had significantly more frequent hard tissue bridge formation at 28 days than the GIC and VLC Dycal groups (H= 11.989, P=0.009). The measured thickness of the hard tissue bridges with the pure Portland and light cured TheraCal LC groups were statistically greater than that of the other two groups (H= 15.849, P=0.002). In addition, the occurrence of pulpal necrosis was greater with the GIC group than the others. Four premolars, one each treated according to the protocols were analyzed with a microCT machine. The premolar treated with the light cured TheraCal LC demonstrated a complete hard tissue bridge. The premolar treated with the GIC did not show a complete hard tissue bridge while the premolar treated with VLC Dycal had an incomplete bridge. The pure Portland with Chlorhexidine mixture created extensive hard tissue bridging. Conclusion: TheraCal LC applied to primate pulps created dentin bridges and mild inflammation acceptable for pulp capping.

The uric gout has been often diagnosed in cockatiel, birds that has been popularized and frequently shown in the avian practice. This disease is a consequence of renal failure. In the Wildlife Clinic, a specialized facility in Sao Paulo, 15 cockatiel presenting unspecific signs of polyuria and polidipsia were followed and diagnosed as uric gout. These animals had as common feature being lutin breed and fed with commercial pellet diet. In the present work nutritional aspects and causes of this disease are discussed. The main application of laboratory tests in avian practice and uric gout treatment were exemplified

Malformations and possible damages to the urogenital system can be originated in the embryonic period. Moreover, fire guns, knives and accidents, where there is the disruption of the urethra, also cause these lesions. The objective was to analyze the contribution of tissue engineering in the construction of neo-urethra, developed by bioengineering. We performed an urothelial ex vivo expansion of cells in 3D scaffolds (platelet gel matrix and acellular porcine aorta) to assess the contribution of this technique in the construction of a neo-urethra. Mechanical dissociation was made of the inner wall of 10 North Folk rabbit’s bladder, weighing 2.5 to 3.0 kg. After dissociation the cell content was centrifuged and obtained a pellet of urothelial cells. The pellet was ressuspended in culture medium DMEM F12 and cells were maintained in culture for 15 days. Immunohistochemical analysis characterized the urothelial culture. The cells were then implanted in the scaffold - platelet gel. In a second experiment using aortic porcine acellular matrix were implanted urothelial cells alone and urothelial cells on platelet gel, on the inner wall of the scaffold - aorta, with space for setting bordered by a urethral probe. The complex probe - cells - aorta and probe - cells in platelet gel - aorta, were sealed with suture material and culture were maintained in a humidified 37ºC incubator with 5% CO2 in air for 12 days to subsequent histological analysis of urothelium cell adhesion to the scaffolds. By observation under an optical microscope, we could see the growth of cells in the scaffold platelet gel, from a monolayer in to a three-dimensional structure. In the acellular porcine aortic matrix containing the platelet gel, we could observe a few quantity of urothelial cells adhered. However with the acellular porcine aortic matrix in which was implanted only the urothelial cells, we have obtained adhesion to the wall

