Sugarcane workers in Brazil are exposed to various genotoxic compounds, including polycyclic aromatic hydrocarbons (PAHs), derived from an incomplete combustion process of burnt sugarcane fields. The effects of the occupational exposure to sugarcane fields burning were measured in urine samples of sugarcane workers from the northwest of the State of São Paulo when exposed (harvesting) and when non-exposed (non-harvesting). The urinary levels of 1-hydroxypyrene (1-OHP) and the influence of the genetic polymorphisms CYP1A1, GSTM1, GSTT1 and GSTP1 were evaluated. Our results showed that the 1-OHP levels were significantly higher (P < 0.0000) in the exposed sugarcane workers (0.318 mu mol mol(-1) creatinine) than in the non-exposed workers (0.035 mu mol mol(-1) creatinine). In an unvaried analysis, no influence regarding the polymorphisms was observed. However, multivariate regression analysis showed that the CYP1A1*4 polymorphism in the exposed group, and age and the GSTP1 polymorphism in the non-exposed group significantly influenced urinary 1-OHP excretion levels (P < 0.10). The same group of sugarcane workers was significantly more exposed to PAHs during the harvesting period than during the non-harvesting period. (c) 2006 Elsevier B.V. All rights reserved.

Sea anemones contain a variety of biologically active substances. Bunodosoma caissarum is a sea anemone from the Cnidaria phylum, found only in Brazilian coastal waters. The aim of the present work was to study the biological effects of PLA(2) isolated from the sea anemone B. caissarum on the isolated perfused kidney, the arteriolar mesenteric bed and on insulin secretion. Specimens of B. caissarum were collected from the Sao Vicente Channel on the southern coast of the State of São Paulo, Brazil. Reverse phase HPLC analysis of the crude extract of B. caissarum detected three PLA(2) proteins (named BcPLA(2)1, BCPLA(2)2 and BcPLA(2)3) found to be active in B. caissarum extracts. MALDI-TOF mass spectrometry of BcPLA(2)1 showed one main peak at 14.7 kDa. The N-terminal amino acid sequence of BcPLA(2)1 showed high amino acid sequence identity with PLA(2) group III protein isolated from the Mexican lizard (PA23 HELSU, HELSU, PA22 HELSU) and with the honey bee Apis mellifera (PLA(2) and 1POC_A). In addition, BcPLA(2)1 also showed significant overall homology to bee PLA(2). The enzymatic activity induced by native BCPLA(2)1 (20 mu g/well) was reduced by chemical treatment with p-bromophenacyl bromide (p-BPB) and with morin. BcPLA(2)1 strongly induced insulin secretion in presence of high glucose concentration. In isolated kidney, the PLA(2) from B. caissarum increased the perfusion pressure, renal vascular resistance, urinary flow, glomerular filtration rate, and sodium, potassium and chloride levels of excretion. BcPLA(2)1, however, did not increase the perfusion pressure on the mesenteric vascular bed. In conclusion, PLA(2), a group III phospholipase isolated from the sea anemone B. caissarum, exerted effects on renal function and induced insulin secretion in conditions of high glucose concentration. Crotalus durissus cascavella is a snake that is usually found in the scrublands of northeast Brazil. The components of its venom may have effects on the vascular and renal systems. Recently, a new bradykinin inhibitory peptide has been identified in the venom of the Crotalinae family. The aim of the present study was to investigate the renal and vascular effects of the natriuretic peptide isolated from the venom of Crotalus durissus cascavella (NP2_Casca). The chromatographic profile showed the fractionation of substances identified as convulxin, gyroxin, crotoxin and crotamine, as well as fractions V and VI. The electrophoretic profile of fraction V consisted of several bands ranging from approximately 6 kDa to 13 kDa, while fraction VI showed only two main electrophoretic bands with molecular weights of approximately 6 and 14 kDa. Reverse-phase chromatography showed that NP2_Casca corresponds to about 18% of fraction VI and that this fraction is the main natriuretic peptide. NP2_Casca was compared to other natriuretic peptides from other sources of snake venom. All amino acid sequences that were compared showed a consensus region of XGCFGX, XLDRIX and XSGLGCX. The group treated with NP2-Casca showed an increase in perfusion pressure, renal vascular resistance, urinary flow and glomerular filtration rate. The percent of total and proximal tubular transport of sodium was reduced significantly after administration of the peptide. The mean arterial pressure showed a dose-dependent decrease after infusion of NP2_Casca, and an increase in nitrite production. In the aortic ring assay, NP2_Casca caused a relaxant effect in endothelium-intact thoracic aortic rings precontracted with phenylephrine in the presence and absence of isatin. NP2_Casca failed to relax the aortic rings precontracted with an isosmotic potassium Krebs-Henseleit solution. In conclusion, the natriuretic peptide isolated from Crotalus durissus cascavella venom produced renal and vascular effects. NP2_Casca reduced total and proximal sodium tubular transport, leading to an increase in sodium excretion, thereby demonstrating a diuretic action. A hypotensive effect was displayed in an arterial pressure assay, with an increase in nitrite production, suggesting a possible vasoactive action. (C) 2008 Elsevier Ltd. All rights reserved.

