We investigate the diversity of yeasts isolated in gardens of the leafcutter ant Atta texana. Repeated sampling of gardens from four nests over a 1-year time period showed that gardens contain a diverse assemblage of yeasts. The yeast community in gardens consisted mostly of yeasts associated with plants or soil, but community composition changed between sampling periods. In order to understand the potential disease-suppressing roles of the garden yeasts, we screened isolates for antagonistic effects against known microfungal garden contaminants. In vitro assays revealed that yeasts inhibited the mycelial growth of two strains of Escovopsis (a specialized attine garden parasite), Syncephalastrum racemosum (a fungus often growing in gardens of leafcutter lab nests), and the insect pathogen Beauveria bassiana. These garden yeasts add to the growing list of disease-suppressing microbes in attine nests that may contribute synergistically, together with actinomycetes and Burkholderia bacteria, to protect the gardens and the ants against diseases. Additionally, we suggest that garden immunity against problem fungi may therefore derive not only from the presence of disease-suppressing Pseudonocardia actinomycetes, but from an enrichment of multiple disease-suppressing microorganisms in the garden matrix.

The possible roles played by yeasts in attine ant nests are mostly unknown. Here we present our investigations on the plant polysaccharide degradation profile of 82 yeasts isolated from fungus gardens of Atta and Acromyrmex species to demonstrate that yeasts found in ant nests may play the role of making nutrients readily available throughout the garden and detoxification of compounds that may be deleterious to the ants and their fungal cultivar. Among the yeasts screened, 65% exhibited cellulolytic enzymes, 44% exhibited pectinolytic activity while 27% and 17% possess enzyme systems for the degradation of protease and amylase, respectively. Galacturonic acid, which had been reported in previous work to be poorly assimilated by the ant fungus and also to have a negative effect on ants' survival, was assimilated by 64% and 79% of yeasts isolated from nests of A. texana and Acromyrmex respectively. Our results suggest that yeasts found in ant nests may participate in generation of nutrients and removal of potentially toxic compounds, thereby contributing to the stability of the complex microbiota found in the leaf-cutting ant nests.

No presente trabalho foram estudados os ácaros associados a plantas ornamentais de praças, ruas e jardins residenciais, no município de Ilha Solteira, SP. Foram amostradas folhas de 20 espécies de plantas e alguns representantes das morfoespécies de ácaros encontrados foram montados em lâminas de microscopia com o meio de Hoyer, para posterior identificação sob microscópio óptico com contraste de fases. Foram registradas 23 espécies pertencentes a 15 gêneros de oito famílias. Dessas espécies, 13 são fitófagas, nove predadoras e uma de hábito alimentar não determinado. A família com maior riqueza de ácaros foi Tetranychidae, com nove espécies, sendo uma delas registrada pela primeira vez após a descrição original e outra pela primeira vez no Brasil. Lorryia formosa Cooremann, 1958 (Tydeidae) foi a que ocorreu em maior número de hospedeiros. As plantas com as maiores riquezas de ácaros foram Lagerstroemia indica L., Mussaenda alicia Hort. e Tabebuia sp., com seis espécies registradas em cada uma.

