An Arabidopsis thaliana cDNA clone encoding a plant uncoupling mitochondrial protein (AtPUMP1) was overexpressed in transgenic tobacco plants. Analysis of the AtPUMP1 mRNA content in the transgenic lines, determined by Northern blot, revealed variable levels of transgene expression. Antibody probing of Western blots of mitochondrial proteins from three independent transgenic lines showed significant accumulation of AtPUMP1 in this organelle. Overproduction of AtPUMP1 in transgenic tobacco plants led to a significant increase in tolerance to oxidative stress promoted by exogenous hydrogen peroxide as compared to wild-type control plants. These results provide the first biological evidence for a role of PUMP in protection of plant cells against oxidative stress damage.

The present study evaluated stress indicators in pacu exposed to ammonia in water under the following conditions: without NH4Cl (0.00 g/L); with 0.0078 g NH4Cl/L; and with 0.078 g NH4Cl/L (pH 8.3 and 27 ºC). After the salt dilution the water flow was interrupted and reestablished in 24 hours. Sampling occurred prior to the addition of NH4Cl (control) and after 12, 24 and 48 hours. Glycaemia increased only in fish with the highest salt concentration when compared with group control, regardless of time, and at 24 hours, regardless of treatment. Plasma ammonia, highest in fish exposed to the highest NH4Cl concentration, decreased progressively up to 48 hours. Plasma chloride only decreased in fish not exposed to salt when compared with control and osmolality increased after 24 hours. Hematocrit (Ht), number and volume of erythrocytes and hemoglobin did not change when NH4Cl was added; Ht decrease was reported after 12 hours, but it was not followed by the other blood parameters. The results show tolerance of the pacu to ammonia in the environment.

Sugarcane is a very important economic crop in Brazil. The effects of abiotic stresses cause negative reduction of the productivity in the sugarcane industry. In order to identify indicators of stresses tolerance, two physiological variables were evaluated, nitrate reductase activity and chlorophyll contents in young plants of sugarcane, cv. IAC91-5155. The simultaneous effect of abiotic stresses of high occurrence in Brazilian soils are, water deficiency and aluminum toxicity. The plants were submitted to three treatments of water availability (% field capacity, FC): no stress (70% FC), moderate stress (55% FC), and extreme stress (40% FC); and three acidity treatments in the soil (base saturation, V%): no acidity (V=55%), average acidity (V=33%), and high acidity (V=23%). The experiment was carried out in greenhouse, with 29.7 +/- 4.3 degrees C and 75 +/- 10% RH. The experimental design was in randomized blocks, in 3x3 factorial arrangement, with four replicates. After 60 days, nitrate reductase activity and chlorophyll contents were evaluated in the diagnostic leaf. The results demonstrate that the response of plants to a combination of drought and aluminum toxicity, similar to the conditions in many natural environments, is different from the response of plants to each of these stresses applied individually, as typically tested in the laboratory. The nitrate reductase activity can be used as a biochemical-physiological marker of water deficiency while chlorophyll contents can be used as a biochemical-physiological marker of both of them, water deficiency or aluminum toxicity in soil. Both parameters can not be as a biochemical-physiological marker for acclimation of young plants of sugarcane cv. IAC91-5155, under the combined stresses.

Silicon has beneficial effects on many crops, mainly under biotic and abiotic stresses. Silicon can affect biochemical, physiological, and photosynthetic processes and, consequently, alleviates drought stress. However, the effects of Si on potato (Solanum tuberosum L.) plants under drought stress are still unknown. The objective of this study was to evaluate the effect of Si supply on some biochemical characteristics and yield of potato tubers, either exposed or not exposed to drought stress. The experiment was conducted in pots containing 50 dm(3) of a Typic Acrortox soil (33% clay, 4% silt, and 63% sand). The treatments consisted of the absence or presence of Si application (0 and 284.4 mg dm(-3)), through soil amelioration with dolomitic lime and Ca and Mg silicate, and in the absence or presence of water deficit (-0.020 MPa and -0.050 MPa soil water potential, respectively), with eight replications. Silicon application and water deficit resulted in the greatest Si concentration in potato leaves. Proline concentrations increased under lower water availability and higher Si availability in the soil, which indicates that Si may be associated with plant osmotic adjustment. Water deficit and Si application decreased total sugars and soluble proteins concentrations in the leaves. Silicon application reduced stalk lodging and increased mean tuber weight and, consequently, tuber yield, especially in the absence of water stress.