Three species of phylogenetically related semi-terrestrial crabs (Superfamily Grapsoidea - Sesarma rectum, Goniopsis cruentata and Neohelice granulata (formerly: Chasmagnathus granulatus) with different degrees of terrestriality were studied to quantify the accumulation of copper (Cu) in hemolymph, gills, hepatopancreas and antennal gland, and its excretion through the faeces. These crabs were fed for 15 days practical diets containing 0 (A), 0.5 (B), 1.0 (C), and 1.5% (D) of added CuCl2 (corresponding to 0, 0.2, 0.5 and 0.7% of Cu2+, respectively). The amount of food ingested was directly proportional to the degree of terrestriality: S. rectum, the most terrestrial species, ate around 2-3 times more than the other crabs, whereas G. cruentata ate 1.5-2 times more than N. granulata, the least terrestrial. The amount of Cu excreted in the feces was proportional to Cu ingestion, and was 76.8% and 64.2% higher for Sesarma fed diet D compared to G. cruentata and N. granulata, respectively. Sesarma also displayed higher Cu concentration in the haemolymph, gills and antennal glands, but not in the hepatopancreas. A detoxifying mechanism followed by elimination was probably present at this last organ, preventing Cu accumulation. More terrestrial crabs, such as Sesarma, may accumulate more Cu in hemolymph and tissues, showing a correlation between metal accumulation and increased terrestriality. In this aspect, contaminated feed sources with Cu may have more impact in conservation of terrestrial crabs. (C) 2008 Elsevier B.V. All rights reserved.

Os objetivos deste trabalho foram determinar a absorção aparente, estimar as perdas endógenas fecais e a absorção real do Mg e determinar a ingestão ad libitum da água de beber e a concentração de Mg no soro sangüíneo de caprinos da raças Anglonubiana (AN) e Saanen (SN). Foram usados doze caprinos, seis de cada raça, com 19,8 kg PV médio. Dietas semipurificadas (baixo teor de Mg) à base de quirera de arroz, glúten de milho e celulose foram suplementadas com MgO, para se obterem os níveis de 0,05 (sem supplementação) 0,20 e 0,35% Mg (%MS). Os níveis de Mg influenciaram os coeficientes de absorção aparente de Mg e Ca, com valores médios de 57,8; 73,9; e 73,2% para Mg e 55,7; 39,6; e 49,5% para Ca, para dietas com níveis 0,05; 0,20; e 0,35% de Mg, respectivamente. Entretanto, para os coeficientes de absorção aparente de P, Na e K, não houve efeito de níveis de Mg na dieta. Os resultados de absorção real de Mg apresentaram interação de níveis de Mg e raças. A média para raça NA, no nível 0,05% Mg, foi de 61,0% e para os níveis 0,20 e 0,35% Mg, 77,2 e 73,2%, respectivamente. Entretanto, para a raça SN, as médias foram 73,3; 75,5; e 76,0%, para os mesmos níveis, sem diferenças. A digestibilidade de matéria seca, proteína bruta e extrato não-nitrogenado diminuiu com os níveis crescentes de Mg nas dietas. As excreções fecais (7,0; 20,8; e 34,4 mg/kg PV0,75.d) e urinárias (3,9; 30,8; e 44,6 mg/kg PV0,75.d) de Mg elevaram-se com o aumento dos níveis crescentes de Mg nas dietas. Houve, também, influência dos níveis de Mg dietético sobre as concentrações de Mg do soro sangüíneo (1,74; 2,23; e 2,80 mg/dL para níveis de 0,05; 0,20; e 0,35% de Mg, respectivamente).

The serum and urine Na+ and K+ levels and their relationship with electrocardiography characteristics,were studied in 15 male adult mongrel dogs, treated twice-a-day,with 2, 4, 8mg/kg of live weight doses of furosemide and furosemide associated with KCl during 35 days. Urine samples were analyzed weekly in order to determine the Na+ and K+ concentrations and to evaluate the urinary excretion of both cations. Electrocardiography was performed simultaneously, using limb derivations, speed 50mm/sec and calibration of 1 cm corresponding to 1mV. Data showed decreased seric concentrations of Na+ and Ki, increase in Na+ fractional excretion between days 7 and 21 of the treatment, slower heart rate and longer PR, QRS and QT intervals in the ECG. Briefly 2mg/kg furosemide associated with KCl was the most suitable treatment since it induced lesser side effects.

