Introduction: Biocompatibility of root canal sealers is important because of the long-term contact of their eluates and/or degradation products with periapical tissues. The literature still lacks studies about the genotoxic effects of these materials and the influence of setting time on biological properties. The cytotoxicity and genotoxicity of an epoxy resin-based sealer (AH Plus), a single methacrylate-based sealer (EndoRez), and a silicone-based sealer (RoekoSeal) were assessed. Methods: Chinese hamster fibroblasts (V79) were cultured and exposed to different dilutions of extracts from the sealers that were left to set for 0, 12, and 24 hours before contact with culture medium. Cell viability was measured by the methyl-thiazol-diphenyltetrazolium assay. Genotoxicity was assessed by the comet assay. Data were statistically analyzed by Kruskal-Wallis and Dunn tests (P < .05). Results: Root canal sealers were statistically more cytotoxic than the untreated control group, except for the silicon-based sealer. Cell viability ranking was the following (from the most to the least cytotoxic): methacrylate-based > epoxy resin-based > silicone-based. The setting time influenced the epoxy resin-based sealer cytotoxicity (decreased at 12 hours) and the general genotoxicity (increased at 24 hours). DNA damage ranking was the following (from the most to the least genotoxic): methacrylate-based > silicone-based = epoxy resin-based. Conclusions: The setting time had influence on the cytotoxicity of the epoxy resin-based sealer and genotoxicity of all tested sealers. The methacrylate-based sealer was the most cytotoxic, and the silicone-based sealer was not cytotoxic. Genotoxicity was observed for all sealers. © 2013 American Association of Endodontists.

Central α2-adrenoceptors and the pontine lateral parabrachial nucleus (LPBN) are involved in the control of sodium and water intake. Bilateral injections of moxonidine (α2-adrenergic/imidazoline receptor agonist) or noradrenaline into the LPBN strongly increases 0.3 M NaCl intake induced by a combined treatment of furosemide plus captopril. Injection of moxonidine into the LPBN also increases hypertonic NaCl and water intake and reduces oxytocin secretion, urinary sodium, and water excreted by cell-dehydrated rats, causing a positive sodium and water balance, which suggests that moxonidine injected into the LPBN deactivates mechanisms that restrain body fluid volume expansion. Pretreatment with specific α2-adrenoceptor antagonists injected into the LPBN abolishes the behavioral and renal effects of moxonidine or noradrenaline injected into the same area, suggesting that these effects depend on activation of LPBN α2-adrenoceptors. In fluid-depleted rats, the palatability of sodium is reduced by ingestion of hypertonic NaCl, limiting intake. However, in rats treated with moxonidine injected into the LPBN, the NaCl palatability remains high, even after ingestion of significant amounts of 0.3 M NaCl. The changes in behavioral and renal responses produced by activation of α2-adrenoceptors in the LPBN are probably a consequence of reduction of oxytocin secretion and blockade of inhibitory signals that affect sodium palatability. In this review, a model is proposed to show how activation of α2-adrenoceptors in the LPBN may affect palatability and, consequently, ingestion of sodium as well as renal sodium excretion.

The objective of this work was to evaluate the development, survival, reproductive capacity, and longevity of the Asian ladybug Harmonia axyridis in comparison with Cycloneda sanguinea and Hippodamia convergens. Coccinellid larvae and adults were fed daily with Schizaphis graminum. Ten couples of each species were isolated for evaluation of the adult phase. The duration of the larval stage of H. axyridis is the longest (10.2 days) and its adults are the heaviest (29.7 mg) compared with C. sanguinea and H. convergens. The three species showed similar percentages of survival during the developmental stages. An average of 82% of C. sanguinea, H. axyridis, and H. convergens larvae reached adulthood, which indicates that temperature (25°C) and the offered prey are favorable to coccinellid development. Harmonia axyridis produces a higher total number of eggs per female (1,029.2) than the other evaluated species. However, H. axyridis, which lives for an average of 147.2 days, does not show a significantly greater longevity than C. sanguinea (87.2 days) and H. convergens (134.3 days).

Despite the importance of plant pathogens belonging to the Order Pucciniales (=Uredinales), popularly called rusts, certain peculiarities of its biology is poorly known, mainly due to the difficulty of growing them in culture media and also by the lack of appropriate methodologies for the development of research. Therefore, this study was to verify the efficiency of apparatus called germinatelia to obtain data about the basidiospores production biology of different Pucciniales species. Were used in the study Puccinia pampeana, P. psidii, P. pelargonii-zonalis, P. arachidis, P. mogiphanis, P. emiliae, P. hetereospora, P. malvacearum and P. cnici-oleracei. The use of germinatelia have revealed high basidiospores production for a period up to 5 months at temperatures between 12 degrees and 21 degrees C. Furthermore, in P. malvacearum was possible to record the germination by repetition (survival strategy) in 4.5% of basidiospores produced. All species were favored to mild temperatures. These results therefore demonstrate that the use of germinatelios is efficient, assisting in obtaining data about the biology and cycles of different Puccinia species, enabling better understanding of these pathogens, resulting in management and control measures most appropriate/accurate.

