The bee Apis mellifera has a great importance because it is the most economically valuable pollinator for crops worldwide, ensuring cross-pollination and increasing fruit yield. Moreover, agriculture increasingly develops chemicals to control weeds, fungi and insect pests to ensure productivity. Insecticides are used on a large scale in the state of São Paulo, in cultures of citrus for control of greening. Applications are usually made by aircraft and as a result of the effect derives a significant mortality is observed in apiaries near the plantations. Honey bees can get in contact with such chemical agent through their activities of water harvesting, plant resins, pollen and nectar. Intoxication resulting from this exposure can be lethal, which is easily detectable, or cause effects on the physiology and behavior of the insect. These, in turn are hardly detectable, such as paralysis, disorientation, behavioral changes, but can compromise the entire social structure of the colony, therefore aimed to study the effects of the insecticide Thiamethoxam behavior of honeybees A. mellifera. Newly emerged individuals and with 10 days of age were tested. Applications of 1 μL de Thiamethoxam, diluted in acetone, were made on the dorsal thorax with a microapplicator. Preliminarily, it was observed LD50 twenty four hours after topical treatment of Thiamethoxam. It was found that the LD50 for newly emerged honeybees is 8 ng/bee and for honeybees with 10 days of age is 18 ng/bee. The behaviors were analyzed 1 hour after application of insecticide at doses corresponding to LD50/100, LD50/50, LD50/10 and LD50, besides the control group. In the test reflex proboscis extension, there was impairment of behavior at doses of 8 and 18 ng/bee workers in newly emerged workers and 10 days of age, respectively. Apis mellifera honeybees are social insects of economic importance, by providing honeybee products, and by the pollination of natural areas of vegetation or agricultural areas. The constant use of pesticides, including the thiamethoxam, which is an insecticide belonging to the class of neonicotinoids with neurotoxic action, is subjecting pollinators to situations of severe stress, which has been evidenced by the decrease in the density of honeybees in many parts of the world. By these considerations, the present study aimed to assess the acute toxicity of thiamethoxam for newly emerged workers of A. mellifera, and to investigate the effect of sublethal doses of this insecticide on the survival time and its cytotoxicity to the brain and midgut to the honeybees. It was established the value of the lethal concentration 50 (LC50) equal to 4.28 ng thiamethoxam/μL of food and from it, the sublethal concentrations of 0.428 ng thiamethoxam/μL (CL50/10) and 0.0428 ng thiamethoxam/μL (CL50/100), which were used in bioassays of intoxication of the honeybees. After the bioassays the bees were dissected and the brain and midguts were collected to analyze possible morphological (staining with Hematoxylin-Eosin) and histochemical alterations (Xylidine Ponceau technique, and Feulgen and PAS reactions) caused by exposure to thiamethoxam, and to calculate the lethal time (LT50) for the workers. The duration of the bioassays was 8 days after beginning of feeding. The results obtained showed that the thiamethoxam is toxic to newly-emerged workers of A. mellifera, causing changes in survival time of individuals. This study also shown that the thiamethoxam causes morphological and histochemical alterations on the midgut and brain of workers. These alterations may be reflected in physiological and behavioral changes that can modify the operation of the colony

The objective of this work was to typify, through physicochemical parameters, honey from Campos do Jordão’s microrregion, and verify how samples are grouped in accordance with the climatic production seasonality (summer and winter). It were assessed 30 samples of honey from beekeepers located in the cities of Monteiro Lobato, Campos do Jordão, Santo Antonio do Pinhal e São Bento do Sapucaí-SP, regarding both periods of honey production (November to February; July to September, during 2007 and 2008; n = 30). Samples were submitted to physicochemical analysis of total acidity, pH, humidity, water activity, density, aminoacids, ashes, color and electrical conductivity, identifying physicochemical standards of honey samples from both periods of production. Next, we carried out a cluster analysis of data using k-means algorithm, which grouped the samples into two classes (summer and winter). Thus, there was a supervised training of an Artificial Neural Network (ANN) using backpropagation algorithm. According to the analysis, the knowledge gained through the ANN classified the samples with 80% accuracy. It was observed that the ANNs have proved an effective tool to group samples of honey of the region of Campos do Jordao according to their physicochemical characteristics, depending on the different production periods.

The technique of osmium imidazol for the ultrastructural detection of lipids in the secretory cells of the venom gland of 14-days old worker bees of Apis mellifera L. demonstrated the presence of these components at various sites of the gland. These lipids were found mainly associated to the external region of the basal lamina and the microvilli, in the intercellular spaces, in the cuticle of the collecting canaliculi and in the secretion contained in the glandular lumen. Therefore, in addition to revealing the presence of lipids in the secretion, this technique also allowed us to attribute an exogenous origin to the lipids in the secretion; they are taken up from the haemolymph.

Oenocytes of adult workers and queens of Apis mellifera L. were studied in different ages or life stages, by means of morphometric and histologic techniques. In workers, the oenocytes were found in the head, near the mandibles and in the abdomen, immersed in the parietal fat body mainly below the sterna, close to the wax gland. In queens, two populations of oenocytes different in size and localization were found within the parietal and visceral fat body, respectively. The oenocytes of workers and queens show the presence of acid lipids and acid phosphatase. The role of these cells in the castes differences is discussed.

Split sting is the name given to a nonfunctional honey bee sting characterized by lancets not attached to the stylet. It has appeared in a mutant line in Brazil, and has provoked interest as a possible means to reduce honey bee colony defensiveness. We induced this alteration in Africanized Apis mellifera L. workers and queens by maintaining pupae at 20 degrees C. In particular, we determined the pupal phase most susceptible to alterations in the sting caused by cold treatment, and we investigated whether this treatment also affected survival to the adult phase and wing morphology. The highest frequency of split sting was detected in workers treated at the pink-eyed pupal phase. The lowest frequency was observed in the bees treated at the oldest worker pupal phase studied (brown-eyed pupae with lightly pigmented cuticle). Both queen pupal phases tested (white and pink-eyed pupae) were equally sensitive and produced high percentages of adults with split sting. However, the 20 degrees C treatment of workers and queens, at the different pupal phases, resulted in high frequencies of adults with deformed wings. Also, fewer workers and queens treated at the earlier pupal stages reached adult emergence. There was also an arrest in developmental time, corresponding to the period of cold treatment.