Na doença renal crônica (DRC) a manutenção da homeostase de água e sódio é o primeiro problema a ser contornado pelo organismo e com o agravamento das lesões renais surgem outros problemas graves relacionados à homeostase de cálcio e fósforo. O presente estudo tem por escopo avaliar a excreção renal de cálcio, fósforo, sódio e potássio, e o perfil sérico destes eletrólitos em cães normais e em cães com DRC naturalmente adquirida. Foram avaliados três grupos de cães adultos, machos ou fêmeas, de raças variadas. Animais normais compuseram o grupo controle (G1) e os cães com DRC foram distribuídos em dois grupos de acordo com os estágios de comprometimento da função renal (G2 e G3, respectivamente, estágios 1-2 e estágios 3-4, descritos pela IRIS 2006 staging CKD). Os cães do G3 apresentaram aumento das concentrações séricas de cálcio ionizado e fósforo, além de diminuição da concentração sérica de sódio. Quanto à excreção renal dos eletrólitos analisados, os animais dos grupos G1 e G2 apresentaram diminuição de carga filtrada e aumento de excreção fracionada, mas as excreções urinárias não variaram significativamente. Os resultados são indicativos de que os rins de cães com DRC podem manter a excreção urinária dos eletrólitos em valores se melhantes aos dos normais. O mecanismo envolve aumento da excreção fracionada na medida em que haja diminuição da filtração glomerular. Esse processo de compensação, entretanto, pode perder a eficiência nos estágios mais avançados da enfermidade no que se refere à manutenção das concentrações séricas de fósforo e sódio.

This trial was conducted to evaluate the utilization of low-protein diets formulated based on the ideal protein concept for broiler chickens from 7 to 21 days of age reared at different environmental temperatures. Nine hundred male Cobb-500® chickens were used. At day seven chicks were distributed according to a completely randomized design in a 3 x 3 factorial with four replications of 25 birds each. It was used three crude protein levels in the diet (21.5; 20.0 and 18.5%) and three environmental temperatures (low, thermoneutral and high). The performance, carcass characteristics (yield and chemical composition), and nitrogen ingestion and excretion were assessed. There was no significant interaction among the factors for the evaluated variables. Environmental temperatures affected differently chicken performance. High environmental temperature resulted in lower weight gain and higher wing fat percentage, whereas cold temperature resulted in higher feed conversion. on the other hand, low-protein diets decreased weight gain, breast yield, nitrogen excretion and influenced breast and wings chemical composition. Birds reared at high environmental temperature showed lower nitrogen intake and excretion. The results showed that the decrease in protein levels from 7 to 21 days of age contributed to lower nitrogen excretion in broiler chickens, but impaired performance and carcass characteristics independent of rearing temperature.

Foi realizado um experimento para avaliar a digestibilidade aparente de dietas e o metabolismo de suínos alimentados com dietas contendo bentonita sódica submetidos a diferentes programas alimentares. Foram utilizados 24 suínos machos castrados, meio irmãos paternos, com peso vivo médio inicial de 42,2kg, alojados em gaiolas metabólicas. O delineamento experimental foi o inteiramente casualizado com três níveis de bentonita sódica (0,0; 0,3 e 0,5%) e dois programas alimentares (restrito e à vontade), com oito repetições cada. A adição de bentonita sódica não alterou (P>0,05) o consumo de ração e os balanços da energia e do nitrogênio. O consumo de ração diferiu (P<0,01) entre a alimentação restrita e à vontade (1,17 vs. 2,19kg d-1). O programa alimentar alterou (P<0,05) o balanço do N, mas não afetou (P>0,05) a retenção de N pelos animais. A alimentação à vontade alterou (P<0,05) o balanço da energia, sobretudo a energia retida (3.825 vs. 3.013kcal d-1). A adição de 0,5% de bentonita sódica nas dietas reduziu em 9% (P<0,01) a excreção fecal de fósforo. A adição de bentonita sódica nas dietas de suínos não altera os balanços da energia e do N nem as digestibilidades aparentes do Ca, Mg, Zn, Cu e Mn. A adição de bentonita sódica reduz a excreção fecal de fósforo. Não há interação entre o programa alimentar e a adição de bentonita sódica nas dietas.

Foram utilizados 50 suínos machos castrados para avaliar o efeito da restrição alimentar qualitativa sobre a digestibilidade dos componentes dietéticos, os órgãos do trato digestório, a composição e a produção fecal. Foram utilizadas cinco dietas experimentais, com níveis de restrição qualitativa de 0, 5, 10, 15 e 20%. Houve redução linear (P<0,001) para todos os coeficientes de digestibilidade, com exceção da fibra em detergente ácido, que apresentou resposta quadrática (P<0,05). Os teores de sólidos totais (P<0,01) e voláteis (P<0,05), e minerais totais (P<0,001) nas fezes aumentaram com os níveis de restrição alimentar, enquanto os níveis de K (P<0,05), Cu (P<0,01) e de N, P, Na, Ca, Mg, Fe e Zn (P<0,001), apresentaram resposta quadrática. A excreção diária de fezes, sólidos totais e voláteis, minerais totais, N, P, K, Mn e Cu (P<0,001), Ca, Na, Mg e Fe (P<0,05) apresentaram aumento em função do nível da restrição alimentar qualitativa. A restrição qualitativa pode ser alternativa para destinação de resíduos da agroindústria, conferindo boas propriedades às fezes suínas, no que diz respeito à utilização para adubação de culturas.
