Throughout the genetic and physiological evolution of microorganisms, the microbiological sciences have been expanding the introduction of new therapeutic trials against microbial diseases. Special attention has been paid to the bacterium Helicobacter pylori, which induces gastric infections capable of causing damage, ranging from acute and chronic gastritis to the development of gastric cancer and death. The use of compounds with natural origins has gained popularity in scientific research focused on drug innovation against H. pylori because of their broad flexibility and low toxicity. The aim of this study was to describe the use of natural products against H. pylori in order to clarify important parameters for related fields. The study demonstrated the vast therapeutic possibilities for compounds originating from natural sources and revealed the need for innovations from future investigations to expand the therapeutic arsenal in the fight against H. pylori infection.

After the isolation of Helicobacter pylori from an injury at the stomach mucosa by Marshall and Wareen, work that was recognized with the Nobel Prize of Medicine or Physiology in 2005, many other works showed the relationship between the presence of H. pylori and diseases at the digestive system, such as gastritis, gastric, duodenal and peptic ulcer, and stomach cancer. The 13C-Urea Breath Test - 13C-UBT is a non-invasive diagnostic method that utilizes the breath of a patient to determine the presence of H. pylori through stable isotopes. This work aimed to find an ideal 13C-UBT Isotopic Ratio Mass Spectroscopy cut-off value (a threshold between positive and negative) to diagnose H. pylori infection at Brazilian population. Patients were selected at the UNESP-Botucatu Clinical Hospital Endoscopy Section. With these results it was possible to indicate that the best cut-off value is between 2.5 to 6 ‰ of Delta Over Baseline (DOB)

O presente trabalho de conclusão de curso visou à otimização e preparação da síntese de Lapdesf-IUNI e avaliação do potencial de inibição de urease de três compostos previamente sintetizados, direcionados especificamente para o tratamento de infecções por Helicobacter pylori, bactéria mundialmente reconhecida como uma das principais causadoras de infecções no mundo, responsável pelos casos de infecções gástricas, como gastrites crônicas, úlceras (gástricas e duodenais) e carcinomas. Lapdesf-IUNI é um composto derivado de metronidazol, com atividade potencialmente inibidora de urease, enzima presente neste microrganismo e essencial a sua sobrevivência no ambiente gástrico. O produto foi obtido, porém com baixo rendimento, por isso sua metodologia ainda necessita ser aperfeiçoada, para que, em seguida este composto possa ser testado. Outros três compostos, um derivado de tiouréia e dois derivados guanidínicos, com potencial atividade inibidora de urease, T-6, R-17 e R-21, sintetizados pela equipe do Prof. Eduardo Perez, foram avaliados com relação a sua atividade inibidora frente à enzima urease e IC50, apresentando bons resultados, que permitem a continuidade dos estudos, comparando inclusive com fármacos utilizados nos protocolos de tratamento padrões para o tratamento das infecções causadas por H. pylori

Helicobacter pylori (H. pylori) is a common gastric pathogen that has infected more than 50% of the population of the world and it has been associated with chronic gastritis, gastric ulcers, duodenal ulcer, and gastric cancer. Although, almost all infected people develop gastritis, there is a variety of clinical outcomes, and only a minority (<1%) of infected individuals develop gastric cancer. There are evidences which suggest that the chronic inflammatory reaction caused by the bacterial infection may be involved in the production of reactive oxygen species or reactive nitrogen species. It may lead to DNA damage, which together with the cellular response could lead to gene mutations, chromosomal aberrations characterizing genomic instability that may represent the early step in gastric carcinogenesis. The extent and severity of gastric mucosal inflammation, as well as the clinical outcome of the infection, depend on a number of factors, including the host genetic susceptibility such SNP T3801 CYP1A1, immune response, age at which the infection was acquired, environmental factors, especially dietary and bacterial virulence factors. Due to the risk of developing gastric cancer in humans infected by H. pylori, we used the Comet Assay to investigate the influence of the SNP T3801C CYP1A1 on levels of oxidative DNA damage in gastric epithelial cells. The study was conducted with biopsies from the gastric antrum and corpus of 103 H. pylori-infected patients and 24 uninfected control patients. Genotype of SNP T3801C CYP1A1 was determined by PCR-RFLP and DNA damage levels were measured in gastric mucosal cells from antrum and corpus by the Comet assay. Levels of DNA damage in gastric mucosa cells from antrum and corpus of H. pylori-infected patients with mild, moderate, severe gastritis, and gastric cancer were significantly higher compared to uninfected normal mucosa cells. A study was conducted to determine the optimal time for collecting the breath examination urea breath marked with the stable isotope 13C. We selected patients before undergoing the examination of endoscopy at the Endoscopy Section of the University Hospital of Botucatu - SP. A screening was performed to determine which patients wanted and could participate. Before performing endoscopy basal sample was collected from the patient and then the labeled urea ingested. The blows were collected in double every 2.5 minutes until an interval of 30.0 minutes after were collected every 5.0 minutes until the time of 45.0 minutes . The samples were analyzed in a mass spectrometer for isotope ratio, located in the center of Stable Isotopes, Institute of Biosciences, UNESP - Botucatu campus. The data were studied and arranged in the form of graphics to better interpretation of results. The association of Helicobacter pylori with different gastric diseases has been continuously discussed in the literature, whereas the last 20 years a large body of publications was intended to characterize the ecological niches and habitats of this pathogen being the mouth a possible habitat for that rod. The present study aimed to discuss the occurrence of this organism as part of the supplemental or transient microbiota of the mouth. Thus, an extensive review of literature covering the period 1990-2014 was carried out with different databases, yielding 5024 articles discussing transmission and occurrence of H. pylori, in English or Portuguese. Articles were selected according to the inclusion and exclusion criteria and data were tabulated and analyzed. Despite the significant heterogeneity of the literature, it was found this microaerophilic has a worldwide distribution, even in the mouth, which behaves as a reservoir for gastric reinfection. The role of oral hygiene and periodontal disease in the distribution of this microorganism remains unclear, but most studies support the hypothesis that oral biofilms and inflammation of periodontium may facilitate the installation of this pathogen in the mouth. Studies suggest that this microorganism could collaborate as a predisposing factor for oral cancer.

