Despite vast efforts and expenditures in the past few decades, malaria continues to kill millions of persons every year, and new approaches for disease control are urgently needed. To complete its life cycle in the mosquito, Plasmodium, the causative agent of malaria, has to traverse the epithelia of the midgut and salivary glands. Although strong circumstantial evidence indicates that parasite interactions with the two organs are specific, hardly any information is available about the interacting molecules. By use of a phage display library, we identified a 12-aa peptide-salivary gland and midgut peptide 1 (SM1)-that binds to the distal lobes of the salivary gland and to the luminal side of the midgut epithelium, but not to the midgut surface facing the hemolymph or to ovaries. The coincidence of the tissues with which parasites and the SM1 peptide interact suggested that the parasite and peptide recognize the same surface ligand. In support of this hypothesis, the SM1 peptide strongly inhibited Plasmodium invasion of salivary gland and midgut epithelia. These experiments suggest a new strategy for the genetic manipulation of mosquito vectorial capacity.

Currently there is very little information on the response of fruiting perennial plants to applied P. This is especially true for tropical production areas where soils have a high capacity of P fixation, and are poor in native phosphorus. An alternative to soil P fertilization, which is inefficient in fixing soils, is to apply phosphorus as a foliar spray. P is quickly absorbed by leaves, and is redistributed quite well through the plants because its phloem mobility, and foliar application may be a viable practice. The purpose of this present work, is to determine the effectiveness of foliar P application on the nutritional status and yield of guava. The experiment was done in a Typic Hapludox, for three consecutive agricultural years, in an adult orchard of 'Paluma' guava. Five treatments were tested: four rates leaf applications of P (0-0.5-1.0 and 2.0% of P2O5) and a control where P was applied to soil (200 g of P2O5/plant). Through the results it was verified that the foliar application of P altered the concentration of the nutrient in the soil (13 to 48 mg dm-3 P-resin), and in the guava leaves (1.2 to 1.8 g of P kg-1), but did not affect the production of fruits. In conclusion, in field conditions, it is viable to combine the phosphorus foliar fertilization with disease control, without increasing the operations and, consequently, the production cost.

Postbloom fruit drop (PFD) of citrus, caused by Colletotrichum acutatum, produces orange-brown lesions on petals and results in premature fruit drop and the retention of calyces. C. gloeosporioides is common in groves and causes postharvest anthracnose on fruit. Both diseases are controlled effectively by the fungicide benomyl in research fields and commercial orchards. Highly sensitive and resistant isolates of C. gloeosporioides were found, whereas all isolates of C. acutatum tested were moderately resistant. In preliminary studies conducted in vitro with three isolates of each, mycelial growth of sensitive isolates of C. gloeosporioides was inhibited completely by benomyl (Benlate 50 WP) at 1.0 μg/ml, whereas resistant isolates grew well at 10 μg/ml. Growth of all isolates of C. acutatum was inhibited by about 55% at 0.1 μg/ml and by 80% at 1.0 μg/ml. Spore germination of C. acutatum was inhibited more at 0.1 μg/ml than at 1.0 μg/ml or higher concentrations. In all, 20 isolates of C. acutatum from 17 groves and 20 isolates of C. gloeosporioides from 7 groves were collected from locations with different histories of benomyl usage in São Paulo, Brazil, and Florida, United States. Benomyl at 1.0 μ.g/ml completely inhibited growth of 133 isolates of C. gloeosporioides, with the exception of 7 isolates that were highly resistant to the fungicide, whereas all isolates of C. acutatum were only partially inhibited at 0.1 and 1.0 μg/ml. Analysis of variance indicated that the sensitivity of the isolates of C. acutatum was not affected by benomyl usage or grove of origin, and country of origin had only minor effects. No highly resistant or sensitive isolate of C. acutatum was recovered. Partial sequencing of the β-tubulin gene did not reveal nucleotide substitutions in codons 198 or 200 in C. acutatum that usually are associated with benomyl resistance in other fungi.

