Validation of Relapse Risk Biomarkers for Routine Use in Patients With Juvenile Idiopathic Arthritis

Objective. The myeloid-related proteins 8 and 14 (MRP-8/MRP-14) and neutrophil-derived S100A12 are biomarkers of inflammation. They can be used to determine the relapse risk in patients with juvenile idiopathic arthritis (JIA) after stopping antiinflammatory treatment. In this study, we tested the performance of different enzyme-linked immunosorbent assays (ELISAs) in order to validate systems available for routine use.Methods. MRP-8/MRP-14 and S100A12 serum concentrations of 188 JIA patients in remission were analyzed. Commercially available test systems were compared to experimental ELISAs established in house. The ability of the assays to identify JIA patients at risk for relapse was analyzed.Results. For MRP-8/MRP-14, the PhiCal Calprotectin and Buhlmann MRP8/14 Calprotectin ELISAs revealed hazard ratios of 2.3 and 2.1, respectively. For S100A12, the CircuLex S100A12/EN-RAGE ELISA revealed a hazard ratio of 3.1. The commercial assays allowed a JIA relapse prediction that was at least comparable to the experimental ELISAs.Conclusion. For the prediction of JIA relapse after stopping medication, the biomarkers MRP-8/MRP-14 and S100A12 can be determined by using assays that are available for routine use. The tested commercial MRP-8/MRP-14 and S100A12 ELISAs showed a performance comparable to well-established experimental ELISA protocols when assay-specific cutoffs for the indication of relapse prediction are thoroughly applied.

Glomerulitis and peritubular capillaritis have been recognized as important lesions in acute renal rejection (AR). We studied glomerulitis and peritubular capillaritis in AR by 2 methods and investigated associations with C4d, type/grade of AR, and allograft survival time. Glomerulitis was measured according to Banff scores (glomerulitis by Banff Method [gBM]) and by counting the number of intraglomerular inflammatory cells (glomerulitis by Quantitative Method [gQM]). Capillaritis was classified by the Banff scoring system (peritubular capillaritis by Banff Method [ptcBM]) and by counting the number of cells in peritubular capillaries in 10 high-power fields (hpf; peritubular capillaritis by Quantitative Method [ptcQM]). These quantitative analyses were performed in an attempt to improve our understanding of the role played by glomerulitis and capillaritis in AR. The g0 + g1 group (gBM) associated with negative C4d (P = .02). In peritubular capillaritis, a larger number of cells per 10 hpf in peritubular capillaries (ptcQM) were observed in positive C4d cases (P = .03). The group g2 + g3 (gBM) correlated with graft loss (P = .01). Peritubular capillaritis was not significantly related to graft survival time. Our study showed that the Banff scoring system is the best method to study glomerulitis and observed that the evaluation of capillaritis in routine biopsies is difficult and additional studies are required for a better understanding of its meaning in AR biopsy specimens of renal allografts.

Some changes in the application of the numeric trapezoidal integration are analyzed for applications considering pi circuits. It is considered numeric and computational proceedings for improving the numeric results obtained with associations of pi circuits. In numeric integration solutions of the linear systems, it is common to represent these associations of pi circuits by only one matrix. This representation introduces undesirable numeric oscillations in simulations of the dynamics of wave propagation in electrical systems. The proposed changes improve the results of application of cascades of pi circuits associated to the trapezoidal integration, avoiding that the numerical oscillations, or Gibb's oscillations, have high values and are slowly damped. For the carried out simulations, different number of pi circuits and voltage sources are checked, confirming the reduction of the influence of the numeric oscillations on the obtained results. Presents a citation analysis of indexing research in the two Proceedings. Understanding that there are different traditions of research into indexing, we look for evidence of this in the citing and cited authors. Three areas of cited and citing authors surface, after applying Price's elitism analysis, each roughly corresponding to geographic distributions.

This work presents the development of a graphical interface to the Lock-in Amplifier, which is used in physiological studies on the motility of the gastrointestinal tract in rats and signal processing. With a simple and low cost instrumentation, the resources offered by the virtual interface of LabVIEW software allows the creation of commands similar to the actual instrument that, through communication via standard serial port, transmits data between a PC and peripheral device performing specific and particular needs in the amplifier. Created for the lock-in amplifier model SR830 Stanford Research Systems, the remote manipulation gives the user greater accessibility in the process of configuration and calibration. And, since the software is installed, there is the advantage of eliminating the need of purchase new devices to upgrade the system. The commands created were made to perform six basic modifications that are used in routine of the Biomagnetism Laboratory. The instrumentation developed has the following controls: Amplitude, Frequency, Time Constant, slope low pass filter, sensitivity and offset

Magazines such as O Cruzeiro are part of the Brazilian collection. In its pages, the female presence is constant. From the concepts of Greimas’ way of life and happening and routine from Zilberberg, we analyse a photoreport which highlights the actress Ilka Soares, comparing her to texts of the session “To the woman”. The analysis of such texts lets us verify the behaviour of a distinguished professional which is different from the traditional model of the housewife of the fifties. Reconstitute the ways of life of these women is to make up for the memory of the country
