The major histocompatibility complex (MHC) contains a set of genes necessary for antigen presentation in the immune system. This gene dense and polymorphic region of the mammalian genome is of considerable interest due to the role of MHC genes in immune function and animal health. Previous cytogenetic studies have indicated that the MHC in river buffalo resides on the short arm of chromosome 2 (BBU2). A 5000-rad radiation hybrid mapping panel was recently generated to enable construction of a whole genome map of river buffalo. To this and, the aims of this project were to elucidate the general organization of the MHC on BBU2, and to compare gene order within this region to the MHC in cattle. PCR primers were selected from the bovine gene map and used with the BBURH(5000) panel to map a set of ten MHC class 11 genes in river buffalo. Analysis indicates that these genes fall into two linkage groups, consistent with organization of the MHC in cattle. This comparison of buffalo and bovine MHC gene order provides the first insight into the organization of the MHC on river buffalo chromosome 2.

Cytogenetic analysis of two local populations of microlepidogaster leucofrenatus showed a basic diploid chromosome number (2N) of 54 in both populations. Some fishes were found to have a 2N = 55 or 56 chromosomes due to the presence of one or two large heterochromatic B chromosomes. Specimens of M. leucofrenatus from the Poco Grande stream had 24 metacentrics, 24 submetacentrics, four subtelocentrics, and one submetacentric homomorphic pair in males and one submetacentric/subtelocentric heteromorphic pair in females, whereas individuals of this species from the Marumbi River had 22 metacentrics, 24 submetacentrics, four subtelocentrics, two acrocentrics, and one submetacentric/subtelocentric heteromorphic pair in females. The occurrence of the heteromorphic pair in the females was due to the presence of an extra C-banded segment on the W chromosome. Ag-NORs in both populations were located interstitially on the short arm of the largest metacentric pair. The Poco Grande population had less constitutive heterochromatin than did the Marumbi River population. The speciation process in this fish species is discussed on the basis of heterochromatin distribution.

Activity profiles of excess Pb-210 measured in four sediment cores from the Corumbatai River basin, São Paulo State, Brazil, provided an opportunity to evaluate sedimentation rates that are helpful for defining appropriate management strategies for the hydrological resources in the basin. This is because Rio Claro city and other municipalities make extensive use of surface waters for drinking water supply. The radiochemical analysis of the sediment cores yielded apparent sediment mass accumulation rates of between 406 and 1014 mg cm(-2) year(-1) for secondary drainage lines, whereas an intermediate value of 546 mg cm(-2) year(-1) was found in the Corumbatai River, the main drainage system of the studied area. These values provided estimates of average linear sedimentation rates of between 3.1 and 16.2 mm year(-1) that are compatible with field evidence, with the highest value corresponding with an area characterized by accumulation of sediment.