Heat stress has negative effects on pregnancy rates of lactating dairy cattle. There are genetic differences in tolerance to heat stress; Bos taurus indicus (B. t. indicus) cattle and embryos are more thermotolerant than Bos taurus taurus (B. t. taurus). In the present study, the effects of sire and sire breed on conception and embryonic/fetal loss rates of lactating Holstein cows during the Brazilian summer were determined. In Experiment 1, cows (n = 302) were AI after estrus detection or at a fixed-time with semen from one Gyr (B. t. indicus) or one Holstein sire (B. t. taurus). Pregnancy was diagnosed 80 days after AI. In Experiment 2, cows (n = 811) were AI with semen from three Gyr and two Holstein sires. Pregnancy was diagnosed at 30-40 and at 60-80 days after AI. Cows diagnosed pregnant at the first examination but non-pregnant at the second were considered as having lost their embryo or fetus. Data were analyzed by logistic regression. The model considered the effect of sire within breed, sire breed, days postpartum, period of lactation, and AI type (AI after estrus versus fixed-time). There was no effect of the AI type, days postpartum or milk production on conception or embryonic loss rates. The use of Gyr bulls increased pregnancy rate when compared to Holstein bulls [9.1% (60/657) versus 5.0% (23/456), respectively, P = 0.008; data from Experiments 1 and 2 combined]. Additionally, in Experiment 2, cows inseminated using semen from sire #4 (Gyr) had lower embryonic loss (10%) when compared with other B. t. indicus (35.3% and 40%) or B. t. taurus sires (18.2% and 38.5%, P = 0.03). In conclusion, the use of B. t. indicus sires may result in higher conception rates in lactating Holstein cows during summer heat stress. Moreover, sire can affect embryonic loss and selection of bulls according to this criterion may result in higher parturition rates in lactating Holstein cows. (c) 2006 Elsevier B.V. center dot Background and Aims Drought is a major environmental constraint affecting growth and production of Coffea canephora. Selection of C. canephora clones has been largely empirical as little is known about how clones respond physiologically to drought. Using clones previously shown to differ in drought tolerance, this study aimed to identify the extent of variation of water use and the mechanisms responsible, particularly those associated morphological traits.center dot Methods Clones (14 and 120, drought-tolerant; 46 and 109A, drought-sensitive, based on their abilities to yield under drought) were grown in 120-L pots until they were 12-months old, when an irrigation and a drought treatment were applied; plants were droughted until the pressure potential (Psi(x)) before dawn (pre-dawn) reached -3.0 MPa. Throughout the drought period, Psi(x) and stomatal conductance (g(s)) were measured. At the end of the experiment, carbon isotope ratio and parameters from pressure-volume curves were estimated. Morphological traits were also assessed.center dot Key Results and Conclusions With irrigation, plant hydraulic conductance (K-L), midday Psi(x) and total biomass were all greater in clones 109A and 120 than in the other clones. Root mass to leaf area ratio was larger in clone 109A than in the others, whereas rooting depth was greater in drought-tolerant than in drought-sensitive clones. Predawn Psi(x) of -3.0 MPa was reached fastest by 109A, followed progressively by clones 46, 120 and 14. Decreases in g(s) with declining Psi(x), or increasing evaporative demand, were similar for clones 14, 46, and 120, but lower in 109A. Carbon isotope ratio increased under drought; however, it was lower in 109A than in other clones. For all clones, Psi(x), g(s) and KL recovered rapidly following re-watering. Differences in root depth, KL and stomatal control of water use, but not osmotic or elastic adjustments, largely explained the differences in relative tolerance to drought stress of clones 14 and 120 compared with clones 46 and 109A.