Small lesions located in the skin might be treated using the laser system. The purpose of this is to report the therapy of benign eyelid tumors using argon laser. Forty-four benign eyelid tumors were treated using argon blue-green laser with spot size of 500 μm, power from 1000 to 1200 mW and 0.3 second exposure time. The eyelid tumors were located mainly in the upper eyelid (65.9%) and the skin tag was the most frequent treated lesion (43.2%). The average number of laser shots to treat the lesions was 165. There was not observed any complication and all patients were satisfied with the results. The authors are considering the argon laser a benefit therapeutic method to treat benign tumors located in the eyelids.

Plan disease control techniques are used through the irrigation water, which reduces the labor and it improves application uniformity with smaller contact of the operator with toxic products, lower environmental impact and lower production cost. In order to control Botrytis cinerea the lisianthus culture, this work aimed to evaluate two fungicide application methods with different treatments. The fungicides were: thiophanate methyl (50 g i.a. L-1), thiophanate methyl + chlorothalonil (50 g i.a. L-1 + 35 g i.a. L-1) and iprodione (50 g i.a. L-1). Number of lesions, number of diseased floral buttons and height of the lisianthus plants were evaluated. It was possible to deduce, that in the growth stage (number of lesions in the plant) as well as in the final stage (number of floral buttons) of the lisianthus culture, the most efficient treatments were 2 (thiophanate methyl + chlorothalonil) and 3 (iprodione). Considering that treatment 2 is a mixture of two fungicides, a systemic and a contact one, independently of application methods, the mixture increased efficiency in relation to treatment 1 (thiophanate methyl). Thus, chemigation was as efficient as spreading technique.

Cysticercosis is one of the most important zoonosis, not only because of the effects on animal health and its economic consequences, but also due to the serious danger it poses to humans. The two main parasites involved in the taeniasis-cysticercosis complex in Brazil are Taenia saginata and Taenia solium. Differentiating between these two parasites is important both for disease control and for epidemiological studies. The purpose of this work was to identify genetic markers that could be used to differentiate these parasites. Out of 120 oligonucleotide decamers tested in random amplified polymorphic DNA (RAPD) assays, 107 were shown to discriminate between the two species of Taenia. Twenty-one DNA fragments that were specific for each species of Taenia were chosen for DNA cloning and sequencing. Seven RAPD markers were converted into sequence characterized amplified region (SCAR) markers with two specific for T. saginata and five specific for T. solium as shown by agarose gel electrophoresis. These markers were developed as potential tools to differentiate T. solium from T. saginata in epidemiological studies. © 2007 Elsevier Inc. All rights reserved.

CAE is caused by a lentivirus. The animals are mainly infected when taking contaminated colostrums and/or milk. This study proposed a CAE control strategy without sacrificing contaminated mothers. Thirty-nine female kids, born to CAE seropositive mothers were isolated from their mothers at birth and fed heat-treated colostrums and pasteurized milk from seronegative goats up to two months of age. All kids were submitted to three-monthly serological tests from birth to 12 months; seropositives were segregated from the herd. The control group consisted of 12 kids born to seropositive mothers that remained with their mothers. Diagnosis was the same, but seropositve animals were not segregated. At the end of 12 months, 34 (87%) animals from the experimental group remained seronegative with 76% to 98% confidence limits; in control group animals, the accumulated negativity rate was 17%, with 0% and 38% confidence limits. These results show that the proposed plan is viable to assure disease control in contaminated herds and that without it contamination can pass to animals born to infected goats.

The aim of this research was to evaluate the white mold severity (Sclerotinia sclerotiorum (Lib.) of Bary), bean production components and yield (Phaseolus vulgaris L.), variety Perola, according to the application of procimidone fungicide (Sialex 500), through fungigation (center pivot) and automotive sprayer (Uniport). The study was carried under field production commercial conditions, in Primavera do Leste - MT - Brazil. The experiment consisted of 5 treatments (with 4 repetitions of 4 ha each), all with two procimidone applications (1.2 kg ha-1 each application, same as, 0.6 kg a.i. per hectare) to the 42 and 52 days after seeding. The water depths of 5.5 and 11.0 mm were tested in the application through central pivot (this had your checked uniformity), providing volumes of 55.000 and 110.000 L ha-1, respectively, and the volumes of 120 and 200 L ha-1 in the automotive sprayer. The severity of disease was evaluated by the percentage of the area affected by plant damage using diagramatic grade scale of white mold severity, as described by Azevedo (1998). The values were used to calculate the area under the disease progress curve (AUDPC). They were also analyzed, the number of the fungus apothecia during the crop cycle and the residual sclerotias weight in harvest. On this occasion, it was also evaluated the crop yield parameters: number of plants per plot (final stand), pods per plant, grains per pod, medium weight of 200 grains and productivity of grains. The AUDPC values, apothecia to 42, 49 and 56 days after seeding, sclerotias in 2 soil kg and the crop productivity parameters were submitted to the variance analysis and Tukey Test at 0.05 of probability. This test was also applied in the comparison among the different fungicide application methods, independent of spray volumes in each one. The statistical processing was accomplished by STAT program. The results showed that weren't differences among application techniques studied in relation to productivity parameters, however, best white mold control, smaller apothecia number to 49 and 56 days after seeding and smaller weight of residual sclerotias in the harvest were obtained with the fungigation, independently of the spray volume used.