Parasitic diseases constitute one of the main problems affecting livestock; however the use of chemical medicaments provides drug resistance residues in animal and environmental contamination. Changes in production concepts require that food must be produced in hygienic conditions, per healthy animals and that are not eliminating antibiotic residues, pesticides or other drugs. This scenario has favored organic production and the use of medicinal plants. For the control of endoparasites, several studies have published the benefits of Azadirachta indica A. Juss., Punica granatum Linn., Musa sp., Operculina hamiltonii G. DON., propolis, among others. However, despite the existence in- vitro studies that demonstrate the pharmacological properties of phytotherapics, there is still need for clinical trials to determine dosage and its effects in- vivo. Investigations of new bioactive natural substances can be of great value for the control of animal health and food safety, which is particularly important for organic production systems in which the use of chemical drugs is a limiting factor for certification.

In this study, we evaluated the efficiency of six isolates of Saccharomyces cerevisiae in controlling Colletotrichum acutatum, the causal agent of postbloom fruit drop that occur in pre-harvest citrus. We analyzed the mechanisms of action involved in biological control such as: production of antifungal compounds, nutrient competition, detection of killer activity, and production of hydrolytic enzymes of the isolates of S. cerevisiae on C. acutatum and their efficiency in controlling postbloom fruit drop on detached citrus flowers. Our results showed that all six S. cerevisiae isolates produced antifungal compounds, competed for nutrients, inhibited pathogen germination, and produced killer activity and hydrolytic enzymes when in contact with the fungus wall. The isolates were able to control the disease when detached flowers were artificially inoculated, both preventively and curatively. In this work we identified a novel potential biological control agent for C acutatum during pre-harvest. This is the first report of yeast efficiency for the biocontrol of postbloom fruit drop, which represents an important contribution to the field of biocontrol of diseases affecting citrus populations worldwide. Rot caused by Fusarium pallidoroseum has had a severely negative impact on the export of melons from Brazil. Uncertainty regarding the health of the fruit due to the quiescent infection of the pathogen has led producers to use fungicides in the postharvest treatment of the fruit, thereby causing contamination and risking the health of consumers. Consequently, there is a demand for clean and safe natural technologies for the postharvest treatment of melons, including biological control. The present study aimed at evaluating bioagents for use in controlling Fusarium rot in 'Galia'melon. The following bioagents were evaluated: two isolates of Bacillus subtilis, B. licheniformis and a mixture of B. subtilis and B. licheniformis, as well as the yeasts Sporidiobolus pararoseus, Pichia spp., Pichia membranifaciens, P. guilliermondii, Sporobolomyces roseus, Debaryomyces hansenii and Rhodotorula mucilagenosa. Treatment with imazalil and water were used as controls. Two experiments were conducted in a completely randomised design with 10 replicates per treatment with four fruit per replicate; the disease incidence was evaluated in the first experiment, and the disease severity was evaluated in the second. Similarity analysis of the temporal evolution profiles of rot incidence caused by F. pallidoroseum allowed the evaluated treatments to be clustered into four groups. In the first experiment, the yeasts P. membranifaciens and D. hansenii produced results similar to that of the fungicide imazalil. The second experiment highlighted the yeasts P. guilliermondii and R. mucilaginosa. Electron microscopy studies confirmed that once applied to the fruit, the yeasts colonised the skin and damaged the pathogen mycelium; the action of the yeasts affected the mycelium of F. pallidoroseum, which had infected wounds on the fruit's surface. Bacillus spp. did not provide good disease control. These results demonstrated that yeasts have the potential to control postharvest rot caused by F. pallidoroseum in 'Galia'melon.

The biological behavior of a new bioactive material composed of calcium-deficient hydroxyapatite, octacalcium phosphate, and beta-tricalcium phosphate was investigated by in vitro indirect and direct cytotoxicity, cell adhesion and proliferation tests, and by in vivo subcutaneous and bone implantation in rats. The results of the in vitro studies showed that the material is biocompatible and no cytotoxic. Slightly poorer initial cell adhesion and lower cell proliferation than in control was observed, which were attributed to the reactivity and roughness of the material surface, In vivo results showed that the material is biodegradable and bioactive in bone tissue, but only biocompatible and partially biodegradable in soft tissue.

The bronze bug is an invasive Australian pest that has reached eucalypt production areas worldwide in <10 years. The fi rst record in South America was in 2005 in Argentina. Collaboration in the region towards a unifi ed strategy for the management of the bronze bug started soon after the dispersal of the pest into Brazil and Uruguay was confi rmed. Here, we present the main achievements of this collaboration in four main topics: 1) biology of the pest, 2) monitoring, 3) biological control, and 4) cooperative networks. Two mass rearing procedures have been implemented in the region with relative success, allowing basic biological studies on the pest. Continuous monitoring in the region for >5 years has provided a reasonable knowledge on seasonal patterns of T. peregrinus. Biological control strategies developed include the use of local natural enemies of T. peregrinus, the development of biopesticides, and the introduction of Cleruchoides noackae, an egg parasitoid of T. peregrinus from Australia. We review the main achievements in each country. Finally, a regional network of institutions, researchers, and students has strengthened in the region, providing a solid background for future collaboration.
