Para o sucesso do plantio direto há necessidade de que a cultura antecessora à principal seja boa produtora e mantenedora de cobertura vegetal. O milheto tem-se constituído em boa opção de cultivo no outono/inverno para rotação com soja, porém não há estudos avaliando o efeito do sistema na qualidade das sementes. O objetivo do trabalho foi estudar o efeito de três épocas de semeadura e cinco manejos com ceifas do milheto sobre a qualidade fisiológica de sementes de soja cultivada em sucessão, por plantio direto, na mesma área, por três ciclos de rotação (1999/2000; 2001/2002 e 2002/2003). As épocas de semeadura constituíram três experimentos (E1, E2 e E3), em delineamento experimental de blocos ao acaso com cinco tratamentos (M1 = ceifa a cada florescimento e retirada da massa vegetal; M2 = ceifa a cada florescimento e permanência da massa vegetal; M3 = ceifa no florescimento e retirada da massa vegetal; M4 = ceifa no florescimento e permanência da massa vegetal e M5 = sem ceifa até a produção de grãos, quando foi cortada a panícula, permanecendo o restante da massa vegetal), e quatro repetições. Os experimentos de milheto foram semeados em 1999: E1 = 05/03, E2 = 25/03 e E3 = 19/04; em 2001 = E1 = 17/04, E2 = 07/05 e E3 = 27/05, e em 2002: E1 = 25/04, E2 = 15/05 e E3. = 05/06. em cada ciclo de rotação, a soja foi implantada nos três experimentos na mesma data após o manejo final do milheto com dessecação química; as colheitas foram realizadas na mesma data, também. As sementes de soja, cv. Empresa Brasileira de Pesquisa Agropecuária (EMBRAPA)-48, foram avaliadas quanto ao tamanho, massa de 100 sementes, teor de água, germinação e vigor. Sementes de soja com melhor qualidade fisiológica foram obtidas, nos três ciclos de sucessão, quando cultivada após a primeira época de semeadura do milheto, independentemente do manejo com ceifas do milheto, cujo efeito foi pouco evidente.

O uso de fontes não convencionais para fornecimento de K às plantas tem sido amplamente estudado, mas os efeitos de materiais alternativos na qualidade fisiológica das sementes não são conhecidos. Este estudo teve como objetivo avaliar a qualidade fisiológica de sementes de soja e trigo em função da aplicação fontes de potássio em uma sucessão de culturas. O delineamento experimental foi o de blocos ao acaso com quatro repetições. Os tratamentos constaram de três fontes de K (KCl, rocha alcalina e fonolito moído, com 58%, 11% e 8.42% de K2O, respectivamente) aplicados em quatro doses (0, 25, 50 e 100 kg K2O ha-1). As doses de potássio foram aplicadas na soja e seu efeito residual foi avaliado na cultura do trigo, cultivado em sucessão. Logo após a colheita, as sementes de soja e trigo foram avaliadas pelos testes de teor de água, massa de sementes, germinação, primeira contagem, condutividade elétrica, comprimento de plântulas e massa da matéria seca de plântulas. Plantas de soja adubadas com fontes alternativas para fornecimento de K produzem sementes com maior massa e menor permeabilidade de membranas comparado às com KCl; maior qualidade fisiológica de sementes de soja e massa de sementes de trigo são obtidas com maiores doses de K2O independente da fonte.

Plant nutrition can positively influence quality of seeds by improving plant tolerance to adverse climate. In this context, silicon is currently considered a micronutrient and it is beneficial to plant growth, especially Poaceaes such as white oat and wheat, thereby improving physiological quality of seeds. This study had the objective of evaluating the effects of silicon leaf application on plant tillering, silicon levels and physiological quality of white oat and wheat seeds besides establishing correlations between them. Two experiments were carried out in winter with white oat and wheat. The experimental design was the completely randomized block with eight replications. Treatments consisted of foliar application of silicon (0.8% of soluble silicon, as stabilized orthosilicic acid) and a control (with no application). Silicon levels in leaves were determined at flowering whereas the number of plants and panicles/spikes per area was counted right before harvest. Seed quality was evaluated right after harvest through mass, germination and vigor tests. Data was submitted to variance analysis and means were compared by the Tukey test at a probability level of 5%. Person's linear correlation test was performed among silicon level in plants, tillering and seed quality data. Silicon leaf application increases root and total length of white oat seedlings as an effect of higher Si level in leaves. Silicon leaf application increases mass of wheat seeds without affecting germination or vigor.

