O objetivo deste trabalho foi avaliar a desnitrificação de efluente de abatedouro de tilápia, em reator em batelada. Para isto, foi avaliado o efluente gerado por uma indústria processadora de mandioca (manipueira) como fonte de carbono. Também foram avaliadas as condições de agitação do sistema. Os experimentos foram conduzidos em escala de laboratório, em que foram testados cinco níveis de agitação (20; 32; 60; 88 e 100 rpm) e cinco níveis de relação DQO/N (0,1; 1,0; 3,2; 5,4 e 6,3), configurando-se um planejamento do tipo Delineamento Composto Central Rotacional (DCCR), com quatro ensaios nos níveis +1 e - 1; quatro ensaios nos níveis dos pontos axiais (-1,414 e +1,414) e mais uma triplicata no ponto central (0), totalizando 11 ensaios. Avaliaram-se o desempenho da desnitrificação através da remoção de nitrato (%) e a remoção de nitrito (%). Durante o processo, também foram monitorados temperatura (ºC), pH e alcalinidade (mgCaCO3.L-1). Os resultados mostraram que a relação DQO/N teve influência significativa, com intervalo de confiança de 95%, sobre o processo de remoção de nitrato e nitrito, com faixa ótima de operação entre 3,2 e 5,4, cujas eficiências de remoção de nitrogênio foram de 100%.

Este trabalho teve por objetivo avaliar o efeito da umidade, temperatura de extrusão e rotação da rosca na produção de produtos extrusados expandidos com a utilização do polvilho doce e da polpa de laranja desidratada como matéria-prima. O processo seguiu delineamento central composto rotacional com três fatores, totalizando 15 tratamentos (oito pontos fatoriais, seis pontos axiais e um ponto central com seis repetições). Os produtos obtidos foram caracterizados quanto ao índice de expansão (IE), ao volume específico (VE), ao índice de solubilidade em água (ISA) e à cor (L*, a* e b*). Os resultados mostraram efeito das condições operacionais do processo sobre as características físicas dos produtos, e produtos claros com maior expansão e menor solubilidade em água foram obtidos nas condições mais elevadas de temperatura (80-90°C), condições intermediárias de umidade (16%) e rotação da rosca (218rpm).

Soil flushing is an alternative remediation technology for soils contaminated with heavy metals, which the main contaminant removing process consists in percolating an extraction solution. This work aimed to use the response surface methodology to point out combinations among the parameters of the extraction solution (Na2EDTA concentration, volume e pH) in order to reduce the concentration of copper in a sandy soil to risk levels lower than the intervention levels for exposure scenarios adopted by the Environmental Company of Sao Paulo State. Thus, a series of tests in leaching columns were carried out using a Fluvisol artificially contaminated (1257,3 mg kg-1). The tests were conducted in triplicate and setup a central composite rotatable design with 15 different parameters combinations of the extraction solution and one replicate in the center point. Using 5% significance level, the adjusted model (R2 = 0,98) indicated combinations of Na2EDTA concentration, pH and volume of the extraction solution which allow reduction of copper concentration below levels reported by environmental agency of Sao Paulo State for industrial, residential, agricultural or maximum exposure scenarios.

A synbiotic yoghurt based on a combination of soymilk and yacon water extract (from yacon root tubers) was developed as a novel food product fermented with a probiotic culture of Enterococcus faecium CRL 183 and Lactobacillus helveticus ssp jugurti 4l6. Response surface methodology (RSM) was used to optimize the independent variables soymilk protein concentration and percentage of yacon extract in the formulation through a Central Composite Rotatable Design (CCRD), consisting of a 22 factorial design with two levels (-1, +1), two central points (0) and four axial points (± a, 0) (0, ± α). The responses were assessed by consumer acceptance tests. The optimization indicated that a formulation with a soymilk protein concentration of 1.74g/L and 25.86% of yacon extract gave the best average values, 5.91 for the taste and 6.00 for the overall impression responses. The formulation with 40% of yacon extract and the same concentration of soymilk protein achieved similar acceptance values: taste (5.94) and overall impression (5.87), however, with the extra yacon, it probably had a greater content of prebiotic fructooligosaccharides. Consequently, both formulations may give useful functional foods, with sensory properties comparable with those of soy yoghurt (control formulation). Extracellular β-1,3-glucanase was produced by Trichoderma harzianum Rifai cultivated in the Agaricus blazei (Agaricus brasiliensis) extract as a substrate in submerged fermentation. A 22-central composite factorial design was developed using the time of culture (x1/day) and Agaricus blazei extract concentration (x2/(g/L)) as variables, and the results were analyzed using response surface methodology (RSM). The results showed that the Agaricus blazei extract concentration was the most important variable in the production of β-1,3-glucanase, and the maximum β-1,3-glucanase activity (0.77 U/mL) was obtained in one day of cultivation. The β-glucan present in the cell wall of Agaricus blazei mushroom proved to be a good substrate for inducing the production of specific β-1,3-glucanase by Trichoderma harzianum Rifai.

This paper proposes a response surface methodology to evaluate the influence of the particle size and temperature as variables and their interaction on the sulfation process using two Brazilian limestones, a calcite (ICB) and a dolomite (DP). Experiments were performed according to an experimental design [central composite rotatable design (CCRD)] carried out on a thermogravimetric balance and a nitrogen adsorption porosimeter. In the SO 2 sorption process, DP was shown to be more efficient than ICB. The best results for both limestones in relation to conversion and Brunauer-Emmett-Teller (BET) surface area were obtained under central point conditions (545 μm and 850 C for DP and 274 μm and 815 C for ICB). The optimal values for conversion were 52% for DP and 37% for ICB. For BET surface area, the optimal values were 35 m2 g-1 for DP and 45 m2 g-1 for ICB. A relationship between conversion and pore size distribution has been established. The experiments that showed higher conversions also exhibited more pores in the region between 20 and 150 Å and larger BET surface area, indicating that the amount of smaller pores may be an important factor in the reactivity of limestones. © 2013 American Chemical Society.

The effects of soybean and castorbean meals were evaluated separately, and in combinations at different ratios, as substrates for lipase production by Botryosphaeria ribis EC-01 in submerged fermentation using only distilled water. The addition of glycerol analytical grade (AG) and glycerol crude (CG) to soybean and castorbean meals separately and in combination, were also examined for lipase production. Glycerol-AG increased enzyme production, whereas glycerol-CG decreased it. A 24 factorial design was developed to determine the best concentrations of soybean meal, castorbean meal, glycerol-AG, and KH2PO4 to optimize lipase production by B. ribis EC-01. Soybean meal and glycerol-AG had a significant effect on lipase production, whereas castorbean meal did not. A second treatment (22 factorial design central composite) was developed, and optimal lipase production (4,820 U/g of dry solids content (ds)) was obtained when B. ribis EC-01 was grown on 0.5 % (w/v) soybean meal and 5.2 % (v/v) glycerol in distilled water, which was in agreement with the predicted value (4,892 U/g ds) calculated by the model. The unitary cost of lipase production determined under the optimized conditions developed ranged from US$0.42 to 0.44 based on nutrient costs. The fungal lipase was immobilized onto Celite and showed high thermal stability and was used for transesterification of soybean oil in methanol (1:3) resulting in 36 % of fatty acyl alkyl ester content. The apparent K m and V max were determined and were 1.86 mM and 14.29 μmol min -1 mg-1, respectively. © 2013 Springer Science+Business Media New York.
