Abstract : The objective of this study was to evaluate quantitatively and to describe qualitatively the process of bone repair in the interface of receptor bed and graft autogenous bone block with or without a e-PTFE membrane, in osteopenia induced rats. To this end, we used forty-eight Wistar rats weighing about 300g, in which, with the aid of 4.1 mm trephine a graft was removed from the parietal bone and fixed to the surface of the left mandibular ramus. The animals were randomly divided into four experimental groups: Group 1 (n=12): SHAM operated and autogenous bone graft only; Group 2 (n=12): SHAM and autogenous bone graft covered by e-PTFE membrane; Group 3 (n=12): ovariectomized rats (OVX) and autogenous bone graft only; Group 4 (n=12): OVX and autogenous bone graft covered by e-PTFE membrane. The animals in each group were sacrificed at three time periods: 21, 45 and 60 days, each time with 4 animals per group. The specimens were decalcified and included, the sections were stained with HE and subjected to histological and histomorphometric analysis in light microscopy. The results of the ANOVA showed that the variables on the condition (OVZ and SHAM), and the time (21, 45 and 60 days) were statistically significant, and can be established with the Tukey test (5%) that the period 21-day differs significantly from the periods of 45 and 60 days, which did not differ among themselves. The descriptive histological analysis showed integration of the graft in all animals. It was concluded that the initial integration of the graft bed was negatively affected in the presence of induced osteopenia, and that the use or not of a e-PTFE membrane did not interfere in the process of integration

Introduction: The aim of this study was to evaluate the effect of raloxifene (RLX) during progression of periapical lesions in ovariectomized (OVX) rats. Methods: Female Wistar rats were OVX or subjected to sham surgery and received vehicle or MX by gavage for 90 days. The treatment groups were as follows: sham surgery and treated with vehicle (SHAM-veh), OVX and treated with vehicle (OVX-veh), and OVX and treated with RLX (OVX-RLX). During treatment, the pulp of lower first molar was exposed to the oral. environment for induction of periapical lesion that was analyzed 7 or 30 days aftdr procedure. Blood samples were taken from jugular vein for measurement of estradiol, and the mandibles were removed and prepared for radiographic, histopathologic, histometric, and immunohistochemical analysis. Results: Estradiol plasma concentration showed hypoestrogenism in OVX rats. The histopathologic analysis of the OVX/RLX group was similar to that of the SHAM-veh group, whereas OVX-veh group showed larger penapical lesions with more intense inflammatory response and more cells positive for tartrate-resistant acid phosphatase. Radiographically, the groups were similar, but lesions on day 7 were smaller than lesions on day 30. Conclusions: The results suggest that hypoestrogenism potentiates the progression of periapical lesions, and such condition was reversed by treatment with RLX.

In order to standardize an experimental model to study the effects of absence of ovarian hormones in maxillary bones compared with long bones, the aim of this research was to analyze the influence of ovariectomy (OVX) on rat alveolar bone and tibiae, in different observation periods. Thirty-six female rats were ovariectomized or sham operated. After 60, 90 or 120 days, the animals were sacrificed and their hemimandibles, maxillae and tibiae were removed and routinely prepared for hematoxylin and eosin staining. The percentage of bone matrix area in bone septum in the first molar furcation region, and in tibial metaphysis was calculated, and data were submitted to statistical analysis (p < 0.05). As regards the histomorphometrical analysis in jaw bones, there was no statistical difference between groups, while the effects of ovariectomy on tibiae were seen as early as 60 days. According to the methods used, there was no significant influence of absence of ovarian hormones on interradicular septum of mandibular or maxillary first molars in the periods studied, despite the reduction in bone matrix area in tibia metaphysis as early as 60 days. The aim of this study was to evaluate the resorption process during the repair of autogenous bone grafts with or without coverage by an expanded polytetrafluoroethylene (e-PTFE) membrane in female rats with estrogen deficiency using the immunohistochemical technique. Eighty female rats were randomly divided into two groups (OVX and SHAM). The 40 female rats in the OVX group were subjected to ovariectomy, and the 40 female rats in the SHAM group were subjected to simulated ovariectomy. The two groups were further divided in subgroup E, which was subjected to surgery for placement of autogenous bone graft (ABG), and subgroup ME, in which the ABG was covered with an e-PTFE membrane. The animals were killed at 0, 7, 21,45 and 60 days. The specimens were analyzed using immunohistochemistry for the bone resorption markers RANK, RANK-L and Osteoprotegerin (OPG). A higher remodeling rate was observed at 7 and 21 days after the autogenous bone grafts, when the markers were more intensely expressed. At the final time point, the specimens presented similar characteristics to those observed at the initial time point. The expression of immunohistochemical markers was not altered by the estrogen deficiency. The presence of the e-PTFE membrane delayed the bone resorption process, influencing the immunohistochemical expression of markers.
