Sixty Piaractus mesopotamicus Holmberg, 1887 (pacu) fry fed a diet containing 0, 50, 100 and 200 mg ascorbic acid/kg dry feed were studied to evaluate the effect on parasitic infestation by the monogenean Anacanthorus penilabiatus Boeger, Husak and Martins, 1995 (Monogenea: Dactylogyridae) for a period of 24 weeks. The temperature of the aquaria was measured daily and remained between 28 and 31oC. At the beginning of the experiment, fish showed 6.15 ± 0.33 cm standard length and 8.64 ± 1.62 g average body weight. A sample of fish was examined and showed 43 ± 17 monogeneans per fish. At the end of the experiment, the gills of control and vitamin C-treated fish were collected for parasite counts. Control fish had 42.5 parasites per fish, a significantly higher number (P<0.05) when compared with fish fed vitamin C, that showed 16.5 parasites per fish. Ascorbic acid fortification in the food promoted an increase in fish resistance to parasites. It is suggested that an optimum level of 139 mg/kg vitamin C supplementation either elicited better nutritional conditions by stimulating the appetite of the fish or improved the immune response.

O valor nutricional do Lentinula edodes (Berk.) Pegler varia em função da linhagem cultivada, do processamento após colheita, do estágio de desenvolvimento do basidioma e do tipo de substrato utilizado. Este trabalho teve como objetivo avaliar nutricionalmente basidiomas de L. edodes em função da linhagem e do tipo de eucalipto cultivado. O delineamento experimental foi inteiramente casualizado, em esquema fatorial 2 x 10 (linhagens de L. edodes x tipo de eucalipto), totalizando 20 tratamentos com 2 repetições, sendo que cada repetição correspondeu a uma amostra de basidiomas desidratados e moídos. de acordo com os resultados obtidos, verificou-se que as propriedades nutricionais do L. edodes (proteína bruta, extrato etéreo, cinzas e fibra bruta) demonstraram sofrer influência da interação eucalipto x fungo. Assim, os melhores resultados foram: Proteína bruta: Linhagem LE-96/18 cultivada em toras de E. urophylla, cuja média foi de 24,3%; Extrato etéreo: Linhagem LE-96/18 cultivada em toras do clone 23, cuja média foi de 3,0%; Cinzas: Linhagem LE-96/18 cultivada em toras de E. paniculata e E. camaldulensis e Linhagem LE-95/01 cultivada em toras de E. citriodora, cujas médias foram de 5,0%; Fibra bruta: Linhagem LE-95/01 cultivada em toras de E. paniculata, cuja média foi de 20,5%.

That the symbiotic fungus of leaf-cutting ants only occasionally produces the sexual phase makes their identification confusing. This has occurred so rarely, either in laboratory nests, or in unbalanced field nests. that the possibility of contamination of the fungal garden by other fungi cannot be disregarded. In this paper we describe the formation of several basidiomata in a healthy and free-living nest of the leaf-cutting ant Acromyrmex hispidus fallax. The cultivation in vitro of the sterile mycelia (isolated from the fungal garden) with their typical inflated tips, and the similarity of both forms confirmed by RAPD analysis of their genomic DNA. The fungus was identified as Leucoagaricus gongylophorus.

Anacanthorus penilabiatus n. sp. is described from the serrasalmid fish, Piaractus mesopotamicus (Holmberg, 1887), cultivated in the Centro de Aquicultura, Universidade Estadual Paulista. The new species is characterized by having a relatively straight copulatory organ with a long ''lip'' on the distal margin and a median longitudinal flap, and a copulatory ligament. The large size of the infrapopulations of this species of parasite indicates that it should be considered a potential agent causing losses in aquaculture of the fish host.

The Agaricus blazei Murill (ABM) mushroom, known as the sun mushroom, is native to Brazil and has become known for its medicinal properties. This study examined the anticlastogenic effect of Agaricus blazei in Chinese hamster ovary cells, CHO-k1, by means of a chromosome aberration test using methyl methanesulphonate (MMS, 10(-4)M) as the DNA damage inducing agent. Two mushroom lines were used, ABM 99/26 and ABM 97/11, and the latter was used in the young (Y) and sporulating (S) developmental phases. The cells were treated for 12 h with MMS alone or combined with aqueous extracts of A. blazei at a final concentration of 0.15%, which were prepared at three different temperatures: (a) hot (60 degreesC), (b) room temperature (25 degreesC) and (c) chilled (4 degreesC). Mushroom extracts showed a marked anticlastogenic effect against DNA damage, as evidenced by a decrease in the number of cells with breaks, regardless of the line used, or the developmental stage or the temperature at which the extract was prepared. Generally, the extracts were more effective in reducing the isochromatid type breaks. The data obtained suggest that extracts of A. blazei mushroom are anticlastogenic under the conditions tested, mainly during the G1 and S stages of the cell cycle, where chromosome breaks of the isochromatid type are produced by the MMS agent. The present work evaluated the efficacy of mebendazole (MBZ) treatment against infections with the monogenean helminths Anacanthorus penilabiatus, gill parasites of young cultivated pacu, Piaractus mesopotamicus. A short-term bath treatment using 100, 200 and 500 mg MBZ/l of water for 10 and 30 min and a long-term-bath using 1, 10 and 100 mg MBZ/l of water for 24 h were utilized. Seven days after, fish were sacrificed and parasites counted. Concentrations of 500 and 200 mg MBZ/l for 10 and 30 min showed reduced efficacy (0.0 and 0.7%) and (14.2 and 11.0%), respectively. Nevertheless, 100 mg MBZ/l (10 min) and 10 nag MBZ/l (24 h) showed better efficacy (79.6 and 8 1.4%, respectively). Treatments for 24 h provoked increases in hematocrit, hemoglobin concentration and leukocyte number when compared to those untreated fish. Mebendazole treatments also provoked alterations in the defense blood cells especially in lymphocyte and thrombocyte numbers, when the fishes were submitted to 10 and 30 min baths.