The goal of this work was to establish comparisons among environmental degradation in different areas from Southern Spain (Gulf of Cadiz) and Brazil (Santos and Sao Vicente estuary), by using principal component analyses (PCA) to integrate sediment toxicity (amphipods mortality) and chemical-physical data (Zn, Cd, Pb; Cu, Ni, Co, V, PCBs, PAHs concentrations, OC and fines contents). The results of PCA extraction of Spanish data showed that Bay of Cadiz, CA-1 did not present contamination or degradation; CA-2 exhibited contamination by PCBs, however it was not related to the amphipods mortality. Ria of Huelva was the most impacted site, showing contamination caused principally by hydrocarbons, in HV-1 and HV-2, but heavy metals were also important contaminants at HV-1, HV-2 and HV-3. Algeciras Bay was considered as not degraded in GR-3 and -4, but in GR-3' high contamination by PAHs was found. In the Brazilian area, the most degraded sediments were found in the stations situated at the inner parts of the estuary (SSV-2, SSV-3, and SSV-4), followed by SSV-6, which is close to the Submarine Sewage Outfall of Santos - SSOS. Sediments from SSV-1 and SSV-5 did not present chemical contamination, organic contamination or significant amphipod mortality. The results, of this investigation showed that both countries present environmental degradation related to PAHs: in Spain, at Ria of Huelva and Gudarranque river's estuary areas; and in Brasil, in the internal portion of the Santos and Sao Vicente estuary. The same situation is found for heavy metals, since all of the identified metals are related to toxicity in the studied areas, with few exceptions (V for both Brazil and Spain, and Cd and Co for Brazilian areas). The contamination by PCBs is more serious for Santos and Sao Vicente estuary than for the investigated areas in Gulf of Cadiz, where such compound did not relate to the toxicity. The evolution of elemental Hg from its environmental compounds has already been supposed to be an important process within the global mercury cycle. The present study characterizes the abiotic reduction of Hg(II) ions by typical river humic substances (HS) conventionally pre-isolated by the adsorbent XAD 8 from the Rio Negro near Manaus, Brazil. For the investigation of this reduction process a special reaction and Hg(0) trapping unit combined with cold-vapor atomic absorption spectrometry (CVAAS) was developed. Preconcentration of traces of mercury(II), if required, was obtained by a home-made FIA system using microcolumns filled with the Hg(II)-selective collector CheliteS(R) (Serva Company). The effect of environmentally relevant parameters such as the pH value, the Hg(II)/HS ratio and the HS concentration on the I-IE;(II) reduction process was studied as a function of the time. The Hg(0) production was highest at pH 8.0 and in the case of decreasing HS amounts (0.5 mg) when about 65% of initially 1.0 mug Hg(H) was reduced within 50 h. Moreover, the reduction efficiency of HS towards HE;(II) strongly depended on the HS concentration but hardly on the Hg(II)/HS ratio. The reduction kinetics followed a relatively slow two-step first-order mechanism with formal rate constants of about 0.1 and 0.02 h(-1), respectively. Based on these findings the possible relevance of the abiotic evolution of mercury in humic-rich aquatic environments is considered. (C) 2000 Elsevier B.V. B.V. All rights reserved.

Presented here is a study about the capability of a sensing unit to detect changes in river water quality. In order to determine its accuracy, water quality was monitored at 11 points along the Veado River in Presidente Prudente, Brazil. To have a basis for comparison, a water quality index (WQI) was developed following methods previously applied in different watersheds. Results showed an accurate relationship between WQI and electric impedance readings detected by the sensing unit. Principal components analysis (PCA) was used to derive results in a form that can be correlated with WQI calculated for each sample point, which showed the potential application of this device.

This study aimed to evaluate the infection by the Austrodiplostomum compactum metacercariae in fishes from the Nova Avanhandava Reservoir, low Tiete river, São Paulo State, Brazil. The parasites were collected from eye (aqueous and vitreous humor), fixed in AFA solution and stained with carmine. The morphometric analysis was performed using a computerized system for analysis of images QWin Lite 2.5 (Leica). Prevalence, mean intensity of infection and abundance of infected fish were calculated. of the 22 species of fish registered, five were infected by metacercariae: Hoplias malabaricus, Metynnis maculatus, Plagioscion squamosissimus, Satanoperca pappaterra and Schizodon imparts. of the 627 fish evaluated, 34% were infected. A higher prevalence was observed in P. sqnamosissionis and S. pappaterra. Schizodon nasutus and M. maculatus are new hosts reported for A. compactum metacercariae.

Two projects of water treatment for public water supply were developed and operated by using combined systems of constructed wetlands. One of the projects was carried out in the town of Analandia, Sao Paulo, Brazil and wetlands with floating aquatic plants associated to the HDS system were used. Nearly 6480 inhabitants were supplied. The other conducted project was an experimental station in partnership with SABESP (Sao Paulo State Sanitation Agency/Brazil), for the pretreatment of 1700 l.s-1 of waters from the Cotia River, which is used for the population's supply after conventional treatment at the Lower Cotia Water Treatment Station. For this pilot project, wetlands with emergents and floating plants associated to the HDS system were used. The proposed objectives were achieved in both projects.

