Nematropica 41: 150-153.Medicinal plants can be attacked by pests, diseases and nematodes, which can compromise the quality and quantity of their healing properties and their yield. The aim of this study was to analyse the reaction of 15 medicinal plant species to the nematode Meloidogyne spp. The seedlings were produced by seed germination or cuttings under greenhouse conditions. A completely random experimental pattern of 15 treatments and 10 replications was chosen for the study. The seedlings were inoculated with approximately 5000 eggs + J(2)/plant 20 days after planting. Plant height and fresh and dry leaf weight, were measured 60 days after planting. The roots were collected, thoroughly washed and stained with Philoxine B and then processed to extract the eggs to determine the reproduction factor. For M. incognita the results showed that Chamomile was susceptible with RF = 1,64 making it a good host, and the other plants were resistant (RF < 1), and for M. javanica that all the plants showed resistance (RF < 1), Myrrh, Rue and Balsam demonstrating immunity (RF = 0).

Coccoloba mollis (Family Polygonaceae) is a medicinal plant popularly used in cases of memory loss, stress, insomnia, anemia, impaired vision, and sexual impotence, but the scientific literature, to date, lacks studies on the biological effects of this species, particularly with regard to cytotoxicity and induction of DNA damage. The aim of the present study was to assess in vitro (in hepatic HTC cells) ethanolic extracts of the roots and leaves of C. mollis for cytotoxicity, genotoxicity, and induction of apoptosis. For these evaluations the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) cytotoxicity assay, comet assay, micronucleus test with cytokinesis block, and an in situ test for detection of apoptotic cells with acridine orange staining were used. The results showed that the extract obtained from the roots of C. mollis is more cytotoxic than that obtained from the leaves and that the reduction in cell viability observed in the MTT assay was a result, at least in part, from the induction of apoptosis. Both extracts induced DNA damage at a concentration of 20 mu g/mL in the comet assay, but no genotoxicity was detected with any of the treatments carried out in the micronucleus test.

The hexane-soluble fractions from the rhizomes or from the leaves from five Dorstenia (Moraceae) species (D. bahiensis Kl., D. bryoniifolia Mart ex. Miq., D. carautae C.C.Berg, D. cayapiaa Vell. and D. heringerii Car. & Val) were analysed by HRGC-MS (high-resolution gas chromatography - mass spectrometry). Pentacyclic triterpenes, steroids and furocoumarins were identified. HRGC-MS is shown to be a valuable tool for the analysis of terpenoidal compounds from Dostenia species. These compounds may be related to the folk utilization of Dorstenia species as antiophidicals.

The biotechnology, biochemical characterization, and protective effects of the himematsutake and shiitake mushrooms were studied for the Thematic Project, from 1998 to 2003. A new species, Agaricus Brasiliens is Wasser et al. (= A. blazei Murrill ss. Heinem.), was proposed for the cultivated lineages in Brazil. Interactions among lineages, substrates, casing layers, and fructification inductions (temperature alternations) significantly increased the productivity of A. brasiliensis in Brazil (from 40 g to 200 g of fresh mushroom kg -1 moist compost). However, pests and diseases (mainly Sciaridae flies and false truffle), drastically reduced the productivity of A. brasiliensis (below 50 g kg -1). Biochemically for each mushroom species, the polar extracts, no polars, and medium polarity presented the same organic substances; however, their concentrations differed as a result of the lineages, maturation of the fruit bodies, and cultivation conditions. In this aspect, concentration of linoleic acid in A. brasiliensis (added to animal food) was related directly to the chemical protection against carcinogenic drugs in mice. Aqueous extracts of Lentinus edodes (= Lentinula edodes (Berk.) Pegler) and A. brasiliensis may be preventive chemical protectors against mutagenic and carcinogenic drugs, depending on the lineage and extraction method (tea or juice). However, immunomodulator effects and tumor reduction were only observed with concentrated fractions (hexanic, methanolic, and ATF extracts). Aqueous extracts of A. brasiliensis and L. edodes have antibiotic-like substances and substances able to act as elicitors of resistance responses in some plants (local and systemic) and show a potential to be used in the alternative control of plant pathogens. © 2005 by Begell House, Inc.

Isotope screening is a simple test for determining the photosynthetic pathway used by plants. The scope of this work was to classify the photosynthetic type of some herbs and medicinal plants through studies of the carbon isotope composition (δ13C). Also, we propose the use of carbon isotope composition as a tool to control the quality of herbs and medicinal plants. For studies of δ13C, δ 13C‰ = [R (sample)/R (standard) - 1] × 10-3, dry leaves powdered in cryogenic mill were analyzed in a mass spectrometer coupled with an elemental analyzer for determining the ratio R = 13CO2/12CO2. In investigation of δ13C of 55 species, 23 botanical families, and 44 species possessed a C3 photosynthetic type. Six species found among the botanical families Euphorbiaceae and Poaceae were C4 plants, and 5 species found among the botanical families Agavaceae, Euphorbiaceae, and Liliaceae possessed CAM-type photosynthesis. Carbon isotope composition of plants can be used as quality control of herbs and medicinal plants, allowing the identification of frauds or contaminations. Also, the information about the photosynthetic type found for these plants can help in introducing and cultivating exotic and wild herbs and medicinal plants.