This study aimed to evaluate the effect of substituting chemical nitrogen (N) fertilization for equivalent N levels from sewage sludge of Wastewater Treatment Plant (WTP) on sunflower plant development. Nutrient levels in physiologically mature leaves and seeds, besides nutrient exportation during a 130-day assay, were also assessed. The experiment was carried out in 100 m(2) permanent plots at Sao Manuel Farm, which belongs to School of Agronomical Sciences, São Paulo State University-UNESP, Botncatu, São Paulo State, Brazil. The farm is located in the municipality of Sao Manuel, São Paulo State. Experimental design was in randomized blocks including 5 treatments and 5 replicates. Treatments were: T1 - chemical N fertilization according to the recommendation for the culture; T2 - 50% N from sewage sludge and 50% N from chemical fertilization; T3 - 100% N from sewage sludge; T4 - 150% N from sewage sludge; T5 - 200% N from sewage sludge. For all treatments, equal amounts of P and K fertilization were applied. Treatments differed for plant height from 21 to 64 days, stern diameter from 28 to 57 days, and leaf number from 21 to 38 days. Seed nutrient levels slightly varied; however, the quantities of exported N, P, Mg, Fe and Zn varied as sewage sludge levels increased.

O objetivo deste trabalho foi avaliar a influência de doses de calcário e zinco e, a interação destes sobre o desenvolvimento da cultura da mamona. A hipótese deste trabalho é que o aumento de doses de calcário diminui a disponibilidade de zinco no solo para a planta. O experimento foi desenvolvido em estufa para cultivo protegido, sem controle de temperatura, em vasos com 17 dm³ de Latossolo Vermelho distrófico, textura média. O delineamento experimental foi em blocos casualizados, com quatro repetições. Os tratamentos foram arranjados num esquema fatorial 3 x 5, sendo os fatores: três saturações por bases (40, 60 e 80%) e cinco doses adicionadas de Zn (0,5; 1,0; 2,0; 4,0 e 8,0 mg dm-3 de solo). Apenas uma planta por vaso foi conduzida até 174 dias após emergência (maturidade fisiológica do racemo primário), quando foi avaliada a produção. Maiores doses de calcário resultaram em maior número de grãos por planta e MS dos grãos por planta. Houve interação V% e Zn na altura da planta, massa seca da raiz, comprimento da parte masculina e total do racemo. No entanto, nenhum destes incrementos refletiu em diferença significativa na massa de 100 grãos da mamoneira.

BACKGROUND: Baru (Dipteryx alata Vog.) is a fruit distributed throughout the Brazilian savanna and contains a seed with a high protein content, whose properties have been rarely explored. The purpose of this study was to characterize this protein, especially by isolation and quantifying its fractions and measuring some of its molecular properties.RESULTS: Baru seeds contain 244 g kg(-1) protein on a dry weight basis. Solubility profiles showed a preponderance of globulins. This fraction dominated the seed composition, with 61.7 wt% of the total soluble proteins. Albumins and glutelins accounted for 14 and 3.3 wt%, respectively. SDS-PAGE resolution of albumin and globulin showed main bands with molecular weights of 84 kDa and 64,66 and 73 kDa, respectively. The total protein of the flour and the globulin showed values of in vitro digestibility of 85.59% and 90.54%, relative to casein. Total globulin produced only one chromatographic peak, both on Sepharose CL-6B gel filtration and on DEAE-cellulose ion-exchange columns, eluted at a concentration of 0.12 mol L(-1) NaCl.CONCLUSION: The baru seed had high protein content with large quantities of storage proteins. The chromatographic and solubility profiles indicate the predominance of a fraction with characteristics of a legumin-type protein. (C) 2011 Society of Chemical Industry

The guava seed protein isolate ( PI) was obtained from the protein precipitation belonging to the class of the gluteline (Ip 4.5). The conditions for the preparation of the PI were determined by both the solubility curve and simultaneous thermogravimetry-differential thermal analysis (TG-DTA): pH 11.5, absence of NaCl and whiteners and T=( 25 +/- 3) degrees C. Under these conditions a yield of 77.0 +/- 0.4%, protein content of 94.2 +/- 0.3, ashes 0.50 +/- 0.05% and thermal stability, T= 200 degrees C, were obtained. The TG-DTA curves and the PI emulsification capacity study showed the presence of hydrophobic microdomains at pH 11.5 and 3.0 suggesting a random coil protein conformation and, to pH 10.0, an open protein conformation. The capacity of emulsification (CE), in the absence of NaCl, was verified for: 1 - pH 3.0 and 8.5, using the IP extracted at pH 10.0 and 11.5, CE >= 343 +/- 5 g of emulsified oil/g of protein; 2 - pH 6.60 just for the PI obtained at pH 11.5, CE >= 140 +/- 8 g of emulsified oil/g of protein.

