A utilização de recursos naturais e a necessidade de recuperação de áreas degradadas têm assumido importância global, sendo que a estimativa de custos garante a viabilidade econômica, para implantação de projetos de conservação ou recuperação. Foram estimados os custos para implantação de um hectare da espécie arbórea Gonçalo-Alves (Astonium fraxinifolium Schott), com adubação verde, em Latossolo Vermelho degradado, como se segue: Gonçalo-Alves + Feijão-de-porco; Gonçalo-Alves + crotalária; Gonçalo-Alves + fórmula NPK (04-14-08); e Gonçalo-Alves + braquiária + lodo. A estimativa do custo de implantação de Gonçalo-Alves, para a região leste do Estado do Mato Grosso do Sul, variou entre R$ 2.829,13 e R$ 3.872,21. O tratamento com braquiária + lodo de esgoto foi o mais econômico, considerando-se um período de cinco anos, além dos benefícios ambientais relacionados ao descarte viável deste resíduo, por estações de tratamento de esgoto.

Snake venom glands are a rich source of bioactive molecules such as peptides, proteins and enzymes that show important pharmacological activity leading to in local and systemic effects as pain, edema, bleeding and muscle necrosis. Most studies on pharmacologically active peptides and proteins from snake venoms have been concerned with isolation and structure elucidation through methods of classical biochemistry. As an attempt to examine the transcripts expressed in the venom gland of Bothrops jararacussu and to unveil the toxicological and pharmacological potential of its products at the molecular level, we generated 549 expressed sequence tags (ESTs) from a directional cDNA library. Sequences obtained from single-pass sequencing of randomly selected cDNA clones could be identified by similarities searches on existing databases, resulting in 197 sequences with significant similarity to phospholipase A(2) (PLA(2)), of which 83.2% were Lys49-PLA(2) homologs (BOJU-1), 0.1% were basic Asp49-PLA(2)s (BOJU-II) and 0.6% were acidic Asp49-PLA(2)s (BOJU-III). Adjoining this very abundant class of proteins we found 88 transcripts codifying for putative sequences of metalloproteases, which after clustering and assembling resulted in three full-length sequences: BOJUMET-I, BOJUMET-II and BOJUMET-III; as well as 25 transcripts related to C-type lectin like protein including a full-length cDNA of a putative galactose binding C-type lectin and a cluster of eight serine-proteases transcripts including a full-length cDNA of a putative serine protease. Among the full-length sequenced clones we identified a nerve growth factor (Bj-NGF) with 92% identity with a human NGF (NGHUBM) and an acidic phospholipase A2 (BthA-I-PLA(2)) displaying 85-93% identity with other snake venom toxins. Genetic distance among PLA(2)s from Bothrops species were evaluated by phylogenetic analysis. Furthermore, analysis of full-length putative Lys49-PLA(2) through molecular modeling showed conserved structural domains, allowing the characterization of those proteins as group II PLA(2)s. The constructed cDNA library provides molecular clones harboring sequences that can be used to probe directly the genetic material from gland venom of other snake species. Expression of complete cDNAs or their modified derivatives will be useful for elucidation of the structure-function relationships of these toxins and peptides of biotechnological interest. Convulxin (CVX), a C-type lectin, isolated from the venom of the South American rattlesnake Crotalus durissus terrificus, causes cardiovascular and respiratory disturbances and is a potent platelet activator which hinds to platelet glycoprotein GPVI. The structure of CVX has been solved at 2.4 Angstrom resolution to a crystallographic residual of 18.6% (R-free =26.4%). CVX is a disulfide linked heterodimer consisting of homologous alpha and beta chains. The heterodimers are additionally linked by disulfide bridges to form cyclic alpha(4)beta(4)heterotetramers. These domains exhibit significant homology to the carbohydrate-binding domains of C-type lectins, to the factor IX-binding protein (IX-bp), and to flavocetin-A (Fl-A) but sequence and Structural differences are observed in both the domains in the putative Ca2+ and carbohydrate binding regions. The venom proteome of Daboia russelli siamensis, a snake of medical importance in several Asian countries, was analysed by 2-D electrophoresis, subsequent MS/MS and enzymatic assays. The proteome comprises toxins from six protein families: serine proteinases, metalloproteinases, phospholipases A(2), L-amino acid oxidases, vascular endothelial growth factors and C-type lectin-like proteins. The venom toxin composition correlates with the clinical manifestation of the Russell's viper bite and explains pathological effects of the venom such as coagulopathy, oedema, hypotensive, necrotic and tissue damaging effects. The vast majority of toxins are potentially involved in coagulopathy and neurotoxic effects. The predominant venom components are proteinases capable of activating blood coagulation factors and promoting a rapid clotting of the blood, and neurotoxic phospholipase A(2)s. The analysis of the venom protein composition provides a catalogue of secreted toxins. The proteome of D. r. siamensis exhibits a lower level of toxin diversity than the proteomes of other viperid snakes. In comparison to the venoms of Vipera ammodytes ammodytes and Vipera ammodytes meridionalis, the venom from D. r. siamensis showed quantitative differences in the proteolytic, phospholipase A2, L-amino acid oxidase and alkaline phosphatase activities. Lotus tetragonolobus lectin (LTA) is a fucose-specific legume lectin. Although several studies report a diverse combination of biological activities for LTA, little is known about the mechanisms involved in L-fucosyl oligosaccharide recognition. The crystal structure of LTA at 2.0 angstrom resolution reveals a different legume lectin tetramer. Its structure consists of a homotetramer composed of two back-to-back GS4-like dimers arranged in a new mode, resulting in a novel tetramer. The LTA N-linked carbohydrate at Asn4 and the unusual LTA dimer-dimer interaction are related to its particular mode of tetramerization. In addition, we used small angle X-ray scattering to investigate the quaternary structure of LTA in solution and to compare it to the crystalline structure. Although the crystal structure of LTA has revealed a conserved metal-binding site, its L-fucose-binding site presents some punctual differences. Our investigation of the new tetramer of LTA and its fucose-binding site is essential for further studies related to cross-linking between LTA and complex divalent L-fucosyl carbohydrates. A adubação verde é uma das formas de aporte de matéria orgânica ao solo. O sistema de cultivo consorciado de culturas pode ser uma alternativa para aumentar a reciclagem de nutrientes e melhorar a produtividade. Para avaliar o sistema consorciado de adubos verdes com o milho, foram estudadas as características químicas do solo, a produção de matéria seca, a composição mineral de adubos verdes e o rendimento de grãos de milho, num experimento realizado em campo, entre 1995 e 1997, em solo classificado como Nitossolo Vermelho eutrófico. O milho foi semeado no espaçamento de 90 cm nas entrelinhas, perfazendo, aproximadamente, 50.000 plantas por hectare. Os tratamentos constaram de quatro espécies de adubos verdes: mucuna anã [Mucuna deeringiana (Bort.) Merr], guandu anão (Cajanus cajan L.), crotalária (Crotalaria spectabilis Roth) e feijão-de-porco (Canavalia ensiformis L.) e um tratamento-testemunha, sem cultivo consorciado. Essas espécies foram semeadas sem adubação, no meio da entrelinha, em duas épocas: simultânea ao milho e 30 dias após. O delineamento experimental foi o de blocos ao acaso em parcelas subdivididas, com quatro repetições. O feijão-de-porco apresentou maior produção de fitomassa e acúmulo de N, P, K, Ca, Mg e S. No primeiro ano de cultivo, o rendimento de grãos de milho não foi influenciado pelo cultivo consorciado com adubos verdes; no entanto, no segundo, a produção foi beneficiada pelo consórcio com feijão-de-porco.