LASSBio-767 [(-)-3-O-acetyl-spectaline] and LASSBio-822 [(-)-3-O-tert-Boc-spectaline] were recently described as cholinesterase inhibitors derived from the natural piperidine alkaloid (-)-spectaline, obtained from the flowers of Senna spectabilis (Fabaccae). We investigated their mechanism of inhibition of acetylcholinesterase and their efficacy in reversing scopolamine-induced amnesia. Competition assays with the substrate acetylthiocholine showed a concentration-dependent reduction in rat brain cholinesterase V-max without changes in apparent K-m. The kinetic data for LASSBio-767 and LASSBio-822 were best fit by a model of simple linear noncompetitive inhibition with K-i of 6.1 mu M and 7.5 mu M, respectively. A dilution assay showed a fast and complete reversal of inhibition, independent of incubation time. Simulated docking of the compounds into the catalytic gorge of Torpedo acetylcholinesterase showed interactions with the peripheral anionic site, but not with the catalytic triad. Anti-amnestic effects in mice were assessed in a step-down passive avoidance test and in the Morris water maze 30 min after injection of scopolamine (1 mg/kg i.p.). Saline, LASSBio-767, or LASSBio-822 was administered 15 min before scopolamine. Both compounds reversed the scopolamine-induced reduction in step-down latency at 0.1 mg/kg i.p. LASSBio-767 reversed scopolamine-induced changes in water maze escape latency at 1 mg/kg i.p. or p.o., while its cholinergic side effects were absent or mild up to 30 mg/kg i.p. (LD50 above 100 mg/kg i.p.). Thus, the (-)-spectaline derivatives are potent cholinergic agents in vivo, with a unique profile combining noncompetitive cholinesterase inhibition and CNS selectivity, with few peripheral side effects. O Planejamento Paisagístico do Campus da Faculdade de Ciências Agronômicas da Universidade Estadual Paulista (UNESP) em Botucatu - SP, foi elaborado a partir de um Plano Diretor já existente e considerado o comportamento da paisagem, com edificações e circulações já estabelecidas, dando ênfase ao projeto de ajardinamento do entorno da Central de Salas de Aulas. Os objetivos principais do planejamento visam a proteção do solo, arborização adequada e ajardinamento do encontro dos prédios principais. O traçado proposto é simples, com a finalidade de dar uma escala humana ao Campus, considerando a necessidade dos usuários e de favorecer a utilização de máquinas para manutenção e tratos culturais, em face da pequena disponibilidade de mão-de-obra, sendo mais elaborado somente nos entornos dos prédios principais. A estrutura vegetal é formada de árvores, arbustos, gramados e outras forrações que foram selecionadas por sua rusticidade, resistência, aclimatação ao local e disponibilidade nos Viveiros Experimentais da Faculdade de Ciências Agronômicas-UNESP. O organofosforado diclorvós impregnado em coleiras plásticas é um recurso utilizado em medicina veterinária que visa ao controle de ectoparasitas de cães e gatos. O objetivo deste trabalho foi avaliar os efeitos do uso de coleiras plásticas impregnadas com diclorvós (8,37%) em ratas Wistar durante o período de gestação e lactação, como possível fonte de alterações comportamentais e da atividade colinesterásica cerebral dos filhotes. Na desmama, não houve diferença na atividade colinesterásica cerebral entre as mães tratadas com diclorvós e o grupo controle, bem como entre os respectivos filhotes. O tratamento com diclorvós também não influenciou no comportamento geral dos animais, avaliado no campo aberto, nem no nível de ansiedade testado no labirinto em cruz elevado, ambos aos 35 dias pós-natal.

Syagrus schizophylla (Mart.) Glass. belongs to the Arecaccae family. This palm is native of Brazil and presents great potential for use in gardens and cultivation in pot. Palms species, with relatively few exceptions, can only be propagated from seeds; even so, there are no reports in the literature about the germination of this palm seeds. The seed maturity is a factor that interferes in the success of the germination process. For some species, studies showed that palm seeds germinated better when the fruits were completely ripe (showing full color) and for other, when they were with green coloration. Several species of the Arecaccae family presents physical dormancy of seeds in varied degrees, demanding treatments for improve germination. The objective of this work was to study the effects of maturation and of the scarification on seed germination of S. schizophylla. The experimental design used was a factorial 3 x 2 (3 maturation stadiums: green, half-ripe - yellow and completely ripe - red; and mechanical scarification: with and without), entirely casualized, with four replications of 15 seeds per plot. The seeds (with 32,43% of humidity) were placed in plastic boxes with sand (60% of humidity, placing water according to weight in each three days), under controlled conditions of alternated temperature of 25-35 degrees C, photoperiod of 12 hours. The percentage of germination and the speed germination index (SGI) were evaluated until 79 days. It was observed that, so much for germination percentage as for SGI, there was not significant difference for the interaction among the two factors, maturation stadium and scarification, however, there were significant differences among the maturation stadiums and between seeds scarified or not. The seeds from green fruits presented lower germination percentage and slower germination when compared with seeds from yellow or red fruits; the seeds from yellow or red fruits didn't differ statistically to each other. To seeds scarificated, independently of the maturation stadium, presented germination percentage significantly larger and the germination was significantly faster when compared with the seeds without scarification.