Strychnos pseudoquina St. Hil. is a native plant of the Brazilian Savannah, used in popular medicine to treat a number of conditions. Since it contains large quantities of alkaloids with proven antiulcer activity, we tested the genotoxic potential of crude extracts and fractions containing alkaloids and flavonoids from the leaves of this plant, on Salmonella typhimurium and performed the micronucleus test on peripheral blood cells of mice treated in vivo. The results showed that the methanol extract of the leaves of S. pseudoquina is mutagenic to the TA98 (-S9) and TA100 (+S9, -S9) strains of Salmonella. The dichloromethane extract was not mutagenic to any of the tested strains. Fractions enriched with alkaloids or flavonoids were not mutagenic. In vivo tests were done on the crude methanol extract in albino Swiss mice, which were treated, by gavage, with three different doses of the extract. The highest dose tested (1800 mg/kg b.w.) induced micronuclei after acute treatment, confirming the mutagenic potential of the methanol extract of the leaves of S. pseudoquina. In high doses, constituents of S. pseudoquina compounds act on DNA, causing breaks and giving rise to micronuclei in the blood cells of treated animals. © 2006 Elsevier Ltd. All rights reserved.

The scope of this work was to classify the photosynthetic type of Melissa officinalis L. and Cymbopogon citratus [DC.] Stapf, through studies of the carbon isotope composition (δ13 C), and we are proposing the use of carbon isotope composition results as a tool to control the quality of medicinal plants. For studies of δ 13C (13 C% = [R (sample)/R (standard) - 1] × 10 -3), dried, powdered leaves were analyzed in a mass spectrometer coupled with an elemental analyzer for determining the ratio R (R = 13CO2/12CO2). As results, M. officinalis presented a C3 photosynthetic type, and C. citratus presented a C4 photosynthetic type. The carbon isotope composition from this study can be used as quality control of M. officinalis adulterants.

A simple method to determine Cu, Fe, Mn and Zn in single aliquots of medicinal plants by HR-CS FAAS is proposed. The main lines for Cu, Mn and Zn, and the alternate line measured at the wing of the main line for Fe at 248.327 nm allowed calibration within the 0.025 - 2.0 mg L-1 Cu, 1.0 - 20.0 mg L-1 Fe, 0.05 - 2.0 mg L-1 Mn, 0.025 - 0.75 mg L-1 Zn ranges. Nineteen medicinal plants and two certified plant reference materials were analyzed. Results were in agreement at a 95% confidence level (paired t-test) with reference values. Limits of detection were 0.12 μg L-1 Cu, 330 μg L-1 Fe, 1.42 μg L-1 Mn and 8.12 μg L-1 Zn. Relative standard deviations (n=12) were ≤ 3% for all analytes. Recoveries in the 89 - 105% (Cu), 95 - 108% (Fe), 94 - 107% (Mn), and 93 - 110% (Zn) ranges were obtained.

The Iguape and Cananéia Lagoon-Estuarine Complex is a biodiversity hotspot in southeastern Brazil. In recent decades, the region has become an important destination for recreational fishing. The objective of this study was to analyze the socioeconomic characteristics of visiting anglers and fishing guides working in the Cananéia-Iguape-Peruíbe Environmental Protected Area and their views on fisheries management. Data were collected through semi-structured interviews conducted between January 2009 and January 2010. We interviewed 278 anglers, who were predominantly male (93%) with a mean age of 47 years and from the state of São Paulo. The targeted species were snooks (Centropomus undecimales and Centropomus parallelus) and weakfishes (Cynoscion leiarchus and Cynoscion acoupa). Only half the anglers had the mandatory fishing license, and many of them lacked knowledge about catch quotas and minimum size requirements for specific species. The fishing guides (n = 80) were all male, with a mean age of 39 years and extensive experience. Most of the guides believe that the study area is somewhat degraded due to the removal of riparian vegetation, siltation, pollution, and especially the depletion of fish stocks. The opinions of the stakeholders (anglers and guides) converge on the high priority needs of the fishery and possible management actions regarding recreational fishing, such as improved fisheries enforcement (first in order of importance), proper training of fishing guides, zoning of fishing areas, and the definition of a maximum size limit. © 2013 Elsevier Ltd.

Casearia sylvestris Swartz (Salicaceae) é uma planta utilizada na medicina tradicional, cujos extratos de folhas demonstraram importantes ações farmacológicas. A espécie apresenta variação morfológica, genética e química. Duas variedades são consideradas devido a diferenças morfológicas: C. sylvestris var. sylvestris e var. lingua. Há dificuldades na definição destas variedades. O objetivo deste trabalho é avaliar diferenças morfo-anatômicas e químicas entre as variedades de C. sylvestris que permitam sua diferenciação com aplicação farmacêutica ou botânica. Seções transversais e paradérmicas de folhas foram preparadas para análises morfo-anatômicas, histoquímicas e microscopia quantitativa (indices de estômatos e paliçada). Análises cromatográficas (CLAE-DAD e CCD) foram realizadas para obter o perfil de diterpenos clerodânicos. Os resultados das análises morfo-anatômicas demonstraram diferenças significativas entre as variedades somente em cortes paradérmicos: var. sylvestris - paredes celulares epidérmicas poligonais e hipoestomática, var. lingua - paredes celulares epidérmicas arredondadas e anfiestomática. Os índices de estômatos não revelaram diferenças; os valores dos índices de paliçada foram de 2,8 para var. lingua e 3,9 para var. sylvestris. As análises cromatográficas confirmaram resultados prévios, demonstrando predomínio de diterpenos na var. sylvestris. Este trabalho sugere que análises cromatográficas e morfo-anatômicas podem ser ferramentas aplicáveis na distinção das variedades da espécie.