O presente trabalho teve por finalidade avaliar os possíveis efeitos alelopáticos de três plantas daninhas (Cyperus rotundus, Sorghum halepense e Cynodon dactylon), uma leguminosa (Canavalia ensiformis) e colza (Brassica napus), na germinação das sementes do tomateiro (Lycopersicon esculentum cv. Santa Cruz) utilizado como indicador. Tubérculos de tiririca, rizomas de capim massambará, o sistema radicular de grama seda, além de folhas e raízes de feijão-de-porco e colza, foram homogeneizados em solução aquosa, submetidos a filtração e centrifugação. A aplicação do sobrenadante no substrato de germinação das sementes de tomateiro mostrou afetar o processo germinativo e o desenvolvimento da radícula e do hipocótilo. C. rotundus, S. halepense e raízes de C. ensiformis possuem substâncias altamente inibitórias à germinação do tomateiro 'Santa Cruz'', sendo que a planta indicadora também mostrou-se sensível às substâncias presentes nas folhas de B. napus e em C. dactylon.

Parkia platycephala lectin 2 was purified from Parkia platycephala (Leguminosae, Mimosoideae) seeds by affinity chromatography and RP-HPLC. Equilibrium sedimentation and MS showed that Parkia platycephala lectin 2 is a nonglycosylated monomeric protein of molecular mass 29 407 +/- 15 Da, which contains six cysteine residues engaged in the formation of three intramolecular disulfide bonds. Parkia platycephala lectin 2 agglutinated rabbit erythrocytes, and this activity was specifically inhibited by N-acetylglucosamine. In addition, Parkia platycephala lectin 2 hydrolyzed beta(1-4) glycosidic bonds linking 2-acetoamido-2-deoxy-beta-D-glucopyranose units in chitin. The full-lengthamino acid sequence of Parkia platycephala lectin 2, determined by N-terminal sequencing and cDNA cloning, and its three-dimensional structure, established by X-ray crystallography at 1.75 angstrom resolution, showed that Parkia platycephala lectin 2 is homologous to endochitinases of the glycosyl hydrolase family 18, which share the (beta alpha)(8) barrel topology harboring the catalytic residues Asp125, Glu127, and Tyr182.

The venom of Bothrops insidaris snake, known in Brazil as jararaca ilhoa, contains a variety of proteolytic enzymes such as a thrombin-like substance that is responsible for various pharmacological effects. B. insularis venom chromatography profile showed an elution of seven main fractions. The thrombin-like activity was detected in fractions I and 111, the latter being subjected to two other chromatographic procedures, so to say DEAE and Hi Trap Benzamidine. The purity degree of this fraction was confirmed by analytical reverse phase HPLC, which displayed only one main fraction confirmed by SDS-PAGE constituting fraction III. About 5 mu g of fraction III protein potentiated the secretion of insulin induced by 2.8mM of glucose in rats isolated pancreatic beta-cells treated; the increase being around 3-fold higher than its respective control. B. insidaris lectin (BiLec; 10 mu g/mL) was also studied as to its effect on the renal function of isolated perfused rat kidneys with the use of six Wistar rats. BiLec increased perfusion pressure (PP), renal vascular resistence (RVR), urinary flow (UF) and glomerular filtration rate (GFR). Sodium (%TNa+) and chloride tubular reabsorption (%TCl-) decreased at 120 min, without alteration in potassium transport. In conclusion, the thrombin-like substance isolated from B. insularis venom induced an increase in insulin secretion, in vitro, and transiently altered vascular, glomerular and tubular parameters in the isolated rat kidney.