The present study investigated the effects of intranigral MPTP (1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine) infusion on rats treated with phosphatidylserine and evaluated in two memory tasks and on striatal dopamine levels. The results indicated that MPTP produced a significant decrease in the avoidance number in comparison to sham-operated and non-operated rats submitted to a two-way avoidance task. MPTP-lesioned rats exhibited an increase in the latencies to find the platform in cued version of the water maze in comparison to sham-operated and non-operated animals. The tested toxin reduced striatal dopamine levels in comparison to sham-operated and non-operated groups. A final surprising result was that phosphatidylserine was unable to reverse the cognitive deficits produced by MPTP or the reduction of striatal dopamine levels. In conclusion, the data suggest that MPTP is a good model to study the early impairment associated with Parkinson's disease and phosphatidylserine did not improve the memory impairment induced by MPTP. Wistar dams were exposed to 500 ppm of Pb, as Ph acetate, or 660 ppm Na acetate in drinking water during pregnancy and lactation. Male pups at 23 (weaned) or 70 days (adult) of age were submitted to behavioral evaluation and Pb determination. The behaviors evaluated were: locomotor activity (open-field test), motor coordination (rotarod test), exploratory behavior (holeboard test), anxiety (elevated plus maze and social interaction tests), and learning and memory (shuttle box). Ph levels were measured in the blood and cerebral regions (hippocampus and striatum) of dams and pups. The results of the present report demonstrated that exposure to Ph during pregnancy and lactation induces in weaned pups hyperactivity, decreased exploratory behavior, and impairment of learning and memory. These alterations were observed at blood Ph levels in the range that may be attained in children chronically exposed to low levels of Pb (21 +/- 3 mug/dl). Regarding adults, the results demonstrated that the regimen of exposure adopted induces anxiety in these animals at nondetectable blood Ph levels. (C) 2001 Elsevier B.V. All rights reserved.

There are conflicting results on the function of 5-HT in anxiety and depression. To reconcile this evidence, Deakin and Graeff have suggested that the ascending 5-HT pathway that originates in the dorsal raphe nucleus (DRN) and innervates the amygdala and frontal cortex facilitates conditioned fear, while the DRN-periventricular pathway innervating the periventricular and periaqueductal gray matter inhibits inborn fight/flight reactions to impending danger, pain, or asphyxia. To study the role of the DRN 5-HT system in anxiety, we microinjected 8-OH-DPAT into the DRN to inhibit 5 HT release. This treatment impaired inhibitory avoidance (conditioned fear) without affecting one-way escape (unconditioned fear) in the elevated T-maze, a new animal model of anxiety. We also applied three drug treatments that increase 5-HT release from DRN terminals: 1) intra-DRN microinjection of the benzodiazepine inverse agonist FG 4172, 2) intra-DRN microinjection of the excitatory amino acid kainic acid, and 3) intraperitoneal injection of the 5-HT releaser and uptake blocker D-fenfluramine. All treatments enhanced inhibitory avoidance in the T-maze. D-Fenfluramine and intra-DRN kainate also decreased one-way escape. In healthy volunteers, D-fenfluramine and the 5-HT agonist mCPP (mainly 5-HT2C) increased, while the antagonists ritanserin (5-HT2A/(2C)) and SR 46349B (5-HT2A) decreased skin conductance responses to an aversively conditioned stimulus (tone). In addition, D-fenfluramine decreased, whereas ritanserin increased subjective anxiety induced by simulated public speaking, thought to represent unconditioned anxiety. Overall, these results are compatible with the above hypothesis. Deakin and Graeff have suggested that the pathway connecting the median raphe nucleus (MRN) to the dorsal hippocampus promotes resistance to chronic, unavoidable stress. In the present study, we found that 24 h after electrolytic lesion of the rat MRN glandular gastric ulcers occurred, and the immune response to the mitogen concanavalin A was depressed. Seven days after the same lesion, the ulcerogenic effect of restraint was enhanced. Microinjection of 8-OH-DPAT, the nonselective agonist 5-MeO-DMT, or the 5-HT uptake inhibitor zimelidine into the dorsal hippocampus immediately after 2 h of restraint reversed the deficits of open arm exploration in the elevated plus-maze, measured 24 h after restraint. The effect of the two last drugs was antagonized by WAY-100135, a selective 5-HT1A receptor antagonist. These results are compatible with the hypothesis that the MRN-dorsal hippocampus 5-HT system attenuates stress by facilitation of hippocampal 5-HT1A-mediated neurotransmission. Clinical implications of these results are discussed, especially with regard to panic disorder and depression.

A population boom of the millipede Plusioporus setigerhas been recorded in coffee crops in the district of Patrocinio-MG, Brazil on the last five years. The millipedes spread over dwelling houses and to vegetable gardens, orchards and small plantations. It was suggested that population increase occurred as a consequence of the organic materials in high scale and also periodic irrigation in the coffee crop.
