The comet assay is a method of DNA damage analysis widely used to quantify oxidative damage, crosslinks of DNA, apoptosis and genotoxicity of chemicals substances as chemical, pharmaceuticals, agrochemicals products, among others. This technique is suitable to detect DNA strand breaks, alkali-labile sites and incomplete excision repair sites and is based on the migration of DNA fragments by microeletroforesis, DNA migrates for the anode forming a “tail”, and the formed image has the appearance of a comet. The slides can be stained with fluorescence or silver, having differences in the microscopy type used for the analysis and the possibility of storage of the slides, moreover, the first one is a stained-method with more difficulties of accomplishment. The image analysis can be performed by a visual way, however, there is a disadvantage as the subjectivity on the results, that can be minimized by an automated method of digital analysis. This process was studied in this report with the aim to perceive the validation of the digital analysis turning it a quantitative method with larger reproductibility, minimizing the variability and imprecision due to the subjective analysis. For this validation we selected 50 comets photographed in a standardized way and printed, afterwards, pictures were submitted to three experienced appraisers, who quantified them manually. Later, the images were processed by free software ImageJ 1.38x, printed and quantified manually by the same appraisers. The intraclass correlation was higher to comet measures after image processing. Following, an algorithm of automated digital analysis from the measures of the comet was developed; the values obtained were compared with those 12 estimated manually after the processing resulting high correlation among the measures. Além de ser bom bioindicar a espécie Prochilodus lineatus (Characiformes, Prochilodontidae) também apresenta importância econômica, ecológica e ampla distribuição na região neotropical. A ecotoxicologia estuda a interação da química ambiental e a biota, sendo assim, este estudo teve como objetivo identificar alterações quantitativas de bile e glicogênio no fígado dos peixes, órgão responsável pelo processo de detoxificação, biotransformação e também pela reserva de nutrientes como o glicogênio e por secretar a bile. Os grupos foram separados em três tratamentos com o objetivo de comparar os danos causados aos peixes que vivem em áreas perturbadas, (como é o caso do Lago Azul – Rio Claro-SP), e os que vivem em águas com detergentes biodegradáveis diluídos, como na maioria dos nossos cursos d’água, comparados a um grupo controle (água clorada do poço artesiano da UNESP- Campus Rio Claro). Análises histológicas foram feitas com a utilização das técnicas de H-E e PAS, sendo o material coletado após sete e 20 dias de experimento, visando identificar os efeitos agudos de tais poluentes. A identificação de alterações na estrutura histológica foi realizada com o auxilio do programa ImageJ com o qual foi feita a quantificação da área ocupada pela bile e pelo glicogênio

In the second half of the last century the automobile industries suffered from the petroleum crisis caused mainly by the wars in the Middle East. These crises led the automakers rethink their vehicles. One of the most important events after that was the adoption of new steels by the industry. One example is the TRIP steels (Transformationinduced plasticity). It is known that the macroscopic behavior of a material is strongly dependent on its microstructure and therefore the quantitative metallography is important to understand and relate the material properties to its microstructure. In this work, different specimens of TRIP steels were etched using LePera reagent. The obtained images were analyzed using digital processing. Using the ImageJ software the methods threshold and watershed were studied as well as a comparison with the ASTM E562 standard. The methods were compared and finally the morphological characteristics and volumetric fraction of the retained austenite and martensite phases were analyzed. The results showed that the threshold led to a higher number of identified grains with lower mean area and total area fraction than the watershed method and ASTM standard

Insects of the suborder Heteroptera are known for their odor, for being pests, or for being disease carriers. To gain better insight into the cytogenetic characteristics of heteropterans, 18 species of terrestrial Heteroptera belonging to eight families were studied. The presence of heteropycnotic corpuscles during prophase I, terminal or interstitial chiasmas, telomeric associations between chromosomes, ring disposals of autosomes during metaphase, and late migrations of the sex chromosomes during anaphase were analyzed. These features showed identical patterns to other species of Heteroptera previously described in the literature. Another studied characteristic was chromosome complements. The male chromosome complements observed were 2n = 12 chromosomes [10A + XY, Galgupha sidae (Amyot & Serville) (Corimelaenidae) and Pachycoris torridus (Scopoli) (Scutelleridae)]; 2n = 13 [10A + 2m + X0, Harmostes serratus (Fabricius), Harmostes apicatus (Stål), Jadera haematoloma (Herrich-Schaeffer), Jadera sanguinolenta (Fabricius), Jadera sp. (Rhopalidae)], and Neomegalotomus parvus (Westwood) (Alydidae); 2n = 13 [12A + X0, Stenocoris furcifera (Westwood) (Alydidae); 2n = 14 [12A + XY, Dictyla monotropidia (Stål) (Tingidae)]; 2n = 19 [18A + X0, Acanonicus hahni (Stål) (Coreidae)]; 2n = 21 [18A + 2m + X0, Acanthocephala sp. (Dallas) (Coreidae)]; 2n = 27 [24A + 2m + X0, Anisoscelis foliacea marginella (Dallas) (Coreidae)]; 2n = 18 [16A + XY, Oncopeltus fasciatus (Dallas) (Lygaeidae)]; 2n = 17 [14A + X1X2Y, Oxycarenus hyalinipennis (Costa) (Lygaeidae)]; 2n = 16 [12A + 2m + XY, Pachybrachius bilobatus (Say) (Lygaeidae)]; 2n = 26 [24A + XY, Atopozelus opsinus (Elkins) (Reduviidae)]; and 2n = 27 [24A + X1X2Y, Doldina carinulata (Stål) (Reduviidae)]. The diversity of the cytogenetic characteristics of Heteroptera was reflected in the 18 studied species. Thus, this study extends the knowledge of these characteristics, such as the variations related to chromosome complements, sex chromosome systems, and meiotic behavior.

Seeking to meet the requirements: relatively low cost of materials and wide applicability in the automotive industry. The best option was the steel Dual Phase (DP), because that is lighter, have high formability, meet the cost requirements and applicability, steel Dual Phase (DP) came to meet these requirements with its two-phase microstructure, ferrite and martensite microstructure who claim to respect and mechanical properties. In this context, the aim of this study was to correlate the microstructure revealed in metallography to the mechanical properties observed in hardness and tensile tests. The microstructure is revealed by etching in 2% nital and then captured images of the sample were processed in ImageJ software to aid in determining the volume fraction of the phases present. Therefore, the mechanical properties were evaluated with respect to volume fraction of the steel layers and analyzed DP 600 together with the mechanical properties obtained by Rockwell hardness test and tensile test. With the values of the mechanical properties calculated and tested, it was possible to describe the method of metallography, as the attack phase and counts, so that it can use this relationship tested/calculated property as a qualitative analytical tool. The method used for the correlation between the microstructure and mechanical properties confirmed the importance of the phases present in the Dual Phase steel to obtain the desired mechanical properties in the application of the steel