It has been reported that the phage therapy is effective in controlling the number of colony-forming unit (CFU) of Salmonella spp. in chicken gut. This paper describes the protective effect of phage and Lactobacilli administration on Salmonella infection in 1-day-old chicks. We administered the bacteriophage P22 in a single dose and a probiotic mixture of four species of bacteriocin-producing Lactobacillus once a day for one week. Samples were analyzed every 48 hours, and intestinal eradication of S. Typhimurium was confirmed after treatments. We observed an increase in the size of duodenal villi and cecal crypts, as well as an increase in body weight in groups that received daily doses of Lactobacilli. This study confirms the efficiency of bacteriophage therapy in controlling salmonellosis in chicks and the beneficial effect of Lactobacilli mixtures in the weight gain of the birds.

A busca por novos agentes antimicrobianos é muito importante, uma vez que as bactérias possuem um alto potencial de recombinação genética, levando a um aumento de cepas resistentes aos fármacos antimicrobianos já existentes. Nesse contexto, os cogumelos podem ser uma alternativa, uma vez que extratos e constituintes químicos destes apresentam atividade frente a diversos isolados patogênicos e utilizados na medicina popular. Dentre as espécies de maior interesse estão o Agaricus blazei (cogumelo do sol), Agaricus bisporus (champignon de Paris) e Lentinus edodes (shitake). O Helicobacter pylori é responsável por doenças gastrointestinais de extrema importância para a saúde pública, como úlceras, gastrites até tumores malignos. O tratamento mais utilizado nos controles deste micro-organismo não é eficaz. Os cogumelos aparecem como alternativa para novas pesquisas neste ramo podendo vir a ser utilizados no desenvolvimento de novo fitoterápico ou mesmo como adjuvante deste tratamento. Existem diversos métodos para determinar a atividade antimicrobiana, entre eles a microdiluição. Esta técnica vem sendo muito utilizada principalmente devido à sua sensibilidade e pequena quantidade de reagentes e amostras, o que possibilita um maior número de réplicas, aumentando a confiabilidade dos resultados. Em continuidade ao estudo do nosso grupo de pesquisa este trabalho objetiva determinar o potencial anti-H. pylori de espécies de cogumelos comestíveis instigados pela possibilidade de encontrar uma alternativa para o tratamento desse micro-organismos dada a importância do mesmo na saúde pública. This work had as objective tests therapeutic treatments seeking the monogenea eradication in fingerlings Florida Pompano (Trachinotus carolinus) servants in cage net in the area of Ubatuba, State of São Paulo. The fingerlings presented an accentuated weigh loss and strong discoloration and through observation of scraped of gills under light microscope were identified a high amount of monogenea. Three treatments were tested in form of baths: T1 (fresh water for 5 minutes); T2 (formalin: 1: 1.000 for 20 minutes) and T3: (formalin: 1:4.000 for 30 minutes). The treatments were appraised through scraped of gills, mounted among you laminate and laminulas and observed to the light microscope. It was observed that in the tested conditions all of the treatments were efficient in the elimination of the monogenea without presenting lethality to the fish. The treatment is recommended T I (take a bath in fresh water for 5 minutes) for the facility in the application and for the absence of use of chemical products.