In tropical conditions such as Brazil, depending on the local altitude, potatoes can be planted and harvested in every month of the year successively, which favors pest and disease incidence, especially aphids transmission viruses. Therefore, obtaining good yield depends on constant pest and disease control and quality potato seed acquisition. One of the main strategies to increase a healthier seed potato multiplication rate and production is the use of hydroponic systems, with or without substrates, in channels, pots or boxes. In 2005 and 2008, researches investigated several hydroponic systems with and without the use of substrates. In the hydroponic systems without substrate, the aeroponic system resulted in the highest multiplication rate, with 47 tubers plant-1 compared to NFT and DFT, 35 and 37 tubers plant-1, respectively. With the use of substrates, the pot system obtained better results, reaching 12 tubers plant-1, followed by the capillary system and boxes, with 8 and 7 tubers plant-1, respectively. Potato seed production was influenced by hydroponics systems. Among hydroponic systems with substrate, the pots were shown as the best option. However, higher yields were obtained in the hydroponics systems without substrate, with the aeroponic system that provided the best results.

Purpose: To assess the safety and efficacy of transitioning patients whose intraocular pressure (IOP) had been insufficiently controlled on prostaglandin analog (PGA) monotherapy to treatment with travoprost 0.004%/timolol 0.5% fixed combination with benzalkonium chloride (TTFC). Methods: This prospective, multicenter, open-label, historical controlled, single-arm study transitioned patients who had primary open-angle glaucoma, pigment dispersion glaucoma, or ocular hypertension and who required further IOP reduction from PGA monotherapy to oncedaily treatment with TTFC for 12 weeks. IOP and safety (adverse events, corrected distance visual acuity, and slit-lamp biomicroscopy) were assessed at baseline, week 4, and week 12. A solicited ocular symptom survey was administered at baseline and at week 12. Patients and investigators reported their medication preference at week 12. Results: Of 65 patients enrolled, 43 had received prior travoprost therapy and 22 had received prior nontravoprost therapy (n = 18, bimatoprost; n = 4, latanoprost). In the total population, mean IOP was significantly reduced from baseline (P = 0.000009), showing a 16.8% reduction after 12 weeks of TTFC therapy. In the study subgroups, mean IOP was significantly reduced from baseline to week 12 (P = 0.0001) in the prior travoprost cohort (19.0% reduction) and in the prior nontravoprost cohort (13.1% reduction). Seven mild, ocular, treatment-related adverse events were reported. Of the ten ocular symptom questions, eight had numerically lower percentages with TTFC compared with prior PGA monotherapy and two had numerically higher percentages with TTFC (dry eye symptoms and ocular stinging/burning). At week 12, TTFC was preferred over prior therapy for 84.2% of patients (48 of 57) by the patients themselves, and for 94.7% of patients (54 of 57) by their physicians. Conclusion: When TTFC replaced PGA monotherapy in patients whose IOP had been uncontrolled, the outcome was a significant reduction in IOP and an acceptable safety and tolerability profile. The angular leaf spot of dry bean caused by the fungus Phaeoisairopsis griseola is a very important disease on dry bean crops in Brazil. In propitious weather conditions, susceptible cultivars have significant losses due to this disease. In these conditions, fungicide usage is an important step for angular leaf spot management. Since the effects of this method of control on plant physiology are not known, this study had the following objectives: a) Verify the action of fungicide in physiological changes in dry beans (photosynthesis rate and stomatal conductivity); b) evaluate the efficacy of some fungicides for the control of angular leaf spot; and c) examine the effects of these fungicides on other agronomic parameters; weight of 100 grains, number of grains per pod and number of pods per plant. The dry bean cultivar used in this study was Perola. Four trials were conducted, two in a net greenhouse and two trials under field conditions. A spray of piraclostrobin, tebuconazol and tebuconazol+ trifloxistrobin increased photosynthesis rates, although stomatal conductivity was not changed. In addition, sprays of tebuconazol, piraclostrobin, metiram+piraclostrobin and azoxistrobin provided better levels of disease control and also higher weight of 100 grains, with the exception of azoxystrobin for this parameter.