The final levels of ethanol (levels of ethanol produced plus that added initially to the media) reached by the thermotolerant yeasts were highest (16.5-20.3%, v/v) at 8% initial ethanol. The thermotolerant yeasts were found to have the following characteristics: constant levels of ethanol formation (10.5-12.3%, v/v), fog additions of external ethanol within the range 2-8% (v/v) of initial ethanol; constant values of product coefficients when initial ethanol was in the range of 2-6%, which increased or decreased, depending on the strain, when initial ethanol exceeded 6%; growth activity was inhibited at different levels of addition of external ethanol when final biomass and specific rate of growth were compared; significant differences among the yeast strains in the amount of external ethanol capable of reducing biomass formation by one half. In addition, the viability of the strains (early stationary phase) varied with the amount of external ethanol, the lowest viabilities occurring at concentrations of initial ethanol ranging from 4 to 7% and the highest in the range of 7 to 8% (v/v). The relative levels of trehalose (with/without 7% ethanol added initially) in the yeast strains (the stationary phase) ranged from 1.03 to 1.75, suggesting that the effect of produced ethanol on trehalose accumulation was stronger than that of external ethanol. The levels of final ethanol shown by the yeast strains were also correlated with the cellular levels of glycerol-3-phosphate dehydrogenase (increase in enzyme levels with decrease in final ethanol) for cells harvested at the stationary phase.

Aqueous extracts of the sporophores of eight mushroom species were assessed for their ability to prevent H2O2-induced oxidative damage to cellular DNA using the single-cell gel electrophoresis (Comet) assay. The highest genoprotective effects were obtained with cold (20°C) and hot (100°C) water extracts of Agaricus bisporus and Ganoderma lucidum fruit bodies, respectively. No protective effects were observed with Mushroom Derived Preparations (MDPs) from Flammulina velutipes, Auricularia auricula, Hypsizygus marmoreus, Lentinula edodes, Pleurotus sajor-caju, and Volvariella volvacea. These findings indicate that some edible mushrooms represent a valuable source of biologically active compounds with potential for protecting cellular DNA from oxidative damage. © 2002 Wiley-Liss, Inc.

Agaricus blazei Murill is a mushroom largely consumed due to its medicinal properties. Effects of aqueous extract from its lineage AB97/11 in 2 fruiting body development stages (closed and opened pileus) were evaluated on chinese hamster V79 cells using cytokinesis blocking micronucleus (CBMN) and comet assays. The cells were treated at 0.15% concentration of aqueous extract prepared at different temperatures: ice-cold (4°C), room temperature (21°C) and warm (60°C). The extracts were applied in mutagenicity and antimutagenicity protocols (simultaneous, pre-incubation and continuous). The results showed that the aqueous extracts of Agaricus blazei lineage AB97/11 obtained at the 3 temperatures and both development stages did not present mutagenic or antimutagenic effect in V79 cells either in CBMN or comet assay.

The use of biosolids in horticulture could contribute to recycle residues produced by men. This study analyzed concentrations of Cu, Mn and Zn in the compost during fermentation, in the soil amended with the composts and in the tomato plant materials. Five composts were produced using sugar-cane bagasse, biosolid and cattle manure in the proportions: 75-0-25; 75-12.5-12.5; 75-25-0; 50-50- 0 and 0-100-0 (composts with 0; 12.5; 25; 50 and 100% biosolid), respectively. These composts were used in an experiment with 6 treatments (the 5 composts and a control with mineral fertilization) in a design of randomized blocks with a split plot design. The control and the treatment of 0% biosolid received inorganic nitrogen. All the treatments received the same amount of N, P and K. Two tomato plants were cultivated in each 24 L pot, in a greenhouse at the Technology Department of the Faculdade de Ciências Agrárias e Veterinárias of the Universidade Estadual Paulista in Jaboticabal County, São Paulo State, Brazil. The concentrations of Cu, Mn and Zn were evaluated in the compost 7, 27, 57, 97 and 127 days after composting began, in the soil 0 and 164 days after the compost applied, and in the plants. Compost, soil and plant samples were subjected to digestion with HNO3, H 2O2 and HCl and the metals were determined by AAS. There were positive and significant correlations between Mn in the compost and Mn uptake by the plant (0.46 p>0.05), and between Zn in the compost and Zn concentration in the plant (0.78 p>0.05). Cu, Mn and Zn concentrations increased during composting. The biosolid in the compost supplied Cu and Zn to tomato plants, and the cattle manure supplied Mn to the plants.

The behavior of two hybrids of muskmelon ('Bônus no 2' and 'Don Carlos') in relation to the productivity and quality of its fruits, making use of fertigation in sand substrate in protected environment, was studied. Amianthus cement structures of the Canalete runlet type were used to keep the plants 0.30m from each other and 1.0m between lines. 'Don Carlos' hybrid had fruit mean weight, longitudinal and transversal diameter, pulp mean thickness, and total acidity substantially superior in relation to 'Bônus no 2'. Considering the total soluble solid content, 'Bônus no 2' had better results. However, there was no significant difference in fruits/plant or pulp thickness between the two hybrids.