Stem cells are defined as cells capable of self-renewal and differentiation into specialized cells when submited to external signalings in the enviroment. Among adult stem cells, mesenchymal cells occupy an important position because they can differentiate into mesodermal cells such as osteoblasts, adipocytes and chondrocytes. Cell therapy consists in the use of mesenchymal stem cells (MSC) in the treatment of degenerative diseases and harmed tissue reconstruction. Due to the longstanding and costly procedure for cultivation of MSC, it was proposed the use of low power light sources, such as light emitting diodes (LED), to optimize these factors. Recent works have shown a series of results from the influence of LED light on biological tissues such as increased rate of cell proliferation, increased RNA, DNA and ATP synthesis rate. The purpose of this study is to compare the biomodulator effect of LED light set at wavelengths 630nm ± 10nm and 805nm ± 10nm on the mesenchymal stem cells proliferation. For this, the mesenchymal stem cells culture adopted the procedure used in the Departament of Animal Reproduction and Veterinary Radiology of the Faculty of Veterinary Medicine and Animal Sciences of Botucatu. MSC were obtained from an adult horse bone marrow, and isolated by density gradient separation, with the FICOLL reagent and by centrifugation. The pellet containing the stem cells was removed and these were placed in low glucose DMEM culture medium, containing 10% fetal calf serum and antibiotics. The material was observed daily by inverted microscopy for monitoring the progression of the cells and subsequently the amount of cells were counted in a Neubauer counting chamber. The amount of MSC was obtained by cell culture seeded in 24 wells culture plate and segregated into three distinct groups: Group 1 was irradiated with wavelength set at 630nm ± 10 nm, Group... Amino acids play essential roles in both metabolism and the proteome. Many studies have profiled free amino acids (FAAs) or proteins; however, few have connected the measurement of FAA with individual amino acids in the proteome. In this study, we developed a metabolomics method to comprehensively analyze amino acids in different domains, using two examples of different sample types and disease models. We first examined the responses of FAAs and insoluble-proteome amino acids (IPAAs) to the Myc oncogene in Tet21N human neuroblastoma cells. The metabolic and proteomic amino acid profiles were quite different, even under the same Myc condition, and their combination provided a better understanding of the biological status. In addition, amino acids were measured in 3 domains (FAAs, free and soluble-proteome amino acids (FSPAAs), and IPAAs) to study changes in serum amino acid profiles related to colon cancer. A penalized logistic regression model based on the amino acids from the three domains had better sensitivity and specificity than that from each individual domain. To the best of our knowledge, this is the first study to perform a combined analysis of amino acids in different domains, and indicates the useful biological information available from a metabolomics analysis of the protein pellet. This study lays the foundation for further quantitative tracking of the distribution of amino acids in different domains, with opportunities for better diagnosis and mechanistic studies of various diseases.

The Bi-Sr-Ca-Cu-O system has been one of the most studied superconducting ceramic materials for industry applications. The most of the studies with this aim are on silver/ceramic composites, due to the benefits and great compatibility of this metal with the oxide. In this paper we describe a systematic and comparative study on Ag/BSCCO composite, made by the citrate route, in which the ceramic pellets are sintered in the presence of silver powder using several proportions and having several granulations. It was observed that the introduction of fine (0.5 and 2 μm) silver powder in the proportions of 5 wt. % always implies in a better critical current density compared to the no silver pellet. According to the results, the silver powder in excess of 5 wt.% may not promote best electrical properties, depending on the size of the silver particles.

Fertigation management of gerbera crop has been many times performed inadequately, and it has been worsened when mixtures of substrates with different physical and chemical characteristics are used. Aiming at evaluating the production and quality of potted gerbera in two substrates and different levels of fertigation, the experiment was conducted in the greenhouse of the DRN/Soil Science, FCA/UNESP, Botucatu (SP). A 5 x 2 factorial randomized block design (5 levels of fertigation and 2 substrates) was adopted with 4 replications. Levels of fertigation corresponded to maintenance of 100% available water (AW) in the substrate; 100 to 80% of AW; 100 to 60% of AW; 100 to 40% of AW and 100 to 20% of AW. The substrates were as follows: 1- mixed coconut fiber (50% pellet coconut fiber and 50% coir fiber), 2- 40% red soil, 40% decomposed pine bark, 10% composition 1 (40% decomposed pine bark, 30% vermiculite and 30% carbonized rice husk) and 10% composition 2 (75% decomposed pine bark and 25% needles of pine). Plants at the marketing stage were evaluated according to the number of leaves; diameter of leaf surface; leaf area; fresh and dry phytomass of leaves, inflorescence and total plant; inflorescence number and diameter, stem diameter, plant height, leaf area rate and electrical conductivity of the substrate solution. Plants of better quality were obtained when they were maintained in levels of 100% available water and the mixed coconut fiber was used as substrate.