Este trabalho teve como objetivo avaliar o efeito do estresse salino sobre a eficiência fotoquímica do fotossistema II (PSII) nas cultivares de manga 'Haden', 'Palmer', 'Tommy Atkins' e 'Ubá' enxertadas sobre o porta-enxerto 'Imbu'. Foi utilizada solução nutritiva de Hoagland modificada contendo 0; 15; 30e 45 mmol L-1 NaCl. Aos 97 dias após a exposição ao estresse salino, foram avaliados os parâmetros da fluorescência da clorofila (F0, Fm, Fv, F0/Fm, Fv/Fm, Fv'/Fm', ΦPSII = [(Fm'-Fs)/(Fm')], D = (1- Fv'/Fm') e ETR = (ΦPSII×PPF×0,84×0,5). Aos 100 dias, foram avaliados a emissão foliar, a área média de folhas (cm²), o índice de toxidez nas folhas e o índice de abscisão foliar. em todas as cultivares, em graus diferenciados, ocorreram decréscimo na eficiência fotoquímica do fotossistema II, na emissão de folhas, e aumento nos índices de toxidez e abscisão foliar, intensificados nas concentrações a partir de 15 mmol L-1 NaCl. As plantas cultivadas em 45 mmol L-1 NaCl apresentaram decréscimos na razão Fv/Fm de 27,9; 18,7; 20,5 e 27,4%, incremento no índice de toxidez foliar de 33,0; 67,5; 41,6 e 80,8% e no índice de abscisão foliar de 71,8; 29,2; 32,5 e 67,9% para as cultivares 'Haden', 'Palmer', 'Tommy Atkins' e 'Uba', respectivamente. Os decréscimos na razão Fv/Fm foram acompanhados de redução na emissão de folhas e aumento no índice de toxidez foliar, mostrando, portanto, o potencial da fluorescência da clorofila na detecção precoce de estresse salino em mangueira.

Uma das utilizações da técnica de cultura de tecidos para o melhoramento vegetal é a identificação de linhas de células que apresentem tolerância ao estresse salino. Para se estudar os mecanismos bioquímicos envolvidos na expressão genética da tolerância a salinidade, calos oriundos de eixos embrionários de quatro cultivares de feijão (Phaseolus vulgaris L.; cultivares IAC - carioca, IAPAR 14, JALO-EEP 558, BAT - 93), foram cultivados em meio sólido Murashige & Skoog (1962), suplementado com NaCl nas concentrações de 0, 20, 40, 60 e 80 mM. Após 14 dias de incubação, os calos foram coletados e analisados quanto aos padrões isoenzimáticos e de atividade das peroxidases. Os cultivares BAT e IAPAR apresentaram duas zonas de atividade em comum na região anódica e apenas uma zona enzimática específica a cada um deles (migração mais rápida).Possivelmente as duas zonas anódicas intermediárias sejam produtos do mesmo loco enzimático, porém com alelos diferentes, consequentemente diferentes mobilidades eletroforéticas. O cv. JALO apresentou duas zonas anódicas de atividade em comum com os cultivares IAC e IAPAR com uma zona anódica exclusiva de migração mais lenta, a qual apresentou atividade mais intensa de todos os cultivares analisados. Este cultivar revelou ainda uma zona catódica provavelmente dimérica e heterozigota nos indivíduos de todos os tratamentos aplicados. Provavelmente, esta é a mesma zona que ocorre em homozigose com fixação do alelo lento para os indivíduos de todos os tratamentos efetuados nos cultivares BAT e IAPAR. O cv. IAC apresentou duas bandas anódicas em comum com os cv. IAPAR e JALO. Apresentou também a banda anódica mais rápida em comum com o cv. IAPAR e uma banda anódica exclusiva de migração mais lenta. Curiosamente, os indivíduos deste cv. mantidos em meio suplementado com 20 mM de NaCl não apresentaram atividade nas três zonas anódicas mais lentas. Ocorreu no cv. IAC uma única zona de atividade catódica, dimérica e heterozigota para os indivíduos provenientes de todos os tratamentos, composta provavelmente de dois alelos diferentes da zona correspondente ao cv. JALO. Amostras provenientes dos tratamentos 40 e 60 mM de NaCl, desta zona catódica, apresentaram maior atividade enzimática. A análise da atividade da peroxidase no extrato bruto, revelou que os cultivares responderam diferentemente ao aumento da concentração salina no meio de cultura, com aumento pronunciado dessa atividade nos cultivares IAC e JALO.