Fungicide application is one of the control strategies of fungal diseases in corn leaves. In Brazil, there are no fungicides recorded for the control of corn macrospora leaf spot (MLS). The aim of this study was to assess the efficacy of 16 fungicides on MLS control in a protective, curative and eradicant form. Fungicides of the chemical groups of benzimidazoles, strobilurins and triazoles were used alone or in mixture, in completely randomized block design, with six replicates of five plants, totaling 30 plants per treatment. The experiments were carried out in a greenhouse with the single-cross hybrid AS 1565 in phenological stage of two to six expanded leaves, using an isolate of S. macrospsora from the same hybrid. The inoculum was deposited into the cartridge of plants at 48 hours after, 48 hours before and 10 days before fungicide applications for preventive, curative and eradicant action, respectively. The experiment was repeated twice. The data underwent analysis of variance (p<0.05), and the means of treatments were compared by using the Scott-Knott test (p<0.05). Severity was estimated at 21 days after inoculations. All fungicides significantly differed from the control treatment in the preventive, curative and eradicant action. For the preventive action, mean disease control was 85%. The mixture of triazoles plus strobilurins controlled, on average, 75% of the disease severity, while the isolated products such as strobilurins reduced it by 62%, benzimidazoles by 55% and triazoles by 38% for the curative action. The lowest control was obtained for the eradicant action, with mean reduction of 40.1% in the disease severity and no significant difference among fungicides.

Background: Obesity in infancy and adolescence has acquired epidemic dimensions worldwide and is considered a risk factor for a number of disorders that can manifest at an early age, such as Metabolic Syndrome (MS). In this study, we evaluated overweight, obese, and extremely obese adolescents for the presence of MS, and studied the prevalence of single factors of the syndrome in this population. Methods. A total of 321 adolescents (174 females and 147 males) aged 10 to 16 years, attending the Adolescent Outpatient Clinic of Botucatu School of Medicine, Brazil, between April 2009 and April 2011 were enrolled in this study. Adolescents underwent anthropometric evaluation (weight, height, and abdominal circumference) and Body Mass Index (BMI) was estimated according to age and gender, following Disease Control and Prevention Centers recommendations (CDC, 2000). Blood pressure was measured and individuals with BMI ≥ 85§ssup§th§esup§ percentile were submitted to laboratory evaluation for Total Cholesterol, HDL and LDL Cholesterol, Triglycerides, Fasting Insulinemia, and Fasting Glycemia to identify MS factors, according to the criteria suggested by the International Diabetes Federation. Insulin resistance was calculated by HOMA-IR, Quicki, and Fasting Glycemia/Fasting Insulinemia (FGI). Results and discussion. Of the 321 adolescents, 95 (29.6%) were overweight, 129 (40.2%) were obese, and 97 (30.2%) were extremely obese. Around 18% were diagnosed with MS. The most prevalent risk factors were abdominal circumference ≥90§ssup§th§esup§ percentile (55%), HDL < 40 mg/dL (35.5%), High Pressure ≥130/85 mm/Hg (21%), Triglycerides ≥150 mg/dL (18.5%), and Fasting Glycemia ≥100 mg/dL (2%). Insulin resistance was observed in 65% of the adolescents. Conclusion: An increased prevalence of overweight and obesity, together with cardiometabolic risk factors such as dyslipidemia and abnormal blood pressure, were observed in adolescents, contributing to the onset of metabolic syndrome at younger ages.