Variation in seed size is often observed in samples of eucalypt seeds and this leads to heterogeneous populations of plants, principally through variation in the early stages of plant development. It follows that samples of seeds more uniform in size could produce more uniform populations of plants. In studies of Eucalyptus globulus ssp. globulus it was of interest to determine whether or not the genetic diversity within a population, through the use of isozyme markers, was altered in the subpopulations developed from seeds of different size classes. A commercial sample of seed was separated by seed size into three subpopulations and the percentage germination and mean fresh weight of the seedlings were determined. Proteins extracted from leaves of the seedlings were separated by electrophoresis and tested for activity of eight different enzymes. These eight enzymes showed activity at 20 loci and mean genetic diversity and fixation index were determined using 13 of these loci. The subpopulation of the smallest seeds contained a greater proportion of abnormal seeds and had a lower percentage germination and plant weight compared to the other subpopulations. No significant differences were found in the number of alleles per locus, percentage of polymorphic loci, mean heterozygosity. The major part of the endogamy, indicated by F statistic, was found within the subpopulations: F-(IS) = 0.518; F-(ST) = 0.010 and F(IT) = 0.523. We conclude that the use of seeds of uniform size will lead to more uniform germination and plant growth without alteration in overall genetic diversity.

The mature dentinoenamel junction (DEJ) is viewed by some investigators and the current authors, not as a fossilized, sharp transition between enamel and dentin, but as a relatively broad structural transition zone including the mantle dentin and the inner aprismatic enamel. In this study, the DEJ structure in bovine incisors was studied with synchrotron microComputed Tomography (microCT) using small cubes cut parallel to the tooth surface. The reconstructions revealed a zone of highly variable punctate contrast between bulk dentin and enamel; the mean linear attenuation coefficients and their standard deviations demonstrated that this zone averaged less mineral than dentin or enamel but had more highly variable structure than either. The region with the punctuate contrast is, therefore, the mantle dentin. The thickness of the mantle dentin seen in a typical data set was about 30 mu m, and the mantle dentin-enamel interface deviated +/- 15 mu m from the average plane over a distance of 520 mu m. In the highest resolution data (similar to 1.5 mu m isotropic voxels, volume elements), tubules in the dentin could be discerned in the vicinity of the DEJ. Contrast sensitivity was high enough to detect differences in mineral content between near-surface and near-DEJ volumes of the enamel. Reconstructions before and after two cubes were compressed to failure revealed cracks formed only in the enamel and did not propagate across the mantle dentin, regardless of whether loading was parallel to or perpendicular to the DEJ. Background and Aims the occurrence of stomata in seed coats is uncommon and there is limited information about their function(s). The aim of this study was to verify the distribution of stomata in seed coats of Swietenia macrophylla and to relate it to the imbibition process and aspects of the structure of the outer integument layers.Methods For the structural and ultrastructural studies, the seeds were processed using the usual techniques and studied under light and scanning electron microscopes. Histochemical tests were employed to identify the cell wall composition in the different seed coat portions. To assess the role of the stomata in the imbibition, non-impervious seeds were compared with partially impervious ones, in which only the embryo, median or hilar regions were left free. Further, the apoplastic pathway marker was employed to confirm the role of the stomata as sites of water passage during imbibition.Key Results A positive relationship was observed between seed coat thickness and stomata density. The stomata were devoid of movement, with a large pore. They occurred in large numbers in the embryo region and extended with lower frequency towards the wing. Imbibition rates were related to stomata density, suggesting that the stomata act as preferential sites for water entry in the S. macrophylla seeds.Conclusions At maturity, the stomata in the seed coat play a significant role in seed imbibition. The data may also infer that these permanently opened stomata have an important role in gas exchange during seed development, aiding embryo respiration.