A rapid and sensitive method was developed to determine trace levels of Cd2+ ions in an aqueous medium by flame atomic absorption spectrometry, using on-line preconcentration in a mini-column packed with 100 mg of 2-aminothiazol modified silica gel (SiAT). The Cd2+ ions were sorbed at pH 5.0. The preconcentrated Cd2+ ions were directly eluted from the column to the spectrometer's nebulizer-burner system using 100 μL of 2 mol L-1 hydrochloric acid. A retention efficiency of over 95% was achieved. The enrichment factor (calculated as the ratio of slopes of the calibration graphs) obtained with preconcentrations in a mini-column packed with SiAT (A = -1.3 × 10-3 + 1.8 × 10-3 [Cd2+]) and without preconcentrations (A = 4 × 10-5 + 3.5 × 10-3[Cd2+]), was 51 and the detection limit calculated was 0.38 μg L-1. The preconcentration procedure was applied to determine trace levels of Cd in river water samples. The optimum preconcentration conditions are discussed herein.

Bryconamericus turiuba, a new species from the upper Rio Paraná system (Ostariophysi: Characiformes)

Bryconamericus turiuba, new species, is described from the upper Rio Paraná system in Brazil. It differs from all congeners in the following combination of characters: 15-20 branched anal-fin rays; four or five scales in a transverse series from the dorsal-fin origin to the lateral line; shallow body (depth 23.6-30.6% of SL); 37-43 perforated lateral line scales; first, third, and fifth or, more rarely, first and fourth outer premaxillary tricuspid teeth projecting anteriorly; two to five tricuspid to pentacuspid maxillary teeth; dentary with three or four tricuspid or pentacuspid large teeth, followed by five to seven smaller conical to tricuspid teeth; a conspicuous dark, vertically elongate humeral spot, extending to below the lateral line; a dark dorsal stripe extending from the supraoccipital spine to the caudal peduncle with a gap at the adipose-fin base; caudal-fin lobes without conspicuous markings, uniformly pigmented along rays; mature males lacking hooks on the pelvic- and anal-fin rays. The new species is compared with the Bryconamericus species described from the basins of the Rio Paraná, Rio Paraguay, Rio São Francisco, and coastal Brazilian rivers. © 2005 by the American Society of Ichthyologists and Herpetologists.

Feeding, seasonal changes in visceral fat and condition factor were compared in two species of characidiin fishes, Characidium lauroi and C. alipioi from Ribeirão Grande system, southeastern Brazil. Five streams of Ribeirão Grande system were sampled (22° 47' 08 S, 45° 28' 17W). The samples were taken four times per site, from July, 2001 to April, 2002: winter (July 2001), spring (October 2001), summer (February 2002) and autumn (April 2002). Quantitative collections were made with an electro-fishing device powered by a generator with maximum capacity of 1,500 V and 8.7 A of 60 Hz alternating current. Ephemeroptera nymphs, Diptera larvae (Chironomidae, Simuliidae), Plecoptera nymphs, Trichoptera larvae (Hydroptilidae, Psychoyiidae), terrestrial insects (Coleoptera, Isoptera, Hemiptera [Heteroptera, Homoptera]), Megaloptera larvae (Corydalidae), Arachnida, Ostracoda and vegetal debris were found in both species' diets. Visceral fat declined in February, coinciding with the decline of the condition factor in both species. The increased feeding from summer to fall provides fat accumulation. During subsequent seasons, fish may utilize visceral fat reserves for maintenance and reproduction.

Numerous potentially mutagenic chemicals have been studied mainly because they can cause damaging and inheritable changes in the genetic material. Several tests are commonly used for biomonitoring pollution levels and to evaluate the effects of toxic and mutagenic agents present in the natural environment. This study aimed at assessing the potential of a textile effluent contaminated with azo dyes to induce chromosomal and nuclear aberrations in Allium cepa test systems. A continuous exposure of seeds in samples of the textile effluent in different concentrations was carried out (0.3%, 3%, 10%, and 100%). Cells in interphase and undergoing division were examined to assess the presence of chromosome aberrations, nuclear changes, and micronuclei. Our results revealed a mutagenic effect of the effluent at concentrations of 10% and 100%. At lower concentrations, the effluent (3% and 0.3%) did not induce mutagenic alterations in the test organism A. cepa. These findings are of concern, since cell damage may be transmitted to subsequent generations, possibly affecting the organism as a whole, as well as the local biota exposed to the effluent discharge. If the damage results in cell death, the development of the organism may be affected, which could also lead to its death. © 2008 Elsevier Ltd. All rights reserved.
